Gray mold is a destructive disease caused by Botrytis cinerea , a pervasive plant pathogen, which poses a threat to both tomato growth and postharvest storage. The utilization of induced resistance presents a potential strategy for combating plant pathogenic attacks. ZNC (zhinengcong), an extract derived from the endophytic fungus Paecilomyces variotii , has been discovered to play a vital role in preventing diverse forms of bacterial infections. Nevertheless, the precise mechanism behind its ability to enhance tomato resistance to fungi remains unclear. In this study, we found that the exogenous spraying of ZNC could significantly improve the resistance of tomato plants to B. cinerea . The results of both the metabolomic analysis and high-performance liquid chromatography (HPLC) demonstrated that tomato plants responded to ZNC treatment by accumulating high levels of rutin. Additional transcriptome analysis uncovered that rutin enhances tomato resistance possible by initiating the generation of reactive oxygen species (ROS) and phosphorylation of mitogen-activated protein kinases (MPKs) related genes expression during the initial phase of invasion by B. cinerea . In addition, we also found that rutin might activate plant immunity by eliciting ethylene (ET) and jasmonic acid (JA)-mediated pathways. Therefore, plant immune inducer ZNC and rutin has bright application prospects and high utilization value to control gray mold. Graphical Abstract

In this study, using a facile one-pot approach, we constructed a series of multifunctional and tunable pesticide formulation based on NKMOF-101-M (M = Mg, Mn, Zn, Co, or Ni) for azoxystrobin (AZOX), aiming to efficiently control gray mold disease. The physicochemical properties of AZOX@NKMOF-101-M, including particle size, zeta potential, release profile, and drug adhesion, were systematically characterized. The pesticide loading capacity and encapsulation efficiency reached up to 0.14 mg/mg and 90 %, respectively. Release studies demonstrated a pH-responsive behavior, attributed to the presence of weakly acidic groups in NKMOF-101-M. In vitro antifungal activity assays revealed that the efficacy of AZOX@NKMOF-101-M (M = Mg, Mn, Zn, Ni) was comparable to that of free AZOX. Notably, AZOX@NKMOF-101-Co exhibited significantly enhanced antifungal activity, which might be attributed to the synergistic interaction between AZOX and cobalt ions. In pot trials, AZOX@NKMOF-101-Co demonstrated approximately 80 % efficacy in controlling Botrytis cinerea infection. Fluorescence tracking of NKMOF-101-Co and biosafety experiments showed that NKMOF-101-Co could be absorbed by plant leaves and transported to various parts of tomato seedlings and that AZOX@NKMOF-101-Co was relatively safe for silkworms, zebrafish, and 293 T cells. Furthermore, these formulations could promoted seed germination and tomato growth, thereby functioning as effective fertilizers to enhance crop yield. This work highlights the potential of NKMOF-101-M as a multifunctional platform for the development of novel controlled-release pesticide formulations, offering a promising approach for sustainable agricultural practices.

Botrytis cinerea is a phytopathogenic fungus that utilizes multiple infection pathways and is known for causing gray mold on crops. This fungal infection results in the deterioration of crop quality and substantial economic losses. ZH-5 , a phenolic acid compound derived from the methanol extract of the twigs of Zanthoxylum armatum DC., exhibited significant inhibitory effects on the mycelial growth, spore germination, and germ tube elongation of B. cinerea in in vivo experiments. The EC 50 of ZH-5 was determined to be 58.96 mg L −1 , which surpassed the effectiveness of commonly used carbendazim and was comparable to the plant-derived pesticide eugenol. Our study focused on investigating the mechanisms underlying the antifungal activity of ZH-5 . Our findings have demonstrated that ZH-5 has the ability to disrupt the integrity of cellular structures, including mitochondria. This disruption affects energy metabolism and induces oxidative stress, ultimately ZH-5 might be interfere with cellular activity or inhibit growth. In summary, we reported that ZH-5 demonstrated efficacy as a plant-derived compound against B. cinerea , making it a potentially valuable environmentally-friendly chemical agent for the management of gray mold.

