Anti-eczema potential of three tea extracts: mechanisms of anti-inflammatory, antibacterial, antioxidant, and immunomodulatory effects

Zeting Huang,Lanyue Zhang,Jie Xuan,Duoling Xu,Jiyu Weng,Bing Yu,Weihua Peng

Background: Tea, with a long history in China, is known for its anti-inflammatory and antioxidant properties. Limited research exists on its use in eczema treatment. This study explores the effects and mechanisms of three tea extracts—Camellia sinensis var. assamica (Royle ex Hook.) (Theaceae) (CS), Camellia ptilophylla Hung T. Chang (Theaceae) (CP), Camellia arborescens Hung T. Chang, F. L. Yu & P. S. Wang (Theaceae) (CA). On eczema induced by 1-chloro-2,4-dinitrobenzene (DNCB) in mice and to explore the underlying anti-inflammatory and immune regulatory mechanisms. Methods: The metabolites of tea extracts were analyzed using ultra-high performance liquid chromatography/quadrupole Orbitrap mass spectrometry (UHPLC-Q-Orbitrap-MS). A DNCB-induced dermatitis model in mice was established, with histological staining and immunohistochemistry to assess eczema lesions and cytokine expression. In vitro tests were performed on RAW 264.7 cells and HaCaT cells to analyze the effects on inflammation, immune regulation, and cell migration. Results: All three tea extracts alleviated DNCB-induced epidermal thickening, reduced mast cell infiltration, and decreased TNF-α and IL-1β levels. The extracts suppressed nitric oxide (NO), reactive oxygen species (ROS), and NF-κB expression. Additionally, they downregulated immune-related factors such as IL-1, IL-6, IFN-γ, and TGF-β. Scanning electron microscopy revealed morphological changes in Escherichia coli and Staphylococcus aureus. Conclusion: CS, CP, and CA show potential for treating eczema, demonstrating anti-inflammatory and immune-regulatory effects. These tea extracts could serve as promising natural treatments for eczema and related inflammatory skin conditions.

Mongolian medicine Wenguanmu ointment treats eczema by inhibiting the CKLF-1/NF-κB pathway

Li Zhu,Xiao-jia Li,Chao Gu,Yuan Gao,Chun-sheng Zhang,Lu-yu Wang,Nai-hong Chen,Gang Li

Ethnopharmacological relevance The main clinical manifestations of eczema include itching, erythema, swelling and pain. Currently, allergies and TH1/TH2 cytokine imbalances are significant causes of eczema. TCM believes that eczema is mainly caused by incongruity between dry and wet. Wenguanmu ointment is a classic Mongolian medicine, which mainly composed of Xanthoceras sorbifolia Bunge , Coptis chinensis Franch and Bezoar. These ingredients can clear heat and dampness, dispel wind and dehumidification, anti-inflammatoryad analgesic. In this study, it was found that Wenguanmu ointment can treat eczema with anti-inflammatory, analgesic and antipruritic. Aim of the study In this study, the content of main components in Wenguanmu ointment was tested. Moreover, the therapeutic effect and mechanism of Wenguanmu ointment on eczema model mice were studied. Materials and methods Kunming mice (25 ± 2 g) were randomly divided into 6 groups: Control group; Model group; Vehicle group; Wenguanmu ointment group; Compound dexamethasone acetate cream group; Chushizhiyang ointment group. The eczema mouse model was established by DNCB. HPLC and TLC tests were used to determine the content of the main components in Wenguanmu ointment. HE staining was used to assess skin damage in mice. In order to detect the anti-inflammatory effect of Wenguanmu ointment on eczema, The levels of IgE, TNF-α, IFN-γ, COX-2 and IL-4 in serum was measured by ELISA. Genecards and Online Mendelian Inheritance in Man databases were used to analyze potential target gene predictions, and it was speculated that Wenguanmu ointment was associated with NF−κB signaling pathway and chemokine signaling pathway. To detect this inference, RT-qPCR and western blotting were used to detect protein and mRNA levels of CKLF-1, IκB-α, and NF-κB. Results Wenguanmu ointment can repress the symptoms of eczema caused by 2, 4-dinitrochlorobenzene, and inhibit the level of serum immunoglobulin E. Simultaneously it restrain the elevation of miscellaneous pro-inflammatory cytokines and chemokines, as well as reducing the expression of CKLF-1 and NF-κB protein in the nucleus, and increasing the protein expression of IκB to improve eczema. Conclusions The ameliorating effect of Wenguanmu ointment on eczema lesions can play a importment role by inhibiting the CKLF-1/NF-κB pathway.

