Taurine in several aqueous binary solvents: Solubility prediction, measurement, modeling, solvent effect, and thermodynamics

Weizhong Gong,Pan Li,Sohrab Rohani

The crystallization of taurine in pure water results in a low yield. Introducing anti-solvents during the cooling crystallization process would increase the yield. Therefore, measuring and evaluating the solubility of taurine in different binary aqueous solvents is necessary. In this work, the qualitative trend of the solubility of taurine in the binary solvent systems of 1,2-propanediol-water, acetonitrile–water, and ethylene glycol–water was first predicted using Hansen Solubility Parameters (HSPs), and then experimentally determined using isothermal gravimetric method. The experimentally obtained solubility is correlated with the modified Apelblat model as a linear model, and NRTL, UNIQUAC, Wilson, and λh model as nonlinear models. The solubility of taurine in binary solvents is further analyzed using the KAT-LSER model to investigate the solvent effect on solubility. For all researched systems, the standard molar enthalpy and the standard molar Gibbs energy changes are computed and found to be positive, indicating the dissolution of taurine is an endothermic process.

Effects of dietary taurine on growth, taurine metabolism and Vibrio crassostreae resistance in juvenile clam Ruditapes philippinarum

Yang Liu,Kai Liao,Yuxiang Zhu,Yang Zhang,Hailong Huang,Hui Ge,Jilin Xu

Taurine serves an important role in all aquatic animals. The purpose of this experiment was to investigate the effects of exogenous taurine on growth, endogenous taurine metabolism, and Vibrio crassostreae resistance in juvenile clam Ruditapes philippinarum . Microencapsulated taurine concentrations of 0, 0.75, 1.5, 3 and 6 mg/g clam wet weight were designated as the T0, T0.75, T1.5, T3 and T6 groups. After 2 weeks of feeding, the results demonstrated that exogenous taurine did not promote the growth of clams ( P  > 0.05). The taurine content of clams was increased significantly with the increase of exogenous taurine content ( P  < 0.05). The gene expression of taurine transporter ( TauT ) was significantly elevated in the T3 and T6 groups compared to the T0 group ( P  < 0.05). With the addition of exogenous taurine, mRNA expressions of cysteine sulfinic acid decarboxylase ( CSAD ) , 2-aminoethylsulfide dioxygenase ( ADO ) , cysteine dioxygenase ( CDO ) , cystathionine β-synthase ( CBS ) , cystathionine γ-lyase ( CSE ) and serine dehydratase ( SDH ) were suppressed ( P  < 0.05). The survival rate of clams challenged with Vibrio crassostreae was significantly higher in the T0.75 group compared to the T0 group on the fifth day ( P  < 0.05). Additionally, the activities of antioxidant enzymes and lysozyme were significantly elevated, immune-related and anti-inflammatory factor genes were upregulated, while pro-inflammatory factor genes were downregulated in the T0.75 group compared to the T0 group ( P  < 0.05). Dietary taurine supplementation regulates endogenous taurine metabolism and exhibits potential anti- Vibrio effect in clam R. philippinarum .

Comparison of the effects of taurine and methionine supplementation on the nitrogen metabolism of beef steers elucidated through plasma metabolome profiling

