PHYTOCHROME INTERACTING FACTORS PIF4 and PIF5 promote heat stress induced leaf senescence in Arabidopsis

Li Na,Bo Cunpei,Zhang Yuanyuan,Wang Lei

Leaf senescence can be triggered by multiple abiotic stresses including darkness, nutrient limitation, salinity, and drought. Recently, heatwaves have been occurring more frequently, and they dramatically affect plant growth and development. However, the underlying molecular networks of heat stress-induced leaf senescence remain largely uncharacterized. Here we showed that PHYTOCHROME INTERACTING FACTOR 4 (PIF4) and PIF5 proteins could efficiently promote heat stress-induced leaf senescence in Arabidopsis. Transcriptomic profiling analysis revealed that PIF4 and PIF5 are likely to function through multiple biological processes including hormone signaling pathways. Further, we characterized NAC019, SAG113, and IAA29 as direct transcriptional targets of PIF4 and PIF5. The transcription of NAC019, SAG113, and IAA29 changes significantly in daytime after heat treatment. In addition, we demonstrated that PIF4 and PIF5 proteins were accumulated during the recovery after heat treatment. Moreover, we showed that heat stress-induced leaf senescence is gated by the circadian clock, and plants might be more actively responsive to heat stress-induced senescence during the day. Taken together, our findings proposed important roles for PIF4 and PIF5 in mediating heat stress-induced leaf senescence, which may help to fully illustrate the molecular network of heat stress-induced leaf senescence in higher plants and facilitate the generation of heat stress-tolerant crops.

Cinnamic acid pretreatment enhances heat tolerance of cucumber leaves through modulating antioxidant enzyme activity

Ai-Hua Dai,Yong-Xin Nie,Bin Yu,Qian Li,Ling-Yun Lu,Ji-Gang Bai

To investigate the physiological mechanism of heat stress mitigated by exogenous cinnamic acid (CA), seedlings of a cucumber cultivar ( Cucumis sativus cv. Jinchun no. 4) were pretreated with CA for 2   d and then were exposed to normal (25/18   °C) and elevated (42/38   °C) temperatures for 3   d. 50   μM CA was chosen since it improved growth more than other CA concentrations under heat stress. At 2   d, supply of 50   μM CA elevated the activities of antioxidant enzymes superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), guaiacol peroxidase (GPX, EC 1.11.1.7), glutathione peroxidase (GSH-Px, EC 1.11.1.9), ascorbate peroxidase (APX, EC 1.11.1.11), monodehydroascorbate reductase (MDHAR, EC 1.6.5.4), dehydroascorbate reductase (DHAR, EC 1.8.5.1) and glutathione reductase (GR, EC 1.6.4.2) in cucumber leaves, and it also increased the contents of reduced glutathione (GSH) and ascorbate (AsA). When the CA-pretreated cucumber seedlings have been exposed to heat stress, their antioxidant activities were changed further and were higher than heat treatment alone. Compared to heat treatment, the combination of CA pretreatment and heat also increased relative water content (RWC) of leaves, enhanced levels of proline and soluble sugars, elevated contents of CA and vanillic acid (VA) in leaves, and decreased levels of malonaldehyde (MDA), superoxide radical (O 2 − ) and hydrogen peroxide (H 2 O 2 ), while it resulted in 8.33% of leaf edges being dried up. Meantime, heat had adverse effects on plant growth, RWC, CA and VA contents in leaves, and it as well increased the levels of MDA, O 2 − and H 2 O 2 and made the dried leaf edges to be 79.17%. We conclude that pretreatment with 50   μM CA elevates CA and VA contents in leaves, enhances antioxidant activities under heat stress and decreases lipid peroxidation to some extent, accompanying with increasing proline and soluble sugar levels in heat-stressed leaves, thus the pretreatment enhances heat tolerance of cucumber seedlings.