Plant fungal diseases can lead to yield reduction and quality degradation in crops, which usually cause serious economic losses. Additionally, chemical fungicides used in the prevention and control of plant diseases are increasingly restricted due to resistance development and high toxicity. Therefore, biogenic fungicides such as chitosan with low toxicity and good biocompatibility are receiving increasing attention. This study found that the acid swelling chitosan pretreatment method can accelerate the rate of the specific oxidation of chitosan catalyzed by the TEMPO-NaBr-NaOCl system. This study proved that OCTS induces plant disease resistance, and the control efficiencies achieved in protection and treatment experiments against Botrytis cinerea were 80.6 % and 83.4 %, respectively, at 400 μg/mL OCTS. In addition, OCTS can promote plant growth and enhance plant defense enzyme activities. This research has realized a forward-looking exploration of the application of OCTS in the agricultural field.

Cuticular wax has been reported to play an essential role in resisting pathogens in various fruits. This study investigated the antifungal ability of the components in blueberry cuticular wax. We showed that the cuticular wax of blueberry inhibited the growth of Botrytis cinerea and ursolic acid (UA) was the key antifungal compound. UA inhibited B. cinerea growth in vitro and in vivo . Furthermore, UA increased extracellular conductivity and cellular leakage in B. cinerea , deformed the mycelial morphology, and destroyed cell ultrastructure. We also demonstrated that UA stimulated the accumulation of reactive oxygen species (ROS) and inactivated ROS scavenging enzymes. These results indicate that UA may exert antifungal effects against B. cinerea by disrupting cell membrane integrity. Thus, UA has significant potential as an agent for the control of gray mold in blueberry.

Tomato gray mold, caused by Botrytis cinerea , is one of the most devastating diseases affecting tomato plants. In this study, Pantoea jilinensis D25 fermentation broth (PJFB) was used to inhibit B. cinerea growth. The optimum fermentation time was 7 days. PJFB antifungal activity decreased when PJFB was heated to ≥60°C, whereas it was the strongest at pH 8. Moreover, ultraviolet treatment for a long time reduced its antifungal effect. PJFB, applied at 10 % (v/v), could inhibit B. cinerea growth by inhibiting its mycelial diameter, conidial production, and mycelial dry weight with the inhibition rates of 57.3 %, 51.2 %, and 78.6 %, respectively. PJFB could also increase conductivity and malondialdehyde (MDA) content in B. cinerea . Furthermore, PJFB inhibited B. cinerea on not only the leaf tissues but also the seedlings of tomato. The secondary metabolites in PJFB were determined through gas chromatography–mass spectrometry. Through laboratory toxicity analysis, we found that the major compounds in PJFB were 1,2-benzenedicarboxylic acid bis(2-methylpropyl) ester (14.3098 %), 2,4-di- tert -butylphenol (3.8036 %), 2,6,10,14-tetramethyl pentadecane (2.9947 %), and 1,3-dimethyl naphthalene (1.8586 %).

Gray mold , caused by Botrytis cinerea is an intractable fungal disease that causes extensive damage to agricultural products. In the search for novel antifungal active ingredients, we discovered a linear pyranocoumarin Pd-D-V was effective against B. cinerea in both in vitro and in vivo assays. Furthermore, this study investigated the effects of Ca 2+ and the Ca 2+ -calcineurin signaling pathway on its antifungal activity against B. cinerea . The results indicated that Pd-D-V reduced the concentration of Ca 2+ in the mycelia of B. cinerea ; CaCl 2 , the Ca 2+ channel blocker verapamil , or the calcineurin inhibitor cyclosporin A could affect the sensitivity of Pd-D-V against B. cinerea ; the expression of genes ( Bccch1, Bcmid1, BccnA, Bccnb1, Bcpmc1, and Bcpmr1 ) of the Ca 2+ -calcineurin signaling pathway decreased after Pd-D-V treatment. In summary, Pd-D-V is compound for developing fungicides against B. cinerea . Pd-D-V can reduce intracellular Ca 2+ concentration and disturb Ca 2+ homeostasis . The Ca 2+ -calcineurin signaling pathway is important in the antifungal activity of Pd-D-V against B. cinerea .

