Effects of a commercial whitening toothpaste containing hydrogen peroxide and citric acid on dentin abrasion and erosion

Kim Jae-Heon,Kim Soyeon,Park Young-Seok

Background Hydrogen peroxide (HP) and citric acid (CA), key contributors to toothpaste acidity, can lead to dental loss. This study aimed to compare the amount of abrasion or loss of dentin based on pH, buffering, and concentration of HP and CA in commercial and experimental toothpastes after toothbrushing or immersion. Methods Bovine dentin specimens were randomly assigned to nine solutions. The prepared solutions included two commercial toothpastes (whitening toothpaste [WT] with HP and CA; conventional toothpaste [CT] without HP and CA), reference slurry (RS), two CA solutions (1.92%, CAS1; 0.001%, CAS2), basic solution (7.16% sodium phosphate dibasic [SPDS]), CA phosphate buffer solution (3.58% SPDS and 0.96% CA [CAPB]), HP solution (4%, HPS), and distilled water (DW). Dentin specimens were performed in two treatments: one with only abrasion (10,000 brushings) and one with only immersion (1 h). After treatments, the amount of dentin loss and surface images were measured and observed using noncontact profilometry. Data were analyzed using an one-way analysis of variance and the Tukey test as a post hoc analysis ( p  < 0.05). Results WT with pH 5.0 had lower dentin abrasion than CT and RS after brushing but had higher dentin loss than both after immersion. The dentin surfaces of CAS1, CAPB, and WT were damaged after immersion, whereas HPS, CAS2, CT, SPDS, RS, and DW remained intact after soaking. CAS2 and HPS, which had a pH of 5.0 like WT, did not significantly differ from those of DW after brushing. Conclusions WT containing HP and CA did not cause significant dentin abrasion but may cause additional dentin loss even without brushing. After brushing or immersion, the CA concentration may affect the dentin surface more than the HP concentration included in WT. The amount of abrasion or loss of dentin after brushing or soaking can vary based on the composition, concentration, and buffer in the solution, even if the pH of the solution is similar to pH 5.0.

Is whitening toothpaste safe for dental health?: RDA-PE method

Jae-Heon KIM,Soyeon KIM,Van Mai TRUONG,Joo Won LEE,Young-Seok PARK

The relative dentin abrasivity-profilometry equivalent values were compared using non-contact profilometry with three subtypes of regular toothpaste and two subtypes of whitening toothpaste containing sodium bicarbonate and 35% hydrogen peroxide. Bovine dentin specimens were assigned to six groups: regular toothpaste (R): R1 (BAMBOO SALT GUM OINTMENT); R2 (MEDIAN TARTAR ORIGINAL); R3 (PERIOE Alpha), Reference slurry: RS (calcium pyrophosphate), whitening toothpaste (W): W1 (NET. WT); W2 (Vussen 28 WHITENING). Relative dentin abrasion–profilometry equivalent (RDA-PE) was determined by brushing 10,000 times (n=8). The pH of the toothpaste was measured (n=5) and the abrasive constituents of the toothpaste was analyzed by FE-SEM and EDS. The RDA-PE values ranged from 26 to 166, and the pH level ranges were 4.928–9.153. The RDA-PE value of the whitening toothpaste containing hydrogen peroxide was not high compared with that of the regular toothpaste. The RDA-PE values of whitening toothpaste could vary depending on the mechanism and ingredients of the whitening agents.

Coffee-stained tooth enamel color restoration and surface abrasion with whitening and regular toothpaste

Soyeon Kim,Yeon-Jee Yoo,Franklin Garcia-Godoy,Young-Seok Park

Purpose: To evaluate the effectiveness of whitening toothpaste in restoring tooth color after coffee staining and its potential impact on enamel surfaces compared with regular toothpaste. Methods: Bovine tooth enamel specimens were prepared and stained with coffee solutions before undergoing brushing simulation with different toothpaste slurries (whitening, regular, reference). For precise evaluation, spectrophotometric measurements were taken at intervals to assess color changes using the CIELAB (Commission Internationale de l'Éclairage Lab*) color space. Additionally, profilometric measurements were taken to determine the impact of toothpaste type on the roughness and abraded depth of the enamel surface. To understand the effects of toothpaste and brushing on color change, surface roughness, and abraded depth, while also considering correlations between these factors, the findings were analyzed using mixed-effects models. Results: The whitening toothpaste group demonstrated the highest recovery rate (71%) after 10,000 brushstrokes, followed by the regular toothpaste group (48%) and the reference slurry group (43%). The mixed-effects model analysis revealed that the reference group had a smaller change in lightness (ΔL) than those in the regular toothpaste group. The whitening toothpaste group showed a greater change in lightness on average than those in the regular toothpaste group, with an increase in lightness as the number of brushstrokes increased. According to the roughness and abraded depth data, the whitening toothpaste group was least affected by brushing, while the reference and regular toothpaste groups showed higher levels of roughness and abraded depth at all intervals. Clinical significance: Gaining a thorough understanding of the effectiveness of whitening toothpaste and its impact on the enamel surface plays a crucial role in refining toothpaste formulations and advancing tooth whitening techniques in dental care.

