Influence of Heat- and Cold-Stressed Raw Milk on the Stability of UHT Milk

Nan Li,Zhigang Yang,Zhiyu Yuan,Zizhu Zhen,Xinna Xie,Danqing Zhu,Gang Lu,Feng Zhao,Bo Qu,Bingli Qi,Yujun Jiang,Qianyu Zhao,Chaoxin Man

This study investigated the variations and alterations in the concentrations of plasmin system components in raw and UHT (ultra-high-temperature) milk under cold stress (WCT ≤ −25 °C), heat stress (THI ≥ 80), and normal (THI < 70 and WCT ≥ −10 °C) circumstances. The findings indicated elevated amounts of plasmin system components in cold-stressed raw milk. While storing UHT milk at 25 °C, the concentrations and activity of plasmin in the milk exhibited an initial increase followed by a decrease, peaking around the 30th day. The maximum plasmin level and activity in cold-stressed milk were 607.86 μg/L and 15.99 U/L, respectively, with the beginning of gelation occurring around day 60. The higher activity of plasmin in cold-stressed milk led to the poorer stability and sensory assessment of the milk. However, heat-stressed milk is not such a problem for UHT milk as cold-stressed milk. The findings indicate shortcomings in the quality of cold-stressed milk and its adverse effects on the stability of UHT milk, underscoring the necessity of preventing cold stress in the herd and refraining from utilizing cold-stressed milk as a raw material for UHT production.

Naringenin alleviates heat stress-induced liver injury in Ningdu yellow chickens by decreasing RIPK3 and PDC binding

Zhenni Liu,Ruoyun Tang,Qiurong Qi,Siting Lin,Ping Liu,Gaofeng Cai,Zhanhong Zheng,Xiaoquan Guo,Xiaona Gao

Naringenin, a flavonoid extract, possesses anti-inflammatory, antioxidant, hepatoprotective, antitumor, and antineurotoxic properties. This study investigated the anti-heat stress effects in broilers by adding 200mg/kg naringenin to the diet of Ningdu yellow chicken under heat stress conditions. Heat stress conditions was controlled at 37 ± 2°C (7:00 a.m. to 7:00 p.m.) and 24 ± 2°C (7:00 p.m. to 7:00 a.m.) at humidity maintained at 60-65%. The results suggest that naringenin elevated the body weight and the ratio of liver mass to weight of Ningdu yellow chicken significantly. Additionally, naringenin significantly reduces heat stress level, improves liver function and antioxidant capacity. Meanwhile, the levels of necroptosis indexes (CYLD, RIPK1, RIPK3 and MLKL) and oxidative stress indexes (PDC, PYGL, GLUL and GLUD1) are downregulated by naringenin. Naringenin mitigated liver damage by decreasing inflammatory indexes caused by heat stress, including NF-κB, IL-1β, IL-18 and HMGB1. This anti-inflammatory effect arose through the downlink binding of the necroptosis index (RIPK3) and the oxidative stress index (PDC) as shown in results of fluorescence co-localization and co-immunoprecipitation. The use of naringenin in poultry may be a possible feed additive to address clinical heat stress.

Rutin alleviates heat stress induced hepatic abnormal lipid metabolism of broilers via improving antioxidant capacity to maintain mitochondrial homeostasis

Longfei Ma,Huijuan Liu,Zhongyu Ge,Bing Bai,Jianle Zhao,Shun Chen,Binbin Zhou,Jiaqi Zhang,Tian Wang,Chao Wang

