Improving laccase activity and stability by HKUST-1 with cofactor via one-pot encapsulation and its application for degradation of bisphenol A

Rongzheng Zhang,Lei Wang,Juan Han,Jiacong Wu,Chunmei Li,Liang Ni,Yun Wang

Enhancing the catalytic activity and stability of enzymes is of great importance in the development of green chemical and cost-effective application, with removal of bisphenol A (BPA) as a prominent example. Engineering immobilization carriers and immobilization methods of enzymes endows great potential to achieve above goal. Until now, these reports have focused on employing the metal-organic frameworks (MOFs) to increase the stability and reusability of enzymes, an enhancement in its catalytic activity has yet to be addressed. This work introduced a biomimetic mineralization process for facile synthesis of laccase@HKUST-1 biocomposite under mild condition. By exploiting the activity of laccase@HKUST-1, we demonstrated, for the first time, that the integration of laccase and HKUST-1 containing cofactor Cu 2+ ions leaded to 1.5-fold enhancement in the catalytic activity compared with free laccase, which was due to the synergistic enhancement of substrate oxidation. Indeed, the laccase@HKUST-1 biocomposite could function as active biocatalysts under biologically challenging conditions, such as acidic condition, high temperature, organic solvent, and continuous operation. The oxidation of phenols, such as BPA, with laccase@HKUST-1 reached higher catalytic performance than free laccase, and gave 100% degradation efficiency within 4 h. This study provides a feasible method to improve the activity and stability of laccase, which enable completely remove of BPA from the environment.

Construction of a bioinspired laccase-mimicking nanozyme for the degradation and detection of phenolic pollutants

Jinghui Wang,Renliang Huang,Wei Qi,Rongxin Su,Bernard P. Binks,Zhimin He

Nanozymes, defined as nanomaterials with enzyme-like activity, have attracted extensive interest in both fundamental and applied research. Laccases are members of the multi-copper oxidases, which are utilized as green catalysts in the environmental catalysis and biochemical industry. In this paper, we report a facile strategy for the preparation of a new class of nanozyme (denoted as CH-Cu) with laccase-like activity inspired by the structure of the active site and the electron transfer pathway of laccase via the coordination of Cu + /Cu 2+ with a cysteine (Cys)-histidine (His) dipeptide. The CH-Cu nanozymes exhibit excellent catalytic activity, recyclability and substrate universality and have a similar K m (Michaelis constant) and a higher v max (maximum rate) than laccase at the same mass concentration. They are robust under a variety of conditions, such as extreme pH, high temperature, long-term storage and high salinity, which can cause severe loss in the catalytic activity of laccase. Higher efficacy of the CH-Cu nanozymes compared with laccase in the degradation of chlorophenols and bisphenols is also demonstrated in a batch reaction. Furthermore, a method for the quantitative detection of epinephrine by a smart phone is established based on the CH-Cu nanozymes. We believe that this nanozyme has promising applications in environmental catalysis and rapid detection and expect that combining key peptides as metal ligands with metal ions to mimic the structure of the catalytic center of a natural enzyme will be a general and important strategy for the design and synthesis of a new type of nanozyme that can be used in various applications.

Pyrogallol-Stabilized Iridium Nanoclusters With Laccase-Like Activity for Dual Mode Detection of 2,4-Dichlorophenol

Lin Zhou,Xuan Chen,Zhanghong Guo,Haining Cui,Jinxin Ma,Qijun Song

Metal nanoclusters (MNCs) have attracted widespread attention due to their unique subnanometer size, tunable fluorescence emission, and excellent biocompatibility. However, the enzyme-like activity and the optical properties of MNCs are rarely exploited simultaneously. Here, iridium nanoclusters (Py-IrNCs) with excellent fluorescence properties and the laccase-like activity were synthesized by the reduction of IrCl 3 with NaBH 4 in the presence of pyrogallol. The laccase activity of Py-IrNCs exhibited in the catalytic transformation of 2,4-dichlorophenol (2, 4-DP) is substantially superior to that of natural laccase, which is manifested by the fact that the amount of Py-IrNCs was only 2.5% of natural laccase to achieve the same catalytic performance. In the coexisting of 4-aminoantipyrine (4-AP), a distinct color change and quench of fluorescence were observed; thus, a sensitive colorimetric and fluorescence dual-mode determination of 2,4-DP was achieved with a linear range of 0–40 μM and a limit of detection of 0.5 μM. The dual-mode detections can be mutually verified, which effectively improved the reliability of the analysis, demonstrating a promising application potential of Py-IrNCs in pollutant determination.

