Wound-induced suberization is an essentially protective healing process for wounded fruit to reduce water loss and microbial infection. It has been demonstrated that abscisic acid (ABA) could promote wound suberization, but the molecular mechanism of ABA regulation remains little known. In this study, the transcript level of Achn030011 (designated as AchnKCS), coding a β-ketoacyl-coenzyme A synthase (KCS) involved in suberin biosynthesis, was found to be significantly upregulated by ABA in wounded kiwifruit. A bZIP transcription factor (Achn270881), a possible downstream transcription factor in the ABA signaling pathway, was screened and designated as AchnbZIP12 according to its homology with related Arabidopsis transcription factors. A yeast one-hybrid assay demonstrated that AchnbZIP12 could interact with the AchnKCS promoter. Furthermore, significant trans-activation of AchnbZIP12 on AchnKCS was verified. The transcript level of AchnbZIP12 was also upregulated upon treatment with ABA. These results imply that AchnbZIP12 acts as a positive regulator in ABA-mediated AchnKCS transcription during wound suberization of kiwifruit.

The objective of this study is to investigate the effects of carvacrol on the postharvest storage quality and cell wall metabolism of the ’Guichang’ kiwifruit. For this purpose, the ’Guichang’ kiwifruit is selected as the test material and treated with 0.03 mg/mL of a carvacrol solution for dipping. The storage quality and cell wall metabolism of the fruit are then evaluated under ambient conditions (20 ± 1 °C). The results indicate that treatment with carvacrol effectively reduces the postharvest respiration rate and weight loss in the kiwifruit, while preserving the levels of vitamin C (VC), soluble protein, total phenols, flavonoids, soluble solids, solid–acid ratio, and soluble pectin. Additionally, it delays the decline in the starch content and mitigates the increase in soluble sugars. Furthermore, carvacrol significantly decreases amylase activity and inhibits the elevation of polygalacturonase (PG), pectin methylesterase (PME),β-galactosidase (β-Gal), and cellulase (Cx) activity. Consequently, this treatment effectively maintains fruit firmness as well as protopectin (PP) and cellulose content at elevated levels. A correlation analysis shows that firmness is highly and negatively correlated with the cell wall degrading enzymes PG, PME, β-Gal, Cx, and α-L-Af, with R values of −0.88, −0.83, −0.98, −0.94, and −0.93, respectively. The negative correlation values for the control group are all lower than 0. In conclusion, treatment with carvacrol effectively preserves a high level of postharvest ripening quality in the kiwifruit, attenuates the metabolic processes of the cell wall, and delays fruit softening and senescence.

The kiwi berry (Actinidia arguta) has been widely studied because of its rich phenolic, flavonoid, and vitamin C contents. Numerous reports have demonstrated that fruit peels contain higher phenolic content and antioxidant activity than that of flesh. In this study, the phytochemical content and antioxidant activities of peel and flesh extracts of six kiwi berries were analyzed from four regions (namely, Dandong, Benxi, Taian, and Tonghua) in China. The antioxidant activity was determined using the peroxyl radical scavenging capacity (PSC) and cellular antioxidant activity (CAA) assays. The phenolic, flavonoid, and vitamin C contents of kiwi berry peel were 10.77, 13.09, and 10.38 times richer than that of kiwi berry flesh, respectively. In addition, the PSC and CAA values of kiwi berry peel were higher than those of kiwi berry flesh. The analysis of the separation and contents of phenolics were performed by the high-performance liquid chromatography (HPLC)-diode-array detectormass spectrometry/mass (DAD-MS/MS) system, and the results illustrated that protocatechuic acid, caffeic acid, chlorogenic acid, and quinic acid were the major phenolic compounds. In conclusion, this study indicated that kiwi berry peel contains a rich source of antioxidants. These data are of great significance for the full development and utilization of kiwi berries in these four regions of China to produce nutraceutical and functional foods.

Purpose: Pseudomonas syringae pv. actinidiae causes bacterial canker of kiwifruit and is responsible for severe economic losses and emergence of drug-resistant bacteria. Bacteriophages are viruses that infect target bacterial hosts and may be the best strategy to prevent and control kiwifruit canker disease. The objective of this experiment was to monitor the prevalence of Pseudomonas syringae pv. actinidiae and provide insight for the use of phages in biological control.Materials and methods: In this study, 52 strains of Pseudomonas syringae pv. actinidiae were isolated from 68 stem samples of kiwi plant (cv. Hongyang & Jinkui). Following polymerase chain reaction (PCR) analysis, 15 isolates belonging to biovar 3 were identified, one of which was named XWY0007 and used as the target strain to isolate the phages. Thirty-six phages were isolated and purified from a total of 51 surface water samples collected in Shanghai. All phages were identified by transmission electron microscopy (TEM) and their host ranges were evaluated. Three phages, designated φXWY0013, φXWY0014 and φXWY0026 were selected and further characterised using one-step growth curve and stability at different temperatures and pH.Results and conclusions: The isolated phages are promising for use as antimicrobials against bacterial canker in kiwi. This report is regarding Pseudomonas syringae pv. actinidiae and its phages from major areas of kiwifruit cultivation.

