The cigarette beetle, Lasioderma serricorne (F.), presents a significant management challenge as a stored product pest. The beetle's increasing resistance to conventional fumigants necessitates the search for alternative solutions to mitigate losses in agro-based industries. Here, we constructed a small molecule compounds library containing twenty-eight methyl benzoate-related compounds (MBCs) and evaluated their repellent and toxic effects against cigarette beetles by Y-tube olfactometer behavioral assays, free-choice box assays and fumigation assays. We found that six MBCs (methyl 3-methoxybenzoate, methyl benzoate, methyl 3-formylbenzoate, methyl 2-aminopyridine-3-carboxylate, methyl 2-aminobenzoate and methyl 4-amino-2-methoxybenzoate) act as repellents for cigarette beetles. Notably, methyl benzoate (MeBA) was effective across a broader range of concentrations (200–4000 ppm) compared to other MBCs. Four compounds (methyl 3-formylbenzoate, methyl 2-aminopyridine-3-carboxylate, methyl 2-aminobenzoate and methyl 4-amino-2-methoxybenzoate) required higher minimum concentrations (500–4000 ppm) than MeBA for repellence, while methyl 3-methoxybenzoate was effective at a concentration (10 ppm) lower than that for MeBA. Mixtures of MeBA with methyl 3-methoxybenzoate and methyl 3-formylbenzoate enhanced the repellent effect against cigarette beetles when compared to MeBA alone. Additionally, exposure to varying concentrations of MeBA, methyl 3-methoxybenzoate, methyl 3-formylbenzoate, and methyl 2-aminobenzoate resulted in 100% mortality of cigarette beetles. Our findings indicate that specific MBCs offer the potential for their use as repellents and fumigants for this problematic stored product pest.

Stored products have been damaged by insects. Multiple approaches for pest management are employed. Among these approaches, botanical insecticide is an emerging one. This work investigated the pest management potential of Magnolia coriacea and Magnolia macclurei essential oils (EOs) to three major stored-product insects, namely the red flour beetle, cigarette beetle and booklouse. Magnolia coriacea and M. macclurei EOs showed promising contact toxicity to the cigarette beetle, with LD50 values of 11.7 and 12.3 μg/adult. The contact toxicity of M. coriacea EOs to the booklouse (LC50 = 95.5 μg/cm2) was much stronger than that of M. macclurei EOs (LC50 = 245.4 μg/cm2). To explore the contribution of individual compounds to insecticidal activity of EOs, chemical analysis was performed by GC-MS. Results showed that nerolidol (27.84%), agarospirol (18.34%), elixene (15.84%) and helminthogermacrene (12.69%) were major compounds of M. coriacea EOs, β-guaiene (60.31%) and elixene (20.42%) dominated in M. macclurei EOs. Nerolidol and β-guaiene showed contact activity to three insect species. Nerolidol showed stronger contact toxicity to the red flour beetle and cigarette beetle than M. coriacea EOs did, both samples were similar to the booklouse. β-Guaiene was much stronger to the red flour beetle and booklouse, but weaker to the cigarette beetle than M. macclurei EOs did. The repellent effects of EOs and compounds were at various levels. Generally, results suggested that the contact toxic potential of samples could serve as management for the cigarette beetle and booklouse, while repellent effect would be used to control the red flour beetle.

In insects, glutathione S -transferases (GSTs) play a pivotal role in the detoxification of a wide range of pesticides. The cigarette beetle, Lasioderma serricorne , is an economically important pest insect of stored products. Recently, pyrethroid insecticides have been used to control this pest. However, little is known concerning the responses and functions of GSTs in L. serricorne under pyrethroid exposure. In this study, transcriptome sequencing was performed on L. serricorne , and a total of 14 GSTs were identified by retrieving the unigene dataset. Of these, 13 predicted GSTs fell into six cytosolic classes, namely, delta, epsilon, omega, sigma, theta, and zeta, and one was assigned to an “unclassified” group. The GST genes were differentially expressed in various larval tissues and at different developmental stages. Exposure to the pyrethroid insecticide lambda-cyhalothrin (LCT) caused oxidative stress in L. serricorne larvae and led to significantly elevated expression levels of six genes, among which LsGSTe1 was the most upregulated. Recombinant LsGSTE1 protein displayed LCT-metabolizing activity. Furthermore, LsGSTE1 protects cells against oxidative stress. Moreover, knockdown of LsGSTe1 by RNA interference dramatically increased the susceptibility of L. serricorne larvae to LCT treatment. The results from this study provide sequence resources and expression data for GST genes in L. serricorne . Our findings indicate that LsGSTE1 plays a dual role in LCT detoxification by metabolizing the pesticide and by preventing LCT-induced oxidative stress. Thus, the LsGSTe1 gene could be used as a potential target for sustainable management of the cigarette beetle.

In this study, four kinds of chemical substances (2,3,5,6-tetramethylpyrazine, β-ionone, citronellal, and paeonol), three kinds of plant essential oils (tea tree essential oil, lavender essential oil, and myrrh essential oil), and their combinations were selected to explore their synergistic effects on tobacco beetle [Lasioderma serricorne (Fabricius) (Coleoptera: Anobiidae)] adults by the behavioral test and laboratory simulation test. Behavioral test results showed that some of the combinations revealed a synergistic effect on tobacco beetle adults, especially the sexual attractant +2,3,5,6-tetramethylpyrazine + β-ionone + citronellal + paeonol (SABCD, one portion of sexual attractant, and 1 mg/L synergistic substances) combination and the food attractant +2,3,5,6-tetramethylpyrazine + paeonol (FAD, 1 ml of food attractant and 1 mg/L synergistic substances) combination showed the best behavioral effect on tobacco beetle adults with average dwell times of 120.97 and 126.74 s, respectively, compared to those of other combinations. Meanwhile, SABCD had the highest selection rate [89.47%, about 1.5 times that of the sexual attractant (S)] on tobacco beetle adults compared with those of other combinations. In addition, laboratory simulation test results showed that the SABCD combination had the highest average selection rate (37.31%, about 2 times that of S) on tobacco beetle adults at 1 mg/L. However, our results showed that there was no significant difference in the indoor simulation results of food attractant synergistic substances. Our results will provide guidance for the development of new pesticides for tobacco beetle adults.
