Stiffness-damping balance, high specific mechanical performance, and corrosion resistance are demanded for metals. Here, we developed an enamel-inspired ceramic (EIC) coating strategy involving hydrothermal growth of a tooth enamel-like ZrO₂ nanorods on Zr foil and amorphous ZrO₂ intergranular phase formation via controlled hydrolysis. The EIC coated Zr foil (Zr-EIC) achieved a high viscoelastic figure of merit (4.6 GPa); as well as excellent specific stiffness and hardness owing to the lightweight of ceramics, surpassing the values of Zr and previously reported Zr-based alloys or Zr-based composites. This was due to the vertically oriented stiff nanorods and unique energy dissipation derived from deformation at the crystalline/amorphous interfaces. It demonstrated exceptional NaCl corrosion resistance. This approach showed universality across Ti, Zn, and Cu substrates, enhancing both mechanical properties and corrosion resistance. By constructing an enamel-like hierarchical structure on the metal surface, the strategy overcomes traditional metal limitations without causing an increase in density.

Remineralization is a common strategy for the repair of early demineralized tooth enamels, but the harsh dynamic oral environment often hampers its efficacy. Rapid remineralization is expected to address this challenge, however, the stabilizers of remineralization materials often resist their transformation required for repair. Here, by dissolving the ions of calcium and phosphate in glycerol-dominant solvents, we obtain the calcium phosphate clusters (1–2 nm), which are stabilized by glycerol (with high viscosity and affinity to clusters), but can perform a fast enamel repair via the water-triggered transformation in both static and dynamic environments. Upon the in vitro and in vivo (female Sprague-Dawley rats) studies, the clusters swiftly enter the nano-/micro-sized enamel defect sites, then form a compact hydroxyapatite repair layer within a short time (30 min, much faster than the conventional materials), and significantly recovers mechanical properties. This material is promising for large-scale preparation and applications in dental remineralization.

Enamel demineralization is a common complication of orthodontic treatment. Studies related to enamel demineralization are usually conducted in the laboratory using in vitro models. Detection technology relies on large instruments, which are not conducive to its popularization and promotion. We designed a fluorescence sensing system using a complex formed by 3,4,9,10-tetra-(4-trimethylaminohexyloxy-carbonyl)-perylene (PDI-HTMA) and casein phospho-peptides (CPPs) that can rapidly detect calcium ions in enamel. In this study, we investigated the feasibility of using PDI-HTMA/CPPs as probes for detecting enamel demineralization. We used Ca 2+ from enamel demineralization as the detection target for the fluorescent probe and verified its stability, specificity, and linear range. This method exhibits strong linear relationships in the Ca 2+ concentration range of 30–500 μM, with a limit of detection of 3.89 μM. The PDI-HTMA/CPPs were applied to human enamel specimens. We compared the detection results with those of existing enamel demineralization evaluation methods, and the results showed a trend consistent with that of ICDAS grading and laser fluorescence caries detection. This fluorescent probe assay is expected to be applied for the early detection of enamel demineralization, which has strong application advantages. To the best of our knowledge, this is the first fluorescent probe assay applied for the in vitro detection of enamel demineralization.

Dental caries, a prevalent oral disease, is caused by acid- producing plaque microorganisms-notably Streptococcus mutans ( S. mutans )-which drive enamel demineralization. Current prevention strategies, such as fluoride and chlorhexidine, primarily target planktonic bacteria but exhibit limited efficacy against biofilms and neglect enamel remineralization, thereby reducing their therapeutic efficacy. To address these limitations, we developed dual-functional nanoparticles (AXCP NPs) that can eradicate S. mutans biofilm and promote enamel remineralization. The amphiphilic polymer Arg-XOS-CPP (AXC) was synthesized by covalently conjugating oligosaccharide carbonyl groups to the amino residues of L-arginine and casein. Self-assembly of AXC with calcium and phosphate ions yielded hybrid nanoparticles (AXCP NPs). Experimental results indicated that AXCP NPs had no effect on the viability of HaCaT cells. Under acidic conditions, protonation of arginine triggered a surface charge reversal from negative to positive, enabling targeted binding to negatively charged biofilms. As expected, AXCP NPs exhibited potent anti- S. mutans activity (MIC: 640 μg/mL) and remarkable biofilm eradication efficacy (>80% clearance). In vitro release and remineralization assays demonstrated that AXCP NPs continuously released calcium and phosphorus ions, regulating the mineralization process and effectively restoring enamel functionality. Furthermore, in mice caries model, localized application of AXCP NPs significantly suppressed biofilm accumulation on tooth surface and restored mineral density in demineralized enamel, inhibiting caries progression.

