Water-distilled essential oil from the dried bulbs of Allium chinense (Liliaceae) was analyzed by gas chromatography–mass spectrometry (GC–MS). Eighteen compounds, accounting for 98.4% of the total oil, were identified and the main components of the essential oil of A. chinense were methyl allyl trisulfide (30.7%), dimethyl trisulfide (24.1%), methyl propyl disulfide (12.8%) and dimethyl disulfide (9.6%) followed by methyl allyl disulfide (3.4%) and methyl propyl trisulfide (3.6%). The essential oil exhibited contact toxicity against the booklice ( Liposcelis bostrychophila ) with an LC 50 value of 441.8 μg/cm 2 while the two major constituents, dimethyl trisulfide and methyl propyl disulfide had LC 50 values of 153.0 μg/cm 2 and 738.0 μg/cm 2 against the booklice, respectively. The essential oil of A. chinense possessed strong fumigant toxicity against the booklice with an LC 50 value of 186.5 μg/l while methyl allyl trisulfide (LC 50 = 90.4 μg/l) and dimethyl trisulfide (LC 50 = 114.2 μg/l) exhibited stronger fumigant toxicity than methyl propyl disulfide (LC 50 = 243.4 μg/l) and dimethyl disulfide (LC 50 = 340.8 μg/l) against the booklice. The results indicated that the essential oil and its major constituents have potential for development into natural insecticides or fumigants for control of insects in stored grains.

Purpose Inactivating filamentous fungi (molds) reduce toxin production and transmission. This study evaluated the efficacy of formic acid (FA) fumigation to identify optimal conditions and its effects on fungal morphology. We aim to develop an efficient inactivation protocol for molds and provide valuable insights into laboratory techniques for fungal inactivation. Methods Initially, 238 clinical isolates from 23 species of molds were evaluated for the inactivation efficacy of formic acid, formaldehyde, and peracetic acid. After identifying the optimal sterilizing agent, 188 isolates from 16 clinically significant species were tested. Four concentrations (30%, 50%, 70%, and 100%) and six-time intervals (4 to 14 hours) were tested to optimize fumigation conditions. Morphological changes were analyzed using Aspergillus fumigatus ATCC MYA-3626 as the standard strain. Results The inactivation rates of 238 molds fumigated with formic acid, formaldehyde, and peracetic acid for 24 hours were 99.16%, 89.08%, and 90.35% (P < 0.05), indicating that formic acid was more effective than formaldehyde and peracetic acid. Subsequently, 188 isolates from 16 species were fumigated with formic acid at concentrations of 30%, 50%, 70%, and 100% for 24 hours, resulting in inactivation rate of 73.9%, 98.4%, 100%, and 100% (P < 0.05). Inactivation rate increased significantly with longer exposure times: 51.1% at 4 hours, 80.9% at 6 hours, 89.4% at 8 hours, 94.7% at 10 hours, 100% at 12 hours, and 100% after 14 hours (P < 0.05). After fumigation, A. fumigatus ATCC MYA-3626 growth ceased. Electron microscopy revealed irregular folds and roughness on spore surfaces, as well as collapsed or dead spores. Conclusion Formic acid demonstrated superior inactivation efficacy compared to formaldehyde and peracetic acid. At a concentration of 70% and a fumigation duration of 12 hours, the inactivation of molds was most effective.

