The study of a new detergent (octyl-glucopyranoside) for decellularizing porcine pericardium as tissue engineering scaffold

Jiaoming Dong,Yu Li,Xiumei Mo

Background The use of extracellular matrix (ECM) derived from decellularized tissue is increasingly frequent in regenerative medicine and tissue engineering . However, it is recognized that currently used decellularization procedures have negative effects on ECM integrity. The objective of this study was to investigate the impact of a decellularization protocol with a new detergent on the ECM integrity of porcine pericardium (PP) compared with other traditional detergents. Materials and methods Fresh PP were decellularized by sodium deoxycholate in combination with Triton X-100 (SDT), sodium dodecyl sulfate (SDS), and octyl-glucopyranoside (OGP), respectively. Histologic analysis and scanning electron microscopy were performed to confirm the removal of cells and to examine the structure of ECM. DNA content was examined by the method of DNA extraction . Mechanical properties and biochemical compositions of ECM were also studied. Results Histologic analysis and DNA determination demonstrated that SDS and OGP completely removed the cells, and the major ECM structure was preserved well for PP treated with 1% (wt/vol) OGP but disrupted for PP treated with SDS; whereas treatment with SDT was insufficient to remove cells from PP. Uniaxial tensile tests showed that PP decellularized by OGP had similar mechanical properties to native PP, whereas the mechanical properties of PP decellularized by SDS and SDT decreased. The biochemical compositions of PP decellularized by OGP were also well conserved, except that glycosaminoglycans markedly decreased. Moreover, the results obtained in the MTT study further indicated that the cytotoxicity of PP decellularized by OGP was significantly lower than that decellularized by SDS and SDT. Conclusion It is suggested that the environmentally friendly and nontoxic OGP can be used as a decellularizing agent. The OGP method could achieve both complete removal of cells from native PP and preservation of the matrix structure; thus, it might be a suitable approach to preparation of tissue engineering heart valve scaffold.

Porcine pericardium crosslinked with POSS-PEG-CHO possesses weakened immunogenicity and anti-calcification property

Xiaobo Yu,Jingli Ding,Yingjie He,Shunbo Wei,Xing Chen,Qiujie Luo,Yuqing Zhang,Chen Qian,Jiahui Wang,Mengjie Hu,Xiang Zhang,Cuifen Lu,Jinping Liu,Jianliang Zhou

Valvular heart disease (VHD) poses a thorny problem in cardiovascular diseases. The most effective treatment for VHD is heart valve replacement. Biological heart valve (BHV) is more favored than mechanical heart valve due to the maturity of transcatheter heart valve replacement (THVR) and the absence of the need for lifelong anticoagulant use. However, traditional commercial BHV suffers degeneration within 10 to 15 years because of calcification caused by the cross-linking reagent, glutaraldehyde. Considering the remarkable properties of POSS, PEG, and the star-like eight-arm structure, we fabricated POSS-PEG-PP, which is a decellularized porcine pericardium (DPP) crosslinked by a star-like eight-arm cross-linker octafunctionalized POSS of benzaldehyde-terminated polyethylene glycol (POSS-PEG-CHO) based on the Schiff's base reaction. POSS-PEG-PP exhibits more intense fiber arrangement and better mechanical properties than GLUT-PP (glutaraldehyde crosslinked DPP). The results also show that the cytocompatibility, endothelialization, and hemocompatibility of POSS-PEG-PP are outstanding in vitro. Subsequently, in vivo assessments demonstrate that POSS-PEG-PP has anti-inflammatory and anti-calcification abilities. Furthermore, RNA sequencing analysis of subcutaneous implants suggests that the intervention of AMPK and IL-17 signaling pathways plays an important role in the inflammatory and immune responses regulation of POSS-PEG-PP. Therefore, POSS-PEG-PP is an excellent substitute material for BHVs and is expected to be clinically transformed.