Boosting laccase activity by synergistic drive of near infrared laser, surfactant, and sodium chloride for healthcare coatings with UV resistance, anti-oxidation, and photothermal conversion

Guolin Zheng,Yifan Cui,Zhe Jiang,Man Zhou,Ping Wang,Yuanyuan Yu,Qiang Wang

Natural enzymes as green and efficient biocatalysts are essential in constructing multifunctional organic coatings for meeting the green chemistry processes. Herein, the laccase activity was for the first time enhanced by synergistic drive of near infrared (NIR) laser, dodecyldimethyl (3-sulfopropyl) ammonium hydroxide internal salt (SDDAB), and sodium chloride (NaCl). The SDDAB activation on laccase activity is unpredictable, which is advanced compared with conventional predictable ones, including adding mediator and improving reactive temperature. The NaCl can again boost the laccase activity by 29.8 % in the presence of SDDAB. Moreover, NIR laser irradiation again promotes laccase activity by 16.3 % in laccase/SDDAB/NaCl. The total specific activity of laccase exhibits an improvement of 29.2 %, indicating an advancement compared with the activities decrease in immobilized and encapsulated laccases. The Vm of laccase has been improved to 150.9 %. Besides, the hydrogen bonds and van der Waals force between laccase and SDDAB have been verified. Thus, laccase/SDDAB/NaCl was adopted to catalyze the polymerization of gallic acid to create eco-friendly UV-resistant, antioxidative, and photothermal coatings for healthcare. The ultraviolet protection factor (UPF), free radical scavenging ability, and photothermal temperature are 126.28, 95.0 %, and 94.2 °C, respectively. The multifunctional coating not only exhibits the response of sunlight-like photothermal conversion, but also possesses the photothermal response mediated by NIR laser. With the intensity increase of NIR laser irradiation, the surface temperature of modified cotton fabric is gradually strengthened from 29.5 to 43.8 °C. The surface characteristic and chemical composition of the multifunctional coatings were investigated by modern technology. The smartphone was adopted to quickly detect the red, green, and blue (RGB) values for demonstrating the successful fabricating of composite coatings , compared with traditional K/S characterization. Briefly, this work provided a novel, feasible, and elegant idea to boost laccase activity, inspiring more scholars to create advanced organic coatings with natural enzyme as the high-performance green catalysts for meeting green chemistry processes.

Ammonia-Etched Laccase-Mimicking Nanofiber-like Enzyme with Fluorescent Properties for the Detection of Oxytetracyclin

Luxi Huang,Huizhu Fan,Zhiying Jiang,Yin Dai,Jie Liu,Ling Dai,Junli Hong

A nanofiber-like enzyme, e-Cu-BN, was synthesized, exhibiting enhanced laccase-like activity compared to Cu-BN without ammonia etching. This study utilized the laccase-like activity and fluorescent properties of e-Cu-BN to develop a dual-mode fluorescence-colorimetric sensor for the detection of oxytetracycline (OTC). Specifically, e-Cu-BN catalyzes the oxidation of 2,4-dichlorophenol (2,4-DP), resulting in the formation of red quinone imine dye (QI) in conjunction with 4-aminophenol (4-AP), which exhibits strong UV absorption at 510 nm, while the OTC inhibits this reaction. Meanwhile, the fluorescence intensity of e-Cu-BN decreases as the concentration of OTC increases. Utilizing the two signal changes, we successfully achieved the detection of OTC. In addition, a smartphone-assisted hydrogel-based simple sensing platform was designed. The real-time detection of OTC was realized by the smartphone RGB value analysis software, and we added logic gates to simplify the detection process. This approach offers a rapid and cost-effective solution for OTC detection. This study not only developed a dual-mode sensor for OTC detection but also provided insights for further research into multifunctional nanozymes.

Construction of biomimetic nanozyme with high laccase- and catecholase-like activity for oxidation and detection of phenolic compounds

Jinghui Wang,Renliang Huang,Wei Qi,Rongxin Su,Zhimin He

Despite the rapid development of nanozyme , it is still challenging to develop multifunctional nanozyme with high oxidase-like activity. In this work, a novel type of amorphous imidazole-Cu nanozyme (I-Cu) with high laccase- and catecholase-like activity was prepared by the water induced precipitation of Cu 2+ and imidazole to mimick the N-Cu coordinated environment in their active site. I-Cu shows similar K m but 3.7-fold higher V max than laccase at the same mass concentration. The possible catalytic mechanism of I-Cu nanozyme, which is composed of substrate binding, substrate oxidation , oxygen binding and oxygen reduction, is proposed. In addition, it also displays high stability under various pH, temperature, long-term storage and high salinity . The oxidation efficiency of I-Cu nanozyme for environmental phenolic pollutants (2,4-dichlorophenol) is 91.8% at 10 h, which is higher than that of laccase (68.8% at 10 h). Furthermore, the colorimetric and smart phone detection of dopamine by I-Cu nanozyme is developed with the detection limit of 0.412 μM. Therefore, we expect this amorphous nanozyme is promising in the various application of bioremediation and biosensor .