Heat stress (HS) leads to abnormal lipid metabolism in broilers, while rutin as a natural flavonoid with multiple biological functions shows potential to regulate antioxidant capacity and lipid metabolism. Therefore, this study aimed to investigate the effects of rutin supplementation on antioxidant and lipid metabolism functions in heat-stress broilers and to further explore its potential mechanisms. A total of 192 male broilers were randomly divided into three groups and treated with normal control (CON), heat stress (34 °C, HS), and HS with 500 mg kg -1 rutin supplementation (HS + Rutin), respectively. The results showed that dietary supplementation of 500 mg kg -1 rutin could significantly reverse the HS-induced liver damage. Rutin also mitigated HS-induced increases in liver triglyceride content and decreases in high-density lipoprotein and fatty acid binding protein contents. This might be achieved by increasing the expression of PPARα and decreasing the expression of SREBP1, which regulated the expression of downstream factors (such as CPT1, ACOX1, and ACC). Compared with the HS group, rutin increased mitochondrial DNA copy number and maintained mitochondrial homeostasis by regulating the expression of related genes (such as AMPK, PGC1-α, and SIRT3). In addition, rutin increased the activity of antioxidant enzymes, decreased the content of malondialdehyde, and activated the nuclear respiratory factor 2 (Nrf2) signaling pathway in the liver, which alleviated HS-induced oxidative stress. The effect of rutin in alleviating abnormal lipid deposition in HS broiler livers were correlated with indicators that reflected the antioxidant capacity and mitochondrial homeostasis. In conclusion, dietary 500 mg kg -1 rutin alleviates heat stress-induced hepatic abnormal lipid metabolism of broilers via improving antioxidant capacity to maintain mitochondrial homeostasis.

Environmental Concentrations of Herbicide Prometryn Render Stress-Tolerant Corals Susceptible to Ocean Warming

Yanyu Zhou,Qiuli Li,Quan Zhang,Meile Yuan,Xiaoshan Zhu,Yuanchao Li,Qipei Li,Craig A. Downs,Danwei Huang,Loke-Ming Chou,Hongwei Zhao

Global warming has caused the degradation of coral reefs around the world. While stress-tolerant corals have demonstrated the ability to acclimatize to ocean warming, it remains unclear whether they can sustain their thermal resilience when superimposed with other coastal environmental stressors. We report the combined impacts of a photosystem II (PSII) herbicide, prometryn, and ocean warming on the stress-tolerant coral Galaxea fascicularis through physiological and omics analyses. The results demonstrate that the heat-stress-induced inhibition of photosynthetic efficiency in G. fascicularis is exacerbated in the presence of prometryn. Transcriptomics and metabolomics analyses indicate that the prometryn exposure may overwhelm the photosystem repair mechanism in stress-tolerant corals, thereby compromising their capacity for thermal acclimation. Moreover, prometryn might amplify the adverse effects of heat stress on key energy and nutrient metabolism pathways and induce a stronger response to oxidative stress in stress-tolerant corals. The findings indicate that the presence of prometryn at environmentally relevant concentrations would render corals more susceptible to heat stress and exacerbate the breakdown of coral Symbiodiniaceae symbiosis. The present study provides valuable insights into the necessity of prioritizing PSII herbicide pollution reduction in coral reef protection efforts while mitigating the effects of climate change.

RNA-seq revealed the effects of heat stress on different brain regions of Leiocassis longirostris

Senyue Liu,Qiang Li,Yongqiang Deng,Zhongwei Wang,Yang Feng,Han Zhao,Zhongmeng Zhao,Lu Zhang,Yuanliang Duan,Zhipeng Huang,Jian Zhou,Chengyan Mou

Understanding how distinct brain regions of Leiocassis longirostris molecularly adapt to heat stress is vital for improving aquaculture sustainability and guiding conservation strategies in a warming climate. To elucidate the region-specific molecular mechanisms underlying heat stress responses in the brain of L. longirostris , we exposed L. longirostris to acute heat stress (32°C) for 24 h and performed RNA-seq and WGCNA on five brain regions (OB: olfactory bulb, FB: pituitary, hypothalamus, forebrain, MB: mesencephalon, CB: cerebellum, and SC: spinal cord). The results showed that, after heat stress, the FB region significantly activated the ER stress pathway, and the abnormal proteins were synergically cleared by HSP-mediated UPR (such as Hsp70, Hsp90, IRE1α, Perk, ATF6) and UPS-mediated ERAD (such as UBE2, UBE3, TRIM63). Meanwhile, the SC region showed marked downregulation of lipid metabolism and PPAR signaling pathway, suggesting energy conservation as a compensatory strategy. WGCNA further highlighted the FB as the hub for ER stress and the SC for metabolic suppression. In conclusion, our study suggests that distinct brain regions of L. longirostris adopt different strategies under heat stress, in which the FB region mediates protein quality control and the SC region drives metabolic inhibition. These findings highlight the adaptation strategies of the L. longirostris brain to heat stress and provides a potential target for improving its survival under global warming.

