Vibrio alginolyticus is a ubiquitous marine opportunistic pathogen that can infect various hosts in marine environment. In the present study, V. alginolyticus strain Wz11 was isolated from diseased cuttlefish , Sepia pharaonis , with 20% of promoted death and high survival capacity in skin mucus and tissue liquid. Its growth, siderophore production, and expressions of haemolysin and swarming related genes were characterized under iron limited conditions. The minimal inhibitory concentration (MIC) of 2,2′-dipyridyl (DP) to V. alginolyticus strain Wz11 was 640 μM. While growth of V. alginolyticus strain Wz11 was inhibited by DP , production of iron-seizing substances, haemolytic activity and swarming motility were increased. Moreover, expressions of haemolysin related genes tlh , tdh and vah and flagellar related genes flgH , fliC , fliD and fliS were also characterized using real-time reverse transcriptase PCR. Expression of tdh was up-regulated to 7.7-fold, while expressions of tlh and vah were down-regulated to 0.016-fold and 0.03-fold, respectively. The expression of fliC , flgH , fliD and fliS was up-regulated to 4.9-, 3.8-, 8.6- and 4.5-fold, respectively. Concluded from our results suggested that V. alginolyticus strain Wz11 was considered as a potential pathogen of S. pharaonis , and iron level played an important role in the production of iron-seizing substances, and activities of haemolysin and bacterial swarming as well as their related gene expressions.

The antiglycemic drug metformin, which is widely prescribed as a first-line drug for the treatment of type 2 diabetes, has become a concern for emerging pollutants in natural ecosystems. However, its effects on bacterial swimming motility remain unclear. In this study, we showed that metformin promotes bacterial surface aggregation by tracking swimming and by measuring the density distribution of Escherichia coli cultured with metformin near a surface in a homogeneous environment. Flagella are essential for the promotion of bacterial surface aggregation by metformin. Swimming motility, which is mediated by flagella, determines bacterial surface aggregation. The promotion of bacterial surface aggregation by metformin is caused by a reduction in swimming motility, which is governed by a decrease in the proton motive force. Our results reveal that metformin has a pronounced effect on flagellated bacterial motility associated with surface sensing and aggregation.

By employing external fertilization (broadcast spawning) as a mating strategy, the gametes and subsequent fertilization of various marine invertebrates are directly subjected to pollution. Although microplastics (MPs) are ubiquitous in marine environments, their potential effects on the fertilization of broadcast spawners remain largely unknown. Therefore in this study, the impacts of polystyrene MPs on the fertilization success of broadcast spawning bivalve ( Tegillarca granosa ) were investigated. In order to reveal the underlying mechanisms affecting fertilization, the sperm swimming performance, sperm ATP status, sperm viability, DNA integrity, gamete collision probability, gamete fusion efficiency, enzymatic antioxidants, and key ion transport enzyme activities were analyzed. The results showed that MPs weakened the sperm swimming performance through reducing ATP production and cell viability, thus leading to the decreased probability of gamete collision. Furthermore, MPs affected ion transport in the gametes by inducing oxidative stress, which resulted in gamete fusion failure. In conclusion, this study demonstrates that MPs could significantly decrease the fertilization success of T. granosa through reducing gamete collision and lowering gamete fusion efficiency.

Bacteria-inspired magnetic helical micro-/nanoswimmers can be actuated and steered in a fuel-free manner using a low-strength rotating magnetic field, generating remotely controlled 3D locomotion with high precision in a variety of biofluidic environments. They are therefore envisioned for biomedical applications related to targeted diagnosis and therapy. In this article, a porous hollow microswimmer possessing an outer shell aggregated by mesoporous spindle-like magnetite nanoparticles (NPs) and a helical-shaped inner cavity is proposed. The fabrication is straightforward via a cost-effective mass-production process of biotemplated synthesis using helical microorganisms. Here, Spirulina -based fabrication is demonstrated as an example. The fabricated microswimmers are superparamagnetic and exhibit low cytotoxicity. They are also capable of performing structural disassembly to form individual NPs using ultrasound when needed. For the first time in the literature of helical microswimmers, a porous hollow architecture is successfully constructed, achieving an ultrahigh specific surface area for surface functionalization and enabling diffusion-based cargo loading/release. Furthermore, experimental and analytical results indicate better swimming performance of the microswimmers than the existing non-hollow helical micromachines of comparable sizes and dimensions. These characteristics of the as-proposed microswimmers suggest a novel microrobotic tool with high loading capacity for targeted delivery of therapeutic/imaging agents in vitro and in vivo.