Natural laccase guided synthesis of copper-doped carbon dots with enhanced laccase-like activity

Zhanghong Guo,Lin Zhou,Haining Cui,Jin-Xin Ma,Chan Wang,Qijun Song

Natural laccase, a member of the copper blue oxidase family, has its catalytic sites are classified as type1 (T1), type2 (T2) and type3 (T3) Cu. The electron transfer distance between the T1 to T2/T3 Cu has a great effect on the electron transfer efficiency in the laccase-catalyzed reaction. To mimic the active sites of laccase, a copper-doped carbon dots (CuCDs) was synthesized by a one-step hydrothermal treatment of copper sulfate and riboflavin. Experiment characterizations and theoretical simulations revealed that the catalytic sites in CuCDs consist of N, O coordinated copper and quinoxaline structure, which resemble the coordination environment of T2 Cu and the function of the T3 Cu center, respectively. The short electron transfer distance of 4.8 Å in catalytic sites of CuCDs accelerates the oxidation of the substrate dopamine (DA) to polydopamine (PDA) with a reaction rate being 40 times faster than that of laccase. Galvanic cell experiments proved that electron transfer from DA to oxygen was achieved in the presence of CuCDs. Therefore, this study not only could provide an efficient and eco-friendly nanozyme for hair dyeing in ambient condition, but also could inspire more and deeper researches in the field of mimicking natural enzyme with carbon nanomaterials.

Linxue Zhen,Zhichun Bi,Xinyue Zhang,Peize Jin,Donghui Song,Hui Huang,Yongxin Li

A nanozyme sensor array was developed combining with machine learning algorithms for the effective identification and differentiation of multiple antibiotic classes and antibiotic individuals. The four channel sensor array was composed by four copper-based nanozymes with laccase-like activity. The significant structural differences between different classes of antibiotics (such as Aminoglycosides, Tetracyclines, β-lactams, Nitroimidazole, Sulfonamides, Quinolones) resulted in the different effect on the nanozymatic reactions through interact with copper、substrate and so on. In addition, the differences between antibiotics in the same class also made certain effect on the nanozymatic reactions. Based on the above affect and differences, we combined the machine learning algorithms to successfully realize the identification of six major classes and sixteen individuals of antibiotics simultaneously for the first time. The proposed sensing strategy realized the facile identification of major categories of antibiotics, which has good application prospects in the rapid antibiotic identification in environmental water bodies.

2-Methylbenzimidazole-copper nanozyme with high laccase activity for colorimetric differentiation and detection of aminophenol isomers

Yingjiang Wang,Mingjing Li,Lingbo Qu,Lanlan Yu,Zhaohui Li

Laccase is well-known for its eco-friendly applications in environmental remediation and biotechnology, but its high cost and low stability have limited its practical use. Therefore, there is an urgent need to develop efficient laccase mimetics. In this study, a novel laccase-mimicking nanozyme (MBI-Cu) was successfully synthesized using 2-methylbenzimidazole (MBI) coordinated with Cu 2+ by mimicking the copper active site and electron transfer pathway of natural laccase. MBI-Cu nanozyme exhibited excellent catalytic activity and higher stability than laccase, and was utilized to oxidize a series of phenolic compounds. Environmental pollutant aminophenol isomers were found to display different color in solution when catalytically oxidized by MBI-Cu, which provided a simple and feasible method to identify them by the naked eye. Based on the distinct absorption spectra of the oxidized aminophenol isomers, a colorimetric method for quantitatively detecting o-AP, m-AP, and p-AP was established, with detection limits of 0.06 μM, 0.27 μM, and 0.18 μM, respectively. Furthermore, by integrating MBI-Cu-based cotton pad colorimetric strips with smartphone and utilizing color recognition software to identify and analyze the RGB values of the images, a portable colorimetric sensing platform was designed for rapid detection of aminophenol isomers without the need for any analytical instrument. This work provides an effective reference for the design of laccase nanozymes and holds significant potential for applications in the field of environmental pollutant monitoring.

