The current widely utilized polymer or C8, C18 end-capped material-based sorbents for solid-phase extraction could not capture alkaloids well only based on “like dissolves like” principle. In this paper, a layer-by-layer functionalized porous Zinc sulfide nanospheres-based solid-phase extraction (SPE) combined with liquid chromatography time-of-flight/mass spectrometry (LC-TOF/MS) and gas chromatography-mass spectrometry (GC-MS) was developed for the specific enrichment and identification of alkaloids from complex matrixes, Crinum asiaticum var. sinicum crude extracts. The functionalized porous Zinc sulfide nanospheres were prepared by the amidation reaction of poly-(acrylic acid) (PAA) homopolymer with amino groups onto the porous ZnS nanospheres. Tandem LC-TOF/MS spectrometry presented that the almost all of the twenty-three main peaks in elution fraction from the SPE could be inferred as alkaloids with ion of mass according to the nitrogen rule and hit formula with Peak View1.2@software from AB SCIEX, and seven alkaloids including two new found chemical entities were directly identified from their GC-MS spectra and retention indices. We believe that this SPE protocol can also be utilized in the future to selectively enrich alkaloids from extracts of other plant species .

Freeze-ultrasonic thawing extraction (FUTE) is a novel, green, and effective method for extracting bioactive compounds from plants, and the Gemini surfactant had the excellent properties especially for extracting hydrophobic and hydrophilic molecules. Therefore, the purpose of this study was to establish an effective method of synthetic Gemini surfactant aqueous system coupled with FUTE to extract the main individual polyphenols from the aerial parts of Empetrum nigrum. Based on identification of the polyphenols in the aerial parts of E. nigrum by ultra-high-performance liquid chromatography (UHPLC)–time of flight (TOF)–mass spectrometry (MS)/MS, the Gemini surfactant– N , N ʹ-bis(dodecyldimethyl)-1,4-butane dibromide (BBD)–FUTE technology was applied to extract the six main individual polyphenols. Under the optimum conditions, the extraction yields of neochlorogenic acid, catechin, rutin, hyperoside, quercetin, and guaijaverin were 8.48, 0.33, 4.23, 2.19, 1.12, and 0.32 mg/g, respectively, which were 1.01–1.96 and 1.15–2.39 times to those of ultrasonic-assisted extraction and freeze-thawing extraction methods, respectively. After cloud-point extraction, a preconcentration factor of 9.1 was obtained. All the results indicated that BBD-FUTE was a green and efficient method in the extraction of polyphenols from the aerial parts of E. nigrum . Therefore, it was suggested that the BBD-FUTE technology can be used as an alternative innovation method to effectively extract bioactive compounds from plant materials.

Developing low-cost and efficient metal-free carbon-based catalysts is of great importance but remains a challenge. In this work, rice straw derived biochar was prepared and doped with heteroatoms (N and S) for the catalytic degradation of metolachlor (MET) through activating peroxymonosulfate (PMS). Adsorption process played a critical role for the subsequent degradation process. Surface ketone C (-C = O) was the catalytic active sites for the PMS decomposition. Interestingly, N-doping positively whilst S-doping negatively influenced the MET degradation process. Through experimental investigation and theoretical calculations, N-doping was demonstrated to be beneficial for the interaction between biochar and PMS, thus accelerating the catalytic degradation process. The reactive oxidative species involved in the degradation process were explored by EPR analysis. The degradation pathways were elucidated based on the liquid chromatography time-of-flight mass (LC-TOF-MS) analysis of the degradation products. This study highlights the applications of biochar for the environmental catalysis.

Purpose: Zhizi-Bopi decoction (ZZBPD) is a classic herbal formula with wide clinical applications in treating liver diseases including hepatitis B. However, the mechanism needs to be elucidated. Methods: Chemical components of ZZBPD were identified by ultra-high-performance liquid chromatography coupled with time-of-flight mass spectrometry (UHPLC-TOF-MS). Then we used network pharmacology to identify their potential targets. Network construction, coupled with protein–protein interaction and enrichment analysis was used to identify representative components and core targets. Finally, molecular docking simulation was conducted to further refine the drug-target interaction. Results: One hundred and forty-eight active compounds were identified in ZZBPD, targeting 779 genes/proteins, among which 174 were related to hepatitis B. ZZBPD mainly influences the progression of hepatitis B through the hepatitis B pathway (hsa05161) via core anti-HBV targets (AKT1, PIK3CA, PIK3R1, SRC, TNF, MAPK1, and MAPK3). Enrichment analysis indicated that ZZBPD can also potentially regulate lipid metabolism and enhance cell survival. Molecular docking suggested that the representative active compounds can bind to the core anti-HBV targets with high affinity. Conclusion: The potential molecular mechanisms of ZZBPD in hepatitis B treatment were identified using network pharmacology and molecular docking approaches. The results serve as an important basis for the modernization of ZZBPD.

