The whitefly Bemisia tabaci (biotype B) is a worldwide pest of vegetables and field crops . We tested the efficacy of imidacloprid (IM) with a root fertilizer ‘Root Feed (RF)’ (9% N, 7% Ca, 1.5% Mg and 0.1% B) sub-irrigated in the growing medium against the whitefly on tomato. Tomato seedlings (3–4 true leaves) were treated with 0, 3, 6 or 12 mg active ingredient (a.i.) seedling −1 of IM and with RF (0, 0.02, 0.04 or 0.08 ml seedling −1 ). The efficacy of 12 mg IM seedling −1 was further evaluated in the greenhouse for 60 days. The survival of B. tabaci adults, nymphs, and egg production were negatively affected by the treatments in a dosage-dependent manner. Treatment of 12 mg IM seedling −1 caused >60% adult and nymph mortality 50 days after treatment (DAT). The active ingredient of IM in tomato leaves also increased with the increase of IM dosage. The RF exhibited a limited effect on B. tabaci . However, the mortality of whitefly adults and nymphs treated with RF (0.02 or 0.04 ml seedling −1 ) positively interacted with IM, and was greater than IM alone. In the greenhouse, 12 mg IM seedling −1 greatly reduced the number of whitefly adults and increased the dry weight of the tomato plants at 30 DAT. In conclusion, application of 12 mg IM seedling −1 on tomato seedlings before transplanting effectively controled B. tabaci for up to 50 days, and the efficacy of IM combined with 0.02 ml RF seedling −1 performed even better. This could be a cost-effective method for managing B. tabaci on tomato and other vegetables.

Elevated ozone (O3) can alter the phenotypes of host plants particularly in induction of leaf senescence, but few reports examine the involvement of phytohormone in O3-induced changes in host phenotypes that influence the foraging quality for insects. Here, we used an ethylene (ET) receptor mutant Nr and its wild-type to determine the function of the ET signaling pathway in O3-induced leaf senescence, and bottom-up effects on the performance of Bemisia tabaci in field open-top chambers (OTCs). Our results showed that elevated O3 reduced photosynthetic efficiency and chlorophyll content and induced leaf senescence of plant regardless of plant genotype. Leaf senescence in Nr plants was alleviated relative to wild-type under elevated O3. Further analyses of foliar quality showed that elevated O3 had little effect on phytohormone-mediated defenses, but significantly increased the concentration of amino acids in two plant genotypes. Furthermore, Nr plants had lower amino acid content relative to wild-type under elevated O3. These results provided an explanation of O3-induced increase in abundance of B. tabaci. We concluded that O3-induced senescence of plant was ET signal-dependent, and positive effects of O3-induced leaf senescence on the performance of B. tabaci largely resulted from changes of nutritional quality of host plants.

Insects rely on odorant-binding proteins (OBPs) for chemical perception, making OBPs a promising target for studying attractants and repellents of pests, such as Bemisia tabaci. However, no reports have reported using B. tabaci OBPs (BtOBPs) as pesticide screening targets. To fill this gap, we obtained BtOBP8 through prokaryotic expression and purification. Then, we confirmed its identity using western blotting and mass spectrometry. Next, we used the sitting drop and hanging drop methods to screen its crystal conditions. Using microscale thermophoresis and isothermal titration calorimetry, we identified the highest affinity ligand, 3l, from 30 compounds. Furthermore, point mutation techniques identified Val119 as a key amino acid residue in binding 31 to BtOBP8. Finally, we tested the bioactivity of B. tabaci Mediterranean and found that 3l more effectively inhibits the bioactivity of B. tabaci MED than imidacloprid. This study presents a new approach for developing green insecticides specific to B. tabaci MED by targeting OBPs. Conclusively, identifying and targeting specific OBPs can create more targeted and effective pest control strategies without relying on toxic chemicals.

