Mxene-based mild hyperthemia microneedle patch for diabetic wound healing

Yuci Gan,Bo Liang,Yan Gong,Lizhen Wu,Peizhe Li,Chuchen Gong,Pei Wang,Ziyou Yu,Lingling Sheng,Da-Peng Yang,Xiansong Wang

The treatment of diabetic wounds remains a serious challenge due to the unfavorable diabetic microenvironment, characterized by high glucose levels, excessive reactive oxygen species (ROS), and chronic inflammation. For decades, several hypoglycemic drugs have been used clinically to decrease blood glucose levels; however, they are unable to regulate in situ hyperglycemic microenvironments. In this study, we introduced a poly (γ-glutamic acid) (γ-PGA)-based microneedle (MN) patch encapsulated with glucose oxidase (GOx)-loaded Ti 3 C 2 MXene nanosheets (MXene) termed MN-PGA-MXene-GOx. GOx, which lowers local glucose and pH by oxidizing glucose, is delivered deep into tissues via the MN patch, while MXene ensures its prolonged release. When the MN patch is exposed to external NIR light, MXene can induce mild hyperthermia, enhance the catalytic activity of GOx, and promote cell proliferation, migration, angiogenesis, and tissue remodeling. Both γ-PGA and MXene possess antioxidant properties, mitigating ROS in the diabetic environment and reducing hydrogen peroxide (H 2 O 2 ) produced during glucose oxidation. In conclusion, this multifunctional microneedle patch provides a novel approach for accelerating diabetic wound healing, utilizing GOx and MXene for in-situ glucose management and mild hyperthermia.

Synergistic inactivation of Escherichia coli O157:H7 by plasma-activated water and mild heat

Qisen Xiang,Wenjie Wang,Dianbo Zhao,Liyuan Niu,Ke Li,Yanhong Bai

This study was designed to evaluate the synergistic antimicrobial effect of plasma-activated water (PAW) combined with mild heat (40, 50, and 60 °C) against Escherichia coli O157:H7. Populations of E. coli O157:H7 were reduced from about 8.28 log CFU/mL to undetectable levels following exposure to the combined treatment of PAW and mild heat (60 °C, 4 min), which was higher than that of individual treatment of PAW (0.77 log CFU/mL) or mild heat (1.78 log CFU/mL). The results demonstrated that PAW had good synergistic activity with mild heat against E. coli cells. Field emission scanning electron microscope (FE-SEM) confirmed that the combined treatment with PAW and mild heat caused marked morphological damage of E. coli cells. The N-phenyl-1-napthylamine (NPN) uptake assay and propidium iodide (PI) staining indicated that the combined treatment with PAW and mild heat synergistically enhanced the permeability of the outer and cytoplasmic membrane of E. coli cells, resulting in the leakage of intracellular components such as proteins and nucleic acids. It can be concluded that membrane damage is a key mechanism involved in the inactivation of E . coli cells by the combined treatment with PAW and mild heat. This synergistic antimicrobial treatment of PAW and mild heat may be used as a potential intervention strategy to reduce the incidence of E. coli and other pathogens entering the food chain.

Self-Supplied Reactive Oxygen Species-Responsive Mitoxantrone Polyprodrug for Chemosensitization-Enhanced Chemotherapy Under Moderate Hyperthermia

Hongjie Zhang,Jing Wang,Ruiying Wu,Benyan Zheng,Yanxiang Sang,Bibo Wang,Lei Song,Yuan Hu,Xiaopeng Ma

Currently, the secondary development and modification of clinical drugs has become one of the research priorities. Researchers have developed a variety of TME-responsive nanomedicine carriers to solve certain clinical problems. Unfortunately, endogenous stimuli such as reactive oxygen species (ROS), as an important prerequisite for effective therapeutic efficacy, are not enough to achieve the expected drug release process, therefore, it is difficult to achieve a continuous and efficient treatment process. Herein, a self-supply ROS-responsive cascade polyprodrug (PMTO) was designed. The encapsulation of the chemotherapy drug mitoxantrone (MTO) in a polymer backbone could effectively reduce systemic toxicity when transported in vivo. After PMTO was degraded by endogenous ROS of the TME, another part of the polyprodrug backbone became cinnamaldehyde (CA), which can further enhance intracellular ROS, thereby achieving a sustained drug release process. Meanwhile, due to the disruption of the intracellular redox environment, the efficacy of chemotherapy drugs is enhanced. Finally, the anticancer treatment efficacy was further enhanced due to the mild hyperthermia effect of PMTO. In conclusion, the designed PMTO demonstrated remarkable antitumor efficacy, effectively addressing the limitations associated with MTO. This article is protected by copyright. All rights reserved

DNA Damage Inducer Mitoxantrone Amplifies Synergistic Mild-Photothermal Chemotherapy for TNBC via Decreasing Heat Shock Protein 70 Expression

