Spatial distribution and time-course of polyphenol accumulation in grape berry (Vitis labruscana cv. ‘Kyoho’)

Ming Qi,Zisheng Luo,Bin Wu,Lei Wang,Mingyi Yang,Xiaochen Zhang,Xingyu Lin,Yanqun Xu,Xihong Li,Li Li

The spatial distribution and time-course evolution of polyphenol compounds in response to exogenous light and temperature in grape berry ( Vitis labruscana cv. ‘Kyoho’) were examined. Results demonstrated that the polyphenol biosynthesis was induced via the up-regulated gene transcription levels during berry ripening. The polyphenol biosynthesis was differentially observed in skins, stems, leaves and seeds, and the more polyphenols were accumulated in seeds; and the polyphenols were significantly distributed in bottom berries of grape clusters. Additionally, the lower temperature (4 ℃) accelerated the polyphenol biosynthesis by 1.3 times after storage of 28 days, whereas the light exposure slightly inhibited the polyphenol biosynthesis of berry during storage at 25 ℃. In conclusion, the data presented in this study shed more light on deeper understanding of the spatial accumulation and time-course evolution of polyphenols in grape berry.

Effects of combined microwave-hot-air-drying on the physicochemical properties and antioxidant activity of Rhodomyrtus tomentosa berry powder

Zhao Guanghe,Hu Congying,Luo Haiyu

Rhodomyrtus tomentosa berries are rich in nutrition, but easily mildew, so it is necessary to dry them with a proper drying technique in time. The effects of the combined microwave-hot-air-drying (CD) on the physicochemical and antioxidant properties of Rhodomyrtus tomentosa berry powder were evaluated in this study, compared with those of hot air drying (HD) and microwave drying (MD). The results showed that the hydration properties and oil holding capacity of CD-berry powder were better than those of HD- and MD-ones. However, CD-berry powder showed lower fillibility and liquidity than HD-one and but similar to MD-one. There was no significant difference among the thermal stability of berry powder produced by the three drying methods. Furthermore, CD was beneficial to maintain the color, nutrition, and active ingredients of Rhodomyrtus tomentosa berry powder. CD-berry powder and MD-one had similar antioxidant properties. In general, CD is more suitable for the processing of Rhodomyrtus tomentosa berry powder.

Preharvest application of alginate oligosaccharides enhances the postharvest storage of kiwiberry by regulating cell wall degradation

Md Saikat Hossain Bhuiyan,Wenxia Wang,Jin Gao,Xiaochen Jia,Hu Zhao,Md Mijanur Rahman Rajib,Heng Yin

The rapid degradation of the cell wall leads to firmness loss in fruit, resulting in a shorter shelf life and quality deterioration. Here, we evaluated the impact of preharvest foliar application of alginate oligosaccharides (AOS) on a perishable fruit kiwiberry ( Actinidia arguta ) over a 42 d storage period using physiological, molecular, and biochemical approaches. The selected plants were sprayed with the AOS solution 15 d before harvest, covering both the leaves and fruit surfaces. The preharvest AOS application significantly reduced decay incidence by about 60 %, maintained nearly twice the firmness, and 22 % less weight loss compared with control fruit after 42 d of storage. Transcriptome analysis reveals that AOS treatment downregulates the genes related to cell wall degradation. Moreover, biochemical analysis indicates that preharvest application suppressed the activation of cell wall degrading enzymes, reduced the solubilization of cell wall polysaccharides, and preserved their monosaccharide composition. These results reveal that preharvest application of AOS ensured the storability of kiwiberry by controlling cell wall degradation enzymes and gene expression. These findings highlight the significant potentiality of AOS preharvest treatments for delaying postharvest softening of kiwiberry.

Development of an enzyme-linked immunosorbent assay and a dipstick assay for the rapid analysis of trans-resveratrol in grape berries

Xiaojiao Chen,Junhu He,Guiyu Tan,Ju Liang,Yuxia Hou,Mian Wang,Baomin Wang

trans -Resveratrol, a natural polyphenol present in grapes, has many beneficial effects to human health. However, measurement of trans -resveratrol is technically complicated and time-consuming. In the present study, we obtained a sensitive and specific monoclonal antibody (mAb), namely 3C9, against trans -resveratrol. An indirect competitive enzyme-linked immunosorbent assay (icELISA) was developed, with 50% inhibitory concentration and working range of 1.0 ng/mL and 0.19–4.9 ng/mL, respectively. A lateral flow immunoassay (LFIA) based on 3C9 was also developed for the semiquantitative detection of trans -resveratrol in an indicator range of 50–100 ng/mL. Average recoveries of trans -resveratrol spiked in red and green grape berries samples were respectively 88–107% and 83–102% by icELISA. The icELISA and LFIA were applied for determination of trans -resveratrol in grape berries. The results were highly consistent with those determined by high-performance liquid chromatography analysis (R 2  = 0.9997). Therefore, we conclude that the immunoassay methods are suitable for the large-scale screening of trans -resveratrol in grape quality breeding.

