Lignin Removal from Tobacco Stem with Laccase Improved by Synergistic Action of Weak Alkali and Tween 80

Chen Cheng,Jiang Linfeng,Ma Guanfeng,Jin Dongxue,Zhao Lisha,Ouyang Xinping

Abstract Lignin removal from lignocellulosic materials plays a great role in the utilization cellulose with high efficiency. In order to improve the quality of reconstituted tobacco sheet, lignin existed in tobacco stem was removed by enzymatic hydrolysis with laccase enhanced by weak alkali treatment together with the addition of Tween 80 into laccase treatment system. It is found that lignin was partially removed by laccase. The treatment of tobacco stem with NaHCO 3 resulted in the dissolution of lignin, and hence increased the porosity and specific surface area of the pretreated tobacco stem. Consequently, laccase could access to the tobacco stem more completely. The surface activity and dispersion of Tween 80 improved laccase activity, solubilization of degraded products of lignin, and contact interface between laccase and tobacco stem. The synergistic action of laccase, weak alkali and Tween 80 improved the ratio of lignin removal significantly, reaching as high as 40.3%. Graphical Abstract

Physical and Combustion Properties of Binder-Assisted Hydrochar Pellets from Hydrothermal Carbonization of Tobacco Stem

Liang Miao,Lu Wenjie,Lei Ping,Wang Le,Wang Bing,Li Bin,Shen Yi,Zhang Ke

Abstract Hydrothermal carbonization (HTC) combined with pelletization is an efficient upgrading process to convert biomass into energy-dense pellets. Binders usually serve an important role in the quality of biofuel pellets. Here, hydrochar was produced from tobacco stem through HTC, and was pelletized in the presence of various binders. The effect of four kinds of binders (K 2 CO 3 , CaCO 3 , xanthan gum and carboxymethyl cellulose) on the basic fuel properties, mechanical strength, moisture adsorption and combustion characteristics was investigated. The results showed that K 2 CO 3 assisted pellet had decreased compressive strength, increased hygroscopicity and high combustion residues. The compressive strength of pellets increased with increasing amount CaCO 3 binder probably due to the hardening effect of enhanced adhesion attraction forces between adjacent particles. Meanwhile, the CaCO 3 assisted pellet exhibited the lowest moisture uptake behavior. In addition, the introduction of organic binders led to improved mechanical strength, especially for xanthan binder, which could be ascribed to the formation of solid bridges and enhancement of adhesion forces between particles. TGA results suggested that the binder-assisted pellets exhibited desirable combustion characteristics such as decreased ignition temperature and increased comprehensive combustibility index. Kinetic parameters of the combustion process of binder-assisted pellets were also determined by employing the model free isoconversional Kissinger–Akahira–Sunose method. This study demonstrated that HTC of tobacco stem combined with pelletization in the presence of appropriate binders provide an alternative for solid biofuel pellets production. Pyrolysis characteristics and kinetic analysis of tobacco stem pretreated with different solvents

Li Xiao,Zhao Qingqing,Han Ming,Zhang Ke,Guo Zhongya,Li Bin,Wang Wei,Wei Xuhe,Liang Miao

In the present study, the effects of pretreatments with different solvents (hydrochloric acid, sodium hydroxide, and ethanol) on the physicochemical structure and pyrolysis behavior of tobacco stem (TS) were investigated. Elemental compositions analysis showed that acid washing increased the carbon content and exhibited a high removal percentage for the inorganic species. SEM, BET, and FTIR analysis showed that the pretreatments exerted different effect on the surface and microstructure. Furthermore, the pyrolysis characteristics analysis indicated that solvent pretreatment increased the initial pyrolysis temperature due to the removal of unstable volatile components, especially for the acid and alkali washing samples. Meanwhile, hydrochloric acid pretreatment increased the maximum pyrolysis rate temperature of cellulose decomposition stage and decreased the char residues due to the large removal of inorganic species, while alkali treatment produced the opposite result. Pyrolysis kinetic analysis based on Coats-Redfern method showed that F1.5 chemical reaction model can be used to describe the pyrolysis stages of TS samples and a decreased activation energy was observed during the decomposition of cellulose stage for the acid and alkali washed samples. The released gaseous products during the pyrolysis process were also monitored by using TG-FTIR technique, and the gaseous release behavior was different from each other.

