Transcriptomic and Functional Analyses of Two Cadmium Hyper-Enriched Duckweed Strains Reveal Putative Cadmium Tolerance Mechanisms

Gui-Li Yang,Lei Huang,Xiao Yang,Zhu Li,Hai-Min Liao,Kang Mao,Zhao-Ju Liu,He-Yan Geng,Qin Cao,Ai-Juan Tan

Cadmium (Cd) is one of the most toxic metals in the environment and exerts deleterious effects on plant growth and production. Duckweed has been reported as a promising candidate for Cd phytoremediation. In this study, the growth, Cd enrichment, and antioxidant enzyme activity of duckweed were investigated. We found that both high-Cd-tolerance duckweed (HCD) and low-Cd-tolerance duckweed (LCD) strains exposed to Cd were hyper-enriched with Cd. To further explore the underlying molecular mechanisms, a genome-wide transcriptome analysis was performed. The results showed that the growth rate, chlorophyll content, and antioxidant enzyme activities of duckweed were significantly affected by Cd stress and differed between the two strains. In the genome-wide transcriptome analysis, the RNA-seq library generated 544,347,670 clean reads, and 1608 and 2045 differentially expressed genes were identified between HCD and LCD, respectively. The antioxidant system was significantly expressed during ribosomal biosynthesis in HCD but not in LCD. Fatty acid metabolism and ethanol production were significantly increased in LCD. Alpha-linolenic acid metabolism likely plays an important role in Cd detoxification in duckweed. These findings contribute to the understanding of Cd tolerance mechanisms in hyperaccumulator plants and lay the foundation for future phytoremediation studies. Keywords:cadmium;duckweed;heavy metals;hyperaccumulator;transcriptome

Deep Learning-Based Automatic Duckweed Counting Using StarDist and Its Application on Measuring Growth Inhibition Potential of Rare Earth Elements as Contaminants of Emerging Concerns

Kevin Adi Kurnia,Ying-Ting Lin,Ali Farhan,Nemi Malhotra,Cao Thang Luong,Chih-Hsin Hung,Marri Jmelou M. Roldan,Che-Chia Tsao,Tai-Sheng Cheng,Chung-Der Hsiao

In recent years, there have been efforts to utilize surface water as a power source, material, and food. However, these efforts are impeded due to the vast amounts of contaminants and emerging contaminants introduced by anthropogenic activities. Herbicides such as Glyphosate and Glufosinate are commonly known to contaminate surface water through agricultural industries. In contrast, some emerging contaminants, such as rare earth elements, have started to enter the surface water from the production and waste of electronic products. Duckweeds are angiosperms from theLemnaceaefamily and have been used for toxicity tests in aquatic environments, mainly those from the genusLemna, and have been approved by OECD. In this study, we used duckweed from the genusWolffia, which is smaller and considered a good indicator of metal pollutants in the aquatic environment. The growth rate of duckweed is the most common endpoint in observing pollutant toxicity. In order to observe and mark the fronds automatically, we used StarDist, a machine learning-based tool. StarDist is available as a plugin in ImageJ, simplifying and assisting the counting process. Python also helps arrange, manage, and calculate the inhibition percentage after duckweeds are exposed to contaminants. The toxicity test results showed Dysprosium to be the most toxic, with an IC50value of 14.6 ppm, and Samarium as the least toxic, with an IC50value of 279.4 ppm. In summary, we can provide a workflow for automatic frond counting using StarDist integrated with ImageJ and Python to simplify the detection, counting, data management, and calculation process. Keywords:duckweed;StarDist;deep learning;cell counting;image segmentation;rare earth element

Salvinia natans-Based Hierarchical Structures for Solar Thermal Clean Water Production from High-Salinity Wastewater

Yuting Zhang,Yimeng Shi,Dongpu Hu,Sha Liang,Wenbo Yu,Fang Xu,Zhen Hu,Jiakuan Yang,Shushan Yuan,Bart Van der Bruggen

Biomaterial-based solar-driven evaporation has great potential for wastewater treatment and seawater desalination with a high energy conversion and utilization efficiency. However, technology gaps still exist for effectively and directly applying multiscale structures and intrinsic water transport channels of natural materials to enhance high-efficiency photothermal evaporation. In this study, a high-performance biomass-derived photothermal evaporative material was obtained using Salvinia natans, a common aquatic floating plant, together with simple poly(m-phenylenediamine) oxidation modification, building a hybrid biomass evaporator. With advantageous natural features of adequate water transport, microscale–nanoscale hierarchical structures, effective water activation, and antisalt-fouling function, the hybrid biomass evaporator achieves a high evaporation rate of 2.24 kg m–2 h–1 under one sun radiation (1 kW m–2). In addition, modified Salvinia natans also demonstrate certain ability to remove heavy metals during the photothermal evaporation of wastewater. This work offers a new perspective on the synthesis of an environmentally friendly and cost-effective solar-driven evaporator material, which has the advantages of low cost, simple process, and high photothermal conversion efficiency, and can be widely applied to seawater desalination and the treatment of wastewater with high salt concentrations.