Yufeng Liu,Cheng Liu,Shuo Zhang,Jinming Hu,Meng M. Li,Guangyong Zhao

The objectives of the experiment were to compare the effects of rumen-protected taurine (RPT) and rumen-protected methionine (RPM) on the nitrogen (N) metabolism, plasma biochemical parameters, and metabolomics in beef steers and to clarify whether taurine plays similar roles as methionine (Met) in improving the regulation of N metabolism in beef steers. Six Simmental steers aged 12 months (liveweight 325 ± 7 kg) were used as experimental animals. The experimental treatments included a basal diet, the basal diet + 70.0 g/d RPT and the basal diet + 74.2 g/d RPM. The treatments were assigned in a replicated 3 × 3 Latin square design. Each experimental period included 15 d of adaptation and 5 d of sampling. The results showed that supplementing the diet with RPT or RPM did not affect the apparent nutrient digestibility ( P > 0.05). Supplementing the diet with RPT or RPM increased the N retention ( P < 0.05) and the N utilization efficiency (NUE) ( P < 0.05) and decreased the urinary excretion of 3-Methylhistidine ( P < 0.05) and the estimated skeletal protein degradation rate ( P < 0.05). Supplementing the diet with RPT increased the plasma concentrations of taurine ( P < 0.001), cysteine ( P = 0.010), valine ( P = 0.013) and total non-essential amino acids (NEAA) ( P = 0.047) and tended to increase the plasma concentrations of essential amino acids (EAA) + NEAA ( P = 0.087), but it did not affect the plasma concentrations of total EAA ( P > 0.05). Supplementing the diet with RPM increased the plasma concentrations of methionine ( P = 0.033), lysine ( P = 0.047), cysteine ( P = 0.007), leucine ( P = 0.046), isoleucine ( P = 0.046), valine ( P = 0.034), total EAA ( P = 0.028), total NEAA ( P = 0.004) and EAA + NEAA ( P = 0.004). The plasma metabolomics profiling revealed that supplementing the diet with RPT upregulated the plasma concentrations of taurine ( P < 0.001), L-Cysteine ( P = 0.004) and some amino acid (AA) analogues ( P < 0.05) and RPM upregulated the plasma concentrations of Met ( P = 0.021), L-Isoleucine ( P = 0.036), L-Tryptophan ( P = 0.006) and some AA analogues ( P < 0.05). In conclusion, taurine has similar impacts to Met in improving the N retention and the NUE in beef steers. Taurine deficiency is one of the main reasons that the NUE of beef steers is low. Supplementation of the diet with taurine is beneficial to the N utilization in beef steers.

Taurine-Induced Fabrication of Nano-Architectured Conducting Polypyrrole on Biomedical Titanium

Jingwen Liao,Haobo Pan,Chengyun Ning,Guoxin Tan,Zhengnan Zhou,Junqi Chen,Shishu Huang

In this article, taurine, one of the small biomolecules associated with bone metabolism, is firstly utilized to induce the fabrication of nano-architectured conducting polypyrrole (NCPPy) on biomedical titanium in diverse pH values of phosphate buffer solution (PBS). Accordingly, the possible mechanism for the fabrication of NCPPy is proposed, which is dependent on the states of polytaurine from the polymerization of taurine, i.e., the inability of forming polytaurine and unordered restricted space results in taurine-incorporated and polytaurine-incorporated tightly packed nanoparticles (pH 6.2 and 8.0), respectively, and however, ordered restricted space constructed by polytaurine chains induces the fabrication of polytaurine-incorporated nanopillars (pH 6.8) and polytaurine-incorporated nanowire networks (pH 7.4).

Investigation on the action mechanisms of taurine on rumen microbial protein synthesis and nitrogen metabolism in beef steers using sodium sulfate as a contrast

