Active compound identification by screening 33 essential oil monomers against Botryosphaeria dothidea from postharvest kiwifruit and its potential action mode

Jie Li,Su Fu,Gang Fan,Dongmei Li,Shuzhen Yang,Litao Peng,Siyi Pan

The antifungal activity of postharvest kiwifruit against the pathogen Botryosphaeria dothidea was evaluated for 33 essential oil monomers . The possible mechanism for the known active compounds were further assessed in this study. The results show all the EO components exhibit inhibitory effects on the pathogen to different degrees except for Farnesol . Carbon chain length and C 2 -C 3 double bonds had a great effect on the antifungal activities of aldehydes. Of all of these, carvacrol had the strongest antifungal activity with EC 50 of 12.58 μL/L and EC 90 of 22.08 μL/L. Carvacrol also exhibits significant inhibitory effects on the pathogen, both in vivo and in vitro . Carvacrol evidently alters the hyphal morphology of B. dothidea and severely damages cell membrane and inhibits the formation of lipid components on the membrane. As cell membrane permeability increases, intracellular homeostasis including ion and biomacromolecules were destroyed by carvacrol. Furthermore, carvacrol appears to significantly inhibit mitochondrial activity and respiration rates, resulting in cell death of B. dothidea . Our results provide evidence that carvacrol could be a very useful compound for controlling postharvest rot soft in kiwifruit .

Development of a LAMP Method for Detecting SDHI Fungicide Resistance in Botrytis cinerea

F. Fan,W. X. Yin,G. Q. Li,Y. Lin,C. X. Luo

Resistance to succinate dehydrogenase inhibitors (SDHI) in Botrytis cinerea is associated with point mutations in the target gene succinate dehydrogenase subunit B ( SdhB ). The substitution from histidine to arginine at codon 272 (H272R) is currently the predominant mutation in SDHI-resistant populations in B. cinerea worldwide. In order to monitor the development of resistance to SDHI, a rapid, simple, and efficient method with high specificity to the H272R point mutation was developed based on loop-mediated isothermal amplification (LAMP). To specifically detect the H272R mutation, a set of four primers was designed based on the sequence of SdhB , and the LAMP reaction was optimized. When SYBR Green I was added after reaction, only samples with the H272R mutation showed the color change (from brown to fluorescent yellow), indicating that this set of primers could successfully discriminate the H272R genotype from other genotypes. Specificity and accuracy tests showed that this LAMP assay had high specificity and accuracy. Moreover, the LAMP method was further simplified with fungal mycelia and conidia as the amplification template which could be prepared within 5 min. Due to the low cost, simplicity, high efficiency, and specificity, the developed LAMP assay may contribute to the monitoring of resistance development to SDHI in B. cinerea , especially in field and high-throughput experiments.

Inhibitory and Curative Effects and Mode of Action of Hydroxychloroquine on Botrytis cinerea of Tomato

Chengyan Xia,Xiansu Wang,Miss Zeqi Qi,Miss Fenghua Liu,Miss Dongxue Li,Miss Xiaolin Zhang,Libo Zhang,Delu Wang,Zhuo Chen

Gray mold is an important disease of crops and is widespread, harmful, difficult to control, and prone to developing fungicide resistance. Screening new fungicides is an important step in controlling this disease. Hydroxychloroquine is an anti-inflammatory and anti-malarial agent, which has shown marked inhibitory activity against many fungi in medicine. This study evaluated the inhibitory activity of hydroxychloroquine against several phytopathogenic fungi, finding a half-maximal effective concentration of 113.82 μg/ml against the hyphal growth of Botrytis cinerea , with significant in-vivo curative effects of 92.37% or 78.37% for gray mold on detached tomato leaves or fruits at 10.0 or 200.0 mg/ml, respectively. Ultrastructural studies indicated that hydroxychloroquine induced collapse of hyphae, with a wrinkled surface, unclear organelle boundaries, and organelle disintegration. Transcriptomic assays revealed that hydroxychloroquine could affect the expression of metabolism-related genes. Molecular docking and molecular dynamics analyses indicated that hydroxychloroquine bound to glucose-methanol-choline oxidoreductase, with low free energy value of −11.4 kcal/mol. Cell membrane permeability assays and hyphal staining confirmed that hydroxychloroquine damaged the cell membrane, causing leakage of hyphal contents and disturbing cell function. Biochemical assays indicated that hydroxychloroquine reduced the concentration of soluble proteins and reducing sugars in the hyphae. In total, hydroxychloroquine disturbed amino acid metabolism, therefore inhibiting the production of biomacromolecules, damaging the cell membrane, and restraining the growth of hyphae, and hence inhibiting gray mold on tomato. This study will explore the use of medicine in the development of agricultural fungicides and their application in managing crop diseases, providing valuable background information.

Improved antifungal activity and reduced aquatic toxicity of fludioxonil by complexation with β-cyclodextrin

Qianqian Geng,Pingyang Sun,Gang Tang,Pingliang Li,Yuxiu Zhai

Poor water solubility results in limited fungicidal bioavailability and poses a great risk to non-target organisms due to the use of organic solvent in formulation processing. Here, the water solubility of fludioxonil (Flu) was improved by generating an inclusion complex with β-cyclodextrin (β-CD) at 1:1 host-guest stoichiometric ratio. The formation of the inclusion complex was confirmed by SEM, XRD, FT-IR, DSC, and 1 H-NMR spectra. Molecular docking simulation found that Flu was imbedded into the β-CD cavity, folded, and centrally aligned inside. In addition, Flu/β-CD displayed increased soil mobility, sustained release behavior, and a 2.75-fold increase of the fungicidal activity against Botrytis cinerea . Importantly, the prepared inclusion complex exhibited decreased acute toxicity to phytoplankton ( Chlorella vulgaris ), zooplankton ( Daphnia magna ), and fish ( Danio rerio ) than technical Flu. This study provided useful information for the rational use of Flu in agriculture, as well as the reduction of ecological risk to the aquatic environment.
