Synthesis of Silver Nanoparticles Using Lysozyme for Scald-Wound Healing

Liang-Liang Chen,Wen-Pu Shi,Tuo-Di Zhang,Wen-Juan Lin,Ya-Qing Zhou,Xiao-Qian Jin,Huan Liang,Wei-Hong Guo,Da-Chuan Yin

The use of natural proteins is a green and effective method for synthesizing silver nanoparticles (AgNPs). Aromatic amino acids of proteins play a reducing role in synthesis, and proteins are coated on the surface of AgNPs to play the role of stabilizers. In this study, we developed an efficient one-step method for synthesizing AgNPs using lysozyme (LYZ) under visible-light irradiation from a xenon lamp in situ. The light conditions and LYZ structure could substantially affect the synthesis. Antibacterial and scald-wound healing results showed that LYZ/AgNPs had broad-spectrum and efficient antibacterial activity and promoted scald-wound healing. Therefore, the proposed synthesis method and LYZ/AgNPs have good practical application prospects.

MiR-133a-3p inhibits scar formation in scalded mice and suppresses the proliferation and migration of scar derived-fibroblasts by targeting CTGF

Abdul Razaq HIRMAN,Lili DU,Shaohang CHENG,Heng ZHENG,Linna DUO,Qianyu ZHAI,Jing XU

Excessive scar formation post burn injury can cause great pain to the patients. MiR-133a-3p has been demonstrated to be anti-fibrotic in some fibrosis-related diseases. However, its possible role in scar formation has not been elucidated yet. In present study, the effect of miR-133a-3p on scar formation was investigated in a scalded model of mice. Moreover, the function of miR-133a-3p on proliferation and migration of scar-derived fibroblasts (SFs) was studied in vitro. It was found that miR-133a-3p was dramatically downregulated in scar tissue of scalded mice. Upregulation of miR-133a-3p by miR-133a-3p agomir obviously inhibited the scar formation in scalded mice. Histological staining showed that upregulation of miR-133a-3p attenuated the excessive deposition of collagen in scar tissue of scalded mice. In vitro study showed that upregulation of miR-133a-3p effectively suppressed the proliferation and migration of SFs. Besides, upregulation of miR-133a-3p attenuated the protein levels of α-smooth muscle actin (α-SMA) and collagen I, indicating that miR-133a-3p could suppress the activation of SFs. The expression of connective tissue growth factor (CTGF), a critical mediator in cell proliferation, migration and extracellular matrix (ECM) synthesis, was also downregulated by the upregulation of miR-133a-3p. Luciferase reporter assay validated that CTGF was directly targeted by miR-133a-3p. In addition, overexpression of CTGF abolished the effect of miR-133a-3p on inhibiting the proliferation, migration and activation of SFs, indicating that miR-133a-3p functioned by targeting CTGF. Therefore, miR-133a-3p might be a promising target for treating pathological scars.

Iodine-responsive poly (HEMA–PVP) hydrogel for self-regulating burst-free extended release

Jinku Xu,Peili Zhao,Yongchun Zhang

Swollen hydrogels with extended iodine release kinetics is highly desirable for burn and scald treatment. In this paper semi-interpenetrating poly (HEMA–PVP) hydrogels were prepared by radical polymerization followed by thermo-treatment to crosslink its PVP component. Incorporation of PVP component endows the hydrogels responsive to loaded iodine undergoing a reversible shrunken/swollen volume transition. This resulted in a self-regulating iodine release model, in which shrunken hydrogel at high iodine loading decreased drug diffusion thereby reducing burst release, and then gradually swollen hydrogel as drug release ensures rapid release of dissociated drug from strong affinity sites on hydrogel backbone, achieving a burst-free extended release. The hydrogels demonstrated 11.5-fold higher iodine loading than pure pHEMA hydrogel and showed a highest 40% volume shrink. Initial burst release of iodine was efficiently decreased from 12,894 μg/day of pure pHEMA hydrogel to 2570 μg/day of pHEMA/PVP hydrogel with 37% PVP content. Iodine-loaded hydrogels showed zero-order release at three time periods of 0–15 h, 15 h–3.5 days and 3.5–23.5 days corresponding to release rate of 2570, 776 and 493 μg/day. The work gained a new insight into swollen hydrogel drug delivery system with burst-free extended drug release kinetics.