We aimed to create a slippery liquid-infused enamel surface with antibiofouling property to prevent dental biofilm/plaque formation. First, a micro/nanoporous enamel surface was obtained by 37% phosphoric acid etching. The surface was then functionalized by hydrophobic low-surface energy heptadecafluoro-1,1,2,2-tetra- hydrodecyltrichlorosilane. Subsequent infusion of fluorocarbon lubricants (Fluorinert FC-70) into the polyfluoroalkyl-silanized rough surface resulted in an enamel surface with slippery liquid-infused porous surface (SLIPS). The results of water contact angle measurement, diffuse-reflectance Fourier transform infrared spectroscopy and atomic force microscope confirmed that the SLIPS was successfully constructed on the enamel surface. The antibiofouling property of the SLIPS was evaluated by the adsorption of salivary protein of mucin and Streptococcus mutans in vitro , as well as dental biofilm formation using a rabbit model in vivo . The results showed that the SLIPS on the enamel surface significantly inhibited mucin adhesion and S. mutans biofilm formation in vitro and inhibited dental plaque formation in vivo .

Tooth enamel possesses excellent mechanical properties owing to its multi-scale highly ordered architecture. To develop tooth enamel-mimetic structural materials (EMSMs) is of great significance for understanding the important role of ordered hydroxyapatite (HAP) nanocrystals in tooth enamel and exploring high-performance materials. However, it is still a great challenge to mimic the unique multi-scale ordered architecture of the natural tooth enamel. Herein, inspired by the multi-scale structure and self-assembly of the tooth enamel, we have developed a novel bottom-up step-by-step assembly strategy to build the EMSMs based on ultralong HAP nanowires, and achieved the multi-scale (from nano- to micro- to macro-scale) highly ordered HAP nanowire structure control. In this work, ultralong HAP nanowires are assembled in sequence into highly ordered HAP nanowire bundles, aligned HAP microfibers, and three-dimensional (3-D) highly ordered HAP nanowire bulk, which perfectly imitate the structure and self-assembly of the tooth enamel. This novel method is suitable to fabricate the EMSMs with arbitrary sizes and well-designed shapes, which are promising for various applications in the biomedical fields such as hard tissue defect repair.

Dental caries have become a major global public health problem. Plaque control and remineralization of initial enamel lesions are paramount for the prevention and control of caries. Polyhexamethylene biguanide (PHMB) is a type of cationic amphipathic antibacterial agent with broad-spectrum antibacterial properties and good biological safety. Fluoride delays demineralization and promotes the remineralization of hard dental tissues. However, a high concentration is needed for it to function as an antibacterial agent. In order to create a PHMB with the benefits associated with fluoride, we synthesized a fluorine-containing cationic polymer, PHMB-F. Fourier transform-infrared spectroscopy and solid state nuclear magnetic resonance characterization confirmed the successful synthesis of PHMB-F. Antibacterial tests showed that PHMB-F had better antiseptic efficacy for Streptococcus mutans compared with just PHMB. Moreover, positively-charged PHMB-F allows fluoride ions to exist closer to the enamel surface with negative potential, which markedly lowers the ion concentrations in the microenvironment adjacent to hard dental tissues needed to maintain equilibrium. Thus, only low concentrations of PHMB-F are required for enamel remineralization.

Purpose: To evaluate the effectiveness of whitening toothpaste in restoring tooth color after coffee staining and its potential impact on enamel surfaces compared with regular toothpaste. Methods: Bovine tooth enamel specimens were prepared and stained with coffee solutions before undergoing brushing simulation with different toothpaste slurries (whitening, regular, reference). For precise evaluation, spectrophotometric measurements were taken at intervals to assess color changes using the CIELAB (Commission Internationale de l'Éclairage Lab*) color space. Additionally, profilometric measurements were taken to determine the impact of toothpaste type on the roughness and abraded depth of the enamel surface. To understand the effects of toothpaste and brushing on color change, surface roughness, and abraded depth, while also considering correlations between these factors, the findings were analyzed using mixed-effects models. Results: The whitening toothpaste group demonstrated the highest recovery rate (71%) after 10,000 brushstrokes, followed by the regular toothpaste group (48%) and the reference slurry group (43%). The mixed-effects model analysis revealed that the reference group had a smaller change in lightness (ΔL) than those in the regular toothpaste group. The whitening toothpaste group showed a greater change in lightness on average than those in the regular toothpaste group, with an increase in lightness as the number of brushstrokes increased. According to the roughness and abraded depth data, the whitening toothpaste group was least affected by brushing, while the reference and regular toothpaste groups showed higher levels of roughness and abraded depth at all intervals. Clinical significance: Gaining a thorough understanding of the effectiveness of whitening toothpaste and its impact on the enamel surface plays a crucial role in refining toothpaste formulations and advancing tooth whitening techniques in dental care.

