Tires are widely used in modern society and various industries. However, the formulations and manufacturing processes of traditional rubber are complex, and it is difficult to simultaneously meet requirements of low rolling resistance, high wet-skid resistance and wear resistance. Herein, we demonstrate a polyurethane elastomer material with high heat resistance and self-repairable ability to overcome the challenges encountered during the development of rubber tires. Results show that the polyurethane elastomer based on polycarbonate glycol, 1,5-naphthalene diisocyanate , and 1,4-butanediol simultaneously exhibits self-repairable ability, high heat resistance, low rolling resistance, high wear resistance, and high wet-skid resistance. The comprehensive performance of this material matches that of existing green tires. Moreover, the material can be recycled several times. This study is expected to solve the existing problems in rubber tires and provides novel ideas for preparing next-generation high-performance tire materials.

Background Photothermal therapy (PTT) is taken as a promising strategy for cancer therapy, however, its applicability is hampered by cellular thermoresistance of heat shock response and insufficient accumulation of photothermal transduction agents in the tumor region. In consideration of those limitations, a multifunctional “Golden Cicada” nanoplatform (MGCN) with efficient gene delivery ability and excellent photothermal effects is constructed, overcoming the thermoresistance of tumor cells and improving the accumulation of indocyanine green (ICG). Results Down-regulation of heat shock protein 70 (HSP70) makes tumor cells more susceptible to PTT, and a better therapeutic effect is achieved through such cascade augmented synergistic effects. MGCN has attractive features with prolonged circulation in blood, dual-targeting capability of CD44 and sialic acid (SA) receptors, and agile responsiveness of enzyme achieving size and charge double-variable transformation. It proves that, on the one hand, MGCN performs excellent capability for HSP70-shRNA delivery, resulting in breaking the cellular thermoresistance mechanism, on the other hand, ICG enriches in tumor site specifically and possesses a great thermal property to promoted PTT. Conclusions In short, MGCN breaks the protective mechanism of cellular heat stress response by downregulating the expression of HSP70 proteins and significantly augments synergistic effects of photothermal/gene therapy via cascade augmented synergistic effects.

The overexpression of heat shock proteins (HSPs) in tumor cells can activate inherent defense mechanisms during hyperthermia-based treatments, inducing thermoresistance and thus diminishing the treatment efficacy. Here, we report a distinct “non-inhibitor involvement” strategy to address this issue through engineering a calcium-based nanocatalyst (G/ [email protected] 3 -PEG). The constructed nanocatalyst consists of calcium carbonate (CaCO 3 )-supported glucose oxidase (GOD) and 2D antimonene quantum dots (AQDs), with further surface modification by lipid bilayers and polyethylene glycol (PEG). The engineered G/ [email protected] 3 -PEG nanocatalyst features prolonged blood circulation, which is stable at neutral pH but rapidly degrades under mildly acidic tumor microenvironment, resulting in rapid release of drug cargo in the tumor microenvironment. The integrated GOD effectively catalyzes the depletion of glucose for reducing the supplies of adenosine triphosphate (ATP) and subsequent down-regulation of HSP expression. This effect then augments the therapeutic efficacy of photothermal hyperthermia induced by 2D AQDs upon irradiation with near-infrared light as assisted by reversing the cancer cells’ thermoresistance. Consequently, synergistic antineoplastic effects can be achieved via low-temperature photothermal therapy. Systematic in vitro and in vivo evaluations have demonstrated that G/ [email protected] 3 -PEG nanocatalysts feature potent antitumor activity with a high tumor-inhibition rate (83.92%). This work thus paves an effective way for augmenting the hyperthermia-based tumor treatments via restriction of the ATP supply.

Tumoral thermal defense mechanisms considerably attenuate the therapeutic outcomes of mild-temperature photothermal therapy (PTT). Thus, developing a simple, efficient, and universal therapeutic strategy to sensitize mild-temperature PTT is desirable. Herein, we report self-delivery nanomedicines ACy NPs comprising a near-infrared (NIR) photothermal agent (Cypate), mitochondrial oxidative phosphorylation inhibitor (ATO), and distearoylphosphatidylethanolamine-polyethylene glycol 2000 (DSPE–PEG2000), which have a high drug-loading efficiency that can reverse tumoral thermal resistance, thereby increasing mild-temperature PTT efficacy. ACy NPs achieved targeted tumor accumulation and performed NIR fluorescence imaging capability in vivo to guide tumor PTT for optimized therapeutic outcomes. The released ATO reduced intracellular ATP levels to downregulate multiple heat shock proteins (including HSP70 and HSP90) before PTT, which reversed the thermal resistance of tumor cells, contributing to the excellent results of mild-temperature PTT in vitro and in vivo. Therefore, this study provides a simple, biosafe, advanced, and universal heat shock protein-blocking strategy for tumor PTT.

Nanomedicine with stable light-heat conversion and spatiotemporally controllable drug activation is crucial for the success of photothermal therapy (PTT). Herein, a metal–organic framework (MOF)-based nanoheater with light-triggered multi-responsiveness is engineered to in-situ and on-demand sensitize cancer cells to local hyperthermia. Well-dispersed platinum nanoparticles synthesized inside nanospaces of the MOF are employed as the near-infrared (NIR)-harvesting unit with stable and high light–heat conversion performance. A conformation switchable polymer shell is constructed as a secondary light-responding unit to gate the targeted activation of a molecular inhibitor against thermoresistance. By cascade transformation of light stimuli to downstream signals, the nanoheater enables inhibitor release to go with local heating at the same time restricted in lesion sites to maximize efficacy and minimize systemic toxicity. The efficient photothermal conversion and the blockage of cellular heat-protective pathways provide a dual-mode of action which selectively sensitizes cancer cells to hyperthermia in a spatiotemporally controlled manner. With NIR as the remote switch, the MOF-based nanosystem demonstrates localized and boosted PTT efficacy against cancer both in vitro and in vivo. These results present nanosized MOFs as tailorable and versatile platforms for synergistic and precise cancer therapy.
