Accumulation behavior and risk assessment of heavy metals and arsenic in tissues of white bream (Parabramis pekinensis) from the Xiang River, southern China

Jia Yuyu,Kong Qian,Yang Zhaoguang,Wang Lin

Aquatic environment suffered from serious heavy metal and As pollutions due to rapid industrialization and urbanization in the last decades. In order to evaluate the heavy metal and As contamination in aquatic organism in southern China, the levels of As, Cd, Co, Cr, Cu, Fe, Mn, Pb, and Zn in muscle, gill, and liver of white bream ( Parabramis pekinensis ) collected from Xiang River were analyzed. The statistically significant differences of the analyzed element concentration (wet weight) between tissues were determined through one-way analysis of variance (ANOVA). The liver presented high affinity for accumulation of Cd, Cu, and Zn due to the binding activity of metallothionein proteins. Mn and Pb mainly accumulated in the gill indicated the major uptake route through the gill. Lack of correlation between analyzed element and fish size might imply no or weak biomagnification of the analyzed elements in fish tissues. The potential health risk associated with exposure to heavy metals and As was evaluated. The uncertainty of target hazard quotient was calculated by Monte Carlo simulation. The results indicated that the analyzed element contents in the white bream muscle posed relatively low-chronic toxic risk to consumers.

Distribution, residue level, sources, and phase partition of antibiotics in surface sediments from the inland river: a case study of the Xiangjiang River, south-central China

Chen Leilei,Li Haipu,Liu Yang,Cui Yue,Li Yue,Yang Zhaoguang

In view of the increasing attention on antibiotic contamination and their scarce data in the inland river (especially for the sediment), the occurrence of 28 antibiotics in sediments from the Xiangjiang River was comprehensively analyzed, and 22 antibiotics were detected with a total concentration ranging from 4.07 to 2090 ng g −1 . The residue was almost at a moderate or higher level in the aquatic environment around the world. Fluoroquinolones and tetracyclines were the dominant detected antibiotics, and the maximum total concentration could reach to 2085 ng g −1 , though that in surface water was just 33.4 ng L −1 . Oxytetracycline and chlortetracycline could be detected with high concentration in areas with lower population density. Usage profile of each antibiotic may be responsible for the spatial variation. Principal component analysis-multiple linear regression model indicated that direct discharge of domestic wastewater and livestock or aquaculture sewage could contribute 94.2% of the pollution. Redundancy analysis was used to screen out the environment variables, which were closely related to the pseudo-partitioning coefficients (K d ) of antibiotics in sediment and surface water for the first time, and showed that the K d was correlated with sediment pH negatively and organic carbon, total phosphorus, and conductivity of the sediments positively. High sedimentary organic carbon was considered to promote the higher K d in this river. This study would deepen the understanding of the occurrence of antibiotics in sediments from the inland rivers and provide scientific support for controlling the antibiotic contamination.

Phenolic endocrine-disrupting compounds in the Xiangjiang River: occurrence and risk assessment

Yang Yu,Li Jing,Xu Jie

The levels of three phenolic endocrine-disrupting compounds (EDCs), NP, BPA and 4-t-OP were determined in water and sediment collected from sites along the Xiangjiang River, Zunyi, China. The NP, BPA and 4-t-OP concentrations ranged from 18.02 to 311.79 ng/L in the surface water, 16.04–408.12 ng/L in the submerged water, and 21.13–892.37 μg/kg dw in the sediment. NP contamination was most severe in both the river water and sediment. The ranges of the three phenolic EDCs were slightly greater in the submerged water than in the surface water ( p  > 0.05). The concentrations in the middle reaches were greater than those in the upstream and downstream reaches in both the water and sediment, and significant differences in content were detected in some reaches. The levels of three phenolic EDCs in the water and sediment had a positive correlation. In addition, the distribution coefficient (K d ) indicated that NP was more likely to adsorb to the sediment, and BPA and 4-t-OP were more likely to adsorb to river water. Moreover, the risk quotient (RQ) and hazard quotients (HQ) were used to reveal the environmental and health risks caused by coexposure to the three phenolic pollutants. The results showed that the current pollution is a threat to the environment of the study area and not a threat to the health of the local population.