Transcriptomic analysis of short-term heat stress response in Pinellia ternata provided novel insights into the improved thermotolerance by spermidine and melatonin

Guangjing Ma,Meide Zhang,Jilei Xu,Wuxian Zhou,Liwen Cao

Heat stress has been a major environmental factor limiting the growth and development of Pinellia ternata which is an important Chinese traditional medicine. It has been reported that spermidine (SPD) and melatonin (MLT) play pivotal roles in modulating heat stress response (HSR). However, the roles of SPD and MLT in HSR of P. ternata , and the potential mechanism is still unknown. Here, exogenous SPD and MLT treatments alleviated heat-induced damages in P. ternata , which was supported by the increased chlorophyll content, OJIP curve, and relative water content, and the decreased malondialdehyde and electrolyte leakage. Then, RNA sequencing between CK (control) and Heat (1 h of heat treatment) was conducted to analyze how genes were in response to short-term heat stress in P. ternata . A total of 14,243 (7870 up- and 6373 down-regulated) unigenes were differentially expressed after 1 h of heat treatment. Bioinformatics analysis revealed heat-responsive genes mainly included heat shock proteins ( HSPs ), ribosomal proteins, ROS-scavenging enzymes, genes involved in calcium signaling, hormone signaling transduction, photosynthesis, pathogen resistance, and transcription factors such as heat stress transcription factors ( HSFs ), NACs , WRKYs , and bZIPs . Among them, PtABI5 , PtNAC042 , PtZIP17 , PtSOD1 , PtHSF30 , PtHSFB2b , PtERF095 , PtWRKY75 , PtGST1 , PtHSP23.2 , PtHSP70 , and PtLHC1 were significantly regulated by SPD or MLT treatment with same or different trends under heat stress condition, indicating that exogenous application of MLT and SPD might enhance heat tolerance in P. ternata through regulating these genes but may with different regulatory patterns. These findings contributed to the identification of potential genes involved in short-term HSR and the improved thermotolerance by MLT and SPD in P. ternata , which provided important clues for improving thermotolerance of P. ternata .

Molecular characterization of a catalase gene from the green peach aphid (Myzus persicae)

Mao-Ye Li,Yun Wang,Xiao Lei,Chuan-Tao Xu,Dong-Dong Wang,Su Liu,Shi-Guang Li

The green peach aphid, Myzus persicae (Sulzer), is a serious agricultural pest with a worldwide distribution. Catalase (CAT), which is encoded by the catalase ( Cat ) gene, is an extremely important antioxidant enzyme that plays a pivotal role in protecting cells against the toxic effects of hydrogen peroxide. The Cat gene has not been characterized in M. persicae ; therefore, this study describes the identification of the Cat ( MpCat ) gene from M. persicae . MpCat contains an open reading frame of 1515 bp and encodes a MpCAT protein consisting of 504 amino-acid residues. MpCAT possesses features typical of other insect catalases, including 7 conserved amino acids involved in binding heme and 15 involved in binding nicotinamide adenine dinucleotide phosphate. Phylogenetic analysis showed that MpCAT was closely related to orthologs from other aphid species. MpCat consisted of nine exons and eight introns, and the number and insertion sites of introns are consistent with those of Cat genes from Acyrthosiphon pisum (Harris) and Aphis gossypii Glover. The mRNA transcripts of MpCat were detected at all tested developmental stages, with the highest mRNA level in alate adults. The expression of MpCat was significantly upregulated when M. persicae was exposed to low and high temperatures, ultraviolet radiation, Beauveria bassiana , and permethrin. The transcription of MpCat and the activity of catalase were suppressed by RNA interference, and knockdown of MpCat significantly reduced the survival rate in M. persicae under heat stress. The results provide valuable information for further study on the physiological functions of MpCat . Research Highlights A catalase gene ( MpCat ) was characterized in Myzus persicae . Transcription of MpCat was responsive to various environmental stresses. Knockdown of MpCat increased mortality in M. persicae under heat stress.

Alleviating Coral Thermal Stress via Inoculation with Quorum Quenching Bacteria

Yang Qingsong,Yang Bing,Yang Bin,Zhang Wenqian,Tang Xiaoyu,Sun Huiming,Zhang Yanying,Li Jie,Ling Juan,Dong Junde