Modification of structure, epitope and allergenicity on heat-stressed ovalbumin by resveratrol

Caiying Yang,Ying Zhang,Xiaoya Zhou,Zhihua Wu,Jinyan Gao,Hongbing Chen,Ping Tong

Allergen modification is a novel strategy for preventing and treating food allergy. Resveratrol (RES) can be used as a modifying ligand for allergens, but the interaction mechanism between RES and allergens is still not detailed, and the ramifications of this interaction on the structure, epitope and allergenicity of allergens are also lacking a clear explanation. In addition, the application of heat can promote protein structure to unfold and expose more binding sites to facilitate interaction. Therefore, in this study, ovalbumin (OVA) was selected as the allergen model and was heated to expose more binding sites. The effects of the interaction between RES and heat-stressed ovalbumin (HOVA) on allergen structure, epitope and allergenicity were investigated by combining routine experimental analysis and computer simulation. Results showed heat stress at 323 K for 15 min could better unfold the molecular structure of OVA. On this basis, the interaction between RES and OVA was strengthened, which showed a static quenching of the ground state complex with binding affinity of 1.25 × 10 6  L/mol. The binding of RES induced the variation of protein microenvironment, and the overall structure tended to disordered direction. Computer simulation had shown RES could directly act on the IgE epitope region (AA251-260) of the OVA. The allergenicity evaluation experiment further indicated the combination of RES alleviated the IgE binding capacity of HOVA. These findings are helpful to further study the mechanism of RES ligand modification to change food allergy.

Dietary rutin improves growth performance, intestinal barrier, mitochondrial homeostasis and redox balance in broilers under heat stress

Huai Shi,Jiaqi Zhang,Huijuan Liu,Shun Chen,Minhang Tu,Gentan Cai,Tong Jin,Tian Wang,Chao Wang

The benefits of rutin administration were evaluated, focusing on effects on broiler growth performance, intestinal barrier, mitochondrial homeostasis, and redox balance under heat stress. The results demonstrated that compared to the HS group, dietary rutin improved growth performance (increasing final body weight, average daily feed intake, and average daily gain), jejunal morphology (expanding villus width and surface area), jejunal barrier (elevating concentrations of Occludin, Claudin-1, and Mucin-2; upregulating mRNA expression of Occludin and Mucin-2), redox balance in both serum and jejunum (boosting total antioxidant capacity, superoxide dismutase activity, glutathione levels, and the glutathione-to-oxidised glutathione ratio; increasing mRNA expression of glutathione S-transferase and nuclear factor erythroid 2–related factor 2), and jejunal mitochondrial homeostasis (amplifying mitochondrial DNA copy number and ATP synthase α-subunit mRNA levels) (p < 0.05). Moreover, dietary rutin reduced the ratio of feed to gain, lowered liver alkaline phosphatase activity, decreased concentrations of serum diamine oxidase and malondialdehyde, reduced jejunal oxidised glutathione levels, and downregulated jejunal mucosa mRNA expression of B-cell lymphoma 2 associated X, kelch-like ECH-associated protein 1, and heat shock protein 90 (p < 0.05). Overall, rutin supplementation (optimal dose of 500 mg/kg) effectively attenuates HS-induced decline in broilers’ growth performance, impairment of intestinal morphology and barrier integrity, disruption of mitochondrial homeostasis, and redox imbalance, potentially through Nrf2-mediated mechanisms.