High citric acid content in kiwifruit wine would lead to bad sensory experience and quality deterioration. It is opportune and crucial to develop an appropriate and feasible method to degrade citric acid for kiwifruit wine. The non-Saccharomycesyeasts confirmed to have the ability to degrade citric acid were screened and used in kiwifruit wine fermentation in the study. A representative number of 23 yeasts with a strong citric acid degradation ability was identified by molecular approaches. JT-1-3, identified to bePichia fermentans, was preferred for high citric acid degradation and strong stress resistance in association with RV002 (commercialSaccharomycescerevisiae). Then it was pure-cultured in kiwifruit juice, and the results indicated that citric, malic and tartaric acids declined significantly from 12.30, 3.09 and 0.61 g/L to 11.00, 2.02 and 0.41 g/L after fermentation, respectively, resulting in the significant decrease in total acid in kiwifruit wine. The analytical profiles for amino acids and volatile compounds showed thatPichia fermentansJT-1-3 could improve amino acids’ proportion and increase the volatile compounds of alcohols, esters and phenols. This work indicated that JT-1-3 has great potential to be applied for fruit wine with high level citric acid.Keywords:non-Saccharomycesyeast;kiwifruit wine;citric acid;deacidification fermentation;GC-MS

To investigate the differences among kiwifruits from six different cultivars ( Actinidia chinensis , Actinidia delicosa and Actinidia arguta ), UPLC-QTOF-MS and UPLC-ESI-MS/MS coupled to multivariate statistical analysis were applied to untargeted metabolites and targeted analysis of kiwifruit samples for the first time. In untargeted analysis, the metabolome of kiwifruits was exhibited by principal component analysis (PCA), a total of 44 up-regulated feature compounds were screened based on the orthogonal partial least squares-discriminant analysis (OPLS-DA) and identified as glycosyl compounds , hydrocinnamic acids and various flavonoids . Furthermore, the contents of 27 targeted procyanidins , flavonoids and hydrocinnamic acids were analyzed by a UPLC-ESI-MS/MS method. It was indicated that phenol glycosides were much higher in A. arguta , responsible for the aroma of kiwifruits, various quercetin glycosides were higher in A. arguta , and much higher flavan-3-ols and hydrocinnamic acids were rich in A. Chinensis cv. Hongyang (red) kiwifruits. In addition, the results of chemometric analysis showed the untargeted metabolites and targeted compounds were responsible for the classification on the kiwifruits, which can benefit the scientific understanding of chemical diversity for kiwifruits, and improve the investigation of new wild kiwifruit cultivars, especially red kiwifruit and A. arguta .

The kiwifruit industry is booming worldwide. As a result, a great deal of kiwifruit waste residue (KWR) containing monosaccharides is produced and discarded. This material shows great potential for the production of platform chemicals. In this study, a series of Nb/Al oxide catalysts were synthesized via a facile and low-cost coprecipitation method, and their structures were characterized using: thermal gravimetric analysis (TGA), XRD, FESEM, TEM, X-ray photoelectron spectroscopy (XPS), NH3-TPD, N2adsorption-desorption, and FTIR-Pyridine adsorption. Experimental results of sugar-to-levulinic acid (LA) conversion revealed that the 20%Nb/Al oxide catalyst provided the highest catalytic performance and durability in terms of LA yield from fructose (74.2%) at 463 K after 10 min and from glucose (47.5%) at 473 K after 15 min. Notably, the 20% Nb/Al oxide catalyst with a 10% dosage is capable of converting kiwifruit waste residue to LA at 473 K after 10 min. In conclusion, the enhanced catalytic performance was obtained due to the high acidity, and large surface areaof Nb/Al oxide catalyst. Keywords:levulinic acid;kiwifruit waste residue (KWR);heterogeneous catalysis;Nb/Al oxide catalyst

Wound-induced suberization is an essentially protective healing process for fruit to reduce water loss and avoid infecting. However, cognate mechanisms that regulate this process are little known. To expand our knowledge of suberization induced by wounding, a wound-healing investigation together with metabolite profiling study was conducted in postharvest kiwifruit ( Actinidia deliciosa ). The development of suberization in wounded fruit was demonstrated by autofluorescence observation and toluidine blue staining at 1–4 day (d) after wounding. Activities of phenylalanine ammonia-lyase (PAL), cinnamyl-alcohol dehydrogenase (CAD) and peroxidase (POD) in wound-healing tissue were enhanced by abscisic acid (ABA). The constituent analysis of suberin including polyphenolics (SPP) and polyaliphatics (SPA) proved that exogenous ABA increased the content levels of total phenols, total flavonoids and alkanes, alkenes, alcohols, alkane acids, olefine acids, esters, glycerides and vitamin E in wound-healing tissue. Results suggested that ABA stimulated suberization through the activation of PAL, CAD and POD to accelerate wound-healing of wounded kiwifruit.