Zuqin Chen,Sunfan Li,Fangzhou Li,Cheng Qin,Xianlei Li,Guangchao Qing,Jinjin Wang,Bozhang Xia,Fuxue Zhang,Liangliang Meng,Xing-Jie Liang,Yueyong Xiao

Patients with triple-negative breast cancer (TNBC) have the worst clinical outcomes when compared to other subtypes of breast cancer. Nanotechnology-assisted photothermal therapy (PTT) opens new opportunities for precise cancer treatment. However, thermoresistance caused by PTT, as well as uncertainty in the physiological metabolism of existing phototherapeutic nanoformulations, severely limit their clinical applications. Herein, based on the clinically chemotherapeutic drug mitoxantrone (MTO), a multifunctional nanoplatform (MTO-micelles) is developed to realize mutually synergistic mild-photothermal chemotherapy. MTO with excellent near-infrared absorption (≈669 nm) can function not only as a chemotherapeutic agent but also as a photothermal transduction agent with elevated photothermal conversion efficacy ( ƞ = 54.62%). MTO-micelles can accumulate at the tumor site through the enhanced permeability and retention effect. Following local near-infrared irradiation, mild hyperthermia (<50 °C) assists MTO in binding tumor cell DNA, resulting in chemotherapeutic sensitization. In addition, downregulation of heat shock protein 70 (HSP70) expression due to enhanced DNA damage can in turn weaken tumor thermoresistance, boosting the efficacy of mild PTT. Both in vitro and in vivo studies indicate that MTO-micelles possess excellent synergetic tumor inhibition effects. Therefore, the mild-photothermal chemotherapy strategy based on MTO-micelles has a promising prospect in the clinical transformation of TNBC treatment.

Mild thermotherapy and hyperbaric oxygen enhance sensitivity of TMZ/PSi nanoparticles via decreasing the stemness in glioma

Zeng Xiaofan,Wang Qi,Tan Xuan,Jia Le,Li Yuwei,Hu Mingdi,Zhang Zhijie,Bai Xicheng,Zhu Yanhong,Yang Xiangliang

Background Glioma is a common brain tumor with a high mortality rate. A small population of cells expressing stem-like cell markers in glioma contributes to drug resistance and tumor recurrence. Methods Porous silicon nanoparticles (PSi NPs) as photothermal therapy (PTT) agents loaded with TMZ (TMZ/PSi NPs), was combined with hyperbaric oxygen (HBO) therapy in vitro and in vivo. To further investigate underlying mechanism, we detected the expression of stem-like cell markers and hypoxia related molecules in vitro and in vivo after treatment of TMZ/PSi NPs in combination with PTT and HBO.Results NCH-421K and C6 cells were more sensitive to the combination treatment. Moreover, the expression of stem-like cell markers and hypoxia related molecules were decreased after combination treatment. The in vivo results were in line with in vitro. The combination treatment presents significant antitumor effects in mice bearing C6 tumor compared with the treatment of TMZ, PTT or TMZ/PSi NPs only. Conclusion These results suggested the TMZ/PSi NPs combined with HBO and PTT could be a potential therapeutic strategy for glioma.

Effects of cuminaldehyde combined with mild heat against Salmonella enterica serovar Typhimurium in powdered infant formula

Hui Li,Keren Agyekumwaa Addo,Yi-gang Yu,Xing-long Xiao

The study investigated the influence of cuminaldehyde (CUM) on cell membrane and genomic DNA of Salmonella enterica serovar Typhimurium ( S . Typhimurium), and revealed its antibacterial mechanism. The minimum inhibitory concentration (MIC) of CUM for S . Typhimurium was determined to be 0.6 μL/mL. CUM could bind to bacterial membrane proteins and change its structure and conformation, leading to the leakage of intracellular substances and the destruction of cell integrity. DNA interaction analysis showed that CUM bound to genomic DNA through groove mode, affecting the normal functioning of S . Typhimurium. In addition, CUM at sub-MIC decreased bacterial motility, prevent cell attachment, restrain metabolic activity, and thus inhibiting S . Typhimurium biofilm formation. Furthermore, mild heat treatment combined with CUM at 1, 2 and 4 MIC could completely kill S . Typhimurium in powdered infant formula. Besides, the combination at 1 MIC of CUM had no obvious influence on the aroma of the powdered infant formula. The combined application of CUM has broad prospects in the control of food-borne pathogens.