A Novel Food Bore Protein Hydrogel with Silver Ions for Promoting Burn Wound Healing

Xiaowen Fu,Gan Hu,Adil M. Abker,Deog-Hwan Oh,Meihu Ma,Xing Fu

Hydrogels have emerged as a promising option for treating local scald wounds due to their unique physical and chemical properties. This study aims to evaluate the efficacy of ovalbumin/gelatin composite hydrogels in repairing deep II-degree scald wounds using a mouse dorsal skin model. Trauma tissues collected at various time points are analyzed for total protein content, hydroxyproline content, histological features, and expression of relevant markers. The results reveal that the hydrogel accelerates the healing process of scalded wounds, which is 17.27% higher than the control group. The hydrogel treatment also effectively prevents wound enlargement and redness of the edges caused by infection during the initial stage of scalding. The total protein and hydroxyproline content of the treated wounds are significantly elevated. Additionally, the hydrogel up-regulates the expression of VEGF (a crucial angiogenic factor) and down-regulates CD68 (a macrophage marker). In summary, this study provides valuable insights into the potential of multifunctional protein-based hydrogels in wound healing.

Crocin-1 laden thermosensitive chitosan-based hydrogel with smart anti-inflammatory performance for severe full-thickness burn wound therapeutics

Xiansen Lv,Hui Li,Ya Chen,Yanting Wang,Jinhua Chi,Shuo Wang,Yan Yang,Baoqin Han,Zhiwen Jiang

Burns are the fourth most common type of civilian trauma worldwide, and the management of severe irregular scald wounds remains a significant challenge. Herein, crocin-1 laden hydroxybutyl chitosan (CRO-HBC) thermosensitive hydrogel with smart anti-inflammatory performance was developed for accelerating full-thickness burn healing. The injectable and shape adaptability of the CRO-HBC gel make it a promising candidate for effectively filling scald wounds with irregular shapes, while simultaneously providing protection against external pathogens. The CRO-HBC gel network formed by hydrophobic interactions exhibited an initial burst release of crocin-1, followed by a gradual and sustained release over time. The excessive release of ROS and pro-inflammatory cytokines should be effectively regulated in the early stage of wound healing. The controlled release of crocin-1 from the CRO-HBC gel adequately addresses this requirement for wound healing. The CRO-HBC hydrogel also exhibited an excellent biocompatibility, an appropriate biodegradability, keratinocyte migration facilitation properties, and a reactive oxygen species scavenging capability. The composite CRO-HBC hydrogel intelligently mitigated inflammatory responses, promoted angiogenesis, and exhibited a commendable efficacy for tissue regeneration in a full-thickness scalding model. Overall, this innovative temperature-sensitive CRO-HBC injectable hydrogel dressing with smart anti-inflammatory performance has enormous potential for managing severe scald wounds.

Liang-Liang Chen,Wen-Pu Shi,Tuo-Di Zhang,Ya-Qing Zhou,Feng-Zhu Zhao,Wan-Yi Ge,Xiao-Qian Jin,Wen-Juan Lin,Wei-Hong Guo,Da-Chuan Yin

Staphylococcus aureus ( S. aureus ) infection, especially its drug-resistant bacterial infection, is a great challenge often faced by clinicians and patients, and it is also one of the most important threats to public health. Finding a safe and effective antibacterial agent is of great significance for the prevention and treatment of S. aureus infection. Lysozyme is known to have antibacterial effects against Gram-positive bacteria including S. aureus . Here, high-quality lysozyme with a purity of more than 99% and an activity of more than 60, 000 U/mg was prepared from egg white, which showed excellent antibacterial activity against three strains of S. aureus , especially against MRSA . Furthermore, an antibacterial cream loaded with lysozyme was prepared and tested in scald wound healing. The lysozyme-loaded cream exhibited the effect of preventing wound infection and promoting wound healing on scalds, and no toxicity was found in animal organs. Overall, lysozyme showed great application potential in the prevention and treatment of infections caused by S. aureus and scalded wound healing. The most remarkable discovery in this work is the unexpectedly powerful inhibitory effect of lysozyme on the drug-resistant bacterial, especially MRSA , which is usually very difficult to deal with using normal antibacterial drugs.

