Transcriptomic and Physiological Analysis of the Effects of Exogenous Phloretin and Pterostilbene on Resistance Responses of Stylosanthes against Anthracnose

Shizi Zhang,Yunfeng Xu,Fang Wang,Liyun Yang,Lijuan Luo,Lingyan Jiang

Anthracnose caused byColletotrichum gloeosporioidesis a destructive disease ofStylosanthes(stylo). Combination treatment of phloretin and pterostilbene (PP) has been previously shown to effectively inhibit the conidial germination and mycelial growth ofC. gloeosporioidesin vitro. In this study, the effects of PP treatment on the growth ofC. gloeosporioidesin vivo and the biocontrol mechanisms were investigated. We found that exogenous PP treatment could limit the growth ofC. gloeosporioidesand alleviate the damage of anthracnose in stylo. Comparative transcriptome analysis revealed that 565 genes were up-regulated and 239 genes were down-regulated upon PP treatment during the infection byC. gloeosporioides. The differentially expressed genes were mainly related to oxidative stress and chloroplast organization. Further physiological analysis revealed that application of PP afterC. gloeosporioidesinoculation significantly reduced the accumulation of O2•−level and increased the accumulation of antioxidants (glutathione, ascorbic acid and flavonoids) as well as the enzyme activity of total antioxidant capacity, superoxide dismutase, catalase, glutathione reductase, peroxidase and ascorbate peroxidase. PP also reduced the decline of chlorophyll a + b and increased the content of carotenoid in response toC. gloeosporioidesinfection. These results suggest that PP treatment alleviates anthracnose by improving antioxidant capacity and reducing the damage of chloroplasts, providing insights into the biocontrol mechanisms of PP on the stylo against anthracnose. Keywords:phloretin;pterostilbene;ROS scavenging;chloroplasts;anthracnose;stylo

Ta4C3 MXene Slows Progression of Fatty Liver Disease through Its Anti-Inflammatory and ROS-Scavenging Effects

Shuying He,Yuerong Lv,Jingnan Qiu,Shudan Cui,Zixian Gao,Liang Peng

Treating metabolic dysfunction-associated fatty liver disease (MAFLD) and reducing the occurrence of MAFLD-associated liver cancer remain challenging. Two-dimensional (2D) tantalum carbide (Ta4C3) MXene nanozymes (MXenzymes) exhibit antioxidant and anti-inflammatory activities and have thus attracted considerable attention in the fields of oncology and engineering. However, the potential mechanism of action and bioactive properties of Ta4C3 in MAFLD remain uncertain. In our study, Ta4C3 not only inhibited lipid accumulation and disrupted lipid metabolism in hepatocytes but also reduced cell death caused by fatty acids by decreasing intracellular reactive oxygen species (ROS) levels, which significantly promoted the polarization of M1 macrophages to M2 macrophages by alleviating oxidative stress and further suppressing inflammatory factor expression. In mice fed a methionine–choline-deficient (MCD) diet, Ta4C3 reduced lipid accumulation, the infiltration of inflammatory cells, and liver cell apoptosis by modulating the cellular microenvironment through its anti-inflammatory and antioxidant properties. Therefore, Ta4C3 can be used as a multifunctional bioactive material to alleviate hepatic steatosis and inflammation in individuals with MAFLD/metabolic dysfunction-associated steatohepatitis (MASH) because of its robust antioxidant and anti-inflammatory effects.

Glucosyltransferase CsUGT78A14 Regulates Flavonols Accumulation and Reactive Oxygen Species Scavenging in Response to Cold Stress in Camellia sinensis

Zhao Mingyue,Jin Jieyang,Gao Ting,Zhang Na,Jing Tingting,Wang Jingming,Ban Qiuyan,Schwab Wilfried,Song Chuankui

Glycosyltransferases (UGTs) play diverse roles in cellular metabolism by altering regulatory metabolites activities. However, the physiological roles of most members of UGTs in crops in response to abiotic stresses are unknown. We have identified a novel glycosyltransferase CsUGT78A14 in tea crops, an important economic crops, whose expression is strongly induced by cold stress. Biochemical analyses confirmed that CsUGT78A14-1 showed the highest activity toward kaempferol and is involved in the biosynthesis of kaempferol-diglucoside, whereas the product of CsUGT78A14-2, which differs from CsUGT78A14-1 by a single amino acid, was identified as 3-O-glucoside. The accumulation of kaempferol monoglucosides and diglucosides was consistent with the expression levels of CsUGT78A14 in response to cold stress, as well as in different tissues and genotypes of tea plants. Down-regulation of CsUGT78A14 resulted in reduced accumulation of flavonols, reactive oxygen species (ROS) scavenging capacity and finally reduced tea plant stress tolerance under cold stress. The antioxidant capacity of flavonols aglycon was enhanced by glucosylation catalyzed by CsUGT78A14. The results demonstrate that CsUGT78A14 plays a critical role in cold stress by increasing flavonols accumulation and ROS scavenging capacity, providing novel insights into the biological role of UGTs and flavonoids in plants.

