Identification of Insecticidal Constituents from the Essential Oil of Valeriana jatamansi Jones against Liposcelis bostrychophila Badonnel

Xin Chao Liu,Ligang Zhou,Zhi Long Liu

The aim of this research was to determine chemical composition and insecticidal activities of the essential oil of Valeriana jatamansi Jones roots against booklice, Liposcelis bostrychophila Badonnel, and to isolate insecticidal constituents from the oil. Essential oil of V. jatamansi was obtained by hydrodistillation and analyzed by GC-MS. A total of 27 components in the essential oil were identified. The major compounds were patchoulol (24.3%), α-bulnesene (13.8%), isovaleric acid (12.9%), α-guaiene (8.7%), and 3-methylvaleric acid (8.4%). Based on bioactivity-guided fractionation, isovaleric acid, 3-methylvaleric acid, and patchoulol were isolated and identified as the active constituents. The essential oil exhibited contact toxicity against L. bostrychophila with an LC50 value of 236.4  g/cm2. Patchoulol (LC50 = 61.35  g/cm2) exhibited stronger acute toxicity than 3-methylvaleric acid (LC50 = 210.69  g/cm2) against the booklice. The essential oil also possessed fumigant toxicity against L. bostrychophila with an LC50 value of 6.0 mg/L, while 3-methylvaleric acid and isovaleric acid had LC50 values of 5.53 mg/L and 5.67 mg/L against the booklice, respectively. The results indicated that the essential oil and its constituent compounds have potential to develop into natural insecticides or fumigants for control of insects in stored grains.

Analysis of the Essential Oil of Illicium henryi Diels Root Bark and Its Insecticidal Activity against Liposcelis bostrychophila Badonnel

Xin Chao Liu,Zhi Long Liu

Water-distilled essential oil from Illicium henryi (Illiciaceae) root bark was analyzed by gas chromatography–mass spectrometry. Thirty-four compounds, accounting for 97.86% of the total oil, were identified. The main components of the essential oil of I. henryi root bark were safrole (46.12%), myristicin (20.39%), and 1,8-cineole (6.17%), followed by a-cadinol (3.784%) and linalool (3.22%). The essential oil had higher levels of phenylpropanoids (66.89%) than of monoterpenoids (14.83%) and sesquiternoids (16.14%). Three constituents were isolated from the oil based on bioactivity fractionation. The essential oil possessed fumigant toxicity against booklice ( Liposcelis bostrychophila ), with a 50% lethal concentration (LC 50 ) of 380.39 mg/liter of air, while the two isolated constituents myristicin and safrole had LC 50 s of 121.95 and 322.54 μg/liter, respectively. Another constituent, 1,8-cineole, showed weaker toxicity, with an LC 50 of 1,120.43 μg/liter. The essential oil also exhibited contact toxicity against L. bostrychophila, with an LC 50 of 96.83 μg/cm 2 . Myristicin (LC 50 , 18.74 μg/cm 2 ) and safrole (LC 50 , 69.28 μg/cm 2 ) exhibited stronger acute toxicity than 1,8-cineole (LC 50 , 1,049.41 μg/cm 2 ) against the booklice. The results indicated that the essential oil and its constituent compounds have potential for development into natural insecticides for control of psocids in stored grains.

Bioactivity of Artemisia vulgaris Essential Oil and Two of Its Constituents Against the Red Flour Beetle (Coleoptera: Tenebrionidae)

Gao Shanshan,Guo Mengyi,Yin Yang,Zhang Xinyi,Zhang Yonglei,Zhang Kunpeng

Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) is one of the most destructive pests of stored products. Although there have been studies on the potential use of essential oils from plants in the Artemisia genus as insecticides, no comprehensive bioactivity data are available on the efficacy of Artemisia vulgaris L. (Asterales: Asteraceae) essential oil and its chemical constituents on stored-product pests. Therefore, in this study, the bioactivity of A. vulgaris essential oil and its chemical constituents, eugenol and terpinen-4-ol, against T. castaneum were determined by contact, fumigant, and repellent bioassays. Analysis of contact and fumigant bioassays showed that A. vulgaris essential oil, eugenol, and terpinen-4-ol have contact and fumigant toxicities against T. castaneum , of which terpinen-4-ol has a strong killing effect on larvae and adults, suggesting that terpinen-4-ol may be the main active component of A. vulgaris essential oil in contact and fumigant effects. Additionally, A. vulgaris essential oil and eugenol have higher repellent activity against T. castaneum larvae and adults, whereas the repellent activity of terpinen-4-ol is low, indicating that the main component of A. vulgaris essential oil in repellence may be eugenol. These results further provide relevant theoretical basis for the development of plant essential oil pesticides.