In the background of global warming, coral bleaching induced by elevated seawater temperature is the primary cause of coral reef degradation. Coral microbiome engineering using the beneficial microorganisms for corals (BMCs) has become a hot spot in the field of coral reef conservation and restoration. Investigating the potential of alleviating thermal stress by quorum quenching (QQ) bacteria may provide more tools for coral microbial engineering remediation. In this study, QQ bacteria strain Pseudoalteromonas piscicida SCSIO 43740 was screened among 75 coral-derived bacterial strains, and its quorum sensing inhibitor (QSI) compound was isolated and identified as 2,4-di-tert-butylphenol (2,4-DTBP). Then, the thermal stress alleviating potential of QQ bacteria on coral Pocillopora damicornis was tested by a 30-day controlled experiment with three different treatments: control group (Con: 29 °C), high temperature group (HT: 31 °C), and the group of high temperature with QQ bacteria inoculation (HTQQ: 31 °C + QQ bacteria). The results showed that QQ bacteria SCSIO 43740 inoculation can significantly mitigate the loss of symbiotic algae and impairment of photosynthesis efficiency of coral P. damicornis under thermal stress. Significant difference in superoxide dismutase (SOD) and catalase (CAT) enzyme activities between HT and HTQQ was not observed. In addition, QQ bacteria inoculation suppressed the coral microbial community beta-dispersion and improved the stability of microbial co-occurrence network under thermal stress. It was suggested that QQ bacteria inoculation can alleviate coral thermal stress via reshaping microbial interaction and maintain community stability of coral microbiome. This study provided new evidence for the probiotic function of QQ bacteria in corals, which shedding light on the development of new microbiological tools for coral reef conservation.

Naturally occurring substitution of an amino acid in a plant virus gene-silencing suppressor enhances viral adaptation to increasing thermal stress

Lina Cai,Mingqing Dang,Yawen Yang,Ruoxin Mei,Fan Li,Xiaorong Tao,Peter Palukaitis,Randy Beckett,W. Allen Miller,Stewart M. Gray,Yi Xu

Cereal yellow dwarf virus (CYDV-RPV) encodes a P0 protein that functions as a viral suppressor of RNA silencing (VSR). The strength of silencing suppression is highly variable among CYDV-RPV isolates. In this study, comparison of the P0 sequences of CYDV-RPV isolates and mutational analysis identified a single C-terminal amino acid that influenced P0 RNA-silencing suppressor activity. A serine at position 247 was associated with strong suppressor activity, whereas a proline at position 247 was associated with weak suppressor activity. Amino acid changes at position 247 did not affect the interaction of P0 with SKP1 proteins from Hordeum vulgare (barley) or Nicotiana benthamiana . Subsequent studies found P0 proteins containing a P247 residue were less stable than the P0 proteins containing an S247 residue. Higher temperatures contributed to the lower stability and in planta and the P247 P0 proteins were subject to degradation via the autophagy-mediated pathway. A P247S amino acid residue substitution in P0 increased CYDV-RPV replication after expression in agroinfiltrated plant leaves and increased viral pathogenicity of P0 generated from the heterologous Potato virus X expression vector system. Moreover, an S247 CYDV-RPV could outcompete the P247 CYDV-RPV in a mixed infection in natural host at higher temperature. These traits contributed to increased transmission by aphid vectors and could play a significant role in virus competition in warming climates. Our findings underscore the capacity of a plant RNA virus to adapt to climate warming through minor genetic changes in gene-silencing suppressor, resulting in the potential for disease persistence and prevalence. Climate change and extreme weather events pose an increased risk for crops to become infected with pathogens, which could severely affect global food security. However, the studies about the effects of climate change on plant virus diseases are still obscure. Yellow dwarf disease is one of the most important diseases of small grain cereals worldwide, which is caused by viruses belonging to the genera Luteovirus and Polerovirus . The gene encoding the P0 protein of poleroviruses has been reported to be the most rapidly evolving ORF of the Polerovirus genus. P0 functions as a viral suppressor of RNA silencing (VSR) and the strength of silencing suppression of P0 is highly variable among CYDV-RPV isolates. However, little is known about the molecular mechanism that determine P0 suppressor activity. Here, we identify a naturally occurring, single-amino-acid substitution in the CYDV-RPV P0 protein that enhances its protein stability to increasing thermal stress. This is the first study to identify a naturally occurring amino acid substitution in a plant virus-encoded protein that determines its adaption to increasing thermal stress. As climate change and extreme weather events occur frequently, we expect that our research can supply insight into how a plant RNA virus evolves to adapt climate change.

