AimsThe essential oil of Melaleuca alternifolia (tea tree) has been evaluated as a potential eco‐friendly antifungal agent against Botrytis cinerea. In this study, we investigated the antifungal activity and mode of action of tea tree oil (TTO) and its components against B. cinerea. Methods and ResultsOf the components we tested in contact phase, terpinen‐4‐ol had the highest antifungal activity, followed by TTO, α‐terpineol, terpinolene, then 1,8‐cineole. As one of characteristic components of TTO, terpinen‐4‐ol treatment led to pronounced alterations in mycelial morphology, cellular ultrastructure, membrane permeability under scanning electron microscope, transmission electron microscope and fluorescent microscope, and also reduced the ergosterol content of fungi. As another characteristic component, 1,8‐cineole caused serious intracellular damage but only slightly affected B. cinerea otherwise. When terpinen‐4‐ol and 1,8‐cineole were used together, the synergistic antifungal activity was significantly higher than either component by itself.ConclusionsThe results of our study confirmed that terpinen‐4‐ol and 1,8‐cineole act mainly on the cell membranes and organelles of B. cinerea, respectively, and when combined are similar to TTO in antifungal activity due to their differences.Significance and Impact of the StudyUnderstanding the mechanism of terpinen‐4‐ol and 1,8‐cineole antifungal action to B. cinerea is helpful for investigation on their synergistic effect and explaining antifungal action modes of TTO.

Benzimidazole fungicides (MBCs) have been widely used in agriculture since 1970s, and resistance to this class of fungicides in Botrytis cinerea is reported worldwide. Resistance to MBCs in B. cinerea is related to mutations in the target β-tubulin gene ( TUB2 ). Compared with the mutation at codon 200, the substitutions from glutamic acid to alanine (E198A), valine (E198V), or lysine (E198K) at codon 198 are currently the predominant mutations in MBC resistant populations of B. cinerea . In this study, a loop-mediated isothermal amplification (LAMP) method was established for rapid detection of benzimidazole resistance in B. cinerea. On the basis of the three mutations at TUB2 codon 198, three sets of LAMP primers were designed, and each of these primer sets was able to specifically amplify the DNA containing its corresponding mutation, while no amplification was detected with other mutated or the wild type DNA. After optimization, the sensitivity and specificity tests illustrated that this LAMP assay had good sensitivity and specificity to detect specific resistant genotypes in B. cinerea . Result also showed that the LAMP assay had good reproducibility. Above all, boiled mycelia could be used as templates, which simplified the process and increased the efficiency of the assay. Considering its rapidity, simplicity, and high efficiency, the LAMP assay developed in this study is a promising tool for the diagnosis of benzimidazole resistance in B. cinerea , and will contribute to the monitoring of resistance development to MBCs in the future.

In this work, a multifunctional food packaging composite coating with transparent, biodegradable, antifogging and antibacterial properties was designed and fabricated by quaternary ammonium salt modified chitosan (HACC) and poly (vinyl alcohol) (PVA) via a facile and environment-friendly solution casting method. A simple quaternization modification enabled the coating simultaneously to achieve excellent antifogging and antibacterial functions. The excellent antifogging property of the HACC/PVA composite coating was attributed to the strong water absorbency of quaternary ammonium chitosan and PVA. A nearly 98% transmittance ratio of coated glasses was achieved during antifogging test. In addition, the inhibition rate of the HACC/PVA composite coating kill against Escherichia coli ( E. coli ), Staphylococcus aureus ( S. aureus ) and Botrytis cinerea were up to ~99%. The antibacterial effect was demonstrated by each group of strawberries after storage for 1, 3, 5 days. The multifunctional coating has broad prospects in the application of fruit and vegetable packaging.

The development of safe, environmentally friendly, edible antimicrobial packaging films represents a promising alternative to conventional plastic packaging for reducing spoilage and extending the shelf life of fresh food. Here, we propose a novel strategy to construct edible β-CD-MOF/carvacrol@zein (BCCZ) composite films by intertwining β-CD-MOF loaded with the antimicrobial essential oil carvacrol, and zein. The resulting BCCZ films exhibit high humidity-triggered, long-lasting bactericidal efficacy, effective fruit preservation, and excellent biosafety. Characterization revealed that BCCZ films possess a compact texture, hydrophilic surface, low water vapor permeability, and high humidity sensitivity. Additionally, this film act as a “storehouse” for carvacrol, enabling humidity-controlled release (96.3% release at 100% RH and only 12.0% at 43% RH on day 7). BCCZ film effectively inhibited Gram-positive Staphylococcus aureus , Gram-negative Escherichia coli , and fungi ( Botrytis cinerea ) through the release of liquid-phase and vapor-phase carvacrols under both direct contact and indirect contact conditions in high humidity. Owing to the film's humidity-triggered, long-term release of carvacrol, strawberries packed in BCCZ films maintained their freshness and appearance significantly better after seven days of storage at 21 °C. This work provides valuable insights and holds promise for the design and commercial application of safe, environmentally friendly freshness-preserving packaging films.