DNA-templated copper nanoclusters as a fluorescent probe for fluoride by using aluminum ions as a bridge

Pang Jiawei,Lu Yuexiang,Gao Xinyu,He Liuying,Sun Jingwei,Yang Fengyi,Hao Zixuan,Liu Yueying

A selective fluorescent on-off-on probe has have designed for the detection of fluoride (F − ) ions based on DNA-templated copper nanocluster (CuNCs) and by using aluminum(III) ions as a bridge. A 40-mer polythymine acts as a template for the reduction of Cu(II) to Cu(0) by ascorbic acid. This result is the formation of red fluorescent CuNCs, with excitation/emission peaks at 340/640 nm. After addition of Al 3+ ions, the fluorescence of CuNCs is quenched because the interaction of Al 3+ and DNA disturbs the formation of DNA-templated CuNCs. Fluorescence is restored on addition of fluoride to the system. This is due to the desorption of Al 3+ from the DNA and the formation of the Al(OH) 3 F − complex. This system displays a fast fluorometric response to fluoride, with high selectivity over other anions. Fluorescence increases linearly in the 2 to 150 μM F − concentration range, and the detection limit is 1.0 μM. This probe has been successfully used for the detection of F − ions in four brands of toothpaste. The method is rapid, cost-effective, selective, and does not require toxic solvents and reagents. Schematic presentation of a method for fluorometric determination of fluoride by using DNA-templated copper nanoclusters (CuNCs) and using aluminum(III) as a bridge. The red fluorescence of the CuNCs is quenched in the presence of Al(III) ions but restored after addition of fluoride.

Effective extraction of parabens from toothpaste by vortex-assisted liquid-phase microextraction based on low viscosity deep eutectic solvent

Jiangping Cao,Lei Shi,Yifei He,Yuwen Liu,Faqiong Zhao

An efficient, green and reliable method, namely deep eutectic solvent-based vortex-assisted liquid-phase microextraction (DES-VALPME) was proposed for the rapid extraction and sensitive high performance liquid chromatography coupled with variable wavelength detector (HPLC-VWD) determination of methylparaben (MP), ethylparaben (EP), propylparaben (PP) and butylparaben (BP) in toothpaste samples as a proof-of-concept example of trace analysis of personal care and cosmetic products. Compared with traditional DES suffering from higher viscosity, a class of six low viscosity DESs were synthesized by simply dissolving quaternary ammonium salts (hydrogen bond acceptors, HBAs) in methanol or ethanol (hydrogen bond donors, HBDs) at ambient temperature. Initial studies indicated that the DES consisting of tetrabutylammonium chloride (TBAC) and ethanol at a molar ratio of 1:5 achieved the highest extraction efficiency for all target analytes. The influence of type, molar ratio and volume of DES, extraction temperature and vortex time on the extraction efficiency was investigated and optimized systematically. Under the selected conditions, the method displayed satisfactory linearity in the range of 0.05–10 μg/g (R 2  ≥ 0.9995), acceptable recoveries for the real samples (varied from 79.4% to 108.6%), good precision (with relative standard deviation ≤ 5.2 %), as well as lower limits of detection (in the range of 0.01–0.02 μg/g). This study demonstrates that the separation and determination of PBs with the developed DES-VALPME-HPLC-VWD technique is simple, efficient, sensitive and inexpensive.

Sandwich-structured electrospun pH-responsive dental pastes for anti-caries

Yuna Lang,Baolin Wang,Ming-Wei Chang,Renyuan Sun,Longfei Zhang

Dental caries is the most common chronic infectious disease in the human oral cavity and current anti-caries drugs may cause drug resistance and microecological imbalance. In this study, a sandwich-structured electrospun pH-responsive dental paste was engineered via the layer-by-layer electrospinning technique. The top and bottom layers of the dental paste were pH-sensitive layers prepared with Methylmethacrylate polymers IV (E100), and the middle layer was the polylactic acid (PLA) membrane loading with sodium fluorides (NaF). Herein, the surface morphology , chemical as well as physical properties, drug release behaviors, biocompatibility and antibacterial properties of the dental paste were investigated. It was found that the sandwich liked structure enhanced the mechanical properties of the dental paste. The drug release curves of NaF-loaded dental pastes showed that NaF released rapidly and massively in the simulated oral fluid with a risk of dental caries. While in the simulated healthy oral fluid, NaF released slowly and in small amounts. And the dental paste maintained excellent hydrophobic , biocompatibility and antibacterial properties against Streptococcus mutans. Therefore, this sandwich-structured dental paste could respond to pH environment and achieve anti-caries smartly, indicating clinical potential in caries prevention.