Aphidius gifuensis Ashmaed is a generalist endoparasitoid that parasitizes a variety of aphid species. In China, it is widely used as a biological control agent to protect vegetables and tobaccos in open fields; control efficiency is largely dependent on its host-seeking ability. In this study, a six-choice olfactometer was used to investigate the olfactory responses of A. gifuensis to tobacco plants that had suffered damage (either varying degrees of mechanical damage or from aphid-feeding at different time intervals) and tobacco volatiles with different dosages. Furthermore, the regularity of A. gifuensis females’ response toward an aphid/tobacco complex was monitored using a Y-tube olfactometer. Our findings suggest that tobacco plants are significantly attractive to A. gifuensis after they have been punctured with 50 holes, or housed with Myzus persicae (Sulzer) at a density of 400 aphids, except at an infestation time of 12 h. Moreover, aphid density had a more significant effect on the response than the time interval since aphid application. Aphidius gifuensis was found to be active during the daytime and preferred to search for their aphid hosts at 14:00 h. Five EAG-active tobacco volatiles (trans-2-hexenal, methyl salicylate, benzaldehyde, cis-3-hexen-1-ol, and 1-hexanal) were found to significantly attract A. gifuensis females at different concentration ranges. The practical implications of these results are discussed in the framework of the sustainable biological control of pest aphids in agricultural production systems.

Neonicotinoid insecticides face a number of challenges in improving the efficacy and mitigating resistance in Bemisia tabaci. The zeolitic imidazolate framework (ZIF) in metal–organic frameworks (MOFs) has attracted much attention in the field of nanopesticide preparation and controlled release due to its simple preparation. However, monometallic ZIFs have a simple structure and low loading capacity. To solve this problem, we doped Fe into ZIF-L and successfully synthesized a cross-stacked bimetallic ZIF nanocarrier, which can effectively load and release dinotefuran (DNF). Subsequent preparation of Zn-Fe-ZIF@DNF@MPN after encapsulation using metal–phenolic networks (MPNs) increased the DNF loading to 23.74% and achieved >99% release within 24 h. Zn-Fe-ZIF@DNF@MPN has greater resistance to UV light, retention on vegetable leaves, and resistance to rainwater washout. Dynamic residue analysis confirmed its effectiveness and persistence. In addition, it achieved 86.95% mortality against Bemisia tabaci without inhibiting cabbage seed germination. This work is of strategic importance for the development of a functional and environmentally friendly smart nanopesticide.

BACKGROUND Bemisia tabaci (Gennadius) is a serious agricultural pest worldwide. Neonicotinoids are the most important new class of synthetic insecticides used in the management of B. tabaci . However, B. tabaci populations have developed resistance to various active ingredients in neonicotinoids following long-term and widespread application. RESULTS Dinotefuran exhibited high toxicity against most B. tabaci field populations. One population (Din-R) with a high level of resistance to dinotefuran (255.6-fold) was first identified in the field. The Din-R population exhibited medium to high levels of resistance to all the tested neonicotinoid insecticides and a high level of resistance to spinetoram. Genetic inheritance analysis revealed that resistance to dinotefuran was incompletely recessive and polygenic. The synergist piperonyl butoxide significantly increased the toxicity of dinotefuran to Din-R. P450 activity in the Din-R population was 2.19-fold higher than in the susceptible population. RNA-sequencing analysis showed that 12 P450 genes were significantly upregulated in the Din-R population, of which CYP6DW5 , CYP6JM1 and CYP306A1 were found to exhibit more than 3.00-fold higher expression in Din-R when using a reverse transcription quantitative real-time polymerase chain reaction. Expression of eight P450 genes was obviously induced by dinotefuran, and CYP6DW5 showed the highest expression level. After knockdown of CYP6DW5 in Din-R, the toxicity of dinotefuran increased significantly. CONCLUSION P450 had a crucial role in dinotefuran resistance in B. tabaci , and CYP6DW5 was involved in the resistance. These results provide important information for the management of resistance in B. tabaci and improve our understanding of the resistance mechanism of dinotefuran. © 2022 Society of Chemical Industry.

Licorice of the taxonomic genus Glycyrrhiza is a traditional Chinese medicinal plant. Its large-scale cultivation is threatened by insect pests, including the whitefly Bemisia tabaci Gennadius (Hemiptera: Aleyrodidae). Our study (1) examined the preference of B. tabaci for G. uralensis Fischer ex de Candolle, G. glabra L., and G. inflata Batalin through caged trials; (2) identified the volatile organic compounds (VOCs) released by the three Glycyrrhiza species using headspace gas chromatography–mass spectrometry (HS-GC-MS); and (3) screened the olfactory behavior responses of B. tabaci to the key VOCs identified. The caged host preference trial showed that B. tabaci had a clear preference for G. uralensis over G. inflata and G. glabra . Through HS-GC-MS analysis, we identified 26, 33, and 31 VOCs from G. uralensis , G. glabra , and G. inflata , respectively. (3 Z )-Hexen-3-ene-1-yl acetate, trans -3-hexen-1-ol, cis -3-hexen-1-ol, 3-carene, and β-pinene were selected for further olfactory response assays on the basis of variable importance projection values and quantity differences. From these, we learned that cis -3-hexene-1-ol and trans -3-hexene-1-ol, abundant in G. uralensis , attract B. tabaci , whereas 3-carene and β-pinene, abundant in G. inflata , repel B. tabaci . The different VOCs from these three species of licorice may be primary factors affecting the preference for and feeding on cultivated licorice by B. tabaci . Our results will facilitate studies on the potential of using selected VOCs for managing B. tabaci , perhaps in push–pull strategies of their use against the pest.