Physicochemical and foam properties of ovalbumin-carboxymethylcellulose mixtures after mild heat treatment: Comparison of electrostatic repulsion and attraction

Ya Li,Liwei Mao,Lifeng Wang,Wenfei Xiong

The effects of electrostatic repulsion (pH6.5) and attraction (pH4.6) between biopolymers on the physicochemical and foam properties of ovalbumin (OVA)/carboxymethylcellulose (CMC) mixtures subjected to mild heat treatment (60–75 °C) were comparatively investigated. It was found that electrostatic binding between OVA and CMC at pH 4.6 resulted in an increase in the peak temperature of thermal denaturation (T d ) of OVA from 71.3 °C to 74.4 °C, while no significant effect was observed at pH 6.5. Thus, 60 and 65 °C below T d were chosen to further investigate the effects of heating on the degree of thermal aggregation, protein conformation and foam properties of OVA/CMC mixtures. Confocal laser scanning microscopy observations and rheological results indicated that electrostatic binding between OVA and CMC at pH 4.6 significantly reduced the degree of protein thermal aggregation, while on the contrary, electrostatic repulsion between proteins and polysaccharides at pH 6.5 promoted OVA thermal aggregation. Raman spectroscopic characterisation indicated that mild heat treatment mainly led to a change in the microenvironment polarity of OVA tyrosine and tryptophan residues, as well as an increase in the content of random coil in the amide III band irrespective of pH. Moreover, after heating at 65 °C for 20 min, The foaming ability of the OVA/CMC mixtures at pH 6.5 and 4.6 increased by approximately 32% and 36% respectively compared to pure OVA. These findings have a positive value for enhancing the heat sterilisation strength of egg white proteins.

Unlocking the Potential of ZIF-67 as an Efficient Electrocatalyst for the Oxygen Reduction Reaction

Yipu Xu,Wenru Zhao,Rui Chen,He Li,Xinyu Liu,Peiran Wu,Hui Yu,Jiansong Wang,Liu-Liu Shen,Gui-Rong Zhang,Donghai Mei

Zeolitic imidazolate frameworks (ZIFs) are widely explored as precursors for constructing electrocatalysts for the oxygen reduction reaction (ORR). However, most studies convert these frameworks into metal-nitrogen-carbon (M-N-C) catalysts via pyrolysis, a process that often compromises their structure integrity and coordination precision. Herein, it is demonstrated that ZIF-67 can serve directly as a highly-active molecular catalyst for ORR. By applying ball-milling to enhance cobalt sites exposure, the optimized ZIF-67 exhibits exceptional ORR activity in alkaline media. Combined spectroscopic and theoretical analyses reveal that this remarkable activity stems from coordinatively unsaturated Co-N 4-x sites, which transform into Co-N 4-x (OH) moieties under reaction conditions, thereby facilitating oxygen adsorption and activation. Furthermore, mild thermal treatment (≤500 °C) enhances the proportion of Co-N 4-x sites up to 48%, as confirmed by X-ray absorption spectroscopy, achieving ORR activity rivals or even exceeds conventional pyrolyzed M-N-C catalysts. This study unlocks the potential of ZIFs as active molecular electrocatalysts upon mild thermal treatment, providing a scalable and efficient strategy for optimizing ZIFs for catalytic applications while preserving their structural precision.

Preparation of silver nanoparticles/polymethylmethacrylate/cellulose acetate film and its inhibitory effect on Cronobacter sakazakii in infant formula milk

Hui Li,Xiaoyan Feng,Xianxiang Zeng,Qixiu You,Wen Li,Hengyi Xu

ABSTRACT Cronobacter sakazakii is a harmful foodborne pathogen, and its contaminated food will pose a huge threat to human health. Prevention of C. sakazakii contamination of food is valuable for food safety as well as for human health. In this study, silver nanoparticles (AgNP) were successfully immobilized on the surface of cellulose acetate (CA) and polymethylmethacrylate (PMMA) composite to obtain AgNP/PMMA/CA film. Through the inhibition zone and growth curve experiments, we found that AgNP/PMMA/CA films has excellent antibacterial activity on C. sakazakii . The AgNP/PMMA/CA film can prolong the lag phase of the growth curve of C. sakazakii from 2 to 8 h. The antibacterial films were found to reduce the survival of C. sakazakii in Luria-Bertani and infant formula by combining it with a mild heat treatment (45°C, 50°C, and 55°C). The AgNP/PMMA/CA film combined with 55°C water bath can completely inactivate C. sakazakii in infant formula within 120 min. Finally, the potential mechanism by which AgNP/PMMA/CA films reduce the heat tolerance of C. sakazakii was investigated by quantitative real-time PCR. The results showed that AgNP/PMMA/CA films could reduce the expression of environmental tolerance-related genes in C. sakazakii . The current research shows that AgNP/PMMA/CA film has strong antibacterial activity, and the antibacterial film combined with mild heat treatment can accelerate the inactivation of C. sakazakii and effectively reduce the harm of foodborne pathogens. The AgNP/PMMA/CA film can be used as a potential packaging material or antibacterial surface coating.