Heat stress enhanced perylenequinones biosynthesis of Shiraia sp. Slf14(w) through nitric oxide formation

Xu Chenglong,Lin Wenxi,Chen Yunni,Gao Boliang,Zhang Zhibin,Zhu Du

Perylenequinones (PQs) are a class of natural polyketides used as photodynamic therapeutics. Heat stress (HS) is an important environmental factor affecting secondary metabolism of fungi. This study investigated the effects of HS treatment on PQs biosynthesis of Shiraia sp. Slf14(w) and the underlying molecular mechanism. After the optimization of HS treatment conditions, the total PQs amount reached 577 ± 34.56 mg/L, which was 20.89-fold improvement over the control. Also, HS treatment stimulated the formation of intracellular nitric oxide (NO). Genome-wide analysis of Shiraia sp. Slf14(w) revealed iNOSL and cNOSL encoding inducible and constitutive NOS-like proteins (iNOSL and cNOSL), respectively. Cloned iNOSL in Escherichia coli BL21 showed higher nitric oxide synthase (NOS) activity than cNOSL, and the expression level of iNOSL under HS treatment was observably higher than that of cNOSL , suggesting that iNOSL is more responsible for NO production in the HS-treated strain Slf14(w) and may play an important role in regulating PQs biosynthesis. Moreover, the putative biosynthetic gene clusters for PQs and genes encoding iNOSL and nitrate reductase (NR) in the HS-treated strain Slf14(w) were obviously upregulated. PQs biosynthesis and efflux stimulated by HS treatment were significantly inhibited upon the addition of NO scavenger, NOS inhibitor, and NR inhibitor, indicating that HS-induced NO, as a signaling molecule, triggered promoted PQs biosynthesis and efflux. Our results provide an effective strategy for PQs production and contribute to the understanding of heat shock signal transduction studies of other fungi. Key points • PQs titer of Shiraia sp. Slf14(w) was significantly enhanced by HS treatment . • HS-induced NO was first reported to participate in PQs biosynthetic regulation . • Novel inducible and constitutive NOS-like proteins (iNOSL and cNOSL) were obtained and their NOS activities were determined .

Multi-omics reveals phenol-based coordinated defense of Sparganium stoloniferum rhizoma

Mengru Sang,Qinan Liu,Dishuai Li,Jingjie Dang,Shilin Dai,Chanchan Liu,Qinan Wu

Sparganium stoloniferum is a widely distributed aquatic medicinal plant that exhibits exceptional tolerance to environmental stress. Interestingly, the phenols in S. stoloniferum which have medicinal properties, are defensive compounds. Studies integrating transcriptome, proteome, and metabolome analyses have highlighted the phenylpropanoid and flavonoid biosynthesis pathways, which are related to the defense mechanisms of S. stoloniferum . During the growth process, S. stoloniferum rhizoma (SL) accumulates lignin, forming a physical defense, and phenylpropanoids and flavonoids derived during the lignin biosynthesis process provide chemical defense. In this study, a map of the molecular processes involved in the coordinated defense of S. stoloniferum was generated. The expression patterns of genes involved in the coordinated defense of SL were consistent with the omics results. The cDNA of the phenylalanine ammonia-lyase ( PAL ) gene from S. stoloniferum ( Ss PAL) was cloned. The recombinant protein Ss PAL showed good affinity and catalytic activity for L-phenylalanine. Heat stress experiments indicated that genes related to phenol biosynthesis were upregulated when S. stoloniferum responded to stress. Therefore, phenols played a crucial defense role when S. stoloniferum was at an early developmental stage or responded to stress in a short time. In contrast, the physical defense mainly relied on lignin formation, which was the main defense mechanism of SL in the late growth stage. Based on these results, we discuss the causes of phenolic pharmacological substance formation in medicinal aquatic plants. These results provide new insights into the defense strategies of aquatic plants.