Manman Fan,Jinming Hu,Cheng Liu,Shuo Zhang,Yufeng Liu,Guangyong Zhao

Taurine is a sulfur-containing nonproteinogenic amino acid. Recent studies have shown that taurine can improve rumen microbial crude protein (MCP) synthesis. This experiment aimed to investigate the action mechanism of taurine on rumen MCP synthesis and nitrogen (N) metabolism in beef steers using sodium sulfate (Na 2 SO 4 ) as a contrast. Six steers (bodyweight 506 ± 17 kg) were assigned to three experimental groups including a basal diet (control), a basal diet supplemented with 45 g taurine/d or 50 g Na 2 SO 4 /d, and were allocated in a replicated 3 × 3 Latin square design. The amounts of sulfur from taurine and Na 2 SO 4 were equal (11.38 g/d). The results showed that, compared with the control group, both taurine and Na 2 SO 4 increased ruminal production of MCP ( P < 0.05) by 37.50% and 29.17%, respectively, and increased the concentration of sulfide (S 2- ) ( P < 0.001). Both taurine and Na 2 SO 4 increased the digestibility of neutral detergent fiber ( P < 0.05). Taurine tended to increase ( P = 0.087) while Na 2 SO 4 decreased plasma urea concentration ( P = 0.049), with the taurine group exhibiting higher plasma urea concentrations than the Na 2 SO 4 group ( P = 0.003). Taurine did not affect urinary urea excretion ( P = 0.246) whereas Na 2 SO 4 decreased urinary urea excretion compared with the control group ( P = 0.002). Compared with the control group, both taurine and Na 2 SO 4 increased urinary allantoin excretion ( P < 0.05), total purine derivatives excretion ( P < 0.05), and the estimated rumen microbial N flow ( P < 0.05). The urinary urea excretion of the taurine group was higher than the Na 2 SO 4 group ( P = 0.019). Compared with the control group, taurine did not affect N excretion, N retention (NR) or N utilization efficiency (NUE) ( P > 0.10), but Na 2 SO 4 decreased urinary N excretion ( P = 0.018) and total N excretion ( P = 0.024), and increased NR ( P = 0.024) and NUE ( P = 0.022). No differences were found in NR and NUE between the taurine and Na 2 SO 4 groups ( P > 0.10). Taurine improved ruminal MCP synthesis by enriching the pathways associated with sulfur and amino acid metabolism while Na 2 SO 4 improved ruminal MCP synthesis by enriching pathways related to nucleotide and purine metabolism. In summary, both taurine and Na 2 SO 4 improved ruminal MCP synthesis by modulating different pathways. Taurine was less effective in decreasing total N excretion than Na 2 SO 4 but no differences in NR and NUE were found between the two groups of beef steers.

Taurine as an additive for improving the fouling resistance of nanofiltration composite membranes

Jinbo Jin,Dongqing Liu,Dandan Zhang,Yuhang Yin,Xinyu Zhao,Yufeng Zhang

A hydrophilic compound, taurine, was investigated as an additive in the interfacial polymerization between piperazine (PIP) and trimesoyl chloride (TMC) to prepare thin-film composite (TFC) membranes. The resulting membranes were characterized by X-ray photoelectron spectroscopy and attenuated total reflectance–Fourier transform infrared spectroscopy. The morphology and hydrophilicity of the membranes were investigated through scanning electronic microscopy and water contact angle measurements. The separation performance of the TFC membranes was investigated through water flux and salt rejection tests. The protein-fouling resistance of the films was evaluated by water recovery rate measurements after the treatment of bovine serum albumin. The membrane containing 0.2 wt % taurine showed the best performance of 92% MgSO 4 rejection at a flux of 31 L m −2 h −1 and better antifouling properties than the PIP–TMC membranes. An appropriately low concentration of taurine showed the same MgSO 4 rejection as the PIP–TMC membranes but a better fouling resistance performance. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. Synthesis of taurine–fluorescein conjugate and evaluation of its retina-targeted efficiency in vitro

Meihong Huang,Jiaqi Song,Bingzheng Lu,Huizhi Huang,Yizhen Chen,Wei Yin,Wenbo Zhu,Xinwen Su,Chuanbin Wu,Haiyan Hu

In this work, retinal penetration of fluorescein was achieved in vitro by covalent attachment of taurine to fluorescein, yielding the F–Tau conjugate. Nuclear magnetic resonance (NMR) and high resolution mass spectrometry (HRMS) were used to confirm the successful synthesis of F–Tau. The cellular uptake of F–Tau in adult retinal pigment epithelial cells (ARPE-19) and human retinal microvascular endothelial cells (hRMECs) was visualized via confocal scanning microscopy. The results indicated an improvement of solubility and a reduction of log P of F–Tau compared with fluorescein. As compared with fluorescein, F–Tau showed little toxicity, and was retained longer by cells in uptake experiments. F–Tau also displayed higher transepithelial permeabilities than fluorescein in ARPE-19 and hRMECs monolayer cells ( P <0.05). These results showed that taurine may be a useful ligand for targeting small-molecule hydrophobic pharmaceuticals into the retina.