Efficient removal of lignin in tobacco stems with choline chloride-based deep eutectic solvents

Saibo Yu,Bingjie Qiu,Yong Jin,Yu Zhao,Wei Luo,Xinhua Qi

Tobacco stems are a by-product of the tobacco industry and a valuable natural resource. To enhance the quality of tobacco stems, the efficiency of five binary and ternary deep eutectic solvents (DES) composed of choline chloride (ChCl), ethylene glycol (EG), urea, lactic acid (LA), and Gamma-Valerolactone (GVL) for the pretreatment of tobacco stems was investigated with the aim of reducing the lignin content. The experimental analysis showed that all the prepared DES were effective in the removal of lignin from tobacco stems. In particular, the ternary DES system composed of ChCl, LA, and EG ([Ch][LA] 5 [EG] 0.6 ) was the most effective for the pretreatment of tobacco stems, achieving a lignin removal efficiency of 85.9 % at 100°C for 4 h. Under these conditions, a tobacco stem residue rich in cellulose fractions was obtained. The pretreatment of tobacco stems with DES can effectively reduce the lignin content in the stems and the phenolics in the combustion smoke of the stems after delignification, leading to an improvement in the sensory quality of tobacco flake cigarette products.

Green Synthesis of Antioxidant and Degradable Biomass-Based Preservative Films for Efficient Strawberry Preservation

Danya Zhan,Qiongfen Yu,Ming Li,Ying Liu,Shengnan Sun,Rong Zhu,Run Zhang,Zhongfan Mo,Runfang Ma

Utilizing biological resources facilitates alleviating energy shortage and environmental pollution problems. This work aims to develop edible packaging material that performs well and can be used for fresh fruit preservation. Tea polyphenol (TP) was successfully added to a composite-forming solution of tobacco stem cellulose (TSC) and xanthan gum (XG) to synthesize greenly the TSC/XG/TP composite film. Subsequently, the effects of TP dosage on the physicochemical and functional properties of TSC/XG/TP composite films were systematically evaluated. Results showed that the tensile strength (7.89 MPa) and elongation at break (5.27%) were maximums when the optimum TP dosage of 0.4% composite film was achieved. In addition, water vapor and oxygen barrier properties were increased by 159.57 and 7.50%, respectively, and the antioxidant activity was also enhanced by 251.59%. The composite film could maintain the strawberries’ appearance excellently and effectively inhibit color deterioration, thereby delaying postharvest decay, weight loss, and softening. It naturally follows that strawberries’ freshness is prolonged up to 14 days. Practical applications of composite films in preserving fresh strawberries have also shown positive results, making it possible as a potential food packaging material.

Preparation of cellulose nano/microfibres with ultra-high aspect ratios from tobacco stem using soda-oxygen delignification and ultrasonication

Zhang Kecan,Li Jing,He Liang,Zhou Huajing,Guan Qingqing,Chen Keli,Shan Shaoyun,Hu Tianding

Cellulose nano/microfibres (CNFs/CMFs) with high aspect ratio were essential for mechanical strength and flexibility properties. In this study, the tobacco stem was employed as raw material for delignification with soda-oxygen (SO). By comparison with the caustic soda-anthraquinone (SAQ) treatment, based on process performance including pulp yield, residual lignin and fibre characteristics, the two-stage delignification process with total alkali 15% was selected for further preparing CNFs/CMFs with ultrasonication. Results showed that the aspect ratio of highly homogeneous CNFs/CMFs could achieve as ultra-high as 17,645. FTIR and XRD results showed that hemicellulose and lignin in CNFs/CMFs were widely removed, and the crystallinity was about 62%. TGA results exhibited that the degradation temperature was significantly enhanced to 330.2 °C. Finally, the CNF/CMF-based film was prepared and its folding endurance, tensile breaking strength, and burst strength could achieve 136, 61.28 N m/g, 278 kPa m 2 /g. The production of CNFs/CMFs from underutilized tobacco stems provided the potential to add value for industrial waste.