Unraveling the metabolic profile regulation of camellia oilseeds under insect and heat stress: Insights into functional effects and mechanistic basis

Qingyang Li,Wei Zhu,Shiman Sun,Maokai Cui,Wei Zhang,Jinping Shu,Runhong Mo,Fubin Tang,Yirong Guo,Yihua Liu

There is very little information on the impacts of pre/post-harvest stresses on oilseeds. Individual and combined insect (pre-harvest) and heat stress (post-harvest) impacts on the metabolic profile of camellia oilseeds (COs) were investigated using a combination of widely-targeted metabolomics and network pharmacology. A total of 1875 metabolites were identified. In response to individual and combined stresses, 169 (insect),149 (heat), and 21 (insect + heat) metabolites were screened as differential metabolic markers (DEMs), Terpenoids, phenolic acids, and flavonoids are the most impacted metabolite species, accounting for almost 49% of total DEMs. Then network pharmacological analysis identifies 98 key active ingredients (AIs) in CO. A single stress may induce CO to impede cardiovascular system function, but the combined stress induced AI-promoting effects of CO in the urinary system. The individual and combined perturbed biological mechanisms were related to the flavonoid biosynthesis and the biosynthesis of various plant secondary metabolites pathway, respectively.

Integrated lipidomic and transcriptomic analysis reveals triacylglycerol accumulation in castor bean seedlings under heat stress

Yanyu Zhang,Yelan Li,Bing Han,Aizhong Liu,Wei Xu

Heat stress is one of the most challenging environmental factors for plants, which have evolved various mechanisms to cope with it. However, the role of lipid remodeling caused by heat stress is not yet fully understood. Here, we integrated UPLC-QTOF/MS-based lipidomic and transcriptomic analyzes to uncover the molecular basis of lipid remodeling under heat stress in castor bean ( Ricinus communis ), an important non-edible oil crop . We detected 297 lipid compounds in castor bean seedlings, of which 54 displayed altered abundances under heat stress. In particular, polyunsaturated triacylglycerols (TAGs) (e.g., TAG54:6, TAG54:7, TAG54:8, and TAG54:9) and diacylglycerols (DAGs) (DAG36:6) significantly increased, while polyunsaturated monogalactosyldiacylglycerols (MGDGs) (MGDG34:3) markedly decreased. When heat stress was subsequently relieved, these lipid molecules recovered to their normal levels. We found that heat-induced TAGs primarily accumulate in the cytosol. Transcriptomic analysis indicated that polyunsaturated fatty acids (FAs) used for TAGs are not derived from de novo synthesis, but likely from the lipids remodeling via lipase activity, and are subsequently esterified into TAGs via diacylglycerol acyltransferase (DGAT). Our results suggest that TAGs may act as intermediates in lipid turnover and provide one mechanism by which plants respond to heat stress, broadening our understanding of how lipid remodeling functions in plant adaptation to heat stress.

Lactic acid accumulation under heat stress related to accelerated glycolysis and mitochondrial dysfunction inhibits the mycelial growth of Pleurotus ostreatus

Yan Zhiyu,Wu Xiangli,Zhao Mengran,Zhang Jinxia

High temperature is a major threat to Pleurotus ostreatus cultivation. In this study, a potential mechanism by which P. ostreatus mycelia growth is inhibited under heat stress was explored. Lactate, as a microbial fermentation product, was found unexpectedly in the mycelia of P. ostreatus under heat stress, and the time-dependent accumulation and corresponding inhibitory effect of lactate on mycelial growth was further confirmed. The addition of a glycolysis inhibitor, 2 -deoxy- d -glucose (2DG), reduced the lactate content in mycelia and slightly restored mycelial growth under high-temperature conditions, which indicated the accumulation of lactate can be inhibited by glycolysis inhibition. Further data revealed mitochondrial dysfunction under high-temperature conditions, with evidence of decreased oxygen consumption and adenosine triphosphate (ATP) synthesis and increased reactive oxygen species (ROS). The removal of ROS with ascorbic acid decreased the lactate content, and mycelial growth recovered to a certain extent, indicating lactate accumulation could be affected by the mitochondrial ROS. Moreover, metabolic data showed that glycolysis and the tricarboxylic acid cycle were enhanced. This study reported the accumulation of lactate in P. ostreatus mycelia under heat stress and the inhibitory effect of lactate on the growth of mycelia, which might provide further insights into the stress response mechanism of edible fungi. Key Points • Lactate can accumulate in Pleurotus ostreatus mycelia under heat stress and inhibit its growth. • The accumulation of lactate may be due to the acceleration of glycolysis and the dysfunction of mitochondria of P. ostreatus mycelia under high-temperature stress. • The glycolysis and tricarboxylic acid cycle of P. ostreatus mycelia were accelerated under high-temperature stress.