Pseudomonas syringae pv. actinidiae (Psa), the pathogen that causes bacterial canker disease in kiwifruit, has brought about substantial losses to the kiwifruit industry. Investigating the infection process at both the cellular and microscopic levels is of great significance for the formulation of effective control strategies against this disease. Thus, the establishment of appropriate chemical imaging analysis methods becomes essential. Confocal Raman microspectral imaging (CRMI), combined with chemometrics, provides an intuitive means to visualize and characterize the spatiotemporal changes of biopolymers in the cell walls of both healthy and infected kiwifruit stems. Raman spectra of different infected stems exhibit clustering effects in Principal component analysis (PCA), and a classification model constructed using Support vector machines (SVM) achieves an accuracy of 97%. Multivariate Curve Resolution-Alternating Least Squares (MCR-ALS) is utilized to resolve spectral matrices and concentration profiles from raw Raman imaging signals. The reconstructed concentration data yields accurate molecular imaging maps of High-methylated pectin (HMP), Low-methylated pectin (LMP), cellulose, hemicellulose, and lignin. The results indicate that, three days after Psa infection, the content of cellulose and HMP in the cell wall increases, while the changes in hemicellulose, lignin, and LMP are minimal. However, five days after infection, the contents of HMP, LMP, cellulose, hemicellulose, and lignin decrease significantly, resulting in the disruption of the cell-wall structure. The chemical imaging method proposed in this study shows great promise as an effective means for studying the bacterial infection process in kiwifruit stems at the cellular level.

The feasibility of simultaneous detection of multiple cheap materials in kiwifruit juice (KFJ) was studied by two fluorescent methods and chemometrics. Because the actual frauds usually involve many known and unknown adulterants, the traditional methods to detect one or more known components are insufficient to identify adulterated KFJ. Therefore, class models of pure KFJ were established using one-class partial least squares (OCPLS) to perform untargeted analysis. To enhance the detection specificity, traditional and double-quantum-dots enhanced fluorescent spectroscopic techniques were combined to characterize pure and adulterated KFJ. By data fusion and using standard normal variables (SNV) transformation, OCPLS model with sensitivity of 0.929 was obtained. The proposed method could detect adulterations of 2% (w/w) or more syrup and artificial fruit powder. The fusion of traditional and quantum dots enhanced fluorescence was demonstrated to provide a rapid and highly sensitive method for untargeted analysis of adulterated KFJ.

Two water-soluble polysaccharides (ACPS1 and ACPS2) were isolated from the roots of Actinidia Chinensis by DEAE-52 cellulose and Sephacryl S300 chromatography. Preliminary structural characterization was conducted by physicochemical property, Fourier transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM) analysis. ACPS1, with an average molecular weight of 5.58 × 10 5 Da, was mainly composed of rhamnose , arabinose , xylose , mannose and galactose in an approximate molar ratio of 1.48:4.28:4.30:1.00:17.83. ACPS2, with a high average molecular weight of 1.23 × 10 6 Da, mainly contained rhamnose, arabinose and galactose in a molar ratio of approximately 1.00:2.33:6.61. Both ACPS1 and ACPS2 exhibited the remarkable antioxidant activity to scavenge the DPPH radical and significant protective effects on H 2 O 2 -induced HEK 293 cells death in a concentration-dependent manner. Meanwhile, in vitro immunomodulatory activities of the two polysaccharides were evaluated. The results showed that treatment with 50–300 μg/mL of the samples could increase NO production and phagocytic activity of macrophages in a dose-dependent manner. The present results suggested that the two polysaccharides from Actinidia Chinensis may be potential antioxidant and immunomodulatory agents for preparing functional foods and nutraceuticals applied in food and pharmaceutical industries.

To improve the functional properties and flavor complexity of kiwi wine using different non- Saccharomyces cerevisiae , Wickeramomyces anomala (Wa), Zygosaccharomyces rouxii (Zr), ZygoSaccharomyces bailii (Zb) and Schizosaccharomyces pombe (Sp) were inoculated sequentially with S. cerevisiae (Sc). The physicochemical and sensorial profiles of the wines were evaluated. The evolution of cells showed that non- Saccharomyces exhibited varying degrees of fermentation vigor and only acted during the first vinification stage. Ethanol content, pH, ΔE and organic acids in the wines varied according to the yeasts used. Compared with the pure Sc fermentation, the sequential fermentations of Wa-Sc and Sp-Sc significantly increased the production of polyphenols. Fifteen volatile compounds with relative odor activity values (rOAV) ≥ 1.0 were identified. Furthermore, principal component analysis (PCA) revealed that Zr-Sc and Sp-Sc were correlated with higher levels of ethyl esters (ethyl hexanoate, ethyl heptanoate, ethyl decanoate), isoamyl acetate and 2-phenyl-1-ethanol in the wines, improving the flower and sweet notes. Zr-Sc also enhanced the tropical fruity aroma. Sequential inoculation with Zb was related to the contents of acetate esters, ethyl butyrate , methyl butyrate and cineole, triggering the tropical fruity odor. In addition, the partial least-squares regression (PLSR) revealed that acetate esters contributed greatly to the tropical fruity note.
