Nitazoxanide protects cats from feline calicivirus infection and acts synergistically with mizoribine in vitro

Zhanding Cui,Dengliang Li,Yinli Xie,Kai Wang,Ying Zhang,Guohua Li,Qian Zhang,Xiaoxueying Chen,Yue Teng,Shihui Zhao,Jiang Shao,Fan Xingmeng,Yanli Zhao,Dongju Du,Yanbing Guo,Hailong Huang,Hao Dong,Guixue Hu,Shuang Zhang,Yongkun Zhao

Feline calicivirus (FCV) is a highly contagious pathogen that causes acute upper respiratory infections and oral disease in cats, thus seriously endangering feline health. Recently, there have been outbreaks of particularly virulent variant strains of FCV, which can cause both acute symptoms and fatal systemic disease. The discovery of effective antiviral agents to treat FCV infection is, therefore, gradually assuming increased importance. In this study, we showed that both nitazoxanide and mizoribine had antiviral activity in F81 cells infected with different strains of FCV and also demonstrated a synergistic effect between the two drugs. Experiments in cats challenged with FCV showed that nitazoxanide significantly reduced the clinical symptoms of FCV infection, reduced viral load in the trachea and lungs, and reduced viral shedding. Our results showed that nitazoxanide and mizoribine could potentially be used as therapeutic agents to treat FCV infection.

Rapid and simultaneous detection of antibodies against three feline viruses using colloidal gold immunochromatographic assay with smartphone integration

Ji Jingjing,Li Jiahao,Wu Minghui,Wang Bin,Lu Yu-nan,Zhang Chuhan,Shan Yanke,Liu Fei

A multiplex colloidal gold immunochromatographic assay (mCGIA) was developed for the simultaneous detection of feline calicivirus (FCV), feline panleukopenia virus (FPV), and feline herpesvirus (FHV) antibodies in feline serum. The expressed and purified recombinant FCV-VP1, FPV-VP2, and FHV-gD proteins exhibited strong antigenic reactivity. The mCGIA test strip achieved rapid (< 15 min) and specific detection, showing no cross-reactivity with other feline viral antibodies. The assay achieved a low visual limit of detection (vLOD) of 1:1 × 10 5 for FCV and 1:1 × 10 4 for FPV and FHV antibodies. Stability testing revealed 90% retention of detection capability after 4 weeks of storage at 37 °C. Reproducibility was robust, with intra- and inter-assay coefficients of variation (CVs) of less than 7% and 13%, respectively. The smartphone-based portable reader enabled quantitative detection with wide linear ranges (from 1:1000 to 1:100000 for FCV, 1:250 to 1:10000 for FPV and FHV) and excellent linearity (FCV: R 2  = 0.992, FPV and FHV: R 2  = 0.991). Compared with indirect ELISA using 164 clinical serum samples, the assay showed 98.17% (FCV), 98.78% (FPV), and 97.56% (FHV) concordance rates, respectively. This novel mCGIA platform provides a rapid, cost-effective tool for sero-surveillance, vaccine evaluation, and assisting clinical diagnosis, enhancing feline health management. Graphical

Cepharanthine: A promising natural compound against feline infectious peritonitis virus infection and associated inflammation

Yao Yao,Rui Jing,Xiang Liu,Li Kang,Pinghuang Liu

Feline infectious peritonitis (FIP), a fatal disease in cats caused by feline infectious peritonitis virus (FIPV), has limited treatment options and lacks effective vaccines. Cepharanthine (CEP), a natural isoquinoline alkaloid, possesses many medicinal properties, including antiviral activities. However, the role of CEP in management of FIPV infection remains poorly understood. Here, we identified that CEP had a potent ability to inhibit FIPV infection in vitro . CEP significantly inhibited FIPV infection when administered at different times, with co-treatment showing the most significant inhibitory effect. Time-of-addition assays demonstrated that CEP exerted antiviral activity during the post-entry stages of the FIPV infection. We also verified that CEP inhibited FIPV infection not through affecting type I interferon expression, and it could decrease pro-inflammatory factors expression induced by FIPV infection. The combination of CEP and GS-441524 exhibited synergistic antiviral effects against FIPV infection. Our findings highlight the therapeutic potential of CEP for treatment of FIP.