With the advantages of a high specificity, a long half-life, and a high safety, the use of antibody biologic drugs, including camrelizumab, has been rapidly increasing in clinical practice. Camrelizumab, an immune checkpoint inhibitor and humanized monoclonal antibody, is used to treat several advanced solid cancers. Measuring its concentration supports personalized dosage adjustments, influences treatment decisions for patients, strengthens the control of disease activity through therapeutic drug monitoring, and helps evaluate and prevent drug interactions in combination therapy. Because antibodies are present in complex biological matrices, quantifying monoclonal antibody drugs is challenging, and must rely on precise, selective, and reliable analytical methods. In this study, a quadrupole time-of-flight mass spectrometry TripleTOF 6600+ (AB SCIEX, Framingham, MA, USA) system equipped with a Turbo V ion source was used for the qualitative analysis of monoclonal antibodies using the data-dependent acquisition (IDA) MS/MS mode, followed by quantitative analysis using a targeted MRMHR workflow. This method showed a good linear relationship within the range of 4–160 μg/mL, with a correlation coefficient of R2≥ 0.996. It demonstrated an acceptable accuracy (88.95–101.18%) and precision (≤15%). Furthermore, the lower limit of quantification was found to be 4 μg/mL, with the lowest detection limit of 0.3217 μg/mL, indicating that this method is rapid, accurate, and reliable for the quantitative analysis of camrelizumab in human serum.

Although ligand fishing has been shown to be an efficient technique for the identification of bioactive components from complex mixtures such as natural products , it cannot be applied to biomedical image processing. Herein, a specific fluorescent ligand fishing combined with in situ imaging approach is presented for the identification of heat shock protein 90 (Hsp 90) inhibitors from complex matrixes, Curcuma longa L. , using N-terminus immobilized Hsp 90α functionalized InP/ZnS quantum dots embedded mesoporous nanoparticles ( i.e. Hsp 90α (NT)-FQDNs) as extraction sorbents and fluorescent tracer. The fished ligands were identified by liquid chromatography time-of-flight/mass spectrometry (LC-TOF/MS) and gas chromatography-mass spectrometry (GC-MS). Moreover, in situ imaging by confocal laser scanning microscopy (CLSM) was applied for evaluating the effect of fished-ligands on bioactivity-induced apoptosis morphologically in HeLa cells. MTT assay verified the bioactivity of the ligands and molecular docking results further provided convincing information to verify the feasible binding mode between ligands and protein. Twelve ligands as potential Hsp 90 inhibitors were ultimately fished and identified from Curcuma longa L. crude extracts. The proposed approach based on Hsp 90α functionalized nanocomposites is superior in the combination of highly specific screening efficiency and concurrent visual in situ imaging, which could have great promise for the development of other plant-derived Hsp 90 inhibitors, and providing a rapid and reliable platform for discovering biologically active molecules in natural products.

In industrial application of polycaprolactam, the content of caprolactam monomer and oligomers , as well as their average molecular weight are the key factors. In this work, a liquid-chromatography/time-of-flight mass spectrometry (LC-TOF-MS) method and an advanced polymer chromatography/multi-angle laser light scattering/refractive index detector (APC-MALLS-RID) hyphenated technique have been developed as a quantitative analysis method for monomer and cyclic oligomers, and a means of determining the molecular weight of polycaprolactam. The melt post-polycondensation reaction characteristics of polycaprolactam films and the variation in monomers and cyclic oligomers (C2–C7) content have been systematically investigated. The results showed excellent repeatability and stability of quantitative test results on monomers and cyclic oligomers in polycaprolactam. The content of monomers and cyclic oligomers in the polycaprolactam decreased from 10.21% to 1.52% at 260 °C and 50 Pa for 60 min, and the average molecular weight increased from 1.53 × 10 4 g/mol to 2.36 × 10 4 g/mol. Additionally, the results of differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA) experiments showed that the thermal stability and crystallization properties of polycaprolactam were improved after the film melt post-polycondensation reaction. Thus, this melt post-polycondensation method can realize the efficient removal of monomers and cyclic oligomers (compared with the traditional hot water extraction method) and afford high-quality polycaprolactam with high average molecular weight.
