Comparative study between cold and hot water extracted polysaccharides from Plantago ovata seed husk by using rheological methods

Peiyuan Zhou,Mohamed Eid,Wenfei Xiong,Cong Ren,Tingyang Ai,Ziyu Deng,Jing Li,Bin Li

Cold and hot water extracted polysaccharides (CP and HP) were isolated from Plantago ovata seed husk at 25 and 85 °C, respectively. Their rheological properties were systematically investigated and compared. Different concentrations and temperatures were studied to evaluate the viscoelastic properties of the two fractions. It was found that CP solutions behaved as viscous fluids, while HP solutions preferred to exhibit as gel-like structure. The sol-gel transition process of HP was evaluated and the gel point was determined to be 4.8 mg/mL by Winter-Chambon method in water at 25 °C. The steady flow sweep investigated the viscosity properties of CP solutions and two critical concentrations were found, which locating the transition from the dilute to semi-dilute region, and from the semi-dilute to concentrated region. In addition, the two fractions could form thermo-reversible weak gels, and the gelation temperatures of them increased with the increase of concentration. It is evident that both polysaccharides have significant potential for application in food industry.

Optimization of ultrasonic extraction of Lycium barbarum polysaccharides using response surface methodology

Jitian Song,Shi Dongqi,Su Hang,Feng Yongxia,Tian Wei

Ultrasonic extraction was a new development method to achieve high-efficiency extraction of Lycium barbarum polysaccharides instead of hot water extraction. In this paper, the single factor method combined with the box Behnken design of response surface method was used to study the influence of extraction time, extraction temperature, material liquid ratio and ultrasonic power on the extraction rate of L . barbarum polysaccharide. The results indicated that the best extraction rate of L . barbarum polysaccharide was 12.54 ± 0.12% under the conditions of 80 min for extraction time, 73 °C for extraction temperature, 1 g:38 mL for material to liquid ratio, and 185 W for ultrasonic power. Under the same operating conditions, the yield of L . barbarum polysaccharide using ultrasonic extraction was 83.3%, which was higher than that hot water extraction. Moreover, the extraction time of ultrasonic extraction was only 47% of that using hot water extraction. This suggested that there was great potentials of using ultrasonic extraction in the realization of high-efficiency extraction of L. barbarum polysaccharide. The results of this study could also provide a theoretical basis for the coupling of ultrasonic extraction and ultrasonic concentration process to develop the integrated equipment of both ultrasonic extraction and ultrasonic concentration.

Optimization of Extraction Process, Preliminary Characterization and Safety Study of Crude Polysaccharides from Morindae Officinalis Radix

Yaxian Chen,Yini Cai,Zhimin Zhao,Depo Yang,Xinjun Xu

In this study, the hot water extraction process of crude polysaccharides from Morindae officinalis radix (cMORP) was conducted and optimized through a single-factor test and orthogonal experimental design. With the optimal extraction process (extraction temperature of 80 °C, extraction time of 2 h, liquid/solid ratio of 15 mL/g, and number of extraction of 1), the cMORP was obtained by the ethanol precipitation method. The chemical properties and preliminary characterization of the cMORP were analyzed by chemical or instrumental methods. Furthermore, to indicate a preliminary study on safety, a single oral dose of 5000 mg/kg body weight (BW) was administered orally to Kunming (KM) mice for acute toxicity, and the cMORP was administered orally to KM mice once a day at doses of 25, 50, and 100 mg/kg BW for 30 days. General behaviors, body weight variations, histopathology, relative organ weights, and hematological and serum biochemical parameters were observed and recorded. The results suggested there were no toxicologically significant changes. Based on the safety study, cMORP can be initially considered non-toxic with no acute oral toxicity up to 5000 mg/kg BW and safe at up to 100 mg/kg BW in KM mice for 30 days. Keywords:Morindae officinalis radix;crude polysaccharides;hot water extraction;optimization;preliminary characterization;safety study

Pressurized hot water extraction, structural properties, biological effects, and in vitro microbial fermentation characteristics of sweet tea polysaccharide

Jing Lei,Wei Li,Meng-Xi Fu,An-Qi Wang,Ding-Tao Wu,Huan Guo,Yi-Chen Hu,Ren-You Gan,Liang Zou,Ying Liu

