In agritechnology applications, the sustained release of pesticides plays significant roles in efficient bioavailability and environmental protection. In this study, hydrophobic derivate of sodium alginate (Ugi-SA), polyacrylamide (PAM) and montmorillonite (MMT) were utilized to construct several stretchable double-network nanocomposite hydrogels, which were used to control the release of λ-cyhalothrin. These nanocomposite hydrogels were fully characterized by Fourier transform infrared spectroscopy (FTIR), thermal gravimetric analysis (TGA), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and X-ray diffraction (XRD). The adoption of Ugi-SA and appropriate incorporation of MMT could significantly improve the tensile properties , pesticide-loading efficiency and sustained release performances of the hydrogels. The swelling behavior of all the hydrogels well fits into the Fickian diffusion model. The maximum fracture strain and loading efficiency of the hydrogels reached 2000% and 81.30%, respectively. The hydrogel with 5% MMT content shows the lowest cumulative release percentage of 6.68% over 87 h, and the pesticide release curves well fit into the Weibull model. These novel nanocomposite hydrogels take advantages of both the gelation property of alginate derivatives and the adsorption capacity of clays, to serve as promising potential carriers and to improve the utilization efficiency of water-insoluble pesticides.

Purpose Peptide drugs have been used in therapy various diseases. However, the poor bioavailability of peptide drugs for oral administration has limited their clinical applications, on account of the acidic environment and digestive enzymes inside the human gastrointestinal tract. To enhance stability in the human gastrointestinal tract, bioavailability, and targeted drug delivery of peptide drugs through oral administration, a vitamin B12-modified amphiphilic sodium alginate derivative (CSAD-VB12) was synthesized.Materials and methods A vitamin B12-modified amphiphilic sodium alginate derivative (CSAD-VB12) was synthesized via the N,N’-dicyclohexylcarbodiimide active method at room temperature, and then characterized using FTIR and 1H NMR spectroscopy. Insulin was used as a model peptide drug and the insulin-loaded CSAD-VB12 (CSAD-VB12/insulin) nanoparticles with negative zeta potentials were prepared in PBS (pH=7.4). Scanning electron microscopy was used to observe CSAD-VB12/insulin as spherical nanoparticles. The CSAD-VB12 derivatives and CSAD-VB12/insulin nanoparticles displayed nontoxicity towards the human colon adenocarcinoma (Caco-2) cells by CCK-8 test. Caco-2 cell model was used to measure the apparent permeability (Papp) of insulin, CSAD/insulin and CSAD-VB12/insulin. Furthermore, confocal was used to confirm the endocytosis of intestinal enterocytes. Type 1 diabetes mice were used to evaluate the intestinal absorption and retention effect of test nanoparticles. Results They were observed as spherical nanoparticles in the size of 30–50 nm. The CSAD-VB12 derivatives and CSAD-VB12/insulin nanoparticles displayed nontoxicity towards the human colon adenocarcinoma (Caco-2) cells. Comparing with insulin and the CSAD/insulin nanoparticles, the CSAD-VB12/insulin nanoparticles exhibited higher permeation ability through intestinal enterocytes in the Caco-2 cell model. Oral administration of the CSAD-VB12/insulin nanoparticles to Type 1 diabetic mice yields higher intestinal retention effect, targeted absorption, and outstanding efficacy. Conclusion CSAD-VB12 derivatives enhance the small intestinal absorption efficacy and retention of peptide by oral administration, which indicated that it could be a promising candidate for oral peptide delivery in the prospective clinical application.

Photosensitizer-encapsulated amphiphilic sodium alginate derivative (Photosan-CSAD) nanoparticles were prepared because of their ability to enhance phototoxicity in the photodynamic therapy of pancreatic cancer . These nanoparticles are spherical, 150–250 nm in size as determined by transmission electron microscopy , and have negative zeta potentials . Upon incubation with human pancreatic cancer cells, the Photosan-CSAD nanoparticles showed high fluorescence activity and reactive oxygen species generation, resulting in strong phototoxicity. However, no dark toxicity was observed. Apoptosis played a leading role in the cell death process induced by the Photosan phototoxicity. These results demonstrate that the Photosan-CSAD nanoparticles are a candidate for the photodynamic therapy of pancreatic cancer.

In order to realize a combination of chemotherapy and selective drug release into tumor cells, novel pH-sensitive prodrugnanoparticles were designed and prepared via the self-assembly of a synthetic amphiphilic macromolecular prodrug for the selective co-delivery of doxorubicin (Dox) and curcumin (Cur). Dox was covalently conjugated to the oxidized sodium alginate through a Schiff base reaction to produce an amphiphilic macromolecular prodrug, and the prodrug was subsequently self-assembled into nanoparticles (Dox-NPs) in an aqueous solution, which were responsive to the acidic environment in tumor cells. Additionally, a second chemotherapeutic agent, Cur, was encapsulated in the core of nanoparticles (Cur–Dox-NPs) via hydrophobic effects, with a significant drug loading capacity. Cur–Dox-NPs exhibited an efficient release of both Dox and Cur in acidic media and further studies of their intracellular uptake and drug release confirmed that Dox-NPs were easily taken up by cells and selectively released the drug into the human breast cancer cell line MCF-7. In vitro cytotoxicity studies of the NPs showed a remarkable efficacy against MCF-7 cell lines, whereas an improved safety profile was observed in the human breast epithelial cell line MCF-10A. Furthermore, in vivo studies in zebrafish further confirmed an efficient absorption of Dox-NPs. In vivo cardiotoxicity experiments on a zebrafish model showed that Dox-NPs exhibited an improved cardiotoxicity profile in comparison with free Dox. This study demonstrated that this novel pH-sensitive prodrug-nanoparticle system may provide a simple and efficient platform for the selective co-delivery of multiple drugs to tumor cells.