Context The traditional drying method, sun drying, for Chrysanthemum morifolium Ramat. cv. Hang-ju (Compositae) (HJ) is widely replaced by sulphur fumigation (SF), which has an unknown effect on its efficacy. Objective To investigate protective effects of nonfumigated HJ (NHJ) and sulphur-fumigated HJ (SHJ) water extracts against oxidative stress and lipid peroxidation.Materials and methods Sprague-Dawley rats were administered high-fat diet to induce hyperlipidaemia and randomly divided into eight groups (n = 6): control, fenofibrate, NHJ and SHJ extracts (1, 2 or 4 g crude drugs/kg/d; intragastric administration for 8 weeks). Serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), superoxide dismutase (SOD) and malondialdehyde (MDA) levels were detected. Human umbilical vein endothelial cells (HUVECs) were treated with NHJ and SHJ extracts (50, 100 or 200 μg/mL) for 24 h, followed by oxidized low-density lipoprotein (ox-LDL, 20 μg/mL) for 2 h in vitro. Cellular reactive oxygen species (ROS), SOD and MDA levels and apoptosis were evaluated. Results NHJ was more effective than SHJ in decreasing serum TG, TC, LDL-C, LDL/HDL and MDA while increasing serum HDL-C and SOD levels at high doses. SHJ (IC50=19.9 mg/mL) suppressed HUVEC growth stronger than NHJ (IC50=186.7 mg/mL). At 200 μg/mL, NHJ was more effective than SHJ in downregulating ROS and MDA levels, reducing HUVECs apoptosis rate and elevating SOD activity in ox-LDL-treated HUVECs.Conclusions SF causes oxidative damage and attenuates antioxidative activity in ox-LDL-treated HUVECs, which promotes lipid peroxidation. SF is not recommended for processing HJ.

Atractylodis Macrocephalae Rhizoma (AMR) is one of commonly used medicinal and edible herbs in China. It is often sulfur-fumigated during post-harvest processing. Carbohydrates are important active components of AMR. However, it is unknown whether sulfur-fumigation would induce changes on carbohydrates. Here, carbohydrates including polysaccharides , oligosaccharides and free monosaccharides were comprehensively analyzed to characterize the quality changes of sulfur-fumigated AMR. Determination of both homemade sulfur-fumigated AMR samples and commercial samples from market revealed that sulfur-fumigation did not affect molecular weight distribution of polysaccharides , but altered polysaccharides content and its ratios of constituent monosaccharides , especially glucose (Glc) and fructose (Fru), as well as the contents of oligosaccharides DP2–10 and free monosaccharide Fru. Moreover, the variations enhanced with the increasing of residual SO 2 content. The potential transformation mechanisms could be due to the hydrolysis of polysaccharides. The research outcomes could provide a chemical basis for the safety and efficacy evaluations of sulfur-fumigated AMR.

Sweet orange oil fractions were prepared by molecular distillation of cold-pressed orange oil from sample A (Citrus sinensis (L.) ‘Hamlin’ from America) and sample B (Citrus sinensis Osbeck ‘Newhall’ from China) respectively, and their fumigant activities against medium workers of red imported fire ants (Solenopsis invicta Buren) were investigated. The volatile composition of the orange oil fractions was identified and quantified using GC–MS. Fractions from sample A (A1, A2, and A3) contained 23, 37, and 48 chemical constituents, and fractions from sample B (B1, B2, and B3) contained 18, 29, and 26 chemical constituents, respectively. Monoterpenes were the most abundant components, accounting for 73.56% to 94.86% of total orange oil fractions, among which D-limonene (65.28–80.18%), β-pinene (1.71–5.58%), 3-carene (0.41–4.01%), β-phellandrene (0.58–2.10%), and linalool (0.31–2.20%) were major constituents. Fumigant bioassay indicated that all orange oil fractions exerted good fumigant toxicity against workers of fire ants at 3, 5, 10, and 20 mg/centrifuge tubes, and B1 had the strongest insecticidal potential, followed by A1, B2, A2, B3, and A3. The fractions composed of more high volatile molecules (A1 and B1) showed greater fumigant effects than others. Compounds linalool and D-limonene, which were the constituents of the orange oil, exhibited excellent fumigant toxicity against red imported fire ant workers. Linalool killed red imported fire ant workers completely at 5, 10, and 20 mg/tube after 8 h of treatment, and D-limonene induced >86% mortality at 8 h of exposure.