Preparation and evaluation of an innovative antibacterial bi-layered composite dressing for skin wound healing

Leilei Sun,Mingbo Li,Tengfei Gong,Jianling Feng

Aim of the study The aim of the current study was to develop collagen-based bi-layered composite dressings with antibacterial property and evaluate the efficiency for wound healing. Materials and methods A bi-layered composite wound dressing was fabricated using two marine biomacromolecules (collagen and chitosan or carboxymethyl chitosan). Non-crosslinked and N-Ethyl-N'-(3-dimethylaminopropyl) carbodiimide/N-Hydroxy succinimide (EDC/NHS) cross-linked collagen sponges fabricated by vacuum freeze-drying technology was used as the inner layer. The medical spun-laced nonwoven coated with chitosan and carboxymethyl chitosan was used as the outer layer. The antibacterial activities against E. coli and S. aureus were evaluated by the inhibition zone assay. Deep second-degree scald model was performed to evaluate the efficiency of bi-layered composite dressings for wound healing. Results In view of comprehensive evaluation of appearance and in vitro antibacterial activity, medical spun-laced nonwoven coated with 3% of chitosan solution was chosen to be used as the optimized preparation conditions to produce the outer layer of composite dressing, which acted as a barrier against microorganisms and provided mechanical support. Furthermore, the results of wound closure and histopathological analysis indicated that EDC/NHS cross-linked collagen-based bi-layered composite dressing was superior to non-crosslinked and commercial products, which stimulated the wound healing process and accomplished deep second-degree scalded skin healing within a time span of 28 days. Conclusion The EDC/NHS cross-linked collagen-based bi-layered composite dressing had immense potential to be applied for an ideal wound dressing for more efficient and faster wound healing. Therefore, the findings provided the essential theoretical basis for great potential of collagen-based composite dressing used in wound healing applications.

Design of an anti-scald photothermal hydrogel for rapid bacteria removal and hemostasis

Nannan Shao,Jingke Yao,Yanxin Qi,Yubin Huang

Applying hydrogels as medical dressing in treating wounds has become a research hotspot in recent years. Photothermal hydrogels with antibacterial properties and favorable biocompatibility offer great potential for treating infected wounds. However, the risks associated with photosensitizer leakage and excessive heat cause edema and vascular damage, cutting off the nutrients and oxygen supply and slowing the wound healing rate. Herein, a bilayer hydrogel (AuDG 2 ) containing the photothermal and heat-absorbing hydrogel layers is developed to generate adequate heat for quick bacteria removal and tissue protection. In this system, Au nanoparticles are firmly fixed to the nano-hydroxyapatite and coated in the photothermal hydrogel layer, effectively avoiding the leakage of the photosensitizer. Near-infrared imaging shows AuDG 2 hydrogel maintains 48–50 °C at the wound by heat exchange between the two layers of it. This property enables AuDG 2 to have an 89.68 % clearance rate against Staphylococcus   aureus ( S. aureus ) and a 73.03 % removal efficiency of Escherichia coli ( E. coli ) within 5 min, increasing the healing area of infected wounds by 26.45 % compared to the normal saline treatment group, without adverse effects from burns, and evaporate water from the blood to stop blooding in a very short time. This anti-scald photothermal hydrogel structure is simple in preparation, offering the possibility to mitigate the side effects of photothermal therapy.

Bio-inspired aFGF modification functionalized piezoelectric chitosan films for promoting scald wound healing

Yuting Luo,Hangqi Luo,Xiaying Yang,Xin Ding,Kun Wang,Miao Zhang,Jiacheng Wei,Ying An,Jie Xu,Huacheng He,Jiang Wu

The application of acidic fibroblast growth factor (aFGF) has shown great potential in the treatment of scald or burn wounds with high morbidity and mortality, especially in promoting the repair of deep partial-thickness wounds. However, its short half-life and instability in vivo do pose challenges for clinical application. Herein, two kinds of bio-inspired modified piezoelectric chitosan (CS) films, namely heparin-coated CS film (HCS) and polydopamine-coated CS film (DCS), are facially fabricated and adopted as controlled-release platforms for local delivery of aFGF. Polydopamine or heparin layers serve as a bridge grafting on chitosan films, facilitating the loading of aFGF and enabling controlled release of aFGF from the piezoelectric film through intermolecular interactions. Additionally, these layers enhance the hydrophilicity and antibacterial properties of the bare CS film due to their inherent biological activities. Furthermore, the polydopamine coating imparts photothermal activity to the CS film. The in vivo experiments ascertained that the synergetic effect of the controlled-released aFGF and low temperature photothermal therapy collectively accelerate scald wound healing outcomes within 14 days by facilitating granulation formation, collagen deposition, re-epithelialization and angiogenesis. This study opens up new possibilities for the development of multifunctional chitosan-based wound dressings and the creation of innovative drug delivery platforms.