Butterfly-Inspired Multiple Cross-Linked Dopamine–Metal–Phenol Bioprosthetic Valves with Enhanced Endothelialization and Anticalcification

Yuqing Zhang,Hai Hu,Yaoxi Zhu,Jie Xiao,Chenghao Li,Chen Qian,Xiaobo Yu,Jinping Zhao,Xing Chen,Jinping Liu,Jianliang Zhou

Valve replacement is the most effective means of treating heart valve diseases, and transcatheter heart valve replacement (THVR) is the hottest field at present. However, the durability of the commercial bioprosthetic valves has always been the limiting factor restricting the development of interventional valve technology. The chronic inflammatory reaction, calcification, and difficulty in endothelialization after the implantation of a glutaraldehyde cross-linked porcine aortic valve or bovine pericardium often led to valve degeneration. Improving the biocompatibility of valve materials and inducing endothelialization to promote in situ regeneration can extend the service life of valve materials. Herein, inspired by the hardening process of butterfly wings, this study proposed a dopamine–metal–phenol strategy to modify decellularized porcine pericardium (DPP). This is a strategy to make dopamine (DA) coordinate trivalent metal chromium ions (Cr(III)) with antiplatelets (PLTs) and anti-inflammatory properties, and then cross-link it with tea polyphenols (TP) to generate a valve scaffold that is mechanically comparable to glutaraldehyde-cross-linked scaffolds but avoids the cytotoxicity of aldehyde and presents better biocompatibility, hemocompatibility, anticalcification, and anti-inflammatory response properties.

Dialdehyde xanthan gum and curcumin synergistically crosslinked bioprosthetic valve leaflets with anti-thrombotic, anti-inflammatory and anti-calcification properties

Mengyue Hu,Xu Peng,Shubin Shi,Chang Wan,Can Cheng,Xixun Yu

Currently commercial glutaraldehyde (GA)-crosslinked bioprosthetic valve leaflets (BVLs) suffer from thromboembolic complications, calcification, and limited durability, which are the major stumbling block to wider clinical application of BVLs. Thus, developing new-style BVLs will be an urgent need to enhance the durability of BVLs and alleviate thromboembolic complications. In this study, a quick and effective collaborative strategy of the double crosslinking agents (oxidized polysaccharide and natural active crosslinking agent) was reported to realize enhanced mechanical, and structural stability, excellent hemocompatibility and anti-calcification properties of BVLs. Dialdehyde xanthan gum (AXG) exhibiting excellent stability to heat, acid-base, salt, and enzymatic hydrolysis was first introduced to crosslink decellularized porcine pericardium (D-PP) and then curcumin with good properties of anti-inflammatory, anti-coagulation, anti-liver fibrosis, and anti-atherosclerosis was used to synergistically crosslink and multi-functionalize D-PP to obtain AXG + Cur-PP. A comprehensive evaluation of structural characterization, hemocompatibility, endothelialization potential, mechanical properties and component stability showed that AXG + Cur-PP exhibited better anti-thrombotic properties and endothelialization potential, milder immune responses, excellent anti-calcification properties and enhanced mechanical properties compared with GA-crosslinked PP . Overall, this cooperative crosslinking strategy provides a novel solution to achieve BVLs with enhanced mechanical properties and excellent anti-coagulation, anti-inflammatory, anti-calcification, and the ability to promote endothelial cell proliferation .

A hydrophobic antifouling surface coating on bioprosthetic heart valves for enhanced antithrombogenicity

Meiling Li,Cheng Zheng,Shumang Zhang,Binggang Wu,Kailei Ding,Xueyu Huang,Yang Lei,Yunbing Wang

Thrombosis is an important factor that causes the failure of artificial biological valves in addition to calcification and immune rejection. A hydrophobic antifouling surface can improve blood compatibility by reducing the absorption of protein. In this study, porcine pericardium was cross-linked with glycidyl methacrylate, and carbon–carbon double bonds were introduced. Then, fluoride monomer was added so that the pericardial surface would become hydrophobic and antifouling. Fluoride modification changed the hydrophilicity of the pericardium surface, and the surface water contact angle increased from 84° to 143°. Compared with unmodified pericardium, the adsorption of bovine serum albumin and fibrinogen decreased by 93.1% and 85%, respectively, and the anti-thrombogenicity was greatly enhanced.