Trans-anethole is a potent toxic fumigant that partially inhibits rusty grain beetle (Cryptolestes ferrugineus) acetylcholinesterase activity

Zhunjing Wang,Yongjian Xie,Maidinai Sabier,Tao Zhang,Jianyu Deng,Xuming Song,Zhihong Liao,Qingguang Li,Shengxiang Yang,Yang Cao,Xingquan Liu,Guoxin Zhou

The safe storage and protection of grain includes the exploitation of plant-based fumigants as a green control technology. This study first determined the fumigant toxicity of 12 traditional Chinese medicinal plants against adults of the rusty grain beetle, Cryptolestes ferrugineus (Stephens), one of the most ubiquitous and pestilent grain storage pests worldwide. Exposure to volatiles released from Illicium verum Hook. f. powder was found to be by far the most lethal. Illicium verum power and its volatiles eluent were then confirmed to be highly effective in killing both larvae and adults of C. ferrugineus . Star anise volatiles also showed good biological activity against Lasioderma serricorne (Fabricius) and Sitophilus zeamais Motschulsky, but not Tribolium castaneum Herbst. No significant differences were found in mortality of the adults of the rusty grain beetle when exposed at different temperatures ranging from 15−35°C. Gas chromatography-mass spectrometry (GC–MS) analysis showed that the main components of the eluent of star anise powder were trans -anethole (62.60%), estragole (23.70%), linalool (3.66%) and d -limonene (2.15%). In the fumigation test using synthetic standards of these four chemicals, only trans -anethole showed effective biological activity with exposure to 30 mL/L trans -anethole for 24 h resulting in 100% mortality of rusty grain beetle adults. Estragole did significantly enhance fumigant efficiency of trans -anethole. We further found that trans -anethole affects the beetles acetylcholinesterase activity, which was inhibited and decreased by 64.48% within 24 h. The results therefore show that trans -anethole, the major component of star anise volatiles contributes to its fumigant toxicity against C. ferrugineus by, at least partly, affecting its insect nervous system. Hence, star anise or its biologically active substances show very good potential to be used as a botanical insecticide to control C. ferrugineus .

Bioactivity of the defensive chemicals of six millipede species in the superorder Juliformia against Solenopsis invicta Buren (Hymenoptera: Formicidae)

Delong Kong,Siquan Ling,Wanru Guo,Eduardo G.P. Fox,Jinzhu Xu,Jiamei Zhong,Desen Wang,Hualong Qiu,Weixin Liu

Millipedes are known to produce a diverse array of chemical defenses against natural predators, notably ants, positioning them as a promising source of natural products useful for pest ant control. A top invasive ant species of international scale is the red imported fire ant, Solenopsis invicta , for which there are numerous studies surveying alternative chemicals, as traditional insecticides have not been able to stop their spread. In this study, we characterized the composition of the defensive secretions from six quinone millipedes and evaluated the fumigant, contact, and repellent activities of some main components against S. invicta workers. In total, 27 components were detected in the defensive secretions of the six millipede species, with 2-methoxy-3-methyl-1,4-benzoquinone being the predominant compound overall. Nine compounds (1,4-benzoquinone, α -pinene, dimethyl trisulfide, β -pinene, 2-methyl-1,4-benzoquinone, 2-methoxy-3-methyl-1,4-benzoquinone, orcinol, 3,4-dimethoxyphenol, and 1-hydroxycyclohexyl phenyl ketone) were selected for bioassays. Among them, the three benzoquinones displayed significant fumigant and contact toxicity, as well as repellent effects against S. invicta . Dimethyl trisulfide exhibited fumigant and repellent activity, while 1-hydroxycyclohexyl phenyl ketone demonstrated just contact toxicity. These findings highlight the potential of millipede-derived compounds as natural agents for S. invicta control, possibly to direct a mixture of chemicals for use against fire ants' infestation in transported goods.

Fumigant Toxicity and Oviposition Deterrent Activity of Volatile Constituents from Asari Radix et Rhizoma against Phthorimaea operculella (Lepidoptera: Gelechiidae)

Wu Mei,Xiong Yan,Han Rui,Dong Wenxia,Xiao Chun

Phthorimaea operculella (Zeller) is a worldwide pest of potato. Plant-borne chemicals would be potential alternatives of synthetic chemical fumigants against P. operculella in the storage. Asari Radix et Rhizoma is derived from the dry roots and rhizomes of Asarum heterotropoides Fr. Schmidt var. mandshuricum, A. sieboldii Miq. var. seoulense, or A. sieboldii. In this study, fumigant toxicity and oviposition deterrent of volatile constituents from ARR, δ-3-carene, γ-terpinene, terpinolene, eucarvone, 3,5-dimethoxytoluene, and methyleugenol were tested against P. operculella. The preliminary verification of preventive and control effects of eucarvone, 3,5-dimethoxytoluene and methyleugenol on P. operculella was carried out by simulating warehouse experiments. The results indicated that the six compounds above had fumigation toxic effects on the adults and eggs of P. operculella. Among them, δ-3-carene, γ-terpinene, and terpinolene had weaker fumigation effects than those of eucarvone, 3,5-dimethoxytoluene, and methyleugenol. The LC50 values of eucarvone, 3,5-dimethoxytoluene, and methyleugenol against adult P. operculella were 1.01, 1.78, 1.51 mg/liter air, respectively. The LC50 values against egg P. operculella were 1.09, 0.55, 0.30 mg/liter air, respectively. The oviposition deterrent experiment showed that only methyleugenol (at 5 and 1 mg/ml) and eucarvone (5 mg/ml) had a substantial oviposition deterrent effect. The simulated warehouse experiment verified that methyleugenol, eucarvone, and 3,5-dimethoxytoluene protected potatoes from P. operculella and demonstrated that methyleugenol had the best preventive and control effects. It was concluded that methyleugenol was the active ingredient with the most potential in the volatiles from ARR on P. operculella control and merit further study as botanic fumigant.
