Identification of Rhizospheric Actinomycete Streptomyces lavendulae SPS-33 and the Inhibitory Effect of its Volatile Organic Compounds against Ceratocystis fimbriata in Postharvest Sweet Potato (Ipomoea batatas (L.) Lam.)

Xuewei Li,Beibei Li,Shurui Cai,Yu Zhang,Mingjie Xu,Chunmei Zhang,Bo Yuan,Ke Xing,Sheng Qin

Black spot disease, which is caused by the pathogenic fungalCeratocystis fimbriata, seriously affects the production of sweet potato and its quality during postharvest storage. In this study, the preliminary identification of the rhizosphere actinomycete strain SPS-33, and its antifungal activity of volatilesin vitroandin vivowas investigated. Based on morphological identification and phylogenetic analysis of the 16S rRNA gene sequence, strain SPS-33 was identified asStreptomyces lavendulae.Volatile organic compounds (VOCs) emitted by SPS-33 inhibited mycelial growth and sporulation ofC. fimbriatain vitroand also induced a series of observable hyphae morphological changes. In anin vivopathogenicity assay, exposure to SPS-33 significantly decreased the lesion diameter and water loss rate in sweet potato tuberous roots (TRs) inoculated withC. fimbriata. It increased the antioxidant enzymes’ activities of peroxidase, catalase, and superoxide dismutase as well as decreased malondialdehyde and increased total soluble sugar. In the VOC profile of SPS-33 detected by a headspace solid-phase micro extraction (HS-SPME) and gas chromatography-mass spectrometry (GC-MS), heptadecane, tetradecane, and 3-methyl-1-butanol were the most abundant compounds. 2-Methyl-1-butanol, 3-methyl-1-butanol, pyridine, and phenylethyl alcohol showed strong antifungal effects againstC. fimbriata. These findings suggest that VOCs fromS. lavendulaeSPS-33 have the potential for pathogenC. fimbriatacontrol in sweet potato postharvest storage by fumigant action. Keywords:Ipomoea batatas(L.) Using essential oils from Citrus paradisi as a fumigant for Solenopsis invicta workers and evaluating the oils’ effect on worker behavior

Zhang Ning,Liao Yihong,Xie Lianjie,Zhang Zhixiang,Hu Wei

The red imported fire ant is one of the world’s most devastating invasive species, adversely affecting humans, wildlife, crops, and livestock. To control infestations, chemical pesticides are deployed extensively around the world. However, their extensive use has led to negative effects on the environment and human health. Essential oils, which are safe and ecofriendly, can potentially be used as alternatives to chemical pesticides. In this study, grapefruit essential oils were used as fumigant agents to control red imported fire ants. The crude grapefruit oil (GO1) contained 28 compounds, and the concentrated grapefruit oil (GO2), which was refined from GO1 by vacuum distillation, contained 20 compounds. D-Limonene was the dominant constituent in both GO1 (70.1%) and GO2 (73.96%), and other important constituents included β-pinene, α-pinene, β-phellandrene, octanal, d-carvone, α-terpineol, and linalool. Both the essential oils and their individual constituents (α-pinene, α-terpineol, β-phellandrene, octanal, and d-carvone) showed strong lethal fumigant effects against workers. Workers were more susceptible to GO2 than GO1, and octanal was more toxic to workers as compared with the other four constituents. When antennas of workers were treated with the two oils or the five constituents, their walking and gripping abilities were significantly suppressed, and there was an obvious bending or breaking phenomenon on the sensilla of the antennas. Fumigant activity by grapefruit essential oils and their main compounds were associated with their effects on the walking and gripping behavior of workers, and this confirmed that grapefruit essential oil is a promising, ecofriendly, and safe fumigant for the control of red imported fire ants.

Stable and Flexible CuInS2/ZnS:Al-TiO2 Film for Solar-Light-Driven Photodegradation of Soil Fumigant

Lili Yan,Zhichun Li,Mingxing Sun,Guoqing Shen,Liang Li

Semiconductor quantum dots (QDs) are suitable light absorbers for photocatalysis because of their unique properties. However, QDs generally suffer from poor photochemical stability against air, limiting their applications in photocatalysis. In this study, a stable solar-light-driven QDs-containing photocatalytic film was developed to facilitate photocatalytic degradation of the soil fumigant 1,3-dichloropropene (1,3-D). Highly stable CuInS2/ZnS:Al core/shell QDs (CIS/ZnS:Al QDs) were synthesized by doping Al into the ZnS shell and controlling ZnS:Al shell thickness; the CIS/ZnS:Al QDs were subsequently combined with TiO2 to form a CIS/ZnS:Al-TiO2 photocatalyst. The optimized ZnS:Al shell thickness for 1,3-D photodegradation was approximately 1.3 nm, which guaranteed and balanced the good photocatalytic activity and stability of the CIS/ZnS:Al-TiO2 photocatalyst. The photodegradation efficiency of 1,3-D can be maintained up to more than 80% after five cycles during recycling experiment. When CIS/ZnS:Al-TiO2 was deposited as photocatalytic film on a flexible polyethylene terephthalate substrate, over 99% of cis-1,3-D and 98% of trans-1,3-D were depleted as they passed through the film during 15 h of irradiation under natural solar light. This study demonstrated that the stable CIS/ZnS:Al-TiO2 photocatalyst both in powder and film form is a promising agent for photodegradation and emission reduction of soil fumigants.

