Solanesol extraction from tobacco leaves by Flash chromatography based on molecularly imprinted polymers

Xiaoqin Ma,Zihui Meng,Lili Qiu,Jing Chen,Yushu Guo,Da Yi,Tiantian Ji,Hua Jia,Min Xue

A novel solanesol extraction method based on molecularly imprinted polymer (MIP) as the Flash chromatography stationary phase was established and evaluated. Spherical MIP particles in a size range of 250–350   μm (d (0.5)   =   320   μm) for solanesol were synthesized by suspension polymerization, with imprinting factor of 3.9. The MIP particles (5.5   g) were packed in common Teflon column as the stationary phase while the sample solution and elution solvent were confirmed as methanol and methanol/acetic acid solution (80/20, v/v), loading at 4   ml/min and eluting 8   ml/min, respectively. Under the optimal chromatographic conditions, the adsorption capacity of the MIP-Flash column was determined as 107.3   μmol/g, and in each process, 370.8   mg purified solanesol (98.4%) could be obtained from the extract (20   mM, 40   ml) of tobacco leaves (14.7   g), and the yield of solanesol was 2.5% of the dry weight of tobacco leaves. The results reported here confirm the feasibility to extract highly purified active ingredients directly from natural products on a large scale by MIP-Flash chromatography.

A simple electrochemical sensor based on rGO/MoS2/CS modified GCE for highly sensitive detection of Pb(II) in tobacco leaves

Chuanen Guo,Chengxiang Wang,Hongyan Sun,Dongmei Dai,Hongtao Gao

High-performance electrode modification materials play a crucial role in improving the sensitivity of sensor detection in electrochemical determination of heavy metals. In this study, a rGO/MoS2/CS nanocomposite modified glassy carbon electrode (GCE) was used to construct a sensitive sensor for detecting lead ions in tobacco leaves. The reduced graphene oxide (rGO) was used to increase the conductivity of the sensor, and the nano-flowered MoS2 could provide a large reaction specific surface area and a certain active site for heavy metal reaction. Chitosan (CS) was used to improve the enrichment ability of heavy metals and increase the electrocatalytic activity of electrode. Thus, an electrochemical sensor with excellent performance in reproducibility, stability and anti-interference ability was established. The stripping behavior of Pb(II) and the application conditions of the sensor were studied by square wave anodic stripping voltammetry (SWASV). The investigation indicated that the sensor exhibited high detection sensitivity in the range of 0.005–0.05–2.0 μM, and the limit of detection (LOD) was 0.0016 μM. This work can provide a fast and effective method for determination of Pb(II) in samples with low content, such as tobacco leaves.

Solid-State Fermentation Improves Tobacco Leaves Quality via the Screened Bacillus subtilis of Simultaneously Degrading Starch and Protein Ability

Ma Lingling,Wang Ying,Wang Xin,Lü Xin

The process of tobacco aging plays a significant role in enhancing the smoking experience by improving the flavor and quality of tobacco leaves. During natural aging, the metabolic activity of the microbes on the surface of tobacco leaves will be greatly changed. Besides, starch and protein are two of the main macromolecular compounds causing the poor smoking quality of tobacco leaves which to be degraded for better tobacco quality. In this study, a bacterium with the simultaneously degrading ability of starch (degradation rate of 33.87%) and protein (degradation rate of 20%) has been screened out from high-class tobacco leaf and then inoculated into low-class tobacco leaf by solid-state fermentation for quality improvement. The changes in components related to carbon and nitrogen showed that the strain had an obvious effect on the quality improvement of tobacco leaves. After that, GC-MS analyses displayed the volatile flavor compounds which become rich and the flavor has been improved. It has been proved that inoculation solid-state fermentation by dominant strain could improve tobacco quality, as well as instead of the traditional natural aging process which greatly shortens the aging process. The work also offers a helpful strategy for solid-state products for deep fermentation.