Novel colchicine ethosomes cataplasm for the treatment of acute gouty arthritis

Yao-Yao Wei,Yang Zhang,Xue Lu,Jie Zhou,Huang-Wei Cheng,Yuan-Yuan Liu,Hua Zhang,Wen Chen

Gout is a disease caused by the deposition of sodium urate (MSU) crystals in the joints and tissues. Colchicine (COL) has become the first-line drug for the treatment of acute gout due to its low price and efficacy. However, colchicine is highly cytotoxic and oral administration is prone to cause severe adverse effects on the gastrointestinal tract, liver and kidney. Therefore, this study aimed to develop a novel dermal delivery formulation for addressing the safety concerns of this drug. The researchers used ethosomes encapsulation technology to improve the skin permeability of COL. In addition, in order to improve the performance of the ethosomes, it was screened and determined that the addition of 1.0–1.5 mg of ceramide III (Cer3) per mL of ethosomes as a modifier could significantly enhance the stability of the ethosomes, Cer3/COL-ethosomes (CCE) were successfully constructed. The CCE was then mixed with a cataplasm matrix to produce a colchicine-carrying CCE cataplasm, which demonstrated the superimposed effect of the advantages of the two dosage forms, the ethosomes and the cataplasm. Compared with the traditional delivery method of COL, this topical formulation is an attractive alternative for the treatment of gout as it can achieve effective blood levels without causing fluctuations in blood levels, and has good efficacy and higher safety profile.

Variation in fatty acid composition of giant panda (Ailuropoda melanoleuca) milk fats during different lactation stages

Xi Cao,Miao Qiao,Yayu Pan,Qingbin Bao,Guiquan Zhang,Yikun Xian

The fatty acid (FA) composition of giant panda colostrum and the variation during lactation were determined. Of the 35 FAs detected in giant panda milk fat , the most abundant were oleic acid , palmitic acid and linoleic acid . There were 14 very long-chain and 5 short- and medium-chain FAs detected. The ratio of n-6 to n-3 FAs was 3.60–7.21. Hierarchical clustering analysis demonstrated there was an unusually slow transition from colostrum to mature milk, probably an adaptive strategy. The oleic acid/linoleic acid ratio was higher in colostrum than mature milk, which may benefit digestion. Most saturated FAs were more abundant in mature milk than colostrum. Arachidonic acid accounted for 48.49% of very long-chain polyunsaturated FAs in colostrum, which was 3.34 fold that in mature milk, and for docosahexaenoic acid , 2.75 fold. During the lactation, Spearman correlations within unsaturated and saturated FAs were generally positive; correlations between them were generally negative.

Pharmacokinetics of ceftiofur sodium in cats following a single intravenous and subcutaneous injection

Mei Zhang,Fang Yang,Hua-jie Yu,Tian-jing Kang,Yong-hui Ding,Meng-li Yu,Qian-kun Wang,Yuan-xin Zhu,Fan Yang

Ceftiofur, a third-generation cephalosporin antibiotic, is being extensively used by pet doctors in China. In the current study, the detection method was developed for ceftiofur and its metabolites, desfuroylceftiofur (DCE) and desfuroylceftiofur conjugates (DCEC), in feline plasma. Then, the pharmacokinetics studies were performed following one single intravenous and subcutaneous injection of ceftiofur sodium in cats both at 5 mg/kg body weight (BW) (calculated as pure ceftiofur). Ceftiofur, DCE, and DCEC were extracted from plasma samples, then derivatized and further quantified by high-performance liquid chromatography. The concentrations versus time data were subjected to noncompartmental analysis to obtain the pharmacokinetics parameters. The terminal half-life ( t 1/2λz ) was calculated as 11.29 ± 1.09 and 10.69 ± 1.31 hr following intravenous and subcutaneous injections, respectively. After intravenous treatment, the total body clearance (Cl) and volume of distribution at steady-state ( V SS ) were determined as 14.14 ± 1.09 ml hr -1 kg -1 and 241.71 ± 22.40 ml/kg, respectively. After subcutaneous injection, the peak concentration ( C max ; 14.99 ± 2.29 μg/ml) was observed at 4.17 ± 0.41 hr, and the absorption half-life ( t 1/2ka ) and absolute bioavailability (F) were calculated as 2.83 ± 0.46 hr and 82.95%±9.59%, respectively. The pharmacokinetic profiles of ceftiofur sodium and its related metabolites demonstrated their relatively slow, however, good absorption after subcutaneous administration, poor distribution, and slow elimination in cats. Based on the time of drug concentration above the minimum inhibitory concentration (MIC) (T>MIC) calculated in the current study, an intravenous or subcutaneous dose at 5 mg/kg BW of ceftiofur sodium once daily is predicted to be effective for treating feline bacteria with a MIC value of ≤4.0 μg/ml.