Tumor immunotherapy boosted by R837 nanocrystals through combining chemotherapy and mild hyperthermia

Zhengjie Meng,Xue Fang,Bowen Fu,Cheng Qian,Zheng Yang,Yunhao Bai,Xinyue Tao,Haixiao Huang,Chenyu Ma,Wenjun Miao,Hao Ren,Aiyun Wang,Xueming Li

Melanoma is a malignant skin cancer that is prone to metastasis in the early stage and has a poor prognosis. Immunomodulatory therapy for melanoma has been a hot research topic in recent years. However, low immune cell infiltration and loss of tumor immunogenicity may occur in tumors, resulting in low response rates to immunotherapy . Thus, immunomodulatory therapy is usually used in combination with chemotherapy and radiotherapy. Development of combined therapeutic strategies with low systemic toxicity, high immune responsiveness and long-term inhibition of metastasis and recurrence of melanoma is the goal of current research. In this study, the insoluble immune adjuvant imiquimod (R837) was prepared as nanocrystals and coated with polydopamine (PDA) to form R837@PDA, which was then loaded into chitosan hydrogel (CGP) to form the drug-loaded gel system, R837@PDA@CGP (RPC), to combine immunomodulation effects, induction of immunogenic cell death (ICD) effects and immune-enhancement effects. After treatment with RPC, ICD in melanoma was induced, and the infiltration rate of cytotoxic T cells (CTLs) in melanoma was also significantly enhanced, which turned the tumor itself into an in situ vaccine and boosted the cancer-immunity cycle at the tumor site. Therefore, melanoma growth, metastasis and recurrence were notably inhibited.

Mild Hyperthermia-Enhanced Enzyme-Mediated Tumor Cell Chemodynamic Therapy

Xinhe Liu,Ying Liu,Junning Wang,Tianxiang Wei,Zhihui Dai

The heterogeneity and diversity of tumors seriously attenuate the curative outcome of single treatment modes. Combined therapy has been demonstrated to be a promising candidate to enhance therapeutic efficacy compared with monotherapy. As an emerging therapeutic strategy, chemodynamic therapy (CDT) has drawn extensive attention in recent years. However, the therapeutic efficiency of CDT is still unsatisfying because the level of intracellular hydrogen peroxide (H2O2) restricts the production of hydroxyl radicals (•OH). In this study, a novel curative strategy which combines glucose oxidase (GOx)-mediated Fe3O4-based Fenton reaction and multiwalled carbon nanotube (MWNT)-produced mild hyperthermia enhancer is proposed, achieving a mild hyperthermia-enhanced enzyme-mediated tumor cell CDT. GOx can catalyze the conversion of glucose into gluconic acid and H2O2, which can elevate acidity in the tumor microenvironment and boost Fe3O4-based Fenton reaction, producing a myriad of •OH to induce tumor cell death. Furthermore, by using the theory that a temperature rise expedites the kinetics of a chemical reaction, producing a higher reaction rate and more resultants per unit time, we integrate MWNTs in this therapy system, which generate mild hyperthermia so as to accelerate the Fenton reaction for increasing the productivity of •OH. Therefore, an amplified CDT can be realized. The therapy platform, mild hyperthermia-enhanced GOx-mediated CDT, provides an effective treatment for cancer and takes CDT a step further.

Hollow silver–gold alloy nanoparticles for enhanced photothermal/photodynamic synergetic therapy against bacterial infection and acceleration of wound healing

Shanshan Lin,Hui Chen,Rui Wang,Tongmeng Jiang,Rui Wang,Fabiao Yu

Bacterial infection seriously restricts the wound healing process due to severe inflammation and delayed wound healing. Unfortunately, the overuse or improper use of antibiotics leads to the advent of multidrug-resistant strains and intractable biofilms, severely affecting the therapeutic effect. Therefore, there is an urgent need to develop antibiotic-free strategies to accelerate the healing process of wounds with bacterial infection. Considering that single photothermal therapy (PTT) or photodynamic therapy (PDT) cannot fully meet the requirements of clinical sterilization and accelerating wound healing, herein, hollow silver–gold alloy nanoparticles immobilized with the photosensitizer molecule Ce6 (Ag@Au-Ce6 NPs) integrated with PTT and PDT are proposed for killing bacteria and accelerating wound healing. The photothermal conversion properties of Ag@Au-Ce6 NPs are obtained using an infrared thermal imager, and the generation of singlet oxygen (1O2) is verified with an 1O2 fluorescent probe DCFH-DA. Manipulated by near-infrared laser triggered mild hyperthermia and limited ROS amount, Ag@Au-Ce6 NPs could effectively kill bacteria that are free and colonized on the surface of wounded skin, promoting epithelium migration and vascularization, further accelerating wound healing, which showed great promise for biomedical application.