The snow samples waste tobacco leaves-derived carbon dots for broad-spectrum and sensitive extraction of organophosphate esters

Shitong Zeng,Xingru Hu,Shihao Sun,Shan Liu,Taibo Liang,Binbin Lu,Yalong Guo,Weichen Zhang,Yu lei,Lin Han,Jianping Xie

The increasing organophosphate esters (OPEs) consumption poses a significant threat to human well-being. The OPEs’ sample pretreatment is often challenging due to their distinct physical and chemical properties. Herein, carbon quantum dots derived from waste tobacco leaves (WTL-CQDs) were synthesized using a one-step hydrothermal method and employed as solid-phase microextraction (SPME) fiber coating for OPEs extraction. Coated fibers exhibited a broad-spectrum and sensitive performance for OPE extraction due to strong hydrogen bonds between amino and hydroxyl groups on WTL-CQDs and P = O groups in OPEs. The density functional theory (DFT) calculations revealed that the TD-CQDs binding energy and three alkyl, halogenated and aromatic substituted OPEs is −16 ∼ -18 kJ/mol. When coupled with a gas chromatography-flame photometric detector (GC-FPD), the developed method demonstrated a wide linear range (2–2000 ng/L) and low detection limits (0.01–20 ng/L). The proposed technique effectively detected trace OPEs in real snow meltwater samples.

Introducing CCD1 into isolated Rhodotorula strain enhances flavor production and improves cigar fermentation

Sida Guo,Yasen Li,Beibei Zhu,Qianying Zhang,Zhen Yang,Yun Jia,Quanwei Zhou,Zhengcheng Zhang,Dongliang Li

Introduction: The fermentation process plays an important role in enhancing the quality of cigar tobacco leaves. Through fermentation, microbial metabolism can degrade aromatic precursors and macromolecules, which increases the content of aroma compounds and reduces irritancy of tobacco leaves. Methods: To further enhance the fermentation effect of cigar tobacco leaves, a Rhodotorula strain (Rh3), capable of producing carotenoids and improving fermentation quality, was isolated from cigar tobacco leaves. Subsequent genetic engineering techniques introduced the carotenoid cleavage dioxygenase 1 (CCD1) gene into the isolated Rh3.Results: The modified Rh3 exhibits a significant increase in carotenoid degradation products compared with the original Rh3 in culture medium (from 0.29 μg/mg to 15 μg/mg). Subsequent cigar tobacco leaf fermentation experiments revealed that the modified Rh3 produced 65.9% more carotenoid degradation products compared to the control group, outperforming the original strain, which achieved a 41.4% increase. Furthermore, the modified strain preserves its ability to improve the intrinsic chemical composition of cigar tobacco leaves.Discussion: We show here that modified Rh3 can increase the content of carotenoid degradation products, thereby enhancing the fermentation effect of cigar tobacco leaves. This study presents a beneficial exploration to improve the quality of cigar tobacco leaves for future use and offers a promising strategy for producing flavor compounds from discarded tobacco leaves.

Waste tobacco leaves derived carbon dots for tetracycline detection: Improving quantitative accuracy with the aid of chemometric model

Yan-Mei Liang,Hui Yang,Bo Zhou,Yang Chen,Min Yang,Ke-Su Wei,Xiu-Fang Yan,Chao Kang