Species-specific bioaccumulation and correlated health risk of arsenic compounds in freshwater fish from a typical mine-impacted river

Yuyu Jia,Lin Wang,Shan Li,Junfei Cao,Zhaoguang Yang

Arsenic (As) speciation and bioaccumulation in fish muscle tissues have been intensively investigated in marine ecosystem. However, little is known about these in freshwater fish. In this study, freshwater fish including 120 specimens and 8 species were collected from the Xiang River, a typical mine-impacted river in China. Six As species including arsenite (AsIII), arsenate (AsV), monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenocholine (AsC) and arsenobetaine (AsB) were simultaneously separated and determined using HPLC-ICP-MS. The mean (±   SD) concentration of total As (tAs) in the dried fish muscle was 0.748   ±   0.651   mg·kg −   1 . AsB was found as the predominant As species in most of the studied fish samples, in accordance with the reports in marine fish. However, the diversity of inorganic/organic As proportion observed in the studied freshwater fish species was larger than that in marine fish species due to greater spatial variability of As contamination, mobilization and origination in the studied catchments. The percentage of AsB (AsB%) in fish muscle was irrelevant to tAs concentration, while the percentage of iAs (iAs%) decreased with tAs concentration in a hyperbolic pattern. This can be attributed to restricted assimilation and accumulation of toxic iAs with increasing tAs concentration in fish. Chronic non-carcinogenic and carcinogenic health risks were evaluated through Monte-Carlo simulation. The result indicated that consuming freshwater fish in the Xiang River could cause considerable carcinogenic risk to local inhabitants.

Ionomic profile and arsenic speciation in Semisulcospira cancellata, a freshwater shellfish from a mine-impacted river in China

Du Fan,Wang Lin,Yang Zhaoguang,Liu Peng,Li Deliang

Freshwater aquatic environment close to cities and industrial areas is more sensitive than marine environment. The freshwater shellfish Semisulcospira cancellate was introduced as a bioindicator to monitor the heavy metal contamination in the river through ionomic profiles and arsenic speciation. The shellfish samples were collected near four cities along the Xiang River in China. The concentrations of elements including Mg, Al, Ca, Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ga, As, Sr, Cd, Sn, Sb, Ba, and Pb were determined using ICP-MS. Multivariate statistical analyses such as Pearson’s correlation analysis and principle component analysis (PCA) were employed to identify the possible sources of the elements in the shellfish samples. Three principle components were extracted from the ionomic matrix and were associated with natural existence, biological pathways, and mining and smelting activities, respectively. The ionomic profiles of the shellfish samples were evaluated through hierarchical cluster analysis (HCA) which was exhibited in the form of heatmap. The shellfish samples were categorized according to the sampling sites with different contamination levels. Six As species including arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenobetaine (AsB), and arsenocholine (AsC) were separated and quantified using HPLC-ICP-MS. The concentrations of As(III) and As(V) were linearly increased with total As concentration increasing. However, the proportion of AsB was decreased with total As while the AsB concentration was irrelevant to total As.

Impact of carbon nanotubes on the mobility of sulfonamide antibiotics in sediments in the Xiangjiang River

Chang Su,Guang-Ming Zeng,Ji-Lai Gong,Chun-Ping Yang,Jia Wan,Liang Hu,Shan-Shan Hua,Yan-Yan Guo

Manufactured in numerous factories, and contained in various consumer products, carbon nanotubes (CNTs) and sulfonamide antibiotics (SAs) may be released into the environment by many pathways. The sorption behavior of CNTs on SAs may increase the environmental and health risks when exposed to SAs–CNTs composites. In this study, we investigated the mobility of SAs in sediment columns in the presence/absence of CNTs. Three kinds of SAs (sulfamethoxazole, sulfapyridine, and sulfadiazine) and two kinds of CNTs (multi-walled carbon nanotubes, MWCNTs and single-walled carbon nanotubes, SWCNTs) in sediments from the Xiangjiang River were investigated in this study. The results showed that the SAs were of high mobility in sediment columns. However, CNTs with a concentration of 4.8 mg g−1 in the sediment could dramatically retain SAs, which might be due to the limited transport of CNTs and their high adsorption capacities of SAs. The percentage of SAs retention in sediment became higher when CNTs existed in the inflow, suggesting that a strong CNTs-associated SAs reaction might occur in the sediment. The findings in this study indicated that CNTs in the sediment environment or river system can reduce the mobility of SAs, which should be taken into account when evaluating the potential environmental risks of SAs and CNTs.