Taurine as a powerful passivator of perovskite layer for efficient and stable perovskite solar cells

Xian Hou,Zhenjia Yuan,Jinlong Liu,Hongzhen Ma,Fucheng Yu

Due to the ionic lattice property and the solution manufacture process of the perovskite light absorbing layer, there are several intrinsic defects (such as vacancies and low coordination Pb2+ and I−) in perovskite films, which cause undesired photon-generated carrier recombination in the perovskite solar cells (PSCs) and seriously affect the power conversion efficiency (PCE) of devices. Defect passivation strategy is one of the most effective ways to eliminate the defects in perovskite films. Herein, a multifunctional Taurine molecule was introduced into CH3NH3PbI3 (MAPbI3) perovskite precursor solution to passivate the defects. It was found that Taurine with sulfonic acid (–SOOOH) and amino (–NH2) groups can bind with uncoordinated Pb2+ and I− ions, respectively, which can significantly reduce the defect density and suppress the carrier non-radiative recombination. Under atmospheric environment, non-hole transport layer FTO/TiO2/perovskite/carbon structure PSCs were prepared. The device with Taurine showed a PCE of 13.19%, which is 17.14% higher than that of the control device (11.26%). With the suppressed defects, the Taurine passivated devices also showed enhanced device stability. The unencapsulated Taurine passivated device stored in ambient air after 720 h (temp. ∼25 °C and RH ∼25%) maintained 58.74% original PCE, while that of the control device was only about 33.98%.

Taurine Prevents Impairments in Skin Barrier Function and Dermal Collagen Synthesis Triggered by Sleep Deprivation-Induced Estrogen Circadian Rhythm Disruption

Qi Shao,Zhaoyang Wang,Yifang Li,Xun Tang,Ziyi Li,Huan Xia,Qihong Wu,Ruxue Chang,Chunna Wu,Tao Meng,Yufei Fan,Yadong Huang,Yan Yang

Sleep deprivation is a prevalent issue that disrupts the circadian rhythm of estrogen, particularly estradiol, thereby significantly affecting women’s skin health and appearance. These disruptions can impair skin barrier functionality and decrease dermal collagen synthesis. In this study, our results demonstrate that topical taurine supplementation promotes the expression of tight junction (TJ)-related proteins and enhances collagen production, effectively restoring skin homeostasis in sleep-deprived female mice. Mechanistically, taurine upregulates the expression ofTMEM38B, a gene encoding the TRIC-B trimeric cation channel, resulting in increased intracellular calcium ion levels. This, in turn, promotes the upregulation of TJ-related proteins, such as ZO-1, occludin, and claudin-11 in epidermal cells, while also enhancing the expression of type III collagen in fibroblasts, thus restoring skin homeostasis. These findings suggest that taurine may serve as an alternative to estradiol, effectively improving skin homeostasis disrupted by sleep deprivation while mitigating the potential risks associated with exogenous estrogen supplementation. Collectively, these results provide preliminary insights into the protective mechanisms of taurine against sleep deprivation-induced skin impairments and establish a foundation for its potential application in treating skin conditions related to estrogen imbalances, such as skin aging in menopausal women.