Although sweet tea is rich in bioactive polysaccharides , the knowledge regarding their structures, bioactivities, and gut microbial metabolism is still limited. Therefore, in order to promote the application of sweet tea polysaccharide (STP) in the food industry, the pressurized hot water extraction (PHWE) of STP was optimized, and its structural properties and biological effects as well as microbial fermentation characteristics were investigated. The maximum extraction yield (4.64 % ± 0.03 %) of STP extracted by PHWE was obtained under the optimal conditions. Both homogalacturonan and arabinogalactan might exist as major polysaccharide fragments in STP. Additionally, STP exerted obviously in vitro antioxidant, anti-diabetic, and immunostimulatory effects, which might be related to its chemical properties, such as uronic acids , conjugated polyphenolics, and esterification degree. Furthermore, STP could be consumed by intestinal microbiota , and its fermentability was about 54 % at the end stage of fecal fermentation. Indeed, STP could modulate the microbial composition via improving the growth of several beneficial microbes, causing the release of beneficial short-chain fatty acids. Collectively, the findings indicate that the PHWE is an efficient method for extracting bioactive polysaccharides from sweet tea, and results can also provide a scientific basis for developing STP into functional foods or functional ingredients.

Comparison on characterization and antioxidant activity of polysaccharides from Ganoderma lucidum by ultrasound and conventional extraction

Qiaozhen Kang,Sisi Chen,Shufang Li,Bo Wang,Xin Liu,Limin Hao,Jike Lu

The polysaccharides of Ganoderma lucidum (GLP) extracted by the methods of ultrasound assisted extraction (UAE) and hot water extraction (HWE) were characterized and the antioxidant activities of GLP UAE and GLP HWE were compared. High performance gel permeation chromatography (HPGPC) showed that the average molecular weight of GLP UAE and GLP HWE were 465.65 kDa and 703.45 kDa, respectively. GLP UAE was composed of mannose , rhamnose , glucose, galactose and arabinose in the molar ratio of 2.58:1.25:11.17:2.5:1, while GLP HWE was composed of the same monosaccharide in the ratio of 3.11:1.11:19.44:2.33:1. GLP HWE showed a relatively higher antioxidant activity than GLP UAE by testing the reducing power, the scavenging ability on 1.1‑diphenyl‑2‑picryl‑hydrazyl (DPPH), hydroxyl radical and cellular protective effect on yeast cells from ultraviolet radiation (UV) damage. GLP UAE and GLP HWE were purified by chromatographic column of DEAE-52 cellulose . GLP UAE , GLP HWE and the purified components could be novel antioxidants for functional food.

The synergy of Box-Behnken designs on the optimization of polysaccharide extraction from mulberry leaves

Shangying Yang,Ying Li,Dongying Jia,Kai Yao,Weijuan Liu

The synergistic effect of hot water extraction (HWE) and enzyme-assisted extraction (EAE) optimized by the Box-Behnken design (BBD) was investigated for a more efficient extraction method for mulberry leaf polysaccharide (MLP) with an improved dried crude MLP yield of 24.04   ±   0.98% under the optimal conditions for HWE (liquid-solid ratio 30, extraction temperature 85   °C and time 60   min) and EAE (pH 6.5, extraction temperature 45   °C and time 50   min), respectively. Furthermore, the static absorption of AB-8 macroporous resin performed the best among decoloring agents for the further crude MLP purification, from which three ethanolic fractions (MLP-40, MLP-60 and MLP-80) were derived with their characterization in carbohydrate, protein and total phenolic contents. The molar ratio of the monosaccharide composition in MLP-80 was 1:14.8:29.3:0.8:0.5:2.2 corresponding to mannose, rhamnose, ribose, glucose, galactose and galacturonic acid, respectively. In addition, the prebiotics effect of MLPs was investigated compared with conventional inulin and oligofructose, in which MLP-80 was finally demonstrated itself as the most promising natural additives in liquid dairy products like yoghourt.