The development of high-performance dental materials that mimic human enamel is urgently required for clinical applications, but remains a key challenge. Herein, we present a biomimetic design strategy for creating inverse enamel-like columnar composites that replicate the mechanical properties and tribological performance of the natural enamel. Inverse to the enamel consisting of hard rods and surrounding relatively soft inter-rod, the composites are assembled from a cellular-structured ceramic scaffold and resin columns. This architecture helps to avoid dramatic stiffness and hardness decreases due to the presence of flexible substances and provides an elegant combination of mechanical properties that strikingly resemble that of natural enamel. Consequently, the inverse enamel-like columnar composites have encouraging tribological performance characterized by an enamel-matched coefficient of friction and wear rate, as well as low abrasive potential to opposing natural teeth, demonstrating great promise for next-generation dental restorations/replacements and other applications. More importantly, the proposed design strategy can be extended to other biological prototypes, thus facilitating the development of various high-performance functional materials.

Dental enamel is a mineralized extracellular matrix, and enamel defect is a common oral disease. However, the self-repair capacity of enamel is limited due to the absence of cellular components and organic matter. Efficacy of biomimetic enamel mineralization using calcium phosphate ion clusters (CPICs), is an effective method to compensate for the limited self-healing ability of fully developed enamel. Preparing and stabilizing CPICs presents a significant challenge, as the addition of certain stabilizers can diminish the mechanical properties or biosafety of mineralized enamel. To efficiently and safely repair enamel damage, this study quickly prepared CPICs without stabilizers using the atomization method. The formed CPICs were evenly distributed on the enamel surface, prompting directional growth and transformation of hydroxyapatite (HA) crystals. The study revealed that the mended enamel displayed comparable morphology, chemical composition, hardness, and mechanical properties to those of the original enamel. The approach of repairing dental enamel by utilizing ultrasonic nebulization of CPICs is highly efficient and safe, therefore indicating great promise.

Objective This study aimed to develop an oral environment-responsive complex with mineralisation-antibacterial dual-activities for preventing and treating early dental caries. Methods The complex was synthesized with phosphorylated starch serving as a template to stabilize calcium phosphate prenucleation clusters (CaP-PNCs) and silver nanoclusters (AgNCs). The binding properties of the complex on human enamel were evaluated using confocal laser scanning microscopy (CLSM). Antibacterial efficacy against Streptococcus mutans , including both planktonic bacteria and biofilms, was assessed through colony counting, scanning electron microscopy (SEM), and live/dead staining. Remineralization effects on artificially demineralized enamel were analyzed using SEM, CLSM, and energy-dispersive X-ray spectroscopy (EDS). Cytotoxicity on MC3T3 cells was tested using CCK-8 assays. Results The CaP-PNCs/AgNCs complex demonstrated strong binding to human enamel. In the presence of α-amylase, the complex exhibited potent antibacterial effects, achieving a 90% reduction in planktonic Streptococcus mutans viability and and significantly disrupted biofilms. Remineralization assays showed a 70-100 μm hydroxyapatite layer resembling natural enamel in structure and composition. Notably, the complex maintained effective remineralization capabilities even under cariogenic bacterial challenge. Cytotoxicity evaluations on MC3T3 cells indicated no adverse effects on viability. Conclusion This complex enables targeted antibacterial action and biomimetic mineralization through responsively release of AgNCs and CaP-PNCs in oral environment, simultaneously addressing biofilm disruption and promoting enamel regeneration for the treatment of early caries. Clinical Significance The oral environmental responsive design provides a minimally invasive strategy to repair demineralized enamel while suppressing pathogenic biofilms, bridging non-invasive prevention and structural restoration in caries management.

Objectives The study investigated the effects of an orthodontic primer containing amorphous fluorinated calcium phosphate (AFCP) nanoparticles on enamel white spot lesions (WSLs). Materials and methods The AFCP nanoparticles were prepared and incorporated into Transbond XT Primer. Thirty-two human enamel slices were highly polished and randomly divided into four groups: no part covered (control), half covered with a primer containing 0 wt%, 25 wt%, and 35 wt% AFCP. Subsequently, samples were challenged by a modified pH-cycling and characterized by color measurement, micro-computed tomography, and scanning electron microscope (SEM). The bonding properties of the primers containing AFCP were assessed using shear bond strength test, and the mouse fibroblasts (L929) were employed to evaluate the cytotoxicity. Results When the enamel was challenged by pH cycling, 25 wt% and 35 wt% AFCP groups exhibited less color change (ΔE) and less mineral loss than the control and 0 wt% AFCP groups. The SEM images showed that the original microstructural integrity and mineral deposition rate of the enamel surface were better in the 25 wt% and 35 wt% AFCP groups. In particular, the 35 wt% AFCP group exhibited the best performance after 3 weeks of pH cycling. The shear bond strength and cell viability revealed no significant difference among the tested groups (P > 0.05). Conclusion Using the primer containing 35 wt% AFCP might be a promising strategy for preventing the occurrence and development of WSLs during orthodontic treatment.