Single stem cell encapsulation by MnO2 doped hydrogel for robust bone regeneration

Yu Tian,Pengxia Zhang,Zhen Ma,Mengya Li,Yali Fan,Rui Guo,YanFeng Li,Yingying Zhang

Mesenchymal stem cells (MSCs) have become an important choice for regenerative medicine because of their specific differentiation ability. However, MSCs are rapidly cleared by direct injection, and living cells need to be embedded in a 3D semipermeable hydrogel matrix to prolong cell retention time to enhance therapeutic effects. Herein, MnO 2 doped alginate hydrogel was prepared for single stem cell encapsulation (SA@MSC-MnO 2 ) with maintaining biological activity. This hydrogel system could accelerate the differentiation potential of MSCs and promoted 60 % repair of bone defects within 8 weeks. Besides, the doping of MnO 2 could react with H 2 O 2 to generate H 2 O and O 2 , thus achieving the goal of reactive oxygen species (ROS) clearing and improving the bone regeneration effects. We believe that this single stem cell hydrogel encapsulation system will further promote the clinical application of stem cell therapy.

Novel Metal-Doped Rutin-Derived Carbon Dots in Injectable Hydrogel for Enhanced ROS Scavenging and Antibacterial Effects in Wound Healing

Jiahui Tong,Yifan Zhao,Xiao Ning,Hongyi Peng,Jingyu Yan,Danlei Qin,Feng Tian,Xiuping Wu,Chunfang Wang,Bing Li

Wound healing is often impeded by bacterial infections, excessive reactive oxygen species (ROS), and inflammatory responses, challenges that traditional dressings inadequately address. Rutin, a natural flavonoid recognized for its antioxidant and anti-inflammatory effects, is limited in biomedical applications because of its low solubility in water. This research presents a novel approach using nitrogen and zinc-doped rutin-derived carbon dots (ZR-CDs), which enhances both antioxidant activity and bioavailability by leveraging the nanoscale properties and surface modifiability of carbon dots. The ZR‑CDs are covalently integrated into an oxidized hyaluronic acid (OHA)/Pluronic F‑127 hydrogel through Schiff‑base chemistry, yielding an injectable, shape‑adaptive composite hydrogel (ZCD@OF), aimed at promoting the healing of infected wounds. In vitro, ZR-CDs exhibit excellent biocompatibility, free-radical scavenging, and protection against ROS-induced cellular dysfunction. The ZCD@OF hydrogel exhibited robust antibacterial efficacy while maintaining favorable rheological properties for wound coverage. In vivo on an infected wound model, ZCD@OF significantly accelerates wound closure, promotes angiogenesis and collagen deposition, and suppresses pro-inflammatory cytokines (IL-6, TNF-α). This study highlights the therapeutic potential of ZCD@OF hydrogel in complex wound treatment, illustrating its capacity to synergistically integrate carbon dot with advanced biomaterial strategies for improved therapeutic outcomes.

Unveiling The Multifunction of Beaded Stream-Like Co9Se8 Catalysts for Water Splitting, Reactive Oxygen Species Scavenging, and Hydrogen Anti-Inflammation

Pan Luo,Honglei Wang,Xue Wen,Xianzhu Luo,Peng Xu

The electrocatalytic preparation of hydrogen at different pH values not only achieves excellent device performance but also promotes the application of reactive oxygen species (ROS) clearance and hydrogen anti-inflammation. However, it is difficult to develop materials that simultaneously achieve these excellent properties. Herein, the preparation of beaded necklace-like Co 9 Se 8 microspheres using a general and facile synthetic strategy is reported. The Co 9 Se 8 -modified electrode has three applications: efficient water splitting for enhanced performance, ROS scavenging, and hydrogen anti-inflammatory activity. Experiments, high-angle-annular-dark-field scanning transmission electron microscopy, and density functional theory calculations indicate that the reconstructed Co(OH) 2 plays a vital role in the oxygen evolution reaction and that the transition from *O to *OOH is the actual rate-determining step. The Co 9 Se 8 material, with its unique beaded necklace-like structure, exhibits exceptional hydrogen production capabilities in phosphate buffer solution (pH 7.4). In particular, hydrogen produce under neutral conditions can effectively reduce ROS levels and significantly inhibit inflammation-related pathological processes, playing a unique antioxidant and anti-inflammatory role at the cellular level. The obtained results indicate that the as-synthesized Co 9 Se 8 is more suitable for water splitting, ROS scavenging, and hydrogen anti-inflammatory applications, outperforming other transition metal electrodes and rendering practical industrial and clinical applications.