Black spot disease, which is caused by the pathogenic fungalCeratocystis fimbriata, seriously affects the production of sweet potato and its quality during postharvest storage. In this study, the preliminary identification of the rhizosphere actinomycete strain SPS-33, and its antifungal activity of volatilesin vitroandin vivowas investigated. Based on morphological identification and phylogenetic analysis of the 16S rRNA gene sequence, strain SPS-33 was identified asStreptomyces lavendulae.Volatile organic compounds (VOCs) emitted by SPS-33 inhibited mycelial growth and sporulation ofC. fimbriatain vitroand also induced a series of observable hyphae morphological changes. In anin vivopathogenicity assay, exposure to SPS-33 significantly decreased the lesion diameter and water loss rate in sweet potato tuberous roots (TRs) inoculated withC. fimbriata. It increased the antioxidant enzymes’ activities of peroxidase, catalase, and superoxide dismutase as well as decreased malondialdehyde and increased total soluble sugar. In the VOC profile of SPS-33 detected by a headspace solid-phase micro extraction (HS-SPME) and gas chromatography-mass spectrometry (GC-MS), heptadecane, tetradecane, and 3-methyl-1-butanol were the most abundant compounds. 2-Methyl-1-butanol, 3-methyl-1-butanol, pyridine, and phenylethyl alcohol showed strong antifungal effects againstC. fimbriata. These findings suggest that VOCs fromS. lavendulaeSPS-33 have the potential for pathogenC. fimbriatacontrol in sweet potato postharvest storage by fumigant action. Keywords:Ipomoea batatas(L.) The red imported fire ant is one of the world’s most devastating invasive species, adversely affecting humans, wildlife, crops, and livestock. To control infestations, chemical pesticides are deployed extensively around the world. However, their extensive use has led to negative effects on the environment and human health. Essential oils, which are safe and ecofriendly, can potentially be used as alternatives to chemical pesticides. In this study, grapefruit essential oils were used as fumigant agents to control red imported fire ants. The crude grapefruit oil (GO1) contained 28 compounds, and the concentrated grapefruit oil (GO2), which was refined from GO1 by vacuum distillation, contained 20 compounds. D-Limonene was the dominant constituent in both GO1 (70.1%) and GO2 (73.96%), and other important constituents included β-pinene, α-pinene, β-phellandrene, octanal, d-carvone, α-terpineol, and linalool. Both the essential oils and their individual constituents (α-pinene, α-terpineol, β-phellandrene, octanal, and d-carvone) showed strong lethal fumigant effects against workers. Workers were more susceptible to GO2 than GO1, and octanal was more toxic to workers as compared with the other four constituents. When antennas of workers were treated with the two oils or the five constituents, their walking and gripping abilities were significantly suppressed, and there was an obvious bending or breaking phenomenon on the sensilla of the antennas. Fumigant activity by grapefruit essential oils and their main compounds were associated with their effects on the walking and gripping behavior of workers, and this confirmed that grapefruit essential oil is a promising, ecofriendly, and safe fumigant for the control of red imported fire ants.

This study was designed to evaluate the acaricidal effect and the mode of action of lemongrass essential oil (LEO) against Aleuroglyphus ovatus. Contact and fumigant mortality indicated that LEO had higher acaricidal activities at a low concentration, and the acaricidal action of LEO was significantly more effective by contact effects than by fumigant bioassay. Repellent rate against A. ovatus at different concentrations of LEO increased with application time. Observation on the toxic symptoms revealed that almost all mites were in immobilized type (IM type) in 30 min, but the symptom of knock-down type (KD type) appeared later. Cuticles of mites killed by LEO exhibited obvious colour change and deformation. Detoxification enzyme activity assay showed that glutathione S-transferase (GST) activity increased in mites treated by LEO compared to the controls. LEO increased catalase (CAT) activity of A. ovatus but inhibited superoxide dismutase (SOD) after 20 h. LEO also increased nitric oxide synthetase (NOS) and inhibited acetylcholine esterase (AChE) activities. Moreover, LEO acted as a fast-acting neurotoxin on A. ovatus. Our results suggest that LEO has an acaricidal potential and is a promising agent in control of A. ovatus.