HSP90 Enhances Mitophagy to Improve the Resistance of Car-Diomyocytes to Heat Stress in Wenchang Chickens

Jiachen Shi,Zeping Ji,Xu Yao,Yujie Yao,Chengyun Li,Qijun Liang,Xiaohui Zhang

Heat shock protein 90 (HSP90) is recognized for its protective effects against heat stress damage; however, the specific functions and underlying molecular mechanisms of HSP90 in heat-stressed cardiomyocytes remain largely unexplored, particularly in tropical species. In our study, Wenchang chickens (WCCs) were classified into two groups: the heat stress survival (HSS) group and the heat stress death (HSD) group, based on their survival following exposure to heat stress. Heat stress resulted in significant cardiomyocyte damage, mitochondrial dysfunction, and apoptosis in the HSD group, while the damage was less pronounced in the HSS group. We further validated these findings in primary cardiomyocytes derived from Wenchang chickens (PCWs). Additionally, heat stress was found to upregulate Pink1/Parkin-mediated mitophagy, which was accompanied by an increase in HSP90 expression in both cardiomyocytes and PCWs. Our results demonstrated that HSP90 overexpression enhances PINK1/Parkin-mediated mitophagy, ultimately inhibiting apoptosis and oxidative stress in heat-stressed PCWs. However, the application of Geldanamycin (GA) reversed these effects. Notably, we discovered that HSP90 interacts with Beclin-1 through mitochondrial translocation and directly regulates mitophagy levels in PCWs. In summary, we have elucidated a novel role for HSP90 and mitophagy in regulating heat stress-induced acute cardiomyocyte injury.

Herbicide prometryn aggravates the detrimental effects of heat stress on the potential for mutualism of Symbiodiniaceae

Yanyu Zhou,Fucun Liu,Meile Yuan,Xianhua Liu,Qipei Li,Hongwei Zhao

Ocean warming threatens the health of corals globally, and superimposed coastal environmental pollution can result in severe and irreversible coral bleaching. However, the responses of the coral symbiont Symbiodiniaceae to multiple stresses remain largely unknown. This study investigated the response of the coral symbiotic algae Cladocopium sp. to short-term exposure (4 days) to an environmentally relevant concentration (1 μg L −1 ) of the photosystem II (PSII) herbicide prometryn under heat stress (32 ℃) through physiological and omic analyses. These results showed that co-stress affected the photosynthetic efficiency of Cladocopium sp. negatively. Overproduction of reactive oxygen species and subsequent oxidative stress under co-stress activated distinct regulatory pathways in Cladocopium sp. Transcriptomic and proteomic analyses revealed that prometryn exacerbated heat stress-induced photosystem damage and reduced the regulatory capacity of Cladocopium sp. Moreover, co-stress disrupted energy metabolism, and further impaired nitrogen assimilation and nutrient transfer processes, potentially compromising the symbiotic potential between corals and Symbiodiniaceae. In summary, this study offers a valuable insight into understanding the molecular responses of Symbiodiniaceae to thermal and prometryn co-stress. It helps uncover the potential toxicity mechanisms induced by herbicide on coral symbionts in the context of climate change.

Drp1 regulated PINK1-dependent mitophagy protected duck follicular granulosa cells from acute heat stress injury.

Chen Yang,Pei Luo,You-tian Yang,Xin-liang Fu,Bing-xin Li,Xu Shen,Dan-ning Xu,Yun-mao Huang,Yun-bo Tian,Wen-jun Liu

ABSTRACT The mitochondrial quality control system is crucial in maintaining cellular homeostasis during environmental stress. Granulosa cells are the main cells secreting steroid hormones, and mitochondria are the key organelles for steroid hormone synthesis. The impact of the mitochondrial quality control system on granulosa cells’ steroid hormone synthesis and survival under heat stress is still unclear. Here, we showed that acute heat stress induces mitochondrial damage and significantly increases the number of mitophagy-like vesicles in the cytoplasm of duck ovary granulosa cells at the ultra-structural level. Meanwhile, we also found heat stress significantly increased mitochondrial fission and mitophagy-related protein expression levels both in vivo and in vitro . Furthermore, by confocal fluorescence analysis, we discovered that LC3 was distributed spot-like manner near the nucleus in the heat treatment group, and the LC3 spots and lysosomes were colocalized with Mito-Tracker in the heat treatment group. We further detected the mitophagy-related protein in the cytoplasm and mitochondria, respectively. Results showed that the PINK1 protein was significantly increased both in cytoplasm and mitochondria, while the LC3-Ⅱ/LC3-Ⅰ ratio increase only occurred in mitochondrial. In addition, the autophagy protein induced by acute heat treatment was effectively inhibited by the mitophagy inhibitor CysA. Finally, we demonstrated that the alteration of cellular mitophagy by siRNA interference with Drp1 and PINK1 inhibited the steroid synthesis of granulosa cells and increased cell apoptosis. Study provides strong evidence that the Drp1 regulated PINK1-dependent mitophagy pathway protects follicular granulosa cells from acute heat stress-induced injury.