Engineered renal-targeting nanozyme achieves sequential treatment of AKI through ROS clearance and immune modulation

Zhiwen Wang,Yue Xie,Mingcun Hu,Min Yang,Xingjian Zou,Chun Zhang

Effective countermeasures for acute kidney injury (AKI) remain unsatisfactory. During AKI progression, the timely and appropriate transition of macrophages from pro-inflammatory to anti-inflammatory states is critical for resolving acute inflammation and promoting tissue repair. Timely clearing of excess reactive oxygen species (ROS) and promotion of M2 polarization of macrophages are key determinants of AKI progression. Accordingly, we report an engineered renal-targeting nanozyme constructed from a novel two-dimensional nanomaterial, MXene nanosheets, via surface conjugation of the anticomplement component 5a aptamer (ac5a-Apt) and magnesium ions (Mg 2 + ). After AKI, the engineered nanozyme can not only target the kidneys but also navigates to inflammatory areas under the guidance of the ac5a-Apt, clear excessive ROS, inhibits the C5a-C5aR complement system activation and promote M2 polarization of macrophages. Importantly, after MXene reacts with ROS and degrades, the surface-loaded Mg 2+ is released, further suppressing the MAPK/ NF-κB signaling pathway to achieve sequential therapy. The engineered renal-targeting nanozyme offers a novel strategy based on ROS clearance and immune modulation for sequential AKI treatment.

Revitalizing osteoporotic bone repair via multilevel ROS scavenging and osteoimmune regulating hydrogel

Yiran Zhang,Qingcheng Song,Shuai Yang,Jiheng Xiao,Fengkun Wang,Chunxu Fu,Xin Xing,Jianhua Wu,Shuo Zhang,Yanbin Zhu,Yingze Zhang

Osteoporotic bone defects (OBDs) pose significant clinical challenges due to impaired tissue regeneration and osteogenic differentiation. Conventional combination therapies using systemic anti-osteoporosis drugs and graft fillings have yielded unsatisfactory results. Excessive reactive oxygen species (ROS) are a hallmark of OBDs, leading to abnormal inflammatory responses and a hostile osteoimmune microenvironment that impedes repair. In this study, we developed a multifunctional injectable hydrogel, termed PTSr@CP, composed of a strontium (Sr) and tannic acid (TA)-based metal–phenolic network core coated with polydopamine (PDA), integrated into a matrix of carboxymethyl chitosan and poly (γ-glutamic acid). The PTSr@CP hydrogel can be easily injected into bone defect sites, where it undergoes rapid in situ crosslinking and exhibits exceptional ROS-scavenging capabilities. Additionally, the hydrogel mediates mild photothermal therapy (MPTT), promoting local tissue repair. Both in vitro and in vivo studies demonstrated its excellent biosafety and confirmed synergistic bone regeneration through enhanced osteogenesis, angiogenesis, and osteoimmunomodulation. This work offers a compelling strategy for OBD treatment by combining mild hyperthermia-enhanced in situ bone repair with sustained delivery of bioactive agents to improve the osteoimmune microenvironment.

Mild hyperthermia enhanced synergistic uric acid degradation and multiple ROS elimination for an effective acute gout therapy

Zhao Pei,Hu Hua-Zhong,Chen Xiao-Tong,Jiang Qi-Yun,Yu Xue-Zhao,Cen Xiao-Lin,Lin Shi-Qing,Mai Sui-qing,Pang Wei-lin,Chen Jin-Xiang,Zhang Qun

Background Acute gouty is caused by the excessive accumulation of Monosodium Urate (MSU) crystals within various parts of the body, which leads to a deterioration of the local microenvironment. This degradation is marked by elevated levels of uric acid (UA), increased reactive oxygen species (ROS) production, hypoxic conditions, an upsurge in pro-inflammatory mediators, and mitochondrial dysfunction. Results In this study, we developed a multifunctional nanoparticle of polydopamine-platinum (PDA@Pt) to combat acute gout by leveraging mild hyperthermia to synergistically enhance UA degradation and anti-inflammatory effect. Herein, PDA acts as a foundational template that facilitates the growth of a Pt shell on the surface of its nanospheres, leading to the formation of the PDA@Pt nanomedicine. Within this therapeutic agent, the Pt nanoparticle catalyzes the decomposition of UA and actively breaks down endogenous hydrogen peroxide (H 2 O 2 ) to produce O 2 , which helps to alleviate hypoxic conditions. Concurrently, the PDA component possesses exceptional capacity for ROS scavenging. Most significantly, Both PDA and Pt shell exhibit absorption in the Near-Infrared-II (NIR-II) region, which not only endow PDA@Pt with superior photothermal conversion efficiency for effective photothermal therapy (PTT) but also substantially enhances the nanomedicine’s capacity for UA degradation, O 2 production and ROS scavenging enzymatic activities. This photothermally-enhanced approach effectively facilitates the repair of mitochondrial damage and downregulates the NF-κB signaling pathway to inhibit the expression of pro-inflammatory cytokines. Conclusions The multifunctional nanomedicine PDA@Pt exhibits exceptional efficacy in UA reduction and anti-inflammatory effects, presenting a promising potential therapeutic strategy for the management of acute gout. An engineered, self-propelled nanozyme as reactive oxygen species scavenger

