Deep-Permeation Nanocomposite Structure of ZIF-8 inside Porous Poly(tetrafluoroethylene) by Flow Synergistic Synthesis

Yangmei Qin,Zeyi Xiao,Shizhao Jian,Yuyang Wang,Senqing Fan,Yinan Wang,Boya Qiu,Jingyun Liu,Zexue Wang,Qidong Wan

It is challenging to fabricate stable, complete, controllable, homogeneous metal–organic framework composites. Hereof, we suggested a versatile method where nanocrystals were assembled inside the porous polymer by synergistic chemical synthesis and permeation flow in and through its pores. The poly(tetrafluoroethylene) (PTFE)/ZnO and PTFE/ZIF-8 were fabricated as the prototype of the composites. For fabricating the composite, ZnO was first synthesized inside the pores followed by ligand vapor treatment. The even and well-distributed ZnO nanorods of 10–30 nm were synthesized inside pores of PTFE and converted into ZIF-8. The ZnO and ZIF-8 were quantified in 281 and 431 mg g–1 against the original PTFE, respectively. The composite is stable and the nanocrystals are not readily washed away. The adsorption uptake (C3 > C2 > C1) of the light hydrocarbon gases for the PTFE/ZIF-8 nanocomposite were preliminarily evaluated. The adsorption selectivities were calculated, where the selectivities for C2H6/CH4 and C3H8/CH4 were 10.9 and 42.8, respectively.

Biofilm-Responsive Polymeric Nanoparticles with Self-Adaptive Deep Penetration for In Vivo Photothermal Treatment of Implant Infection

Congyu Wang,Wei Zhao,Bing Cao,Zhixiong Wang,Qi Zhou,Siyu Lu,Ligong Lu,Meixiao Zhan,Xianglong Hu

Biofilm can protect bacteria from immune attacks and antibiotic inhibition, and bacterial biofilm hosted in implanted materials and medical devices is a serious threat for modern medical system. Herein, we report biofilm-responsive caged guanidine nanoparticles (CGNs) to deeply penetrate and accumulate in bacterial biofilm, and then efficient photothermal eradication of bacterial biofilm is achieved upon NIR laser irradiation via the proof-of-concept formulation of photothermal agents in CGNs. In physiological conditions and blood circulation, CGNs are negatively charged by masking the positive charge of guanidine via covalent modification with acid-cleavable moieties, exhibiting high biocompatibility and minimal hemolysis. Whereas upon blood circulation and passive accumulation at infected implant sites, CGNs are self-adaptive in acidic biofilm to release the protective caging group and expose native guanidine moieties, which can promote nanoparticle deep biofilm penetration and bacteria adhesion as well as membrane fusion. After that, remarkable photothermal effect with a high photothermal conversion efficiency of ∼40.9% can eradicate implant biofilm upon NIR laser irradiation. It can efficiently treat S. aureus biofilm-infected implant catheters in vivo via only one single treatment in a mouse model, exhibiting ∼99.6% bacteria inhibition ratio. Apart from this proof-of-concept work, current guanidine-caged biofilm responsive polymeric nanoparticles are promising general vectors to treat biofilm and resistant pathogens in medicine and daily healthcare.

Tumor-activatable ultrasmall nanozyme generator for enhanced penetration and deep catalytic therapy

Xinping Liu,Zhengwei Liu,Kai Dong,Si Wu,Yanjuan Sang,Tingting Cui,Ya Zhou,Jinsong Ren,Xiaogang Qu

Tumor-activatable ultrasmall nanozyme generation is an unprecedented strategy to overcome intrinsically fatal defects of traditional reactive oxygen species (ROS)-based nanoagents for deep tumor penetration, including limited tissue-penetrating depth of external energy, heavy reliance on oxygen and nonspecific toxicity of therapeutic agents. Here, based on the cascade reaction between glucose oxidase (GOx) and ultrasmall peroxidase nanozyme embedded into acid-dissociable zeolitic imidazolate framework-8 (ZIF-8), such a tumor-activatable ultrasmall nanozyme generator is designed for enhanced penetration and deep catalytic therapy. With the aid of mildly acidic tumor microenvironment , the produced gluconic acid from intratumoral glucose can gradually induce the dissociation of ZIF-8 to release ultrasmall peroxidase nanozyme with significant intratumoral penetration. On the other hand, the generated hydrogen peroxide with relatively long-life can be subsequently catalyzed by penetrated peroxidase nanozyme into toxic hydroxyl radicals for deep catalytic therapy. In this way, the well-designed nanoplatform not only can greatly enhance tumor penetration but also directly induce exogenous ROS without oxygen participation and external energy input, thereby thoroughly avoiding the inactivation of traditional ROS-based nanoagents in the extremely hypoxic tumor center and finally resulting in remarkable deep catalytic therapy.

Enzyme-triggered transcytosis of drug carrier system for deep penetration into hepatoma tumors

Han Yan,Pengchao Xu,He Ma,Yanan Li,Runfeng Zhang,Hailin Cong,Bing Yu,Youqing Shen

In recent years, nano-drug delivery systems have made considerable progress in the direction of tumor treatment, but the low permeability of drugs has restricted the development of nano drugs. To solve this problem, we constructed a nano-drug delivery system with the dual effects of γ-glutamyltransferase (GGT) reaction and high nuclear targeting in tumor microenvironment to promote the deep penetration of drugs. Over-expression of GGT in tumor cells can specifically recognize γ-glutamyl substrate and release amino group from the hydrolysis reaction , which makes the whole system change from negative or neutral to positive charge system. The conjugated complex with positive charge rapidly endocytosis through electrostatic interaction , enhancing its permeability in tumor parenchyma. At the same time, the cell penetrating TAT contains a large amount of lysine, which can be identified by the nuclear pore complexes (NPCs) on the surface of the nuclear membrane , showing excellent nuclear localization function. The active DOX is released in the nucleus, which inhibits the mitosis of cancer cells and enhances the active transport ability of drugs in tumor cells. Therefore, this drug delivery system actively transports adriamycin into the tumor to achieve deep penetration of drugs through enzyme response and nuclear targeting, showing high anti-tumor activity and can be effectively applied to the treatment of liver cancer.
