Wet-Adhesive Hydrogel with Antiswelling Capacity for Strain Sensing

Xiaotao Zhu,Qiran Yu,Wufeng Su,Na Li,Xiao Miao,Yumin Wang,Lide Xu,Jingwei Lu,Guina Ren

The development of functional hydrogels with wet adhesion strength and antiswelling properties is crucial for applications ranging from biomedicine to wearable electronics but remains difficult to realize. Herein, we developed a high-performance TA/PAAc/EMA/C-CNT (TPEC) hydrogel via a solvent displacement process. The TPEC hydrogel showed no observable swelling behavior even after 8 days of immersion in a water bath, demonstrating its excellent antiswelling property. The TPEC hydrogel possesses strong adhesive strength in the underwater environment, allowing it to adhere firmly to rigid substrates and moving fingers, and also acts as a sealing material to prevent liquid leakage underwater. In addition, the TPEC hydrogel also demonstrated improved mechanical properties, good conductivity, and antimicrobial properties. Finally, exploiting its multifunctional properties, the resulting TPEC hydrogel is utilized as a flexible wearable strain sensor for monitoring body motion in both air and water environments. This work is expected to open up new opportunities for developing future generations of high-performance adhesive hydrogels for use in wet environments.

A Janus hydrogel sealant with instant wet adhesion and anti-swelling behavior for gastric perforation repair

Yuqing Liang,Huiru Xu,Qian Han,Meiguang Xu,Jiaodi Zhang,Jiaxin Wang,Xiaofei Liu,Zhanhai Yin,Baolin Guo

Achieving instant adhesion on wet tissues and preventing the concurrently occurred postoperative tissue adhesion remain tough challenges but urgently demanded. And the deteriorated cohesion and adhesion caused by undesired swelling of hydrogel could not be ignored. Herein, we developed a Janus hydrogel with instant wet adhesion and anti-swelling behavior, based on poly (acrylic acid) (PAA), gelatin (GT) and hyperbranched polymers modified with catechol (HBPC). The asymmetric structure is achieved through complexed one-side of the hydrogel with cationic polymers. The strong physical interaction between PAA and GT formed during immersing in water enhanced the mechanical strength of the gels. And the hydrogel exhibits good anti-swelling behavior, which circumvents the limitations of conventional hydrogel caused by swelling. Water drainage induced by hydrophobic component and the following removal of water residuals achieved by hydrophilic PAA synergistically enhanced the instant wet adhesion of the hydrogel. Besides, pH-responsive protonation of carboxyl groups in PAA equipped the adhesive with extreme acid-tolerance. The in vivo evaluation results demonstrated that the asymmetric adhesive hydrogel could promote the repair of gastric perforation while reducing the risk of postoperative tissue adhesion. The hydrogel with asymmetric wet adhesion might provide valuable insights into the establishment of bioadhesives for gastric sealing and repair.

Enhancing wet adhesion in biodegradable hydrogels via water-absorbent integration

Tan Jing Er,Liu Shengnan,Bai Jun,Yu Bingbing,Chen Kuntao,Sheng Xing,Yin Lan

Wet-adhesive hydrogels hold significant promise for biomedical applications, yet achieving long-term adhesion remains a major challenge in biodegradable systems. Here, we present a biodegradable hydrogel incorporating a water-absorbent layer to regulate swelling behavior and water content. Comprising a poly(ethylene glycol) diacrylate (PEGDA)–gelatin matrix with a starch-based absorbent layer, the hydrogel exhibits high wet adhesion energy, elevated burst pressure, effective in vivo hemostasis, and mechanical compatibility with soft tissue. This water-absorbent strategy provides a new avenue for achieving sustained and stable wet tissue adhesion in biodegradable hydrogel systems. Graphical abstract

Bioadhesive Microcarriers Encapsulated with IL-27 High Expressive MSC Extracellular Vesicles for Inflammatory Bowel Disease Treatment

Min Nie,Danqing Huang,Guopu Chen,Yuanjin Zhao,Lingyun Sun

Mesenchymal stem cell (MSC) therapy is a promising candidate for inflammatory bowel disease (IBD) treatment, while overcoming the limitations of naive seeding cells function and realizing efficient intestinal targeting remains a challenge. Here, a bioadhesive microparticle carrying interleukin-27 (IL-27) MSC-derived extracellular vesicles (MSC IL-27 EVs) is developed to treat IBD. The MSC IL-27 EVs prepared through lentivirus-mediated gene transfection technology show ideal anti-inflammatory and damage repair function. By encapsulating MSC IL-27 EVs into dopamine methacrylamide-modified hydrogel, a bioadhesive EVs microcarrier via microfluidic technology is fabricated. The resultant microcarriers exhibit ideal MSC IL-27 EVs sustained release effect and effective wet adhesion property. Furthermore, the therapeutic potential of MSC IL-27 EVs-loaded microcarriers in treating IBD is demonstrated. Through giving IBD rats a rectal administration, it is found that the microcarriers can firmly anchor to the surface of colon, reduce the inflammatory response, and repair the damaged barrier. Therefore, the bioadhesive MSC IL-27 EVs-loaded microcarriers provide a promising strategy for the biomedical application of MSC-derived EVs, and broaden the clinical potential of MSC therapy.

A catechol bioadhesive for rapid hemostasis and healing of traumatic internal organs and major arteries

Guoqing Wang,Xinyue Meng,Peiyan Wang,Xinping Wang,Gaoli Liu,Dong-An Wang,Changjiang Fan

Uncontrolled hemorrhage caused by trauma to internal organs or major arteries poses critical threats to lives. However, rapid hemostasis followed by tissue repair remains an intractable challenge in surgery owing to the lack of ideal internal-use adhesives that can achieve fast and robust wet adhesion and accelerate wound healing. Herein, we develop a robust hemostatic bioadhesive (CAGA) from novel highly-branched aminoethyl gelatin with end-grafted abundant catechol (Gel-AE-Ca). The unique chemical structure of Gel-AE-Ca makes CAGA capable of gelling on wet tissues via synergetic cross-linking of catechol-Fe 3+ chelation and horseradish peroxidase (HRP)/H 2 O 2 -triggered covalent bonds using a dual-channel needle, meeting the key demands of internal medical applications (e.g., instant and strong wet adhesion, injectability , biocompatibility, self-healing, stretching flexibility, infection resistance, and proper biodegradability). It exhibits rapid gelation within 10 s and robust wet tissue adhesion up to 115.0 ± 13.1 kPa of shear strength and 245.0 ± 33.8 mm Hg of sealing strength. In vivo trials demonstrate that CAGA can not only effectively seal anastomosis of the carotid artery, but achieve rapid hemostasis on the sites of liver incisions and penetrating cardiac wounds within 10 s. The wound closure by CAGA and its timely biodegradation promote wound healing of the vital organs.
