Early and accurate diagnosis is considered the key issue to prevent the further spread of viruses and facilitate influenza therapy. Herein, we report a colorimetric immunosensor for influenza A virus (IAV) based on gold nanoparticles (AuNPs) modified with monoclonal anti-hemagglutinin antibody (mAb). The immunosensor allows for a fast, simple, and selective detection of IAV. In this assay, influenza-specific antibodies are conjugated to AuNPs to create mAb–AuNP probes. Since IAV has multiple recognition sites for probes on the surface, the mAb–AuNP probes can be specifically arranged on the virus surface due to their very specific antigen recognition. In this case, this aggregation of the mAb–AuNP probes produces a red shift in the absorption spectrum due to plasmon coupling between adjacent AuNPs, and it can be detected with the naked eye as a color change from red to purple and quantified with the absorption spectral measurements. The aggregate formation is also confirmed with transmission electron microscopy (TEM) imaging and dynamic light scattering (DLS). Under the optimal conditions, the present immunoassay can sensitively measure H3N2 IAV (A/Brisbane/10/2007) with a detection limit of 7.8 hemagglutination units (HAU). This proposed immunosensor revealed high specificity, accuracy, and good stability. Notably, it is a single-step detection using AuNP probes and UV-vis spectrophotometer for readout, and no additional amplification, e.g., enzymatic, is needed to read the result. This assay depends on an ordered AuNP structure covering the virus surface and can be applied to any virus pathogen by incorporating the appropriate pathogen-specific antibody.

Sensitivity and accuracy are the key performance metrics for evaluating immunochromatographic assay (ICA). In this study, we have developed a quantum dot nanobeads-based dual-channel fluorescent ICA (QBs-DICA) designed for the sensitive and accurate detection of both influenza A virus (IAV) and human adenovirus (ADV) antigens. Following systematic assay optimization, we achieved a remarkable improvement in sensitivity, with gains of up to two orders of magnitude at the best. To enhance signal accuracy for IAV and ADV testing, we implemented a temperature-controlled dual-channel detector during sample incubation and the chromatographic process, resulting in an approximate increase of 34.2% and 36.7%, respectively. The Limit of Detection (LOD) for IAV and ADV testing was robustly established at 165 copies/mL and 805 copies/mL, respectively. Furthermore, in the case of IAV-spiked and ADV-spiked samples, we achieved consistently high recoveries ranging from 94.5% to 104.8% and 94.7% to 106.4%, with impressively low coefficients of variation (CV) of below 7.16% and 6.25%, respectively. Our established QBs-DICA system has exhibited a remarkable enhancement in both sensitivity and accuracy, making it exceptionally well-suited for supporting the rapid advancement of clinical diagnosis and treatment strategies.

Respiratory infectious diseases pose a serious threat to human health worldwide. Timely diagnosis is crucial for effective clinical management and infection control. Herein, we developed a dual-channel lateral flow immunoassay (DC-LFIA) for simultaneous sensitive detection of influenza A virus (IAV) and respiratory syncytial virus (RSV) nucleoprotein (NP) antigens. The high brightness latex microspheres (HB-LMs) exhibiting typical absorption peak at 540 nm with uniform size and excellent optical stability, were prepared by a simple surfactant-free emulsion polymerization by mixing Sudan red, which overcame the complexity and color instability met for traditional synthesis methods. After affinity evaluation, two antibody pairs have good affinity for IAV NP and RSV NP were separately identified. Then, combining antibody pairs as probes and HB-LMs as based signal tag, a DC-LFIA was constructed, by which the visual limit of detection is 25 pg/mL, 100 pg/mL for IAV NP and RSV NP, respectively, 10-fold more sensitive than gold nanoparticles based LFIA. The calculated limit of detection (S/N=3) of IAV NP and RSV NP was improved 22.8- and19.5-fold, respectively, in comparison with gold nanoparticles based LFIA. Additionally, the developed sensor shows excellent specificity and reproducibility. Furthermore, the as-proposed method was validated through the analysis of clinical samples, in good agreement with commercial colloidal gold kits. Thus, the developed DC-LFIA has great potential for clinical application in the multiplex detection of respiratory viral antigens.

The rise of drug-resistant influenza A virus (IAV) strains and severe lung injury caused by excessive inflammation highlight the need for new antivirals that combine antiviral and immunomodulatory effects. Adamantane, an established anti-IAV agent, targets the viral M2 matrix protein, but widespread mutations have limited its clinical utility. Previously, we identified that 2,5-dihydroxybenzylalcohol and its derivatives possess anti-IAV activity. In this study, we synthesized a series of adamantane-2,5-dihydroxy-benzylalcohol derivatives via reductive amination. Among these, Amt-1 demonstrated strong antiviral activity against both amantadine-sensitive (H1N1 PR8) and clinical isolates IAV strains (-log IC 50 =6.12–6.51 M). Mechanistic analyses, including immunofluorescence, ITC, qPCR, ELISA, luciferase assays, and Western blotting, revealed that Amt-1 not only interacts with the peptide segment of M2 ion channel protein (-log Kd = 5.08 M) but also activates Nrf2, leading to increased HO-1 expression and reduced production of pro-inflammatory cytokines IL-6, IL-1β, and TNF-α. Additionally, Amt-1 inhibited IAV-induced NF-κB activation, thereby suppressing viral replication. In vivo , Amt-1 reduced lung viral titers and mitigated histopathological damage in mice, surpassing amantadine in controlling inflammation and increased survival to 33%. In summary, Amt-1 is the first adamantane derivative shown to exert anti-IAV effects by modulating Nrf2/HO-1-mediated anti-inflammatory pathways and NF-κB-driven antiviral responses, while retaining M2 ion channel inhibition. This host-directed mechanism addresses drug resistance and limits immunopathology, making Amt-1 a promising therapeutic candidate for resistant influenza infections.

The spread of the FluA virus poses significant public health concerns worldwide. Fluorescent lateral flow immunoassay (LFIA) test strips have emerged as vital tools for the early detection and monitoring of influenza infections. However, existing quantitative virus-detection methods, particularly those utilizing smartphone-based sensing platforms, encounter accessibility challenges in resource-limited areas and among the elderly population. Despite their advantages in speed and portability, these platforms often lack user-friendliness for these demographics, impeding their widespread utilization. To address these challenges, this study proposes leveraging the optical pick-up unit (OPU) sourced from commercial optical drives as a readily available fluorescence excitation module for the quantitative detection of antibodies labeled with quantum-dot fluorescent microspheres. Additionally, we utilize miniaturized and high-performance optical components and 3D-printed parts, along with a customized control system, to develop an affordable point-of-care testing (POCT) device. Within the system, a stepping motor scans the test strip from the T-line to the C-line, enabling the calculation of the fluorescence-intensity ratio between the two lines. This simple yet effective design facilitates rapid and straightforward field or at-home testing for FluA. The proposed prototype platform demonstrates promising performance, achieving a limit of detection (LOD) of 2.91 ng/mL, a total detection time of no more than 15 min, and dimensions of 151 mm × 11.2 mm × 10.8 mm3. We believe that the proposed approach holds great potential for improving access to an accurate influenza diagnosis.