A pilot study of the onset of hepatic encephalopathy (OHE) in mice induced by thioacetamide and the protective effect of taurine by holistic metabolic characterization

Wei Dan-Dan,Wang Jun-Song,Li Ming-Hui,Guo Ping-Ping,Dong Ge,Yang Ming-Hua,Kong Ling-Yi

Hepatic encephalopathy (HE) is a severe neuropsychiatric complication due to acute or chronic liver failure. In this work, an acute thioacetamide induced HE mice model was established to investigate the metabolic events in the onset of HE (OHE) and the protective role of taurine (TAU). Sera for biochemical evaluation, and livers and cortexes for histopathological inspection and NMR-based metabolomic investigation were collected. Principal component analysis and orthogonal partial least squares-discriminant analysis were performed to excavate differential metabolites of OHE, and explore the protective role of TAU. Metabolites pathway analysis was performed on significant metabolites selected based on loading/S-plots and fold change plots to identify biologically meaningful metabolic patterns and relevant pathways. The most affected pathways in OHE mice were those along the liver–brain axis: among them, the alanine, aspartate and glutamate metabolism was the most important. TAU showed better performance in improving the disturbed metabolism in the cortex than in the liver of OHE mice. This pilot study based on metabolomics approach and pathway analysis should help to understand HE systematically, and to develop new therapy for HE.

Modulatory effects of taurine on jejunal contractility

Yao Q.Y.,Chen D.P.,Ye D.M.,Diao Y.P.,Lin Y.

Taurine (2-aminoethanesulfonic acid) is widely distributed in animal tissues and has diverse pharmacological effects. However, the role of taurine in modulating smooth muscle contractility is still controversial. We propose that taurine (5-80 mM) can exert bidirectional modulation on the contractility of isolated rat jejunal segments. Different low and high contractile states were induced in isolated jejunal segments of rats to observe the effects of taurine and the associated mechanisms. Taurine induced stimulatory effects on the contractility of isolated rat jejunal segments at 3 different low contractile states, and inhibitory effects at 3 different high contractile states. Bidirectional modulation was not observed in the presence of verapamil or tetrodotoxin, suggesting that taurine-induced bidirectional modulation is Ca2+dependent and requires the presence of the enteric nervous system. The stimulatory effects of taurine on the contractility of isolated jejunal segments was blocked by atropine but not by diphenhydramine or by cimetidine, suggesting that muscarinic-linked activation was involved in the stimulatory effects when isolated jejunal segments were in a low contractile state. The inhibitory effects of taurine on the contractility of isolated jejunal segments were blocked by propranolol and L-NG-nitroarginine but not by phentolamine, suggesting that adrenergic β receptors and a nitric oxide relaxing mechanism were involved when isolated jejunal segments were in high contractile states. No bidirectional effects of taurine on myosin phosphorylation were observed. The contractile states of jejunal segments determine taurine-induced stimulatory or inhibitory effects, which are associated with muscarinic receptors and adrenergic β receptors, and a nitric oxide associated relaxing mechanism.

Mechanism of Lacticaseibacillus rhamnosus JY027 alleviating hyperuricemia in mice through gut-kidney axis

Chao Miao,Kai Dong,Yu Shen,Yilin Sun,Wenyan Li,Chaoxin Man,Yu Zhang,Qianyu Zhao,Yujun Jiang

Hyperuricemia is associated with various diseases. This study confirmed that intervention with Lacticaseibacillus rhamnosus JY027 ( L. rhamnosus JY027) reduced biochemical markers including uric acid , creatinine, and urea nitrogen in hyperuricemic mice. A combined analysis of gut microbiota , short-chain fatty acids (SCFAs), and metabolites based on the gut-kidney axis revealed that this bacterial intervention significantly enhanced the abundance of beneficial bacteria such as Bifidobacterium , Akkermansia , and Faecalibaculum and increased SCFA levels. The bacterial intervention also regulated the levels of guanosine , creatinine, arginine, choline , taurine and other differential metabolites in the kidneys of mice with hyperuricemia. This intervention ultimately reduced uric acid levels in mice through the modulation of multiple pathways, including arginine biosynthesis, taurine and hypotaurine metabolism, and glutathione metabolism. This study lays the foundation for exploring how L. rhamnosus JY027 alleviates hyperuricemia, providing theoretical support for developing probiotic preparations to mitigate hyperuricemia and their application.