Delphinidin-3-O-sambubioside: a novel xanthine oxidase inhibitor identified from natural anthocyanins

Xie Jiahong,Cui Haoxin,Xu Yang,Xie Lianghua,Chen Wei

ObjectivesThis study was conducted to investigate the xanthine oxidase (XO) inhibitory activities of 18 monomeric anthocyanins from berry fruits and roselle, and to illustrate the underlying mechanism of the most active anthocyanin delphinidin-3-O-sambubioside.Materials and MethodsEighteen monomeric anthocyanins were prepared and purified in our laboratory. The inhibitory properties of anthocyanins were investigated by in vitro inhibitory activity studies and fluorescence quenching studies; the inhibitory mechanism was explored through kinetic studies, fluorescence quenching studies, circular dichroism analysis and computational docking simulations.ResultsXO inhibitory activities of anthocyanins were related to the structures of B rings and glycosides. Among all the tested anthocyanins, delphinidin-3-O-sambubioside showed the most potent inhibitory activity with an IC50 of 17.1 μM, which was comparable to the positive control allopurinol. Spectroscopic results revealed that delphinidin-3-O-sambubioside could spontaneously interact with XO and induce conformational changes. Computational docking study indicated that delphinidin-3-O-sambubioside could bind to XO with a proper orientation, stably formed π–π interactions and hydrogen bonds with key residues, thus preventing the substrate from entering the active pocket.ConclusionsIn brief, our study identified delphinidin-3-O-sambubioside as a potent XO inhibitor from natural anthocyanins, which is potentially applicable for prevention and treatment of hyperuricemia.

A size-controlled green synthesis of silver nanoparticles by using the berry extract of Sea Buckthorn and their biological activities

Simin Wei,Yinghui Wang,Zhishu Tang,Jinhang Hu,Rui Su,Jingjie Lin,Tuan Zhou,Hui Guo,Nan Wang,Rongrong Xu

Green synthesis of silver nanoparticles (AgNPs) has been extensively studied by using a variety of plant extracts for applications in biomedical sciences and engineering. However, there are no reports on the synthesis of AgNPs by utilizing the berry extract of Sea Buckthorn, which is a traditional Chinese medicine and exhibits a wide spectrum of antioxidant, anti-inflammatory and anticancer activities. In this paper, we report an easy and eco-friendly technique for the preparation of AgNPs using the Sea Buckthorn berry extract under ultrasonic radiation at ambient temperature and the evaluation of both biosynthesis parameters and biological activities. The UV-visible spectrum and dynamic light scattering (DLS) analysis showed that the size of AgNPs was sensitive to the biosynthesis parameters, such as the pH of the extract, material proportion and reaction time, offering a size-controlled synthetic method for AgNPs. The X-ray diffraction (XRD), transmission electron microscopy (TEM), selected area electron diffraction (SAED) and DLS studies showed that the AgNPs (pH 10.0; material proportion 1 : 1; 4 h) had a face-centered cubic (fcc) structure and spherical shape with an average particle size of 27.3 ± 0.2 nm covered by anions, and existed in a monodispersed form with a polydispersity index (PDI) of 0.213. The biosynthesized AgNPs showed potent anticancer activity against human colorectal cancer (HCT116 and SW620), hepatoma cancer (HepG2), breast cancer (MCF-7) and cervical cancer (HeLa) cell lines as well as strong antioxidant activity. The IC50 values for these five cell lines were 8.77, 4.61, 14.59, 16.05 and 27.98 μg mL−1, respectively. However, the biosynthesized AgNPs revealed poor inhibition activities for the growth of E. coli and S. aureus. These results confirmed that the Sea Buckthorn could be a low-cost, nontoxic and eco-friendly natural resource for the synthesis of AgNPs, which might be useful for the development of new alternative antioxidant and anticancer agents in biomedicine.