Shuo Zhang,Jun Chen,Mei-Ling Lian,Wen-Sheng Yang,Xu Chen

The development of enzyme-mimicking nanomaterials (nanozymes) with excellent reactive oxygen species (ROS)-scavenging abilities and low biotoxicity is desired for clinical translation. The use of nanomotors as nanozymes for active ROS elimination is a novel strategy that involves rapid ROS diffusion through autonomous movement. Here, we report an engineered, self-propelled nanozyme based on a Janus Pt/SiO 2 nanomotor with Pt nanoparticle active centers, which possess GPx-like, CAT-like and SOD-like enzyme activities as well as ∙ OH scavenging ability. Nanozymes modified with black phosphorous quantum dots (BPQDs) achieved efficient self-diffusiophoretic motion under physiological concentrations of H 2 O 2 , independent of a fuel source. Moreover, the OH and O 2 - removal ability of BPQDs further improved antioxidant ability of this nanozyme. We hope that such strategy involving self-propulsion will facilitate the development of nanozymes and expand biomedical applications of nanomotors.

An Ultrasmall Cu/Cu2O Nanoparticle-Based Diselenide-Bridged Nanoplatform Mediating Reactive Oxygen Species Scavenging and Neuronal Membrane Enhancement for Targeted Therapy of Ischemic Stroke

Jianglong Kong,Rui Zou,Runxuan Chu,Nan Hu,Jiawen Liu,Yuting Sun,Xiaohan Ge,Meiru Mao,Hongrui Yu,Yi Wang

Ischemic stroke is one of the major causes of death and disability worldwide, and an effective and timely treatment of ischemic stroke has been a challenge because of the narrow therapeutic window and the poor affinity with thrombus of the thrombolytic agent. In this study, rPZDCu, a multifunctional nanoparticle (NP) with the effects of thrombolysis, reactive oxygen species (ROS) scavenging, and neuroprotection, was synthesized based on an ultrasmall Cu4.6O NP, the thrombolytic agent rt-PA, and docosahexaenoic acid (DHA), which is a major component of the neuronal membrane. rPZDCu showed strong thrombus-targeting ability, which was achieved by the platelet cell membrane coating on the NP surface, and a good thrombolytic effect in both the common carotid artery clot model and embolic middle cerebral artery occlusion (MCAO) model of rats. Furthermore, rPZDCu exhibited a good escape from the phagocytosis of macrophages, effective promotion of the polarization of microglia, and efficient recovery of neurobiological and behavioral functions in the embolic MCAO model of rats. This is a heuristic report of (1) the Cu0/Cu+ NP for the treatments of brain diseases, (2) the integration of DHA and ROS scavengers for central nervous system therapies, and (3) diselenide-based ROS-responsive NPs for ischemic stroke treatments. This study also offers an example of cell membrane-camouflaged stimuli-responsive nanomedicine for brain-targeting drug delivery.

Room Temperature Preparation of Surface-Clean Hydrogen-Doped Plasmonic Molybdenum Oxide as a High-Efficient and Degradable Reactive Oxygen Species Scavenger

Wang Junren,Li Hua,Yu Haili,He Yi

Considerable efforts have been made to develop reactive oxygen species (ROS) scavengers for removing high level of ROS. However, most of the reported ROS scavengers are nondegradable and involve harsh reaction conditions as well as utilize various surface ligands. In order to overcome these drawbacks, in the present work, we develop a facile and mild synthesis avenue for preparation of surface-clean hydrogen-doped molybdenum oxide (H 0.34 MoO 3 ) via simply mixing MoO 3 dispersion with aluminum foil under an acidic environment without any surface capping reagents at room temperature. The resulting H 0.34 MoO 3 can act as a broad-spectrum ROS scavenger, including . OH, H 2 O 2 , O 2 − , and 1 O 2 as well as 2, 2-diphenyl-1-picrylhydrazyl (DPPH). The free radical scavenging activity of H 0.34 MoO 3 achieves as high as 71.6% and 99.1% for . OH and DPPH scavenging, which is comparable and superior to that of ascorbic acid that is a classic free radical scavenger. More significantly, the resulting H 0.34 MoO 3 is degrade, which can be degraded into molybdate ions under a neutral environment (pH 7.4).