SUMMARY Piercing-sucking insects, such as whiteflies and aphids, cause massive economic losses in major crops around the world. During feeding, the stylets of piercing-sucking insects navigate cuticles, cell walls, epidermal cells, and mesophyll cells; thus, these barriers are vital for the resistance of plants to insects. However, the relationship between insect stylet probing behavior and the composition and structure of these barriers remains unclear. Here, we identified a tobacco Cuticle Related Factor ( NtCRF ), which was induced significantly by whitefly infestation. Bioassays showed that NtCRF positively regulated plant resistance against whiteflies and green peach aphids. Silencing of NtCRF did not affect plant jasmonic acid (JA) and salicylic acid (SA) defenses but shortened the stylet probing time of phloem-feeders. Further studies confirmed that silencing of NtCRF resulted in significant structure destruction of the leaf cuticle and led to increased epidermal permeability. Overexpression of NtCRF in Arabidopsis also significantly enhanced the plant's resistance against whiteflies and green peach aphids. Our findings expand understanding of plant–insect interactions and provide a strategy for genetic improvement of crop resistance against piercing-sucking insects.

Bemisia tabaci is an important agricultural sucking pest, and it develops serious resistance to various insecticides. Although cytochrome P450 was involved in the resistance to cyantraniliprole, limited studies have been conducted on B. tabaci. In the present study, piperonyl butoxide significantly increased the toxicity of cyantraniliprole. P450 activities in two resistant populations were 1.97- and 2.17-fold higher than that in the susceptible population. Among 79 P450 genes, CYP6CX3 expressions in two resistant populations were 3.08- and 3.67-fold higher than that in the susceptible population. When CYP6CX3 was knocked down, the toxicity of cyantraniliprole increased significantly. The LC50 value of cyantraniliprole to the Drosophila melanogaster line overexpressing B. tabaciCYP6CX3 increased 7.34-fold. The content of cyantraniliprole was decreased by 25.74 ± 4.27% after mixing with CYP6CX3 and CPR for 2 h. These results suggested that the overexpression of CYP6CX3 was likely involved in the resistance to cyantraniliprole in B. tabaci.

The whitefly Bemisia tabaci ( B. tabaci ) stands as the exclusive identified vector for tomato yellow leaf curl virus (TYLCV), the causative agent of tomato yellow leaf curl disease (TYLCD). Current disease management strategies face significant challenges due to the absence of effective antiviral chemicals, necessitating heavy reliance on whitefly population control. However, the occurrence of insecticide resistance and the invasiveness of B. tabaci have severely compromised conventional control methods, highlighting an urgent demand for innovative disease management approaches. This study presents a kind of LDH superpolymer as a nano-insecticide to control B. tabaci . It was generated through self-assembly of LDH SLNSs gel with supplement of matrine (MT) to form nanohybrids. We found that the 150 nm LDH nanosheets displayed a high efficiency to permeate tomato leaf stomata, enabling accumulation in intercellular cavities and mesophyll cell cytoplasm. The loading efficiency of MT-LDH nanohybrids achieved to 16 %. Compared to conventional MT formulations, foliar application of 500 mg/L MT-LDH increased 50.39–75.17 % phyllospheric MT retention in tomato leaves. The nanomaterial exhibited efficient showing 2.83-fold bioactivity against B. tabaci adults than that of aqueous MT solutions. Moreover, we revealed that the biocontrol activity of the MT-LDH nanohybrids conferred dual protective benefits: (1) activation of plant defenses to inhibit mitigation of TYLCV; and (2) enhanced systemic acquired resistance against viral infection. These findings establish a novel antiviral paradigm that both killed vector insect and activate host defence. The MT-LDH nanohybrids offered a potential solution to control TYLCV epidemics with reduced pesticide reliance.