Gold Nanoparticles-Based Colorimetric Assay for Cathepsin S Activity

Xianghan Li,Hongyan Shu,Xinna Xu,Yalan Wang,Lingzhi Zhao,Ying Liu,Juanjuan Peng

Cathepsin S (CatS) is a lysosomal enzyme belonging to the papain family of cysteine proteases. Studies have demonstrated that CatS plays a pivotal role in antigen presentation and is associated with Alzheimer's disease, atherosclerosis, and obesity, positioning it as a potential target in related disorders. Consequently, the development of a selective and straightforward method for detecting CatS is crucial. In this study, we introduce a colorimetric approach utilizing gold nanoparticles (AuNPs) and CatS-specific peptides for the detection of CatS activity. The CatS enzyme cleavage peptides, PMGAPC or PMGAPC-NH 2 , can stabilize AuNPs in a hydrosaline solution without aggregation. Upon hydrolysis of PMGAPC or PMGAPC-NH 2 by CatS, the AuNPs aggregate with a color change from red to blue. After optimizing the size of the AuNPs and the concentration of the peptides, the limits of detection for CatS were both to be 10 nM, when PMGAPC or PMGAPC-NH 2 were used to protect the AuNPs. Therefore, this colorimetric assay, which employs peptides and AuNPs, enables not only a selective but also a simple detection of CatS, visible to the naked eye.

Sophoraflavanone G as an Ectosteric Inhibitor of Cathepsin K Attenuates Ovariectomy-Induced Bone Loss by Suppressing Bone Resorption

Xueling Hu,Chunxia Liu,Ziling Tang,Mingyu Pan,Ailing Fang,Ling Li,Xiangbo Meng,Xiyang Tang,Yanzhi Liu,Xinluan Wang,Hao Gao,Jian Zou,Zuocheng Qiu

Background Cathepsin K (CTSK) is a key enzyme in bone resorption, making it a promising target for osteoporosis treatment. Active-site inhibitors of CTSK are effective but have undesirable side effects, while ectosteric inhibitors may provide a safer alternative. Purpose This study investigates whether Sophoraflavanone G (SG), derived from Rhizoma Drynariae , can act as an ectosteric CTSK inhibitor to attenuate osteoporotic bone loss and explores its underlying mechanisms. Study Design SG's effects were evaluated in an ovariectomized (OVX) osteoporotic mice model, with in vitro experiments assessing SG's interaction and binding affinity with CTSK. Methods Micro-CT, histology, and mechanical testing were used to evaluate bone density and strength. CTSK activity and expression were assessed by immunohistochemistry and western blotting. Cell thermal shift assays, isothermal titration calorimetry, CTSK site-specific degradation assays, molecular docking and dynamic simulation were performed to study SG's binding affinity and inhibitory effects. Biosafety, including body weight, uterine histomorphometry, and toxicity of the heart and lung, was also assessed. Results SG improved bone mineral density, microarchitecture, and strength, primarily by inhibiting bone resorption. It inhibited CTSK's enzymatic activity with a strong binding affinity (KD: 8.49 μM) and effectively inhibited osteoclast function. CTSK site-specific assays showed SG inhibited CTSK-mediated degradation of type I collagen. Unlike odanacatib, SG did not affect gelatin or TGF-β1 degradation in fibroblasts. Biosafety assessments revealed no adverse effects. Conclusion SG acts as an ectosteric CTSK inhibitor, offering a safer alternative for postmenopausal osteoporosis treatment by selectively inhibiting bone resorption without the side effects associated with active-site inhibitors.

Generation of Olfactory Compounds in Cat Food Attractants: Chicken Liver-Derived Protein Hydrolysates and Their Contribution to Enhancing Palatability

Yuyan Wei,Ling Xie,Bertrand Muhoza,Qian Liu,Shiqing Song

The present study investigated the impact of four chicken liver protein hydrolysate-based cat food attractants on palatability. Aroma compounds were analyzed in these attractants, which were subsequently sprayed onto four different types of cat foods. Results revealed that CF4 exhibited the highest intake ratio and the first choice ratio, followed by CF2 sample. Orthogonal partial least-squares discriminant analysis (OPLS-DA) demonstrated significant differences among 50 volatile compounds identified from the four cat foods. Using variable importance in projection (VIP) values, we selected 17 key flavor compounds responsible for distinguishing between the four cat foods. Peptides with a molecular mass <180 Da showed correlation with nonanoic acid and cedrol, while those >3000 Da correlated with hexanoic acid ethyl ester. Regression coefficients (RCs) calculated from partial least-squares regression (PLSR) results showed positive correlations between compound content and palatability for six compounds, whereas negative correlations were observed for ten compounds. Validation experiments confirmed that nonanal, 2-propylpyridine, and 3-octen-2-one enhanced palatability and correlated with peptides ranging from 180 to 500 Da; conversely, nonanoic acid ethyl ester and 3-methyl-pentanoic acid reduced palatability and correlated with peptides ranging from 1000 to 3000 Da.

