Dental caries is one of the most prevalent human diseases resulting from tooth demineralization caused by acid production of bacteria plaque. It remains challenges for current practice to specifically identify, intervene and interrupt the development of caries while restoring defects. In this study, inspired by natural dental plaque, a stimuli-responsive multidrug delivery system ( [email protected] ) has been developed to prevent tooth decay and promote enamel restoration. Classic spherical core-shell structures of micelles dual-loaded with antibacterial and restorative agents are self-assembled into bacteria-responsive multidrug delivery system based on the pH-cleavable boronate ester bond, followed by conjugation with salivary-acquired peptide (SAP) to endow the nanoparticle with strong adhesion to tooth enamel. The constructed [email protected] specifically adheres to tooth, identifies cariogenic conditions and intelligently releases drugs at acidic pH, thereby providing antibacterial adhesion and cariogenic biofilm resistance, and restoring the microarchitecture and mechanical properties of demineralized teeth. Topical treatment with [email protected] effectively diminishes the onset and severity of caries without impacting oral microbiota diversity or surrounding mucosal tissues. These findings demonstrate this novel nanotherapy has potential as a promising biomedical application for caries prevention and tooth defect restoration while resisting biofilm-associated diseases in a controlled manner activated by pathological bacteria.

The immobilization technology provides a potential pathway for enzyme recycling. Here, we evaluated the potential of using dextranase immobilized onto hydroxyapatite nanoparticles as a promising inorganic material. The optimal immobilization temperature, reaction time, and pH were determined to be 25 °C, 120 min, and pH 5, respectively. Dextranase could be loaded at 359.7 U/g. The immobilized dextranase was characterized by field emission gun-scanning electron microscope (FEG-SEM), X-ray diffraction (XRD), and Fourier-transformed infrared spectroscopy (FT-IR). The hydrolysis capacity of the immobilized enzyme was maintained at 71% at the 30th time of use. According to the constant temperature acceleration experiment, it was estimated that the immobilized dextranase could be stored for 99 days at 20 °C, indicating that the immobilized enzyme had good storage properties. Sodium chloride and sodium acetic did not desorb the immobilized dextranase. In contrast, dextranase was desorbed by sodium fluoride and sodium citrate. The hydrolysates were 79% oligosaccharides. The immobilized dextranase could significantly and thoroughly remove the dental plaque biofilm. Thus, immobilized dextranase has broad potential application in diverse fields in the future.

Bacterial-driven dental caries and tooth discoloration are growing concerns as the most common oral health problems. Current diagnostic methods and treatment strategies hardly allow simultaneous early detection and non-invasive treatment of these oral diseases. Herein, a wearable multifunctional double network hydrogel combined with polyaniline and barium titanate (PANI@BTO) nanoparticles is developed for oral microenvironment visualized sensing and sonodynamic therapy. Due to the colorimetric properties of polyaniline, the hydrogel displays a highly sensitive and selective response for visualized sensing of oral acidic microenvironment. Meanwhile, the barium titanate in the hydrogel efficiently generates reactive oxygen species (ROS) under ultrasound irradiation, realizing non-invasive treatment in the oral cavity. Through bacterial elimination experiments and tooth whitening studies, the hydrogel can achieve the dual effect of effectively inhibiting the growth of cariogenic bacteria and degrading tooth surface pigments. Owing to the visualized sensing of the oral acidic microenvironment and efficient sonodynamic therapy function, the proposed hydrogel system offers a solution for the prevention of caries and tooth whitening, which is promising in developing the biomedical system targeting the simultaneous sensing and therapy for oral diseases.

Helicobacter pylori ( H. pylori ) is a major pathogen associated with gastric ulcers and gastric cancer, infecting approximately half of the global population. Therefore, developing sensitive, rapid, and cost-effective detection methods for H. pylori is critical for public health. We developed an innovative microscale colorimetric microsensor integrated with smartphone ambient-light sensors for portable, on-site quantification of H. pylori- associated urease activity in dental plaque. This low-cost device (< $ 5 fabrication) employs 3D-printed components and a custom app to measure transmittance changes from urease-catalyzed urea hydrolysis with phenol red, achieving a robust correlation with commercial microplate reader absorbance ( R 2 = 0.9989). The system demonstrates exceptional Analytical sensitivity, with limits of detection of 9.75 U/L for urease And 1.709 × 10 3 CFU/mL for H. pylori , alongside high reproducibility (CV < 10%) And stability over 14 days. When applied to the on-site detection of dental plaque among volunteers, the smartphone-based colorimetric microsensor exhibited detection results that were highly aligned with H. pylori infections confirmed by the urea breath test. It outperformed commercial test strips And achieved 100% sensitivity, 95.5% specificity, And 97.1% accuracy. This miniaturized, smartphone-based platform offers a promising solution for home self-testing and demonstrates significant potential for enhancing H. pylori preliminary screening at the community level.