Dietary rutin improves breast meat quality in heat-stressed broilers and protects mitochondria from oxidative attack via the AMPK/PINK1–Parkin pathway

Shun Chen,HuiJuan Liu,JiaQi Zhang,BinBin Zhou,XiaoFang He,Tian Wang,Chao Wang

BACKGROUND This study was conducted to investigate the effect of dietary rutin on the meat quality, antioxidant status and mitochondrial structure and function in the breast muscle of heat-stressed broilers. A total of 192 male broilers were randomly assigned into three groups and treated with normal control (CON), heat stress (34 °C, HS), and HS with 500 mg kg −1 rutin supplementation (HS + Rutin), respectively. RESULTS Dietary rutin significantly reversed HS-induced decrease in body weight, average daily feed intake, average daily gain, and feed efficiency. Rutin supplementation attenuated HS-induced impaired meat quality by decreasing the lightness, drip loss at 24 and 48 h, the peak time of free water ( T 22 ) and the peak area ratio of free water ( P 22 ), and increasing the pH 24h and peak area ratio of immobilized water ( P 21 ). Rutin supplementation promoted superoxide dismutase, glutathione peroxidase activities and total antioxidant capacity, and decreased malondialdehyde levels compared with the HS group. Moreover, rutin attenuated HS-induced mitochondrial damage by increasing the mitochondrial DNA copy number and improving mitochondrial morphology. Dietary rutin significantly increased mitochondrial biogenesis-related mRNA (proliferator-activated γ receptor coactivator-1α [PGC-1α], nuclear respiratory factor 1 [NRF1], and mitochondrial transcription factor A [TFAM]) expression via the AMP-activated protein kinase (AMPK) signaling pathway. HS significantly increased mitophagy-related genes and proteins (Parkin, PTEN-induced putative kinase 1 [PINK1], microtubule associated protein light chain 3-II [LC3-II]) expression, and dietary rutin significantly reversed these alterations. CONCLUSION Dietary rutin attenuated the HS-induced decline in meat quality and antioxidant capacity of broilers, which may be related to inhibition of the AMPK/PINK1–Parkin signaling pathway to attenuate mitochondrial damage. © 2023 Society of Chemical Industry.

Frequent and mild scrotal heat stress impairs embryo development, implantation and offspring sex ratio in mice

Xiaoyan Wan,Xiaomei He,Qian Liu,Peng Duan,Honggang Li

Research question Frequent and mild scrotal heat stress (fmSHS) often occurs in humans. The aim of this study was to determine the impact of fmSHS on natural fertility , the IVF–embryo transfer (IVF–ET) process and the offspring sex ratio. Design Male mice were randomly divided into four groups: no scrotal heat stress (SHS) (controls) and those subjected to SHS at 37°C, 39°C or 41°C for 30 min once a week for 5 consecutive weeks. The testis, epididymis and sperm quality were assessed to evaluate the effects of different degrees of SHS, to establish an fmSHS model. Then, natural fertility, IVF–embryo transfer (IVF–ET) results, embryo development, offspring sex ratio and the X/Y chromosome-bearing sperm ratio were examined. Results SHS at 39°C and 41°C caused mild impairment to spermatozoa , leading to a phenotype similar to oligoasthenozoospermia in humans. Given that most SHS conditions in humans are close to body temperature, SHS of 39°C was adopted to build the fmSHS model. fmSHS reduced the fertilization rate, impaired on-time development and reduced the implantation rate of the embryos in the IVF–ET process, but it did not affect the development or function of blastocysts . The fmSHS mice produced more Y chromosome-bearing spermatozoa and propagated more male offspring. Conclusions fmSHS not only reduced the fertilization ability of spermatozoa but also influenced their function beyond fertilization, in addition to changing the offspring sex ratio. These results may help to shed new light on the infertility treatment of males with scrotal heat risk and the health concerns of offspring propagated from these fathers.