Laccase activity regulation of platinum-based nanozymes by support engineering strategy and application in heavy metal ions sensing

Hongyi Yin,Yixing Tian,Zheng Liang,Cuiqiong Cao,Fu Wang,Ze Qiao,Dali Wei,Nuanfei Zhu,Zhen Zhang

Platinum nanoparticles (PtNPs) with laccase-mimicking activity have attracted tremendous research interest due to their high efficiency, controllable synthesis, and good stability. However, its application is still limited owing to the self-agglomeration of PtNPs, which easily leads to a reduction in colloidal stability and catalytic activity. Herein, a highly stable PtNPs-based hybrid nanozyme with excellent laccase-like activity was prepared, characterized by its growth on 3-aminophenol formaldehyde resin (APF) microspheres (APF@PtNPs). Benefiting from the large specific surface area and rich functional groups of APF, the catalytically active sites of PtNPs were not only fully exposed but also maximized in number and accessibility, enhancing its overall laccase-mimicking catalytic efficiency and effectiveness. The APF@PtNPs were found to keep laccase-like activity by catalyzing the oxygen to produce O 2 •− and exhibited a higher affinity to laccase-related substrates (K m  = 0.186 mM) than that of PtNPs. Meanwhile, it was observed that APF@PtNPs displayed greater stability at a wide range of temperature and pH levels. More significantly, the introduction of Hg 2+ and Ag + was identified to inhibit the laccase-like activity of APF@PtNPs. Based on this feature, a colorimetric detection method for the dual identification of metal ions was established, along with high sensitivity, selectivity, and satisfactory analytical performance. Also, a hydrogel sensing platform using APF@PtNPs was integrated with a smartphone to allow on-site and visual detection of metal ions. This work provides an effective strategy for reasonable regulation in the laccase-mimicking activity of PtNP hybrid nanozymes, broadening the application prospect of platinum-based nanozymes.

Chiral FeNC single-atom nanozymes with multi-enzyme activity for dye degradation

Guobao Ning,Huan Liang,Linjiang Guo,Xiaokang Lu,Lin Xiao,Tianle Qi,Hui Zhao,Can-Peng Li

The application of single-atom nanozymes in the removal of organic dye pollution by advanced oxidation process (AOPs) have been widely concerned. However, the application of single-atom nanozymes in AOPs is rarely reported. In this work, a kind of Fe-and nitrogen-codoped carbon (Fe-N-C) with peroxidase-like and laccase-like activities was prepared by calcining Fe-doped MOF, then the chiral peptides were introduced into single-atom nanozymes, and chiral single-atom nanozymes (L-Au@FeNC) were successfully prepared. The L-Au@FeNC can effectively activate PMS to degrade different kinds of organic dyes and also shows high cycle stability. In addition, the results of quenching experiment and electron paramagnetic resonance (EPR) test show that 1 O 2 produced during PMS activation plays an important role in the degradation of organic dyes, and O 2 ·- , ·OH, and SO 4 ·- are also produced during PMS activation, suggesting that the degradation of selected organic dyes include radical and non-radical pathways. It should be noted that L-Au@FeNC showed higher activity than D-Au@FeNC in enzyme-like activity (laccase-like and peroxidase-like) experiment and organic dye degradation, the results of molecular docking simulation indicated that it may be due to the better binding force of L peptide to the substrate of enzyme-like reaction and organic dye than D peptide. This chiral single-atom nanozymes provide a new idea for the application of organic dye degradation and enzyme-like catalysis design.

Cellulose / waste Cu2+-activated carbon composite: A sustainable and green material for boosting laccase activity and degradation of bisphenol A in wastewater

Na Li,Fangrui Yu,Hongbin Li,Xiangrui Meng,Chuanbo Peng,Xueru Sheng,Jian Zhang,Shanshan Liu,Qingwei Ping,Huining Xiao

Low enzyme activity is one of the disadvantages of immobilized laccase. In this study, waste Cu 2+ -loaded activated carbon (Cu-AC) was successfully used in preparing a novel composite support, cellulose / Cu 2+ -loaded activated carbon beads (C / Cu-AC), and effectively boosted immobilized laccase activity. To achieve optimum conditions for immobilization of laccase, the immobilization time, pH and laccase concentration were examined. The highest immobilized laccase activity (34.21 U/g) was achieved under optimum conditions ( T  = 4 h, pH = 4, C = 5 g/L), which was increased by 35.86 % compared to control. In addition, the immobilized laccase showed an outstanding performance in thermostability and reusability compared to free laccase. Moreover, the degradation of BPA by immobilized laccase was carried out, and the optimum degradation conditions were explored. Under such conditions: concentration of BPA was 75 mg / L and pH = 4, t  = 1 h, T  = 50 °C,the removal yield of BPA reached a maximum of 79.88 %. Therefore, the utilization of waste Cu-CA is a powerful method to boost immobilized laccase activity and creating a new way to high value treatment of waste Cu-CA.