The recycling and reutilization of biomass wastes are significant for environmental protection and sustainable development. Recently, there have many studies on utilizing biomass wastes to produce carbon dots . Whereas, the spectrum shift effect that occurs in the quantitative application of carbon dots as fluorescent probes limits the accuracy of the quantitative analysis . In this work, waste tobacco leaves were used as the carbon source for synthesizing a novel carbon dots (CDs(WTL)) through a facile hydrothermal method . The CDs(WTL) possess a series of excellent properties, including good water solubility, well stability, and high fluorescence quantum yield . The fluorescent intensity of the CDs(WTL) can be quenched by tetracycline (TC) obviously, but there is a spectrum shift. In order to use the CDs(WTL) as fluorescent probes to quantify TC with higher accuracy, a quantification fluorescence model (QFM) was introduced to overcome this spectrum shift effect that often occurs. The coefficient of determination ( R 2 ) of traditional quantification model (TQ), partial least squares (PLS), and QFM are 0.9672, 0.9834, and 0.9991, respectively; the average relative predictive error (ARPE) of TQ, PLS, and QFM are 8.8%, 4.5%, and 3.9% for the spiked water samples, and 21.9%, 22.0%, and 2.9% for spiked tablet samples, respectively. The obtained results suggest that QFM is more accurate than PLS and TQ for the TC detection. By utilizing QFM, the spike recoveries (mean ± standard deviation) in three kinds of real tablet samples produced by different manufacturers are 98.9 ± 3.6%, 102.5 ± 6.2%, and 98.5 ± 2.7%, respectively; the spike recovery in river water samples is 99.4 ± 5.0%. In addition, high performance liquid chromatography (HPLC) was used as a reference method, the F and t tests suggest that there are no significant differences on the precision and accuracy between QFM and HPLC methods.

Zheng Feng,Xue Yang,Xiaowei Guo,Jiamin Yu,Fan Li,Jimin Shao,Hui Liang,Hong Jiang

A sensitive surface enhanced Raman spectroscopy (SERS) substrate based on SiO2@Ag core-shell with a QuEChERs pretreatment method were constructed for the determination of carbendazim residue in tobacco leaves. The SERS substrate was formed by self-assembly of monolayer homogeneous SiO2 nanospheres onto silicon wafer following by vacuum thermal evaporation of silver film. The QuEChERs pretreatment method was used to eliminate the interfering effects of pigments and organic acids in tobacco leaves. The results for carbendazim in tobacco leaves show the limit of detection (LOD) of the SERS substrate was 0.1 mg/kg and the recovery rate was from 82.56% to 95.93%. The proposed low-cost sensitive SERS substrates provide potential application for tobacco safety monitoring.

Waste tobacco leaf-driven carbon dot nanozymes with high reactive oxygen species scavenging and anti-inflammatory activities for the amelioration of acute lung injury

Hui Yang,Huimin Qiu,Jie Shan,Chao Nie,Yechun Lin,Yizhong Shen,Xianwen Wang

Considering the important role of tobacco in promoting the economic development of agricultural crops, the high-value recycling of its waste has been a hot topic of interest. In this study, we developed a waste tobacco leaf-driven biomass-based carbon dot (Cur@TB CD) nanozyme with high reactive oxygen species (ROS) scavenging and anti-inflammatory activities for the amelioration of acute lung injury (ALI) both in vitro and in vivo . The results showed that the Cur@TB CD manifested good superoxide dismutase (SOD)-like activity and hydroxyl radical (•OH) scavenging ability, making them impressive scavengers of ROS. Moreover, the intracellular analysis of the Cur@TB CD revealed that Cur@TB CD could spontaneously enter the cells via endocytosis and was distributed mainly in the organelles of lysosomes and mitochondria. As a result, intracellular endogenous ROS can be scavenged to inhibit the expression of proinflammatory cytokines ( e.g. , TNF-α, IL-6, and IL-1β), alleviating inflammation in lipopolysaccharide (LPS)-induced ALI in vivo . Importantly, as a biomass-derived product, Cur@TB CD exhibited negligible side effects and acceptable biosafety in vivo , suggesting promising application potential for curing and preventing ALI in clinical applications, with easy-to-prepare, economical, green and environmentally friendly methods.

Evaluation of lipid and metabolite profiles in tobacco leaves from different plant parts by comprehensive lipidomics and metabolomics analysis

Jie Hao,Xusong Wang,Ying Chai,Xuhui Huang,Han Wu,Shanlin Zhang,Xiaojiao Duan,Lei Qin