Characterization and antitumor activities of polysaccharides obtained from ginger (Zingiber officinale) by different extraction methods

Deng-wei Liao,Chen Cheng,Jun-ping Liu,Li-yan Zhao,De-Chun Huang,Gui-tang Chen

Three different extraction technologies including hot water extraction (HWE), enzyme assisted extraction (EAE) and ultrasonic cell grinder extraction (UCGE) were employed to extract crude ginger polysaccharides (GPs) under their respective best parameters, then crude GPs were purified by DEAE cellulose-52 and Sephadex G-200 size-exclusion chromatography in that order. Five GPs fractions (HGP, EGP1, EGP2, UGP1, and UGP2, respectively) were obtained. The differences of five GPs in chemical composition, characterization and antitumor activities were further compared. The molecular weights were different in five GPs, varying from 11.81 to 1831.75 kDa. Mannose and glucose as the main monosaccharide and the glycosidic linkage of →4)-α-D-Glc(1→ and -α-Manp-(1→ existed in both five GPs. While EGP2 and UGP1 possessed specific structure of →6)-β-D-Galp-(1→ and UGP1 contained more sulfate group. Moreover, UGP1 exhibited strong inhibitory effect on three tumor cells especially the colon cancer. The inhibition rates of UGP1 on H1975, HCT116 and MCF-7 were 23.339 ± 2.285%, 56.843 ± 2.405% and 21.061 ± 1.920% respectively. The study indicated GPs extracted by UCGE could reserve more active structure and inhibit colon cancer more significantly.

Enzymatic modification of pomelo pectins for shaping the gut microbiota to a healthy pattern

Yuxin Wang,Jiaxin Liu,Yi Long,Hong Yao,Henk A. Schols,Paul de Vos,Weibin Bai,Lingmin Tian

This study investigated the impact of enzymatic hydrolyzation on the characteristics, composition, and in vitro fermentation patterns of pectins extracted from pomelo ( Citrus maxima ) by thermal treatments. Pectins were obtained using subcritical water extraction (SWE) and hot water extraction with chelator assistance, and subsequently, enzymatically modified using pectinase. The investigation covered constituent monosaccharide composition, molecular weight (Mw) distribution, and other structural parameters of the pectins. The primary monosaccharide present in the extracted pomelo pectins was uronic acid (UA), accompanied by relatively low levels of other neutral monosaccharides. The findings indicate that chelating agents coupled with thermal treatment allowed for the successful extraction of low degree of methylation (DM) pectins from pomelo peel. Enzymatic hydrolyzation reduced the DM and Mw of pectin, while maintaining a nearly unchanged monosaccharide mole ratio compared to the parental pectins. In vitro fermentation models were employed to compare the fermentation patterns of pectin before and after enzymatic modification. Enzymatic modification not only heightened the fermentability of pectins but also altered UA utilization patterns, expediting the production of short-chain fatty acids (SCFAs) during in vitro fermentation. Moreover, enzymatic hydrolyzation induced changes in microbial composition, elevating the relative abundance of potential probiotics such as Bacteroides while suppressing potential pathogens like Escherichia-Shigella . In conclusion, the study posits that enzymatic modification of pectins into low Mw and low DM fragments enhances fermentability and beneficially modifies the microbiota of fermented digesta into a potentially healthy pattern.

Effects of thermal treatments on the extraction and in vitro fermentation patterns of pectins from pomelo (Citrus grandis)

Yuxin Wang,Jiaxin Liu,Li chen,Shuang Jin,Can An,Long Chen,Bao Yang,Henk A. Schols,Paul de Vos,Weibin Bai,Lingmin Tian

Pomelo peel, a by-product of fruit processing, is an excellent source of pectin. We aimed to study the effects of thermal treatments on the extraction and in vitro fermentation patterns of pectins from pomelo peel. We used subcritical water extraction and hot water extraction, with or without chelator (citric acid or EDTA-2Na) assistance to obtain the pectins. The pectin fractions were characterized by their constituent monosaccharide composition and molecular weight (Mw) distribution. The degree of methyl esterification of pomelo pectins was determined using FT-IR spectroscopy. We also compared the fermentation patterns of the pectin using an in vitro simulated human gut fermentation model, monitored Mw changes during fermentation by high performance size exclusion chromatography (HPSEC), and analyzed short-chain fatty acid (SCFA) using gas chromatograph. Our results showed that both chelator assistance and subcritical water treatment increased the pectin yield. Chelator-assisted extraction increased the pectin Mw and decreased the degree of methyl esterification (DM) whereas, subcritical water treatment had the opposite effect. Pectin with low rhamnogalacturonan-I (RG-I) and high homogalacturonan (HG) domain can be obtained from the used pomelo peels by citric acid-assisted extraction. Although gut microbiota intensively utilized uronic acid (UA) of pomelo during early fermentation stages, constituent neutral monosaccharides were consumed even faster than UA. Pomelo pectins enhanced the relative abundance of Bacteroides and Prevotella 9 , and stimulated the production of SCFA, particularly acetic acid. Generally, pomelo pectin could be considered a new source for modulating gut microbiota towards a healthy pattern in functional foods.