An injectable hydrogel dressing for controlled release of hydrogen sulfide pleiotropically mediates the wound microenvironment

Junwei Yang,Xianzhen Dong,Wenying Wei,Kun Liu,Xiaopei Wu,Honglian Dai

The healing of scalded wounds faces many challenges such as chronic inflammation, oxidative stress, wound infection, and difficulties in vascular and nerve regeneration. Treating a single problem cannot effectively coordinate the complex regenerative microenvironment of scalded wounds, limiting the healing and functional recovery of the skin. Therefore, there is a need to develop a multi-effect treatment plan that can adaptively address the issues at each stage of wound healing. In this study, we propose a scheme for on-demand release of hydrogen sulfide (H2S) based on the concentration of reactive oxygen species (ROS) in the wound microenvironment. This is achieved by encapsulating peroxythiocarbamate (PTCM) in the ROS-responsive polymer poly(ethylene glycol)–poly(L-methionine) (PMet) to form nanoparticles, which are loaded into a thermosensitive injectable hydrogel, F127-poly(L-aspartic acid-N-hydroxysuccinimide) (F127-P(Asp-NHS)), to create a scald dressing. The H2S released by the hydrogel dressing on demand regulates the wound microenvironment by alleviating infection, reducing oxidative stress, and remodeling inflammation, thereby accelerating the healing of full-thickness scalded wounds. This hydrogel dressing for the adaptive release of H2S has great potential in addressing complex scalded wounds associated with infection and chronic inflammation.

A novel approach to wound healing: Green synthetic nano-zinc oxide embedded with sodium alginate and polyvinyl alcohol hydrogels for dressings

Tao You,Qixiu You,Xiaoyan Feng,Hui Li,Bo Yi,Hengyi Xu

Wound healing constitutes a formidable challenge within the healthcare system, attributable to infection risks and protracted recovery periods. The pressing need for innovative wound healing methods has spurred the urgency to develop novel approaches. This study sought to advance wound healing by introducing a novel approach employing a composite sponge dressing. The composite sponge dressing, derived from LFL-ZnO (synthesized through the green methodology utilizing Lactobacillus plantarum ZDY2013 fermentation liquid), polyvinyl alcohol (PVA), and sodium alginate (SA) via a freeze–thaw cycle and freeze-drying molding process, demonstrated notable properties. The findings elucidate the commendable swelling, moisturizing, and mechanical attributes of the SA/LFL-ZnO/PVA composite sponge dressing, characterized by a porous structure. Remarkably, the dressing incorporating LFL-ZnO exhibited substantial inhibition against both methicillin-resistant Staphylococcus aureus (MRSA) and Staphylococcus aureus (S. aureus). Hemolysis and cytotoxicity tests corroborated the excellent biocompatibility of the sponge dressing. In vivo evaluation of the therapeutic efficacy of the 1 mg/mL LFL-ZnO composite dressing on scald wounds and S. aureus-infected wounds revealed its capacity to accelerate wound healing and exert pronounced antibacterial effects. Consequently, the composite sponge dressings synthesized in this study hold significant potential for application in wound treatment.

A cost-effective, fast cooling, and efficient anti-inflammatory multilayered topological hydrogel patch for burn wound first aid

Wenqian Zheng,Lina Wang,He Jiao,Zhixin Wu,Qi Zhao,Tao Lin,Hude Ma,Zhilin Zhang,Xinye Xu,Jie Cao,Jiang Zhong,Jingkun Xu,Baoyang Lu

First-aid burn wound patches play an essential role in reducing mortality by preventing hyperthermia and resisting wound infection. However, existing products still suffer from inferior cooling performance, limited therapeutic effect, weaker wound protection, and/or high cost, which hinder their widespread application in market. Herein, we design and fabricate a cost-effective multilayered topological hydrogel-based first-aid burn wound patch (FBP) for rapid cooling, efficient anti-inflammation, and wound protection. Such a hydrogel patch is fabricated from a polyacrylamide-chitosan (PAAm-CS) hydrogel as the soft antibacterial layer integrated with a topological poly(vinyl alcohol)-chitosan (PVA-CS) hydrogel as the stretchable cooling layer, as well as thin polyethylene films as encapsulated layers to prevent dehydration. As-fabricated FBP exhibits a unique combination of intriguing mechanical, thermal, and biological properties, including softness (Young’s modulus of 83 kPa), stretchability (fracture stain over 106%), high thermal conductivity (0.54 W m −1  K −1 ), instant cooling, effective antibacterial activity (96.2% for S. aureus, 92.2% for E. coli), and excellent biocompatibility (94.04% cell viability at 20 mg mL −1 extract in 24 h). A second-degree scald rat model further demonstrates that fast-cooling and anti-inflammatory FBP can effectively reduce the tissue damage and inflammation levels by showing less edema formation and higher collagen/blood vessel ratios. With a set of these interesting performances, the FBP may be useful as a first-aid burn wound product to alleviate patients’ sufferings and even rescue more lives in critical situation.