Discovering potential fumigants against Tribolium castaneum from essential oils using GC-MS and chemometric approaches

Yan Yan Lin,Min Xing Zhang,Miao Sun,Lu Sheng Wan,You Zhou,Jing Sheng Chen

Tribolium castaneum is recognized as one of the most significant global pests affecting stored grains. Due to their general safety, essential oil (EO)-based biopesticides have garnered increasing interest worldwide. To provide a more rapid and objective method for the identification of individual components with potential fumigants from EOs, a GC-MS-based metabolomics strategy was applied in this study. Five EOs— Eucalyptus globulus , Dendranthema indicum , Cinnamomum cassia , Pinus massoniana , and Cinnamomum camphora —demonstrated remarkable toxicity, achieving corrected mortality rates exceeding 90% against Tribolium castaneum . A total of 41 potential fumigants were screened, with 21 of these identified potential fumigants corroborated by existing literature or preliminarily bioassay experiments. Key components included (−)-myrtenol, β-pinene, p -cymene, γ-terpinene, sabinene, β-myrcene, β-ocimene, and α-terpinene. These findings not only highlight promising EO-based fumigants against Tribolium castaneum but also provide lead compounds for future structural optimization. Additionally, this discovery strategy serves as a valuable reference for similar studies in natural products and biopesticides.

Nematicidal Activity of trans-2-Hexenal against Southern Root-Knot Nematode (Meloidogyne incognita) on Tomato Plants

Hongbao Lu,Shuangyu Xu,Wenjuan Zhang,Chunmei Xu,Beixing Li,Daxia Zhang,Wei Mu,Feng Liu

Botanical nematicides have recently received increasing interest because of the high risks of some traditional nematicides to human health and the environment. This study evaluated the nematicidal activity of a plant volatile, trans-2-hexenal, against Meloidogyne incognita. This compound exhibited higher activity in a fumigation experiment than in the aqueous phase in vitro. Both in pot tests and in field trials, trans-2-hexenal showed significant efficacy against M. incognita while maintaining excellent plant growth, especially at doses of 1000 and 500 L ha–1, which were superior to that of abamectin at 180 g ha–1 via hole application treatment but not significantly different from fumigation with 400 kg ha–1 of dazomet. Furthermore, plants treated with 500 L ha–1trans-2-hexenal had fruit yields 20.2 and 45% greater than the control group. On this basis, trans-2-hexenal may be a potential alternative fumigation agent for controlling M. incognita on tomato crops.

Biological Activity of trans-2-Hexenal Against the Storage Insect Pest Tribolium castaneum (Coleoptera: Tenebrionidae) and Mycotoxigenic Storage Fungi

Cui Kaidi,He Leiming,Cui Guangrui,Zhang Tao,Chen Yue,Zhang Tao,Mu Wei,Liu Feng

Grain commodities in postharvest storage often deteriorate because of fungal and insect attacks. With the green consumption requirements of consumers, ecofriendly and safe pesticides are needed for grain storage. The current study investigated the efficacy of the plant volatile compound trans-2-hexenal against the storage insect pest Tribolium castaneum (Herbst) and three commonly occurring storage fungi, viz., Fusarium graminearum, Aspergillus flavus, and Aspergillus niger, to recommend its application as a botanical fumigant for grain commodities. trans-2-Hexenal weakly repels T. castaneum but has favorable insecticidal activity against multiple developmental stages of T. castaneum, ranging in sensitivity as follows: eggs (LC50 = 14.3 µl/l) > adults (31.6 µl/l) > young larvae (42.1 µl/l) > mature larvae (64.5 µl/l) > pupae (70.5 µl/l). Moreover, trans-2-hexenal caused a high malformation rate and high mortality in adults developed from fumigated pupae. In a 7-d grain, trans-2-hexenal at 0.8 µl/ml provided an appreciable efficacy (81.3%), and concentrations ≥ 0.1 µl/ml completely inhibited the offspring of T. castaneum. trans-2-Hexenal was nonphytotoxic to the seed germination and seedling growth of wheat seeds. Furthermore, trans-2-hexenal completely inhibited the growth of A. flavus, F. graminearum, and A. niger at 5, 10, and 10 µl/l, respectively. The favorable biological activity of trans-2-hexenal against T. castaneum and three frequently occurring mycotoxigenic storage fungi indicated the potential of trans-2-hexenal for simultaneously controlling pests and pathogens, which could reduce its application frequency in grains and decrease pesticide resistance risks.

