Effects of elevated temperature and copper exposure on the physiological state of the coral Galaxea fascicularis

Meng Cheng,Yong Luo,Xiao-Lei Yu,Lin-Tao Huang,Jian-Sheng Lian,Hui Huang

The co-occurrence of elevated seawater temperature and local stressors (heavy metal contamination) affects the ecophysiology of phototrophic species, and represents a risk to the environmental quality of coral reefs. Therefore, we investigated the effects of both Cu alone and Cu in combination with elevated temperature (ET) on the physiology of the coral Galaxea fascicularis , and measured the parameters related to the photo-physiology and oxidative state. G.fascicularis is one of the dominant coral species in the South China Sea which exhibits strong adaptability to environmental stress. We exposed the common coral species G.fascicularis to a series of environmentally relevant concentrations of Cu at 29 °C (normal temperature, NT) and 32 °C (elevated temperature, ET) for 96 h. Single polyps were used in the experiments, which reduced individual variability when compared to the coral colonies. The results suggested that: i) Cu or ET had significant negative effects on the actual operating ability of photosystem Ⅱ (PSII), but not on the maximal chlorophyll fluorescence in darkness (Fv/Fm). ii) Symbiodiniaceae density was significantly reduced by high Cu concentrations, for Cu-NT and Cu-ET, a high concentration of Cu (40 μg/L) significantly impacted Symbiodiniaceae density, causing a 75.4% and 81.0% decrease, respectively. iii) the content of malondialdehyde (MDA) in coral tissues increased significantly under Cu-ET. iv) a certain range of copper concentration (25–30 μg/L) increased the pigment content of the Symbiodiniacea. Our results indicated that the combined stressors of Cu and ET made the coral tissue sloughed, caused the coral tissue damaged by lipid oxidation, reduced the photosynthetic capacity of the Symbiodiniacea, and led to the excretion of Symbiodiniacea.

Growth under elevated air temperature alters secondary metabolites in Robinia pseudoacacia L. seedlings in Cd- and Pb-contaminated soils

Y.H. Zhao,X. Jia,W.K. Wang,T. Liu,S.P. Huang,M.Y. Yang

Plant secondary metabolites play a pivotal role in growth regulation, antioxidant activity, pigment development, and other processes. As the global climate changes, increasing atmospheric temperatures and contamination of soil by heavy metals co-occur in natural ecosystems, which alters the pH of rhizosphere soil and influences the bioavailability and mobility of metals. Elevated temperatures in combination with heavy metals are expected to affect plant secondary metabolites, but this issue has not been extensively examined. Here, we investigated secondary metabolites in Robinia pseudoacacia seedlings exposed to elevated temperatures using a passive warming device in combination with Cd- and Pb-contaminated soils. Heavy metals significantly stimulated the accumulation of saponins, phenolic compounds , and flavonoids in leaves and stems; alkaloid compounds increased in leaves and decreased in stems, and condensed tannins fluctuated. Elevated temperatures, alone and in combination with Cd and Pb, caused increases in secondary metabolites in the plant tissues. Phenolic compounds showed the greatest changes among the secondary metabolites and significant interactive effects of temperature and metals were observed. These results suggest that slightly elevated temperature could enhance protective and defense mechanisms of Robinia pseudoacacia seedlings exposed to heavy metals by stimulating the production of secondary metabolites.

Chain microstructure comparison of two transparent polypropylene resins fractionated by temperature rising elution fractionation

Li Pei,Liu Wei,Liu Xue,Tang Yujing,Qiao Jinliang,Zhang Chunbo,Li Chenggang,Wang Xin,Ji Xiangling

Microstructure distribution in polyolefin resins is one of the key factors governing the macroscopic properties of the products. The chain microstructure of two transparent poly(propylene- co -1-butene) (PPB) resins is investigated in this study through temperature rising elution fractionation in the temperature range of 35 − 140 °C. Both resins contain small amount of room temperature soluble component, i.e. less than 3.80 wt% fractions eluted at 35 − 50 °C, implying that the two resins could be used as food containers or food packaging films safely with very low migration of soluble contents. The two resins exhibit remarkable differences in composition distributions as 41.24 wt% of resin PPB-1 was collected at 100 °C, whereas 58.24 wt% of resin PPB-2 was collected at 83 °C. The composition and sequence distribution in each fraction were analyzed by 13 C NMR spectroscopy. All the fractions are random propylene/1-butene copolymers containing 1-butene concentration in the range of 2.7 − 7.6 mol% for resin PPB-1 and 2.2 − 8.5 mol% for resin PPB-2. Individual 1-butene unit is mainly inserted into the polypropylene main chains since the average sequence length of 1-butene ( n B ) is really short (1.0 − 1.3). Resin PPB-1 has more fractions with high molecular weight and isotacticity collected above 105 °C than resin PPB-2, which contributes to a better stiffness of resin PPB-1. Besides, the difference in haze for the films produced from resins PPB-1 and PPB-2 is also demonstrated based on the chain microstructure results. It is found that resin PPB-2 has higher MFR and longer propylene sequences than resin PPB-1, resulting in a higher haze.

Photothermal Soft Nanoballs Developed by Loading Plasmonic Cu2–xSe Nanocrystals into Liposomes for Photothermal Immunoassay of Aflatoxin B1

Xue Li,Lin Yang,Chen Men,Yi Fen Xie,Jia Jun Liu,Hong Yan Zou,Yuan Fang Li,Lei Zhan,Cheng Zhi Huang

Photothermal effects (PTEs) have been greatly concerned with the fast development of new photothermal nanomaterials. Herein we propose a photothermal immunoassay (PTIA) by taking mycotoxins (AFB1) as an example based on the PTEs of plasmonic Cu2–xSe nanocrystals (NCs). By loading plasmonic Cu2–xSe NCs into liposomes to form photothermal soft nanoballs (ptSNBs), on which aptamer of AFB1 previously assembled, a sandwich structure of AFB1 could be formed with the aptamer on ptSNBs and capture antibody. The heat released from the ptSNBs under NIR irradiation, owing to the plasmonic photothermal light-to-heat conversion through photon–electron–phonon coupling, makes the temperature of substrate solution increased, and the increased temperature has a linear relationship with the AFB1 content. Owing to the large amounts of plasmonic Cu2–xSe NCs in the ptSNBs, the PTEs get amplified, making AFB1 higher than 1 ng/mL detectable in food even if with a rough homemade immunothermometer. The proposal of PTIA opens a new field of immunoassay including developing photothermal nanostructures, new thermometers, PTIA theory, and so on.