Dental plaque, a kind of natural biofilm, has attracted much attention because of its role in caries and periodontal disease. The pathogenicity of the dominant bacterial species of S. mutans is based on the formation of extracellular polysaccharides (EPS) and its survival in acidic environments. In addition, the formed plaque biofilm has the characteristics of water insolubility, retention, and low membrane permeability. This is why there are no effective antibiotics or other ways to treat dental caries. Chemodynamic therapy (CDT) is an ideal therapeutic modality with biofilm microenvironment as stimulus. In this paper, a unique nanocomposite, ZnO 2 -Cu@RB NPs, has been prepared which is composed of copper-doped zinc peroxide nanoparticles (ZnO 2 -Cu NPs) and antibacterial agent Rose Bengal (RB). It can quickly (<10 min) and efficiently kill S. mutans (greater than 8.0 log 10 CFU mL −1 ) in acidic biofilm via the in situ enhanced CDT, and significantly reduce the demineralization of apatite. ZnO 2 -Cu@RB NPs can also weaken the acid production of S. mutans and inhibit the formation of EPS. More significantly, excellent therapeutic efficacy against S. mutans in the rat model was observed. Our findings reveal the great potential of metal peroxide nanoparticles as a new strategy for combating biofilm-related diseases.

Biofilms increase bacterial resistance to antibiotics, as conventional antibiotic doses are often ineffective at penetrating the biofilm matrix to eliminate bacteria. Recent research has shown that the Gram-negative predator bacterium Bdellovibrio bacteriovorus can penetrate Gram-positive bacterial biofilms during its predation phase and benefit from them without direct predation. Here, based on the penetration ability of B. bacteriovorus , we constructed antibiotic-loaded liposome-engineered B. bacteriovorus as a drug delivery strategy for biofilm-related diseases. As a “living antibiotic,” B. bacteriovorus can prey on Gram-negative bacteria, penetrate biofilms, and disrupt their dense structure. During this process, the rapid movement of B. bacteriovorus enhances the delivery of antibiotic-loaded liposomes into the biofilm, promoting efficient antibiotic release and improving biofilm eradication. Our findings demonstrate that this engineered living antibiotic strategy significantly improves the control and removal of bacterial biofilms, accelerates the elimination of dental plaque, promotes wound healing, and holds promise as a novel platform for treating biofilm-related infections.

Invisible aligners have been widely used in orthodontic treatment but still present issues with plaque formation and oral mucosa abrasion, which can lead to complicated oral diseases. To address these issues, hydrophilic poly(sulfobetaine methacrylate) (polySBMA) coatings with lubricating, antifouling, and antiadhesive properties have been developed on the aligner materials (i.e., polyethylene terephthalate glycol, PETG) via a simple and feasible glycidyl methacrylate (GMA)-assisted coating strategy. Poly(GMA- co -SBMA) is grafted onto the aminated PETG surface via the ring-opening reaction of GMA (i.e., “grafting to” approach to obtain G- co -S coating), or a polySBMA layer is formed on the GMA-grafted PETG surface via free radical polymerization (i.e., “grafting from” approach to obtain G- g -S coating). The G- co -S and G- g -S coatings significantly reduce the friction coefficient of PETG surface. Protein adsorption, bacterial adhesion, and biofilm formation on the G- co -S- and G- g -S-coated surfaces are significantly inhibited. The performance of the coatings remains stable after storage in air or artificial saliva for 2 weeks. Both coatings demonstrate good biocompatibility in vitro and is not caused irritation to the oral mucosa of rats in vivo over 2 weeks. This study proposes a promising strategy for the development of invisible aligners with improved performance, which is beneficial for oral health treatment.