Coupled use of Fe-impregnated biochar and urea-hydrogen peroxide to simultaneously reduce soil–air emissions of fumigant and improve crop growth

Jiaolong Qin,Daniel J. Ashworth,Scott R. Yates,Guoqing Shen

Reducing the emissions of soil fumigants such as 1,3-dichloropropene (1,3-D) is essential to protecting air quality. Although biochar is useful in reducing such emissions, biochar-adsorbed fumigants may desorb and cause secondary air pollution. This study investigated the degradation of 1,3-D on iron (Fe)-impregnated biochar (FBC) amended with urea-hydrogen peroxide (UHP). The results indicated the degradation rate of trans -1,3-D on FBC-UHP was 54-fold higher than that on pristine biochar (PBC). Electron paramagnetic resonance (EPR) combined with other characterization methods revealed that the presence of semiquinone-type radicals in FBC effectively accelerated the Fe(III)/Fe(II) cycleto maintain enough Fe(IIII) for UHP activation and ·OH generation. ·OH, rather than ·O 2 - , was the dominant active oxidant. Soil column tests showed that application of FBC to the soil surface reduced cumulative 1,3-D emissions from 34.80 % (bare soil) to 0.81%. After the column experiment, the mixing of the FBC with UHP resulted in the residual cis -isomers decreasing from 32.5% to 10.5%. Greenhouse bioassays showed that mixing post-1,3-D degradation FBC-UHP with soil significantly promoted lettuce growth relative to PBC. The findings of this study provide a new approach for biochar application, especially for the emission reduction of hazardous volatile organic compounds from soil.

Carvacrol/β-cyclodextrin inclusion complex as a fumigant to control decay caused by Penicillium digitatum on Shatangju mandarin slices

Yumeng Liu,Luo Weng,Ying Lin,Daijie Lin,Linsheng Xie,Tian Zhong

Preservation and microorganism control of fresh-cut fruit pose a persistent challenge in the food industry. To address this issue, we prepared a β-cyclodextrin (β-CD) inclusion complex containing carvacrol using a coprecipitation method and employed it for the non-contact fumigation of fresh-cut Shatangju mandarin slices. This biodegradable and safe preservative offers an effective means to combat spoilage and ensure product quality. We confirmed the formation of the encapsulated structure of the inclusion complex through various characterization methods, including scanning electron microscopy (SEM), Fourier transform-infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetric analysis (TGA), and differential thermal analysis (DTA). We also demonstrated the inhibitory effect of this preservative on Penicillium digitatum and its associated spoilage both in vitro and in vivo . The incidence and severity were significant lower in the inclusion complex-treated group (75.0% and 46.7%, respectively) compared to the group treated with pure carvacrol (100% and 69.2%, respectively). In addition, fruit freshness parameters and sensory evaluation showed that the inclusion complex treatment effectively maintained the overall quality of the fruit and achieved the highest consumer acceptance.

Dian Huang,Chen Yu,Zongze Shao,Minmin Cai,Guangyu Li,Longyu Zheng,Ziniu Yu,Jibin Zhang

Root-knot nematode diseases cause severe yield and economic losses each year in global agricultural production.Virgibacillus dokdonensisMCCC 1A00493, a deep-sea bacterium, shows a significant nematicidal activity againstMeloidogyne incognitain vitro. However, information about the active substances ofV. dokdonensisMCCC 1A00493 is limited. In this study, volatile organic compounds (VOCs) fromV. dokdonensisMCCC 1A00493 were isolated and analyzed through solid-phase microextraction and gas chromatography–mass spectrometry. Four VOCs, namely, acetaldehyde, dimethyl disulfide, ethylbenzene, and 2-butanone, were identified, and their nematicidal activities were evaluated. The four VOCs had a variety of active modes onM. incognitajuveniles. Acetaldehyde had direct contact killing, fumigation, and attraction activities; dimethyl disulfide had direct contact killing and attraction activities; ethylbenzene had an attraction activity; and 2-butanone had a repellent activity. Only acetaldehyde had a fumigant activity to inhibit egg hatching. Combining this fumigant activity against eggs and juveniles could be an effective strategy to control the different developmental stages ofM. incognita. The combination of direct contact and attraction activities could also establish trapping and killing strategies against root-knot nematodes. Considering all nematicidal modes or strategies, we could useV. dokdonensisMCCC 1A00493 to set up an integrated strategy to control root-knot nematodes. Keywords:virgibacillus dokdonensis;Meloidogyne incognita;volatile organic compound;fumigant;attraction;repellent;integrated strategy