Gray mold caused by Botrytis spp. is a common disease on various hosts, including strawberries. In this study, we obtained 59 Botrytis isolates from strawberries from greenhouses in Zhejiang Province, China. Identification of the sampled isolates at species level was performed by a duplex PCR assay method, the result showed that, in Zhejiang, gray mold on strawberry fruits is caused by a complex of Botrytis groups including B. cinerea group N (47.5 %) and B. cinerea group S (52.5 %). The sensitivities of all Botrytis isolates to pyrimethanil were determined based on discriminatory dose method on L-asparagine-based agar medium plate. Our results showed that the isolates obtained from the greenhouses with continuous use of pyrimethanil developed severe resistance to pyrimethanil, and the resistance frequencies of B. cinerea group N and group S isolates were 89.3 % and 41.9 %, respectively. By sequencing, four different resistance-related point mutations were identified in 38 Botrytis isolates: Bcpos 5 L412F (16 isolates, 42.1 %), Bcpos 5 L412V (14 isolates, 36.8 %), Bcmdl 1 E407K (2 isolates, 5.3 %), and Bcpos 5 L412S ⸱ Bcmdl 1 E407K (1 isolate, 2.6 %). The exogenous addition of methionine could not completely alter the resistance of Botrytis isolates to pyrimethanil. In this study, the pyrimethanil resistance in Botrytis isolates was steadily inherited, and compared to the pyrimethanil-sensitive isolates, the resistant mutants exhibited good fitness in sporulation capacity, spore germination rate, and virulence on strawberries. In conclusion, our results provided a description of the genetic structure of Botrytis groups complex on strawberry fruits and reminded growers to focus on the stable pyrimethanil resistance in Botrytis isolates, caused by point mutations in BcPos5 and BcMdl1.

Lignin, renowned for its renewable, biocompatible, and environmentally benign characteristics, holds immense potential as a sustainable feedstock for agrochemical formulations. In this study, raw dealkaline lignin (DAL) underwent a purification process involving two sequential solvent extractions . Subsequently, an enzyme-responsive nanodelivery system (Pyr@DAL-NPs), was fabricated through the solvent self-assembly method, with pyraclostrobin (Pyr) loaded into lignin nanoparticles . The Pyr@DAL-NPs shown an average particle size of 250.4 nm, demonstrating a remarkable loading capacity of up to 54.70 % and an encapsulation efficiency of 86.15 %. Notably, in the presence of cellulase and pectinase at a concentration of 2 mg/mL, the release of Pyr from the Pyr@DAL-NPs reached 92.66 % within 120 h. Furthermore, the photostability of Pyr@DAL-NPs was significantly improved, revealing a 2.92-fold enhancement compared to the commercially available fungicide suspension (Pyr SC). Bioassay results exhibited that the Pyr@DAL-NPs revealed superior fungicidal activity against Botrytis cinerea over Pyr SC, with an EC 50 value of 0.951 mg/L. Additionally, biosafety assessments indicated that the Pyr@DAL-NPs effectively declined the acute toxicity of Pyr towards zebrafish and posed no negative effects on the healthy growth of strawberry plants. In conclusion, this study presents a viable and promising strategy for developing environmentally friendly controlled-release systems for pesticides, offering the unique properties of lignin.

