Switch-on fluorescent strategy based on N and S co-doped graphene quantum dots (N-S/GQDs) for monitoring pyrophosphate ions in synovial fluid of arthritis patients

Zongen Liu,Jiecheng Xiao,Xianwei Wu,Liqing Lin,Shaohuang Weng,Min Chen,Xiaohui Cai,Xinhua Lin

This work reports a facile and effective approach for preparing graphene quantum dots co-doped with nitrogen and sulfur (N-S/GQDs). The proposed method comprised pyrolysis of citric acid with glutathione and dialysis treatment. The fabricated N-S/GQDs exhibited blue luminescence and a high fluorescence quantum yield of 36.3%. N-S/GQDs can be sensitively quenched by Fe 3+ , which was absorbed on the edge of N-S/GQDs through the coordination interaction between Fe 3+ and N, S elements. The subsequent introduction of pyrophosphate ion (PPi) resulted in the recovery of the fluorescence of Fe 3+ -quenched N-S/GQDs. The stronger coordination reactivity between Fe 3+ and PPi induced changes in fluorescence. PPi concentration was facilely quantified through the recovered amplitudes of the fluorescence intensities of N-S/GQDs. Under the optimized experimental condition, increased fluorescence intensity showed a linear relationship to PPi concentration (1   μM to 1000   μM) in serum samples. Additionally, the proposed method featured sensitive and selectivity for the detection of PPi in synovial fluid, suggesting the potential application for simple and accurate clinical diagnosis of arthritic diseases.

Injectable lubricant-coated hyaluronic acid−dopamine for the repair of early osteoarthritis

Hang Yao,Chirun Wang,Qi Tao,Jian Yang,Dianwei Liu,Xianfeng Shang,Zhonglian Wu,Xu Hu,Zehao Gu,Zhilin Cheng,Jihang Dai,Dong-an Wang

Osteoarthritis (OA) is an irreversible disease of continuous degradation of cartilage, and natural joint super-lubrication is thought to provide hydrated lubrication to articular cartilage. Synovial joint fluid consists of hyaluronic acid (HA) and numerous brush-like macromolecules, exhibits a high affinity for cartilage proteins, and forms a hydrated layer on the cartilage surface. When cartilage friction increases, the surface layer of the cartilage becomes ruptured or damaged, and the cartilage hydration layer is destroyed, subsequently evolving into early OA. In this study, dopamine (DOPA) was coupled with HA to modify it by utilizing the hydrophilicity of HA and the adhesive properties of DOPA. On the one hand, it enhances cartilage lubrication by prolonging the retention time of HA in cartilage and restoring the hydrated layer on the cartilage surface by adhering to the type II collagen network. The successful synthesis of hyaluronic acid−DOPA (HAD) was confirmed by nuclear magnetic resonance (NMR) hydrogen spectroscopy, infrared spectroscopy, and ultraviolet (UV) absorption spectroscopy. The morphology and hydrophilicity of HAD were demonstrated using atomic force microscopy (AFM) and contact angle measurements. Additionally, the interaction of HAD with cartilage matrix proteins was verified through confocal microscopy imaging, while the biocompatibility and feasibility of injection of HAD were confirmed by cellular and animal experiments. On the other hand, the effect of protecting and even regenerating cartilage was achieved by remodeling the cartilage microenvironment. The cartilage protection and even regeneration ability of HAD was confirmed by injecting HAD at different times after OA modeling and characterizing the joints in sections.

Detection of synovial fluid LTF and S100A8 by chemiluminescence immunoassay for the diagnosis of periprosthetic joint infection

Ying Xu,Chenguang Hu,Jiayu liu,Haoran Guo,Jingkai Gao,Chi Wang,Chengbin Wang

Background and Aims Synovial fluid lactoferrin (LTF) and S100 calcium-binding protein A8 (S100A8) have been considered as potential biomarkers for the diagnosis of periprosthetic joint infection (PJI) through our previous research. However, the detection methods of these two proteins are still immature, so a rapid, accurate and cost-effective testing method is warranted. Materials and Methods We developed chemiluminescent immunoassays (CLIA) for the automated detection of synovial fluid LTF and S100A8 and assessed the analytical performance for these two methods. In addition, we recruited 86 patients who were suspected of PJI after total joint replacement (TJA) and examined their synovial fluid using CLIA to explore the clinical application value of these methods and the diagnostic efficiency of synovial fluid LTF and S100A8 for PJI. Results Our established CLIA methods have a wide linear range of 20–10,000 ng/mL for LTF detection system and 5–5000 ng/mL for S100A8 detection system. Performance parameters such as precision, specificity, and recovery rate can meet the industry standards. Then, the established methods were used to detect LTF and S100A8 in synovial fluid samples, which showed excellent diagnostic efficiency for PJI, and the areas under ROC curve (AUC) were 0.954 (95 % CI : 0.909–0.999) and 0.958 (95 % CI : 0.918–0.997), respectively. Conclusion Our established CLIA methods have the advantages of automation, high throughput, low price, and is expected to be widely popularized in clinical applications. Synovial fluid LTF and S100A8 detected through CLIA had efficient diagnostic potentiality for predicting and diagnosing PJI.