A glucomannogalactan from Pleurotus geesteranus: Structural characterization, chain conformation and immunological effect

Zhenhua Liang,Zhenhua Yin,Xiaopeng Liu,Changyang Ma,Jinmei Wang,Yan Zhang,Wenyi Kang

A water-soluble glucomannogalactan was isolated from P . geesteranus fruit bodies by hot water extraction and column chromatography method. The structure, chain conformation and immune effect of PGP-1c (20.9 kDa) were investigated comprehensively based on high purity and neutral sugar content (94.7 ± 0.5%). Confirmed by monosaccharide composition , methylation and NMR analysis, PGP-1c was composed of unsubstituted (1,6- α -Gal and 1,6- α -3- O Me-Gal) and monosubstituted (1,2,6- α -Gal and 1,2,6- α -3- O Me-Gal) galactose units in the backbone through α -(1 → 6) glycosidic bonds and the possible branches that a long-branched chain composed of →3)-Glc-(1 → Man and a few fucoses were connected at the O -2 of galactose with a branching degree of 48%. Through the conformational behavior of the molecular chain, it was observed that PGP-1c might have a long-branched or stacked macromolecular network caused by O -CH 3 groups. Moreover, PGP-1c could promote the secretion of NO and cytokines significantly in a dose-dependent manner, which indicated a good immune-enhancing effect.

Fractionation and structural characterization of polysaccharides derived from red grape pomace

Xiao Dong,Cui-Ping Zhu,Guo-Qing Huang,Jun-Xia Xiao

Polysaccharides were recovered from red grape pomace by hot water extraction and then subjected to deproteinization, decolorization, fractionization, and characterization. The extraction process conferred a crude polysaccharide yield of 8.45 % (w/w) and the highest decolorization rate of 63.12 % was obtained by soaking the crude polysaccharide solution with 0.20 g/mL HPD-300 macroporous resin for 3 h. Separation with the anionic DEAE Fast Flow column generated four fractions, which were negatively charged according to zeta potential measurement. All the fractions contained uronic acid and their weight-average molecular weights were within the range 2 × 10 6 –4 × 10 6 Dal. The most abundant fraction GPP-3 was composed of arabinose, xylose, mannose, glucose, and galactose in molar ratio 8.05:1.37:2.85:1.30:7.81 and rhamnose was also present according to FTIR and NMR analysis. AFM observation revealed that aggregation occurred in GPP-3, while the Congo red and β-elimination tests indicated the existence of triple helix and -O- linkage in the fraction. Hence, red grape pomace is a potential source of water-soluble polysaccharides and their bioactivity needs further study.

Characterization and prebiotic potential of polysaccharides from Rosa roxburghii Tratt pomace by ultrasound-assisted extraction

Zheng-Hao Chen,Xiao-hui Yuan,Ting-Ting Tu,Lei Wang,Yu-Heng Mao,You Luo,Shu-Yi Qiu,Ang-Xin Song

In this study, polysaccharides (RRTPs) were extracted from Rosa roxburghii Tratt pomace by hot water or ultrasound (US)-assisted extraction. The structural properties and potential prebiotic functions of RRTPs were investigated. Structural characterization was conducted through HPAEC, HPGPC, GC–MS, FT-IR and SEM. Chemical composition analysis revealed that RRTPs extracted by hot water (RRTP-HW) or US with shorter (RRTP-US-S) or longer duration (RRTP-US-L) all consisted of galacturonic acid , galactose , glucose, arabinose , rhamnose and glucuronic acid in various molar ratio. US extraction caused notable reduction in molecular weight of RRTPs but no significant changes in primary structures. Fecal fermentation showed RRTPs could reshape microbial composition toward a healthier balance, leading to a higher production of beneficial metabolites including total short-chain fatty acids, curcumin , noopept, spermidine , 3-feruloylquinic acid and citrulline . More beneficial shifts in bacterial population were observed in RRTP-HW group, while RRTP-US-S had stronger ability to stimulate bacterial short-chain fatty acids production . Additionally, metabolic profiles with the intervention of RRTP-HW, RRTP-US-S or RRTP-US-L were significantly different from each other. The results suggested RRTPs had potential prebiotic effects which could be modified by power US via molecular weight degradation.