Ning Sun,Dou Wang,Guoqiang Yao,Xiaomei Li,Ting Mei,Xinke Zhou,Kwok-Yin Wong,Baishan Jiang,Zhiyuan Fang

Background: The intraoperative visualization of tumor cells is a powerful modality for surgical treatment of solid tumors. Since the completeness of tumor excision is closely correlated with the survival of patients, probes that can assist in distinguishing tumor cells are highly demanded.Purpose: In the present study, a fluorescent probe JF1 was synthesized for imaging of tumor cells by conjugating a substrate of cathepsin B (quenching moiety) to Oregon Green derivative JF2 using a self-immolative linker. Methods: JF1 was then loaded into the folate-PEG modified CaCO3 nanoparticles. The folate receptor-targeted, pH-dependent, and cathepsin B activable CaCO3 nanoprobe was test in vitro and in vivo for tumor imaging. Results: CaCO3 nanoprobe demonstrated good stability and fast lighting ability in tumors under low pH conditions. It also showed lower fluorescence background than the single cathepsin B dependent fluorescent probe. The pH-dependent and cathepsin B controlled “turn-on” property enables precise and fast indication of tumor in vitro and in vivo. Conclusion: This strategy of controlled drug delivery enables in vivo imaging of tumor nodules with a high signal-to-noise ratio, which has great potential in surgical tumor treatment.

Glass catfish inspired subaquatic abrasion-resistant anti-fouling window fabricated by femtosecond laser electrodeposition

Jialiang Zhang,Fangzheng Ren,Qing Yang,Qingyun Ma,Jie Liang,Yizhao Meng,Xiaodan Gou,Chongxiao Xia,Feng Chen

Transparent materials utilized as underwater optical windows are highly vulnerable to various forms of pollution or abrasion due to their intrinsic hydrophilic properties. This susceptibility is particularly pronounced in underwater environments where pollutants can impede the operation of these optical devices, significantly degrading or even compromising their optical properties. The glass catfish, known for its remarkable transparency in water, maintains surface cleanliness and clarity despite exposure to contaminants, impurities abrasion, and hydraulic pressure. Inspired by the glass catfish's natural attributes, this study introduces a new solution named subaquatic abrasion-resistant and anti-fouling window (SAAW). Utilizing femtosecond laser ablation and electrodeposition, the SAAW is engineered by embedding fine metal bone structures into a transparent substrate and anti-fouling sliding layer, akin to the sturdy bones among catfish's body. This approach significantly bolsters the window's abrasion resistance and anti-fouling performance while maintaining high light transmittance. The sliding layer on the SAAW's surface remarkably reduces the friction of various liquids, which is the reason that SAAW owns the great anti-fouling property. The SAAW demonstrates outstanding optical clarity even after enduring hundreds of sandpaper abrasions, attributing to the fine metal bone structures bearing all external forces and protecting the sliding layer of SAAW. Furthermore, it exhibits exceptional resistance to biological adhesion and underwater pressure. In a green algae environment, the window remains clean with minimal change in transmittance over one month. Moreover, it retains its wettability and anti-fouling properties when subjected to a depth of 30 m of underwater pressure for 30 d. Hence, the SAAW prepared by femtosecond laser ablation and electrodeposition presents a promising strategy for developing stable optical windows in liquid environments.

Experimental and emulational study on the role of ion in coal adsorbing kerosene: Water–kerosene interface and catenoid characteristics

Yong Zhang,Hongzheng Zhu,Jinbo Zhu,Fan Yang,Hailing He,Zhiqian Qin,Qinghui Shi,Gaochao Pan

Understanding the effect of ions on the interfacial tension (IFT) between kerosene and water is a prerequisite to predicting kerosene–mineral adsorption. Using a high-speed motion acquisition system, we investigated the height evolution of the catenoid with different IFTs. As the IFT increased, the height of catenoid first increased slowly and then increased sharply. The catenoid height of the oil–water interface was larger than was that of the water–gas, whereas the IFT was the opposite, indicating that height was related not only to the IFT, but also to the viscosity of the phases when the ring diameter was constant. The cation valence had a significant influence on the IFT. Interaction between the cation and water molecules led to the generation of cationic clusters. The literature indicates that the higher the valence of ion the easier it would be to form a cationic cluster, resulting in decreasing of the IFT. Our results matched these findings. At low ionic concentration , the IFT increased as the ion concentration increased. At high ionic concentration, the IFT decreased with increasing concentration because of the ionic pairs. The results of molecular dynamics simulation showed that the water molecules only moved toward kerosene molecules without ions. The hydrophilic groups in kerosene molecules spontaneously arranged toward the water–oil interface, whereas the hydrophobic groups stayed away from this interface. The water molecules burst into the kerosene phase in the presence of ions. The number of water molecule entering the kerosene phase increased with increasing ion concentrations. Our results can provide valuable insight into the development of technology for surfactant adsorption and mineral flotation .