Synovial fluid-inspired biomimetic lubricating microspheres: Zwitterionic polyelectrolyte brushes-grafted microgels

Liu Guoqiang,Feng Yang,Gao Xiaohua,Chen Zhuo,Zhao Nan,Zhou Feng,Liu Weimin

Synovial fluid is made up of various biomacromolecules, including hyaluronic acid, aggrecans, lubricins, and phosphatidylcholine lipid, which are assembled onto the surface of articular cartilage in a gel state. Among them, brush-like biomacromolecules or assemblies have a vital effect on human joint lubrication. Inspired by this, the combination of brush-like molecular structures and gel-like assembly may be an efficient approach for the synthesis of biomimetic lubricating matters. Learning from the lubrication system of human joints, poly(2-methacryloyloxyethyl phosphorylcholine) (PMPC) brushes grafted poly(N-isopropylacrylamide-co-acrylic acid) (poly(NIPAAm-co-AA)) microgels, abbreviated as MBs-g-MGs, were synthesized as one kind of biomimetic lubricating additives. It is worth noting that this bionic strategy considered both molecular structure and assembled form, which enabled this hairy microgel to achieve low friction in aqueous medium. Meanwhile, the effective lubrication was still achieved when using MBs-g-MGs at high temperature, indicating that this microgel maintains a good lubricating effect over a wide range of temperature. In addition, this kind of microgel possessed good biocompatibility, which laid the foundation for potential biomedical applications. Looking beyond, these biomimetic microgels may provide an effective lubricating effect for water-based sliding interfaces, especially in biomedical systems.

Enzyme-activated apoptotic bodies-encapsulated NSET biomimetic probe for wash-free detection of intracellular pathogen in synovial fluid and monitoring therapy effect of septic arthritis

Yun Yang,Mingrong Wu,Yaqing Zhang,Lin Wang,Yang Song,Hong Xiao,Erqun Song

The survival of Staphylococcus aureus ( S. aureus ) within host cells leads to recurrences of septic arthritis after conventional treatment, which is a challenging clinical problem. Simple, rapid and sensitive detection of intracellular bacteria in septic arthritis synovial fluid and real-time monitoring of antibiotic efficacy for septic arthritis are important to the management of this disease. Herein, an enzyme-responsive activated nanoparticle surface energy transfer (NSET) biomimetic probe (FP 1 Au@AB) constructed with a fluorescent polypeptide (FP 1 )-gold nanoparticle (AuNPs, Au) complex (FP 1 Au) coated with apoptotic body (AB) was applied to detect intracellular S. aureus in septic arthritis synovial fluid from clinical samples and monitor the treatment of antibiotic of rifampicin (Rif) to septic arthritis in mouse models. The as-prepared NSET bionic probes of FP 1 Au@AB bears AB coating which makes the probes are of good actively targeting to macrophages, and the probes could be specifically tailored by caspase-1 presented in infected macrophages, resulting fluorescent signal on. Based on the FP 1 Au@AB probe, the detection of intracellular bacteria in synovial fluid with a low detection limit of 5 (the average number of S. aureus in one host cell) was realized without the need of washing extracellular bacteria beforehand. Additionally, therapy efficacy of septic arthritis in mouse model could be monitored timely based the proposed probe. Moreover, FP 1 Au@AB probes could be applied to confirm whether the synovial fluid from patients with septic arthritis contained intracellular bacteria or not, which is benefit for the doctor to do the administration. The FP 1 Au@AB probe-based lighting up analytical method shows great potential for the administration and evaluation of therapy of septic arthritis in clinic.

Cationic nanoparticle as an inhibitor of cell-free DNA-induced inflammation

Liang Huiyi,Peng Bo,Dong Cong,Liu Lixin,Mao Jiaji,Wei Song,Wang Xinlu,Xu Hanshi,Shen Jun,Mao Hai-Quan,Gao Xiaohu,Leong Kam W.,Chen Yongming

Cell-free DNA (cfDNA) released from damaged or dead cells can activate DNA sensors that exacerbate the pathogenesis of rheumatoid arthritis (RA). Here we show that ~40 nm cationic nanoparticles (cNP) can scavenge cfDNA derived from RA patients and inhibit the activation of primary synovial fluid monocytes and fibroblast-like synoviocytes. Using clinical scoring, micro-CT images, MRI, and histology, we show that intravenous injection of cNP into a CpG-induced mouse model or collagen-induced arthritis rat model can relieve RA symptoms including ankle and tissue swelling, and bone and cartilage damage. This culminates in the manifestation of partial mobility recovery of the treated rats in a rotational cage test. Mechanistic studies on intracellular trafficking and biodistribution of cNP, as well as measurement of cytokine expression in the joints and cfDNA levels in systemic circulation and inflamed joints also correlate with therapeutic outcomes. This work suggests a new direction of nanomedicine in treating inflammatory diseases.