To extend the shelf life of sweet cherries ( Prunus avium L . ) and considering the environmental problems caused by traditional packaging materials, novel Zein/Gelatin-proanthocyanidins-zinc oxide nanoparticles (ZE/GE-PC-ZnO) and Zein/Gelatin-gallic acid-zinc oxide nanoparticles (ZE/GE-GA-ZnO) protein-based composite nanofiber films were prepared by electrospinning. According to the results, ZE/GE-PC-ZnO and ZE/GE-GA-ZnO films' contact angles were higher than those of Zein/Gelatin film by 28.91% and 21.27%, and their antioxidant activities were 5 and 9 times higher, respectively. Moreover, ZE/GE-PC-ZnO film showed good inhibitory activity against B. cinerea . On the eleventh day of the cherry packaging test, compared to unwrapped cherries, the losses of weight and firmness of wrapped fruit were reduced by more than 20% and 60%, respectively. Respiration time was delayed by 5 days, and the peak of ethylene release was decreased by nearly half. In conclusion, these two nanofiber films were viable packaging materials that fulfilled global strategies for green development.

BACKGROUND Succinate dehydrogenase inhibitor (SDHI) fungicides are an important class of agricultural fungicides with the advantages of high efficiency and a broad bactericidal spectrum. To pursue novel SDHIs, a series of N -substituted dithiin tetracarboximide derivatives were designed, synthesized, and characterized by 1 H NMR, 13 C NMR, and high resolution mass spectrum (HRMS). RESULTS These engineered compounds displayed potent fungicidal activity against phytopathogens, including Sclerotinia sclerotiorum , Botrytis cinerea , and Rhizoctonia solani , comparable with that of the commercial SDHI fungicide boscalid . In particular, compound 18 stood out with prominent activity against S. sclerotiorum with a half-maximal effective concentration (EC 50 ) value of 1.37 μg ml −1 . Compound 1 exhibited the most potent antifungal activity against B. cinerea with EC 50 values of 5.02 μg ml −1 . As for R. solani , 12 and 13 exhibited remarkably inhibitory activity with EC 50 values of 4.26 and 5.76 μg ml −1 , respectively. In the succinate dehydrogenase (SDH) inhibition assay, 13 presented significant inhibitory activity with a half-maximal inhibitory concentration (IC 50 ) value of 15.3 μ m , which was approximately equivalent to that of boscalid (14.2 μ m ). Furthermore, molecular docking studies revealed that 13 could anchor in the binding site of SDH. CONCLUSION Taken together, results suggested that the dithiin tetracarboximide scaffold possessed a huge potential to be developed as novel fungicides and SDHIs. © 2023 Society of Chemical Industry.

Spoilage of industrial fruits and vegetables in storage and transportation due to fungal infection results in significant losses, and new natural antifungal treatments would have a large economic value. The extracts of antifungal compounds and phenolic acids from Jerusalem artichoke ( Helianthus tuberosus L.) leaves were investigated for potential use in enhancing preservation of fruits and vegetables in storage. Either crude leaf extract or n -butanol fraction was active against Botrytis cinerea, Colletotrichum gloeosporioides, Phytophthora capsici Leonian and Rhizoctonia cerealis , with the values of IC 50 (half of the maximum inhibitory concentration) ranging from 2.166 to 2.534 g L −1 for the crude leaf extract and 0.232–1.911 g L −1 for n -butanol fraction. The severity of grey mould caused by B. cinerea was significantly reduced by n -butanol fraction applied at 1 and 2 g L −1 (the control efficiency of 71.3% and 77.8%, respectively, compared with commercial preparation Carbendazim . Following in vitro activity-guided fractionation by bioautography, six phenolic acids were separated from n -butanol fraction. Among them, caffeic acid, 3,4-dicaffeoylquinic acid and 1,5-dicaffeoylquinic acid played a dominant role and were active in bioassays against Gibberella zeae , with respective minimum inhibitory concentrations (MIC) being 108, 60 and 4.2 μg mL −1 . These results imply that Jerusalem artichoke leaves might be a potential source of natural fungicides.
