In order to investigate characterization and the differences of volatile substances in different types of flue-cured tobacco, the volatile components (VOCs) and relative contents of six varieties of flue-cured tobacco were analyzed by headspace gas chromatography–ion mobility spectrometry (HS-GC-IMS) and headspace solid-phase microextraction–gas chromatography–mass spectrometry (HS-SPME-GC-MS). Combined with the relative odor activity value (ROAV), important flavor substances were determined and different aroma components were screened by multivariate statistical analysis. The results showed that 93 and 80 kinds of compounds, such as esters, alcohols, and aldehydes, were detected by GC-IMS and HS-SPME-GC-MS in six flue-cured tobacco varieties, respectively. A total of 19 volatile flavor compounds, such as benzaldehyde, acetic acid, valeraldehyde, and methyl acetate, were selected as compounds to distinguish the aroma of different toasted tobaccos by orthogonal partial least-squares discrimination analysis (OPLS-DA). In this study, more comprehensive information about volatile flavor components of six flavor types of flue-cured tobacco has been obtained by the synergistic analysis of two distinct analytical instruments. Meanwhile, this study clarified the composition and differences of aroma components of six types of flue-cured tobacco, which lays a foundation for the flavor control of different types of flue-cured tobacco.

Background The aging process in the tobacco production, as in other food industries, is an important process for improving the quality of raw materials. In the spontaneous aging, the complex components in flue-cured tobacco (FT) improve flavor or reduce harmful compounds through chemical reactions, microbial metabolism, and enzymatic catalysis. Some believed that tobacco-microbe played a significant part in this process. However, little information is available on how microbes mediate chemical composition to improve the quality of FT, which will lay the foundation for the time-consuming spontaneous aging to seek ways to shorten the aging cycle. Results Comparing aged and unaged FT, volatile and non-volatile differential compounds (DCs) were multi-dimensionally analyzed with the non-targeted metabolomes based on UPLC-QTOP-MS (the ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry), GC–MS (gas chromatography-mass spectrometer) assisted derivatization and HP-SPME-GC/MS (headspace solid-phase micro-extraction assisted GC–MS). Products associated with the degradation pathways of terpenoids or higher fatty acids were one of the most important factors in improving FT quality. With the microbiome, the diversity and functions of microbial flora were analyzed. The high relative abundance function categories were in coincidence with DCs-related metabolic pathways. According to the correlation analysis, Acinetobacter , Sphingomonas and Aspergillus were presumed to be the important contributor, in which Aspergillus was associated with the highest number of degradation products of terpenoids and higher fatty acids. At last, the screened Aspergillus nidulans strain F4 could promote the degradation of terpenoids and higher fatty acids to enhance tobacco flavor by secreting highly active lipoxygenase and peroxidase, which verified the effect of tobacco-microbes on FT quality. Conclusions By integrating the microbiome and metabolome, tobacco-microbe can mediate flavor-related substances to improve the quality of FT after aging, which provided a basis for identifying functional microorganisms for reforming the traditional spontaneous aging.

Early aging is critical for colour formation in tobacco leaves, yet the underlying metabolic mechanisms remain elusive. This study integrated image processing with untargeted and targeted LC-MS to construct a metabolic network characterising colour evolution in flue-cured tobacco (cv. Cuibi-1, CB-1 ) during early aging (0–120 days). Results indicated that leaf colour significantly brightened during the first 30 days, followed by deepening and reddening. Specifically, L* and b* values showed an initial increase followed by a decrease, whereas the a* value exhibited a sustained upward trend. The constructed metabolic network was anchored in saccharide metabolism, connecting downstream phenolic, flavonoid, sphingolipid, and terpenoid pathways. Sixteen differentially accumulated metabolites (DAMs) were identified across categories including alcohols, flavonoids, lignans, nucleotides, phenolic acids, saccharides, and xanthophylls. These DAMs exhibited distinct change patterns and significant correlations with colour parameters (L* and a*, r ranging from −0.88 to −0.58 and 0.56–0.74, respectively; P < 0.05). For the first time, this study elucidated the metabolic mechanisms governing colour changes in CB-1 tobacco leaves during early aging for the first time, providing novel insights for assessing aging progression and maintaining high-value raw material quality.

Tobacco (Nicotiana tabacum L.) is well-known as an economic crop whose quality is evaluated according to its aroma quality. Researchers have found that selenium application can increase the aroma quality of tobacco, but until now, its mechanism is still unclear. So a field experiment was conducted to investigate the regulatory effect of the trace element selenium on enhancing aroma substances in flue-cured tobacco leaves. We compared selenium content, aroma precursors, non-volatile organic acids (NVOAs) content, and neutral aroma substances in tobacco upper leaves that applying three selenium fertilizer types (sodium selenite, organic selenium and nano-selenium) with three levels of pure selenium application rates (5.24 g·hm-2, 10.48 g·hm-2, and 15.72 g·hm-2) through foliar spraying. The results showed that the neutral aroma substances and NVOAs content were significantly increased. Metabolomics analysis of NVOAs showed that biosynthesis of phenylpropanoids pathway was significantly enriched. This indicates that selenium application may be able to regulate the accumulation of phenylpropane degradation products by regulating these NVOAs involved in the biosynthesis of phenylpropanoids pathway. Selenium content was significantly positively correlated with phenylpropane degradation products, NVOAs content, and phenolic compounds, while pyruvate was significantly down-regulated in each selenium application treatment. Therefore, it was speculated that selenium application could enhance the conversion of pyruvate into the biosynthesis of phenylpropanoids pathway. In summary, spraying organic selenium at 5.24–15.72 g·hm-2 or nano-selenium at 5.24–10.48 g·hm-2 could provide a suitable strategy for improving tobacco aroma quality.

An in-depth insight into the pyrolysis characteristics of tobacco is of great significance for the utilization of tobacco wastes. The effects of glycerol addition on the pyrolysis characteristics and distribution of aroma components of four types of tobacco was investigated in this study by using thermogravimetry and self-built rapid heating furnace capture and GC-MS analysis. Further, correlation network analysis was used to study the correlation between pyrolysis characteristics and released aroma components. It was found that the thermal weight loss behavior of different samples exhibited both commonalities and differences, the thermal weight loss process could be divided into four stages. After the application of glycerol, the overall performance of all types of tobacco leaves at stage II showed that DTG max2 and WL 2 increased, and glycerol had a relatively obvious effect on the pyrolysis characteristic parameters of flue-cured tobacco. Pyrolysis kinetic analysis based on Coats-Redfern method showed that the activation energy and pre-exponential factor of tobacco samples were decreased after the addition of glycerol. Under the heating condition, the total amount of released aroma components from each type of tobacco followed the order of burley tobacco > flue-cured tobacco >oriental tobacco > yellow sun-cured tobacco. The release amount of heterocyclic and olefins compounds from burley tobacco were higher, while flue-cured tobacco released higher contents of esters, ketones and furans compounds. The addition of glycerol promoted the release of aroma components. In terms of correlation, T max2 was negatively correlated with esters, aldehydes, and furans compounds, and positively correlated with DTG max3 . T max3 was significantly correlated with a lot of indicators, positively with acids, aldehydes, and furans compounds, but negatively with olefins and heterocyclic compounds.