Tobacco is an agricultural product with extremely high economic value. Manufacturers have realized that the chemical nature of fresh tobacco leaves influences the flavor properties of tobacco products. To explore the key components that cause the differences in tobacco leaves, high-precision mass spectrometry was employed for multi-omics analysis of three groups (upper, middle, and lower parts) of tobacco leaves. The results showed that the fresh leaves from different parts of Yunyan 87 tobacco plant had significant differences. It was not unique compounds that caused these differences, but more likely variations in shared compounds. The species of glycerolipids, glycerophospholipids, and fatty acyls were the most numerous, their numbers account for 76.68% of total lipids components. The number of glycerolipids accounted for 70% of the identified key lipid components, with the majority being polyunsaturated glycerolipids. It suggested that the disparities in polyunsaturated glycerolipids were the primary cause of lipids differentiation in fresh leaves from different parts. Glycerolipids are effective storage forms of carbon and energy. Carbohydrates and carbohydrate conjugates are the main products of tobacco photosynthesis. In key components, they had the highest relative abundance in the lower leaves. In addition, terpenoids, and organic oxygen compounds, all exhibit high relative abundances in the lower leaves. Amino acids, peptides, and analogues were more abundant in the upper leaves. Differences in the chemical composition of fresh tobacco will result in different sensory qualities and styles. In addition to providing insights into the differences and diversity of tobacco leaves in various parts of the plant during growth, the results present potentially rewarding information for directional cultivation of tobacco plants, grading of tobacco leaves, and optimization of resource utilization.

Detection and classification of lambda-cyhalothrin and iprodione residues in tobacco leaves by SERS combined with supervised machine learning

Weifan Pan,Peng Zhang,Qi Wang,Liangguang Du,Xue Yang,Xiaowei Guo,Jiamin Yu

Surface-enhanced Raman spectroscopy (SERS) is a promising technique for the rapid detection of trace pesticide residues in agricultural produce. Currently, research is predominantly centered on utilizing SERS to discern residues characterized by non-overlapping spectral peaks. Yet, the technology encounters challenges in accurately identifying substances when their Raman spectral peaks overlap. In this paper, a silver nanoparticles (Ag Nps) modified SiO 2 nanoparticles (SiO 2 Nps) arrays composite SERS substrate (SiO 2 @Ag Nps-SERS substrate) was prepared for detecting lambda-cyhalothrin (LCT) and iprodione (IPR) residues in tobacco leaves, with detection limits of 0.1 ppm and 1 ppm respectively. And then, supervised machine learning models were used to identify and classify LCT and IPR residues with overlapped Raman characteristic peaks at 1000 cm −1 . Support Vector Machine (SVM), Partial Least Squares Discriminant Analysis (PLS-DA) and Decision Tree (DT) models could all accurately classify the spectral data of LCT and IPR residues, and the classification accuracy of SVM model was the highest, reaching 99.74 %. This capability is a testament to the innovative strength of our approach, highlighting its potential for universal applicability in spectral data analysis.

Low-cost, immediate, general-purpose, and high-throughput (LIGHt) smartphone colorimetric screening assay for water-soluble protein

Yunfei Sha,Yumei Chen,Wenchen Li,Jianhao Zhang,Jiale Wang,Ting Fei,Da Wu,Weiying Lu

An efficient and rapid method for the detection of total soluble protein in tobacco leaves, utilizing a smartphone-based colorimetric approach has been developed. The proposed low-cost, immediate, general-purpose, and high-throughput (LIGHt) smartphone colorimetric screening assay integrates commercially available microplates, enabling on-site, high-throughput screening of tobacco leaf quality. The study involves preparing protein standard solutions and constructing standard curves using both spectrophotometric and smartphone-based methods. The LIGHt smartphone colorimetry yielded an average relative standard deviation of 10.6 %, a limit of detection of 2 μg/mL, and an average recovery of 93 %. The results demonstrated a comparable performance between intensities from the blue channel and the absorbance values in reflecting protein concentrations, validating the feasibility of utilizing smartphone colorimetry for protein concentration determination. Our approach demonstrates the potential for practical implementation in the field, providing a cost-effective and user-friendly solution for rapid quality assessment in the tobacco industry. The LIGHt smartphone colorimetry enhances quality control practices in the tobacco sector and offers a promising tool for on-site production quality testing in various industries, such as fruits and vegetables.