A bioprosthetic heart valve material prepared by copolymerization of 2-Amino-4-pentanoic acid modified pericardium and N, N-Dimethylacrylamide

Kailei Ding,Cheng Zheng,Xueyu Huang,Li Yang,Yang Lei,Yunbing Wang

Bioprosthetic heart valve thrombosis (BHVT) is no longer considered a rare clinical entity in recent years and could cause valve motion abnormalities, which requires reoperation. Currently reported modifications of bioprosthetic heart valves (BHVs) mainly focused on improving the anti-calcification property. However, it was hard to improve their anti-thrombogenicity and crosslinking efficiency at the same time. Here we reported a method that combines improved hemocompatibility , crosslinking efficiency, cytocompatibility, and anti-calcification property for BHVs. Porcine pericardia (PPs) and 2-amino-4-pentanoic acid (APA) were first co-crosslinked with glutaraldehyde to introduce allyl moieties to the PPs, then poly(N, N-dimethylacrylamide) (PDMAA) was covalently immobilized on PPs through radical polymerization . In vitro and in vivo studies demonstrated that the structural stability of PDMAA modified PPs was much superior than that of traditional glutaraldehyde crosslinked PPs (GA). The PDMAA modified PPs exhibited enhanced antithrombogenic performance with a 75% decrease of platelet adhesion compared with GA. Moreover, the calcium content accumulated on the modified PPs after 90-day rat subcutaneous implantation decreased 93.5% compared with that on GA. The PDMAA modified PPs may alleviate BHVT and calcification, offering a potential option for BHVs fabrication in the future.

Visible light–induced cross-linking of porcine pericardium for the improvement of endothelialization, anti-tearing, and anticalcification properties

Fan Yang,Liangpeng Xu,Gaoyang Guo,Yunbing Wang

Population aging and the development of transcatheter aortic valve replacement boost the implantation of bioprosthetic heart valves (BHVs) in patients worldwide. However, the traditional glutaraldehyde cross-linked BHVs fail within 12–15 years mainly due to leaflet tear and calcification defects. In this study, a novel visible light–induced cross-linking of the porcine pericardium (PP) was realized by the photo-oxidation of the furfuryl-modified PP in the presence of Rose Bengal. The resulting material showed comparable collagen stability with the glutaraldehyde cross-linked PP and appropriate biomechanical properties such as tensile strength, modulus, and elongation, suggesting that this material could meet the general requirement for BHVs. Besides, this cross-linked PP showed significantly improved cytocompatibility compared with the Glut–cross-linked PP, with no cytotoxicity to L929 cells and the ability to support HUVECgrowth. Meanwhile, this material showed superior anti-tearing performance and much less calcification than the Glut–cross-linked PP in hope of reducing the risk of BHV failure. Considering these results, the visible light–induced cross-linking method proposed in this study could provide a promising way to construct a biocompatible and robust biomaterial for the fabrication of the BHV.

A functionalized biological heart valve by double bond crosslinking with enhanced biocompatibility and antithrombogenicity

Cheng Zheng,Dajun Kuang,Kailei Ding,Xueyu Huang,Hongsong Fan,Li Yang,Yunbing Wang,Xingdong Zhang

With the advancement of minimally invasive interventional therapy, biological heart valves (BHVs) have been extensively used in clinics. However, BHVs are generally prone to degeneration within 10–15 years after implantation due to defects including cytotoxicity, immune response, calcification and thrombosis, which are closely related to glutaraldehyde-crosslinking. In this work, we prepared a functionalized BHV through the in situ polymerization of methacrylated porcine pericardium and 2-hydroxyethyl methacrylate to avoid and overcome the defects of glutaraldehyde-crosslinked BHVs. The functionalized BHV was proven to be stable against enzymatic degradation and compatible towards HUVECs. After implantation in rats subcutaneously, a significantly mitigated immune response and reduced calcification were observed in the functionalized BHV. With the grafting of hydrophilic 2-hydroxyethyl methacrylate polymers, the antithrombogenicity of BHV was markedly enhanced by resisting the unfavorable adhesion of blood components. Moreover, the hydrodynamics of the functionalized BHV totally conformed to ISO 5840-3 under a wide range of simulated physiological conditions. These results indicate that the functionalized BHV with enhanced biocompatibility, anticalcification property and antithrombogenicity exhibited a low risk of degeneration and should be explored for further application.