Toxic mechanism of two cyanobacterial volatiles β-cyclocitral and β-ionone on the photosynthesis in duckweed by altering gene expression

Siyi Du,Haozhe Xu,Mengdan Yang,Ning Pan,Tiefeng Zheng,Chenyi Xu,Yan Li,Zhaojiang Zuo

Volatile organic compounds (VOCs) promote cyanobacteria dominating eutrophicated waters, with aquatic plant decrease and even disappearance. To uncover the toxic mechanism of cyanobacterial VOCs on aquatic plants, we investigated the growth, photosynthetic pigment levels, photosynthetic abilities and related gene expression in duckweed treated with β-cyclocitral and β-ionone, 2 main components in the VOCs. The levels of chlorophylls and carotenoids gradually declined with raising the concentration of the 2 compounds and prolonging the treatment time. Their decline should result from the down-regulation of 8 genes associated with photosynthetic pigment biosynthesis and up-regulation of 2 genes involved in carotenoid degradation. The reduction was also found in the photosystem II (PSII) efficiency and O 2 evolution rate, which should result from the lowered photosynthetic pigment levels and down-regulation of 38 genes related with photosynthetic process. The frond numbers, total frond area and fresh weight gradually decreased with raising the 2 compound concentration, which may result from the lowered photosynthetic abilities as well as down-regulated expression of 7 genes associated with growth-promoting hormone biosynthesis and signal transduction. It can be speculated that cyanobacterial VOCs may poison aquatic plants by lowering the photosynthesis and growth through altering related gene expression.

Effects of Duckweed Species Diversity on Physiological Responses and Removal Efficiency under Cadmium Stress

Cao Qin,Tan Ai-Juan,Lan Yan,Zou Wen-Bo,Yang Gui-Li

Cadmium (Cd) contamination threatens aquatic ecosystems and human health. Duckweeds are ideal for toxicological studies due to their rapid growth and high metal uptake. While interactions between single duckweed species and Cd has been extensively studied, the impact of duckweed diversity on Cd tolerance remains unknown. This study examines three duckweed species ( Lemna minor , Landoltia punctata , and Spirodela polyrhiza ) under Cd stress, assessing growth, chlorophyll content, Cd accumulation, removal efficiency, and metabolic adaptations in monoculture and mixed-culture systems. The results showed that the growth of duckweeds was inhibited across all cultivation modes under 10 mg L⁻ 1 Cd stress, with growth rates remaining below 0.1 g d⁻ 1 and experiencing more than 80% inhibition. Moreover, chlorophyll content declined with increasing Cd concentration under all cultivation modes, exceeding 64% inhibition under 10 mg L⁻ 1 Cd stress. Cd accumulation peaked in L. minor monoculture at 10 mg L⁻ 1 Cd (1601.33 mg kg⁻ 1 ), with bioenrichment coefficients exceeding 50 in all systems. The combination of S. polyrhiza and L. minor exhibited the highest Cd removal efficiency, achieving 50.60% at a Cd concentration of 1 mg L⁻ 1 . However, the Cd removal rate of all culture modes was below 30% when the Cd concentration increased to 10 mg L⁻ 1 . Elevated antioxidant enzyme activities (POD, CAT) and malondialdehyde (MDA) levels indicated enhanced oxidative stress responses. This study provides support for the application of duckweed polycultures in water contaminated with heavy metals.

Floatable artificial leaf to couple oxygen-tolerant CO2 conversion with water purification

Zhang Zhiyong,Wang Yang,Xie Yangen,Tsukamoto Toru,Zhao Qi,Huang Qing,Huang Xingmiao,Zhang Boyang,Song Wenjing,Chen Chuncheng,Sheng Hua,Zhao Jincai

To enable open environment application of artificial photosynthesis, the direct utilization of environmental CO 2 via an oxygen-tolerant reductive procedure is necessary. Herein, we introduce an in situ growth strategy for fabricating two-dimensional heterojunctions between indium porphyrin metal-organic framework (In-MOF) and single-layer graphene oxide (GO). Upon illumination, the In-MOF/GO heterostructure facilitates a tandem CO 2 capture and photocatalytic reduction on its hydroxylated In-node, prioritizing the reduction of dilute CO 2 even in the presence of air-level O 2 . The In-MOF/GO heterostructure photocatalyst is integrated with a porous polytetrafluoroethylene (PTFE) membrane to construct a floatable artificial leaf. Through a triphase photocatalytic reaction, the floatable artificial leaf can remove aqueous contaminants from real water while efficiently reducing CO 2 at low concentrations (10%, approximately the CO 2 concentration in combustion flue gases) upon air-level O 2 . This study provides a scalable approach for the construction of photocatalytic devices for CO 2 conversion in open environments.