Development of triptolide-nanoemulsion gels for percutaneous administration: physicochemical, transport, pharmacokinetic and pharmacodynamic characteristics

Yang Meng,Gu Yongwei,Yang Dishun,Tang Xiaomeng,Liu Jiyong

Background This work aimed to provide useful information on the use of nanoemulsions for the percutaneous administration of triptolide. Lipid nanosystems have great potential for transdermal drug delivery. Nanoemulsions and nanoemulsion gels were prepared to enhance percutaneous permeation. Microstructure and in vitro/in vivo percutaneous delivery characteristics of triptolide (TPL)-nanoemulsions and TPL-nanoemulsion gels were compared. The integrity of the nanoemulsions and nanoemulsion gels during transdermal delivery and its effects on the surface of skin were also investigated. The penetration mechanisms of nanoemulsions and nanoemulsion gels were investigated by differential scanning calorimetry (DSC) and Fourier transform infrared spectroscopy (FTIR). The transport characteristics of fluorescence-labelled nanoemulsions were probed using laser scanning confocal microscopy. A chronic dermatitis/eczema model in mice ears and the pharmacodynamic of the TPL-nanoemulsion gels were also investigated. Results Compared to TPL gels, significantly greater cumulative amounts of TPL-nanoemulsion gels and TPL-nanoemulsions penetrated rat skin in vitro. The in vivo microdialysis showed the concentration–time curve AUC0–t for TPL-NPs is bigger than the TPL-gels. At the same time, TPL-NPs had a larger effect on the surface of skin. By hydrating keratin and changing the structure of both the stratum corneum lipids and keratin, nanoemulsions and nanoemulsion gels influence skin to promote percutaneous drug penetration. Both hairfollicles and the stratum corneum are also important in this transdermal drug delivery system. Moderate and high dosages of the TPL-nanoemulsion gels can significantly improve the symptoms of dermatitis/eczema inflammation and edema erythematic in mice ears and can reduce the expression of IFN-γ and IL-4. Moreover, the TPL-nanoemulsion gels cause less gastrointestinal damage than that of the Tripterygium wilfordii oral tablet does. Conclusion sNanoemulsions could be suitable for transdermal stably releasing drugs and maintaining the effective drug concentration. The TPL-nanoemulsion gels provided higher percutaneous amounts than other carriers did. These findings suggest that nanoemulsion gels could be promising percutaneous carriers for TPL. The TPL-nanoemulsion gels have a significant treatment effect on dermatitis/eczema in the mice model and is expected to provide a new, low-toxicity and long-term preparation for the clinical treatment of dermatitis/eczema in transdermal drug delivery systems.

Thermal-grinding supramolecular network loaded with paeonol for treatment of skin eczema

Chen Chen,Xiao Yang,Xian-hui Shao,Yue Zhou,Xiao-bin Zhang,Yun-ping Lu,Meng-zhen Xing,Qing-chang Xia,Yuan-xiang Liu,Yu-ning Ma,Ting-ting Zheng,Lin-lin Zhang,Yu-xia Ma,Ji-guo Yang

Lipophilic drugs loaded into hydrogel vehicles for controlled release often encounters drawbacks such as low loading capacity, tedious inclusion methods and invalidity caused by syneresis. Facing this issue, we proposed a non-solvent drug loading system, thermal-grinding supramolecular network (TGSN), in which the monomer, crosslinker and drug molecules in solid state were grinded and then subjected to melting gelation. Paeonol, with a poor water solubility of 0.54 mg/mL, was chosen as a model drug. Both organosulfur-based polymeric network and paeonol aggregates constituted the main structure of TGSNs. Thus, the network had special properties such as stretchability, anti-swelling, elasticity, antibacterial activity, surface hydrophilicity, self-healing and adhesion. In vitro cellular toxicity assay demonstrated that TGSN improved L929 cell proliferation with minimal cytotoxicity. Viewed by transdermal release assay, TGSN could achieve sustained release of paeonol. Proved by skin eczema model in Kunming mice, TGSN possessed an excellent therapeutic effect and a low recurrence probability.