Preparation of cellulose nanocrystal from tobacco-stem and its application in ethyl cellulose film as a reinforcing agent

Shi Junhua,Liu Wenyong,Jiang Xinman,Liu Wenliang

Abstract The brittleness of ethyl cellulose (EC) film limited its full applications, and the addition of plasticizers is one of the most common methods to reduce its brittleness. However, the mechanical properties are normally dropped with the addition of plasticizers. Herein, cellulose nanocrystal (CNC) was chosen as a reinforcing agent to improve its mechanical properties. CNC was firstly extracted from the tobacco-stem and then modified by epoxidized soybean oil (ESO) via ring-opening grafting to obtain epoxidized soybean oil grafting CNC (ECNC). The grafting effect was evaluated by FTIR and XPS. After that, the films of ECNC/EC nanocomposites were prepared by the solution-casting method, and their mechanical, thermal and optical properties and fracture morphology were investigated. The results showed that at 4 phr loading of ECNC, the tensile strength of the nanocomposite film was up to 43.7 Mpa (about twice as much as that of the EC film plasticized by ESO), and the elongation at break was not influenced. Moreover, a higher thermal decomposition temperature was achieved for the ECNC/EC films. Besides, owing to the good dispersion, ECNC had no significant impact on the transparency, and the films presented high light transmittance in the visible light region. Graphic abstract

Beyond electrostatic interaction: Ball milling-induced dominance of pore filling and π-π stacking for methylene blue adsorption on tobacco stem derived activated carbon

Dong-Can Lv,Meng-Ru Chen,Mei-Rong Song,Guang-Ce Jiang,Zhi-Min Wang

Conventional understanding of cationic dye adsorption on carbonaceous materials predominantly emphasizes electrostatic attraction. Herein, we report a ball-milled tobacco stem-derived activated carbon (BMTSAC) that exhibits superior adsorption capacity for methylene blue (MB) via a distinct and more resilient mechanism. Ball milling reduced the particle size to 440 nm and increased the specific surface area to 1171.5 m 2 /g, while concurrently modifying the surface chemistry and pore structure that fundamentally shifted the dominant adsorption pathway. The key finding is the dominance of non-electrostatic interactions, namely pore filling and π-π stacking, over electrostatic attraction. This was evidenced by the high MB uptake (520.7 mg/g) under highly acidic conditions (pH = 2.0), far below the point of zero charge (pH PZC  = 3.0), and the material's robust performance under high ionic strength. The adsorption process followed the pseudo-second-order kinetic and Langmuir isotherm models, with a maximum adsorption capacity of 776.3 mg/g. The revelation of this mechanism shift underscores the superior potential of BMTSAC for treating complex industrial wastewater where pH fluctuates and ionic strength is high. After 5 cycles, BMTSAC maintained excellent reusability with only a 10.6 % capacity loss, further supporting its strong practical application prospects.

Sulfonated tobacco stem carbon as efficient catalyst for dehydration of C6 carbohydrate to 5-hydroxymethylfurfural in γ-valerolactone/water

Feng Huang,Wenzhi Li,Qingchuan Liu,Tingwei Zhang,Shengxin An,Dawei Li,Xifeng Zhu

A carbon solid acid was made by grafting benzenesulfonic acid groups on the tobacco stem-derived porous carbon, which was prepared from waste tobacco stem by simple carbonization . The catalyst was characterized by scanning electron microscope (SEM), N 2 adsorption-desorption, Fourier transform infrared spectroscopy (FT-IR), elemental analysis (EA), pyridine FT-IR spectroscopy and X-ray photoelectron spectroscopy (XPS). The catalyst exhibited excellent catalytic activity for 5-hydroxymethylfurfural (HMF) production from fructose and glucose in γ-valerolactone/water solvent, and high yields of HMF reached 93.7 and 43.8%, respectively. In addition, the high yields of total furans derived from fructose and glucose reached 96.8 and 54.1%, respectively. The benzenesulfonic acid group grafted onto the surface of inner pores of carbon allowed reusing the catalyst for at least five runs without obvious activity reduction. The good performance of the tobacco stem-derived carbon solid acid was determined by the unique surface active sites, such as high density of Brønsted acid, Lewis acid sites, oxygen-containing weak acidic sites and protonated nitrogen-containing functional groups. This study offered a promising approach for efficient carbohydrate conversion.