Sulfonamides impede duckweed growth by inducing an excessive accumulation of salicylic acid

Aolin Ma,Siqi Liu,Zhixuan Du,Longfei Zhu,Guanping Feng

Sulfonamides, owing to their therapeutic properties and cost-effectiveness, are widely employed in clinical settings and the animal husbandry industry. However, the misuse of these compounds has resulted in severe water pollution, thereby posing significant threats to the growth and development of aquatic plants such as duckweed. Our controlled experiments revealed that sulfonamide exposure disrupts the plasma membrane integrity in duckweed root tip cortex cells and inhibits the root growth. Utilizing ESI-HPLC-MS/MS, we identified that sulfonamide exposure elicits an immune response in duckweed, characterized by the generation of substantial amounts of salicylic acid (SA). The exogenous application of SA exacerbated the sulfonamide-induced growth inhibition, whereas the addition of the SA synthesis inhibitor 1-aminobenzotriazole (ABT) mitigated the stress induced by sulfonamides. A loss-of-function mutant of ICS1, a key enzyme in the SA biosynthesis pathway, exhibited complete insensitivity to sulfonamide exposure. Furthermore, transcription factors SARD1 and CBP60g, which regulate ICS1 gene expression, were induced by sulfonamides, and their double mutant showed insensitivity to sulfonamide exposure. Reactive oxygen species (ROS) detection indicated abnormal accumulation of ROS in duckweed roots under sulfonamide exposure, which is likely to induce SA synthesis. These findings suggest that sulfonamide-mediated growth inhibition in plants is dependent on the ICS1-catalyzed SA biosynthesis pathway, thereby enhancing our understanding of the effects and mechanisms of sulfonamide exposure on aquatic plants.

Combined toxic effects of polypropylene and perfluorooctanoic acid on duckweed and periphytic microorganisms

Li Qi,Jiang Jiarui,Lan Yiyang,Kang Shiyun,Yang Yixia,Zhang Jiahui

Microplastics and perfluorooctanoic acid coexist in the aquatic environment. Duckweed was exposed to a range of concentrations (0.1–1000 μg L −1 ) of solutions containing polypropylene (PP) and perfluorooctanoic acid (PFOA) for 14 days to measure their toxicity. The result showed the single and combined PP and PFOA treatments did not significantly influence the growth of duckweed. The greatest PP and PFOA concentrations of combined pollution affect plant chlorophyll. Moreover, the combined treatment of duckweed consistently resulted in increased malondialdehyde (MDA) levels, indicating oxidative damage. As an antioxidant stress response, the combination-treated plants were encouraged to produce superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). Meanwhile, 3519 Operational Taxonomic Units (OTUs) were identified in the duckweed rhizosphere. Proteobacteria was the most predominant microbial community. Shannon, Simpson, and Chao1 discovered that microbial communities changed in response to single and combination PP and PFOA treatments, with decreased diversity and increased abundance. In addition, SEM analysis also revealed that the combined treatment significantly phyllosphere microorganisms. The findings of this investigation add to our knowledge of how PP and PFOA affect duckweed and the rhizospheric microorganisms, expanding the theoretical basis for employing duckweed in complex contamination.

Tissue-specific Responses of Duckweed to Cadmium Stress under Nanoplastic Co-exposure: Differential Accumulation and Toxicity in Roots and Fronds

Zhihao Xue,Xinyi Liao,Jie Hou,Jiang Xu,Daohui Lin

Nanoplastics (NPs), characterized by their small size and widespread presence, are known to interact with other contaminants, potentially modifying their environmental behavior and biological impacts. Herein, we explored the response of duckweed (Spirodela polyrhiza) to Cd (0, 0.01, 0.1, 1 mg/L) stress upon co-exposure to polystyrene NPs (PS-NPs) (0, 0.1, 1, 10 mg/L). Cd exhibits significantly higher toxicity to the duckweed compared to PS-NPs. Notably, the presence of PS-NPs exacerbates Cd toxicity in the roots, while mitigating its adverse effects on the fronds. In-situ analyses using Laser Ablation Inductively Coupled Plasma Mass Spectrometry and Confocal Laser Scanning Microscopy demonstrate that PS-NPs enhance Cd accumulation in the root tips (by up to 19.3%) but reduce its presence in the basal root regions and further translocation to the fronds by up to 26.8%. Metabolomics and gene expression analyses further indicate that PS-NPs elevate organic acid contents and modify gene expressions in duckweed against exogenous Cd stress, thereby inhibiting the root-to-frond translocation of Cd. The findings enhance our comprehension of the complex interactions between NPs and environmental pollutants in aquatic flora.