Effect of g-C3N4 on biodiversity and structure of bacterial community in sediment of Xiangjiang River under tetracycline pressure

Hu Xuemei,Peng Kuan,Chen Yijun,Chen Xiaoyong,Liu Shuguang,Zhao Yunlin,Wu Yaohui,Xu Zhenggang

Photocatalysts have been widely prepared and used in wastewater treatment. Although the influence of photocatalyst application on survival and activity of organisms has been examined, its impact on composition and diversity of microbial community is not fully understood. In this study, the impact of photocatalyst g-C 3 N 4 (Graphitic carbon nitride) on microbial communities in riverbed sediments polluted by antibiotic tetracycline (TC) was investigated. The sediment samples collected from the Xiangjiang River of China were exposed to different concentrations of TC, g-C 3 N 4 and TC/g-C 3 N 4 and the bacterial community were analyzed by Illumina sequencing. The results showed that the dominant bacterial phyla were Acidobacteriota, Proteobacteria, Actinobacteriota , and Chloroflexi in the study site. When compared to the control treatments, the application of TC, g-C 3 N 4 and TC/g-C 3 N 4 exhibited distinguishable effects on bacterial community structure in sediments. The presence of TC had greater influence on bacterial composition, while g-C 3 N 4 and TC/g-C 3 N 4 had less influence on bacteria. The diversity and richness of microorganisms in sediment increased under g-C 3 N 4 application and reached the highest values when g-C 3 N 4 was 75 mg/kg. The photocatalyst g-C 3 N 4 restored bacterial community diversity affected by TC, reduced the TC residues in aquatic environment, and eliminated the side effects of TC application in sediments. Our study indicated that g-C 3 N 4 was an environmentally friendly photocatalyst with lightly negative effects on microbial community in riverbed sediments, and could be used for effective remediation of TC-contaminated environments.

Quantitative determination of ametryn in river water using surface-enhanced Raman spectroscopy coupled with an advanced chemometric model

Yao Chen,Zeng-Ping Chen,Jing-Wen Jin,Ru-Qin Yu

In this contribution, surface-enhanced Raman spectroscopy (SERS) coupled with an advanced chemometric method-multiplicative effects model (MEM SERS ) has been applied to quantitative analysis of ametryn in water samples of the Xiangjiang River (Changsha, China). The adoption of MEM SERS calibration model was to eliminate the detrimental effects caused by variations in the physical properties of enhancing substrates, the intensity and alignment/focusing of laser excitation source. Experimental results showed that the combination of SERS with MEM SERS can provide quite precise concentration predictions for ametryn in water samples of the Xiangjiang River with an average relative prediction error of about 4.8%. The combination of SERS with MEM SERS can compete with LC-MS/MS in terms of precision and accuracy of quantitative results. The limit of quantification was about 0.09   μM. More importantly, no laborious reference methods (e.g., HPLC) were needed to build the MEM SERS calibration model, since the MEM SERS calibration model built on the calibration samples prepared with ultrapure water could provide satisfactory quantification results for the test samples prepared with water collected from the Xiangjiang River. Therefore, it is reasonable to expect that SERS in combination with MEM SERS model would become a competitive alternative in routine quantitative analysis of ametryn in environmental water samples.

Preparation of phosphopeptide-iron chelates from Mylopharyngodon piceus roe phosvitin using high voltage pulsed electric field pretreatment combined with enzymolysis

Wang Xiaoyuan,Wang Ashily Liang,Yang Chenyu,Qiu Weiqiang,Li Li,Jiang Yunzi,Zhao Wenbin,Li Feng,Jin Yinzhe

Peptide–iron chelates were prepared, and their chelating properties were explored. High voltage pulsed electric field (PEF) was used to assist in the extraction of phosvitin (Pv) from Mylopharyngodon piceus roe, and the Pv extract was subsequently treated and enzymolyzed to obtain phosphopeptide (PP). After PEF pretreatment and enzymatic hydrolysis, total antioxidant capacity and Fe 2 ⁺-chelating ability of PP increased by 54.45% and 12.06%, respectively. Peptide–iron chelates (PP 8 -Fe) were prepared using PP with the highest antioxidant activity. Chelate structure determination revealed that PP 8 -Fe was rich in amino acids with strong metal ion chelation capabilities, namely Glu, Asp, and Ser, which accounted for 50.91% of the total amino acids. Compared to PP 8 , PP 8 -Fe exhibited a more loosely structured surface at the microstructural level, increased iron content, a shift in secondary structure from random coil to α-helix and β-sheet, an increase in weighted average particle size from 420.20 ± 2.80 nm to 768.50 ± 1.90 nm, an increase in zeta potential from -33.70 ± 0.60 mV to -3.78 ± 0.46 mV, enhanced hydrophobicity, an increase in ΔH, and good gastrointestinal digestion stability. PP 8 has iron chelating sites, including phosphate, carboxyl, carbonyl, amino groups, and sialic acid residues. Our findings provide a theoretical foundation for peptide preparation and enhancement of their bioactivity via PEF pretreatment, while also providing guidance for future research on their chelation mechanisms with metal ions.