H2O2-enhanced alkaline pretreatment and separation of tobacco stems for biocellulose composite films with potential application in food preservation

Danya Zhan,Qiongfen Yu,Ming Li,Zhenhua Gu,Shengnan Sun,Yinning Li,Aimin Li,Rong Zhu,Zhongfan Mo,Runfang Ma

Utilizing biological resources facilitates alleviating energy shortage and environmental pollution problems, but high-value utilization of these resources requires breaking the biomass structure. This study realized the clean and efficient separation of cellulose, hemicellulose, and lignin from tobacco stems using a sustainable H 2 O 2 -enhanced alkaline pretreatment combined with acid and ethanol precipitation. The purity and thermal stability of the separated cellulose increased from 32.49% to 92.41% and 261 °C to 369 °C, respectively. Changes in the cellulose crystal form and the solubilization mechanisms of hemicellulose and lignin were elucidated. In addition, a food-preservation film was prepared from the obtained biocellulose using the solution-casting method. After adding glycerol and sorbitol as plasticizers , the tensile strength and water-contact angle (a hydrophobicity indicator) of the biocellulose composite film increased from 125 MPa to 154 MPa and from 50.48° to 73.16°, respectively. This study provides a platform for lignocellulosic biomass separation, renewable utilization, and a practical route toward functional biomaterial production.

High-Efficiency Utilization of Waste Tobacco Stems to Synthesize Novel Biomass-Based Carbon Dots for Precise Detection of Tetracycline Antibiotic Residues

Hui Yang,Yunlong Wei,Xiufang Yan,Chao Nie,Zhenchun Sun,Likai Hao,Xiankun Su

Recycling waste biomass into valuable products (e.g., nanomaterials) is of considerable theoretical and practical significance to achieve future sustainable development. Here, we propose a one-pot hydrothermal synthesis route to convert waste tobacco stems into biomass-based N, S-codoped carbon dots (C−dots) with the assistance of carbon black. Unlike most of the previously reported luminescent C−dots, these biomass-based C−dots showed a satisfactory stability, as well as an excitation-independent fluorescence emission at ~520 nm. Furthermore, they demonstrated a pH-dependent fluorescence emission ability, offering a scaffold to design pH-responsive assays. Moreover, these as-synthesized biomass-based C−dots exhibited a fluorescence response ability toward tetracycline antibiotics (TCs, e.g., TC, CTC, and OTC) through the inner filter effect (IFE), thereby allowing for the establishment a smart analytical platform to sensitively and selectively monitor residual TCs in real environmental water samples. In this study, we explored the conversion of waste tobacco stems into sustainable biomass-based C−dots to develop simple, efficient, label-free, reliable, low-cost, and eco-friendly analytical platforms for environmental pollution traceability analysis, which might provide a novel insight to resolve the ecological and environmental issues derived from waste tobacco stems. Keywords:waste tobacco stems;biomass-based C−dots;tetracycline antibiotics;inner filter effect;environmental pollution traceability analysis

Effect of pyrolysis condition on the adsorption mechanism of lead, cadmium and copper on tobacco stem biochar

Zhi Zhou,Zhihui Xu,Qiuju Feng,Denghui Yao,Jingang Yu,Desheng Wang,Shuangqing Lv,Yifei Liu,Nan Zhou,Mei-e Zhong

To promote the application of biochar in environmental pollution decontamination especially the heavy metals amendment, the relationship between the adsorption mechanism and biochar pyrolysis conditions was investigated. Tobacco stem was pyrolyzed at 300–700 °C for various residence time of 2 h, 4 h and 6 h. The adsorption of lead, cadmium and copper in single solute was then analyzed for as-obtained biochars. The change of the crystal structures, the related functional groups, and the exchanged cations were determined to reveal the effect of pyrolysis conditions on the adsorption mechanisms of these three heavy metals. The change of crystal structure confirmed that precipitation was the main mechanism of lead adsorption and that the tobacco stem biochar obtained at 700 °C for around 2 h was excellent adsorbent for lead clearance, since high temperature pyrolysis could promote the formation of carbonates but they would be decomposed under long-time exposure. The alteration of surface functional groups and exchanged cations of both original and demineralized biochars followed an affinity order of Cu(II)>Cd(II)>Pb(II) to oxygen functional groups. The surface complexation should be the primary mechanism of copper adsorption. Thus, the tobacco stem biochars pyrolyzed at 400 °C for 4 h could be effective adsorbents for copper amendment. Despite of the important role of precipitation in its removal, cadmium favores the cation-π interaction best, and the biochars derived from tobacco stem at 700 °C for the residence time of 4–6 h are good adsorbents for cadmium decontamination .