Postharvest decay, primarily occurring after ripening, leads to substantial losses. Ripening increases tomato susceptibility to Botrytis cinerea, linked to desensitization of jasmonate (JA) biosynthesis and defense signaling. During ripening, key JA biosynthesis genes such as allene oxide synthase (AOS) and 12-oxophytodienoate reductase 3 (OPR3) were downregulated in response to wounding or fungal challenges. The Slmyc2 mutants with impaired JA responses showed heightened pathogen sensitivity. Conversely, EIN3-Like sleil1 mutants exhibited elevated SlMYC2 levels, which increased expression of JA biosynthesis genes such as lipoxygenase D (LOXD), OPR3, AOS, and allene oxide cyclase (AOC) alongside defense genes including proteinase inhibitor PI-6, PI-27, polyphenol oxidase, and pathogenesis-related proteins, thereby enhancing resistance. Treatment with DNA methyltransferase inhibitor 5-azacytidine increased JA content, suggesting epigenetic regulation of JA-mediated defense. Collectively, intensified ethylene signaling during ripening, mediated by SlEIL1, suppressed JA-dependent responses against B. cinerea, highlighting ethylene-JA antagonism as a key driver of postharvest decay.

One novel C10polyacetylene rhamnoside, 4,6,8-decatriyne-1-O-α-L-rhamnopyranoside, named xylariside A (1), together with two novel C10polyacetylene quinovopyranosides, 4,6,8-decatriyne-1-O-α-D-quinovopyranoside, xylariside B (2), and 8E-decaene-4,6-diyne-1-O-α-D-quinovopyranoside, xylariside C (3), were obtained from the solid fermentation ofXylariasp. VDL4, an endophytic fungus isolated fromVaccinium dunalianumwight (Ericaceae). Their chemical structures were elucidated through a combination of spectroscopic techniques. The antifungal activities of these compounds were evaluated in vitro against four phytopathogenic fungi (Fusarium oxysporum,Botrytis cinerea,Phytophthora capsici, andFusarium solani). Compound2demonstrated significant antifungal activities, with minimum inhibitory concentration (MIC) values ranging from 3.91 to 7.81 μg/mL. Compound2’s effectiveness levels were similar to those of the reference drugs thiabendazole and carbendazim (each MIC = 0.98−15.62 μg/mL). Xylariside B (2) was further evaluated againstB. cinereain vivo. It exhibited remarkable efficacy in both the prevention and treatment of tomato and strawberry gray mold. Molecular docking studies confirmed the antifungal mechanism of compound2by revealing its binding interactions with key enzyme targets inB. cinerea, thereby supporting the observed in vitro and in vivo results. Additionally, compound2showed effective inhibition of α-glucosidase, with IC50values of 5.27 ± 0.0125 μg/mL.

Gray mold caused byBotrytis cinereais a common postharvest fungal disease in fruit and vegetables. The prevention and treatment of postharvest gray mold has been one of the hot research issues addressed by researchers. This study aimed to investigate the effect of L-methionine and L-arginine onBotrytis cinereain vitro and on cherry tomato fruit. The results of the in vitro experiment showed that L-methionine and L-arginine had significant inhibitory effects on the mycelial growth and spore germination ofBotrytis cinerea, and the inhibitory effects were enhanced with increasing L-methionine or L-arginine concentration. In addition, L-methionine and L-arginine treatment increased the leakage ofBotrytis cinereaelectrolytes, proteins and nucleic acids. The experiment involving propidium iodide staining and malondialdehyde content assay also confirmed that L-methionine and L-arginine treatment could lead to cell membrane rupture and lipid peroxidation. The results of scanning electron microscopy further verified that the morphology of hyphae was damaged, deformed, dented and wrinkled after treatment with L-methionine or L-arginine. Fruit inoculation experiments displayed that L-methionine and L-arginine treatments significantly inhibited the occurrence and development of gray mold in postharvest cherry tomato. Therefore, treatment with L-methionine or L-arginine might be an effective means to control postharvest gray mold in fruit and vegetables.
