Activities of polysaccharide fractions from corn silk: Hemostatic, immune, and anti-lung cancer potentials

Yunwen Zhu,Yaping Li,Xue Li,Tongfei Chen,Hepeng Zhao,Hongli Zhou

Corn silk is the stigma and style of corn and is rich in polysaccharides. Despite the extensive research on its polysaccharides, the hemostatic characteristics of effective parts and the related activities remain insufficiently explored. Corn silk polysaccharide (CSP) was extracted with hot water and purified using a diethylaminoethyl cellulose membrane. Then, it was separated with sephadex G-150 to obtain five fractions. These fractions were investigated for their potential in hemostasis, antioxidant, immune response, and anti-lung cancer activities. CSP-2, CSP-3, and CSP-4 significantly affected the coagulation indicators activated partial thromboplastin time (APTT) and thrombin time (TT) at 125–500 μg/mL. Corn silk flavonoids and saponins at 32.25 μg/mL significantly prolonged APTT, TT, and prothrombin time (PT). CSP-2, with potent antioxidant ability, approaches Vitamin C. At 25 μg/mL, CSPs nearly reached the phagocytosis of neutral red of lipopolysaccharides. The five fractions promoted the proliferation of RAW264.7 cells at 25–800 μg/mL and stimulated NO secretion at 25–100 μg/mL. CSP-2 also showed an 86 % inhibition rate effect on A549 at 200 μg/mL. These results indicate that CSP not only has hemostatic effects but also has immune and anti-lung cancer activities. Thus, it is a potential candidate compound with immune activity for managing bleeding in cancer.

Enhanced Performance in Supercapacitor Supported by Corn Silk-Derived Porous Carbon

Guo Congxiu,Sun Ya,Wang Bing,Wang Yunwei,Wu Jiao,Tong Xili

N, O self-doped hierarchical porous carbon has been successfully synthesized through a facile chemical activation method using corn silk as raw material, where the texture properties of the samples can be tailored via changing the quantity of the Na 2 CO 3 activator. The optimal sample with a BET surface area of 1269.79 m 2  g −1 and a nitrogen content of 2.48 at% exhibited the highest specific capacitance of 158.2 F g −1 and 196.3 F g −1 at 1 A g −1 in basic and acidic solution, respectively. In addition, the high capacitance contributions (86.2% in basic solution and 72.6% in acidic solution at 10 mV s −1 ) imply a capacitive controlled mechanism. These results indicate that the synthesis of heteroatom-doped porous carbon from biomass can be a potential approach for developing effective materials in supercapacitors.

Effects of Different Amounts of Corn Silk Polysaccharide on the Structure and Function of Peanut Protein Isolate Glycosylation Products

Xinxin Han,Yilin Zhao,Sining Mao,Nannan Hu,Dijing Sun,Qi Yang,Zejun Chu,Qihang Zheng,Lin Xiu,Jingsheng Liu

Covalent complexes of peanut protein isolate (PPI) and corn silk polysaccharide (CSP) (PPI-CSP) were prepared using an ultrasonic-assisted moist heat method to improve the functional properties of peanut protein isolate. The properties of the complexes were affected by the level of corn silk polysaccharide. By increasing the polysaccharide addition, the grafting degree first increased, and then tended to be flat (the highest was 38.85%); the foaming, foam stability, and solubility were also significantly improved. In a neutral buffer, the solubility of the sample with a protein/polysaccharide ratio of 2:1 was 73.69%, which was 1.61 times higher than that of PPI. As compared with PPI, the complexes had higher thermal stability and lower surface hydrophobicity. High addition of CSP could made the secondary structure of PPI change from orderedα-helix to disorderedβ-sheet,β-turn, and random coil structure, and the complex conformation become more flexible and loose. The results of multiple light scattering showed that the composite solution exhibited high stability, which could be beneficial to industrial processing, storage, and transportation. Therefore, the functional properties of peanut protein isolate glycosylation products could be regulated by controlling the amount of polysaccharide added. Keywords:peanut protein isolate;glycosylation;corn silk polysaccharide;functional properties

Corn stigma ameliorates hyperglycemia in zebrafish and GK rats of type 2 diabetes

Haowei Liang,Ruiqin Zhang,Li Zhou,Xiaolong Wu,Jingan Chen,Xinyue Li,Jieqiong Chen,Letian Shan,Hui Wang

Ethnopharmacological relevance Corn stigma (CS), derived from the stigma and style of gramineous plant Zea mays . The medicinal use of CS can be traced back to Dian Nan Materia Medica . Lingnan Medicinal Plants Compendium records its effectiveness in ameliorating diabetes. Diabetes is a metabolic disorder characterized by hyperglycemia and the consequent chronic complications of kidney, heart, brain and other organs, which pose a significant threat to human health. CS has shown great potential in relieving hyperglycemia associated with diabetes. However, the mechanism of CS in treating diabetes remains unclear. Aim of the study To explore the pathogenesis of diabetes and the mechanism of CS improving hyperglycemia in diabetes. Materials and methods We measured apigenin and luteolin contents in CS by UPLC/MS/MS method. Selecting Wistar rats as normal group, and GK rats as model group. For rats, we detected glucose and lipid metabolism indicators, including GHb, AST, ALT, U-Glu, UA, U-TP, U-ALB, and ACR after treatment. For zebrafish, we utilized alloxan and sucrose to establish the diabetes model. Measuring zebrafish blood glucose is employed to evaluate the hypoglycemic capability of CS. In order to explore the mechanism of CS in treating diabetes, we sequenced the transcriptome of zebrafish, compared differentially expressed genes of normal, diabetic, and CS-treated group, and validated multiple enrichment pathways by PCR. Results CS can improve blood glucose levels in both GK rats and diabetic zebrafish. For rats, CS partially restored glucose and lipid metabolism indicators. Transcriptome data from zebrafish showed a close correlation with steroid biosynthesis. The RNA-Sequencing was consistent with PCR results, indicating that CS downregulated gene ( fdft1, lss, cyp51 ) expression concerned with steroid biosynthesis pathway in the diabetes model. Conclusion CS effectively improved blood glucose levels, regulated glucose and lipid metabolism by suppressing gene expression in steroid biosynthesis pathway, and ameliorated hyperglycemia. Our research provides valuable insights for CS in the treatment of diabetes, and proposes a new strategy for selecting clinical medications for diabetes.

Improving the flavor, bioactivity, and metabolic characteristics of corn silk by probiotic co-fermentation

Yuhan Yuan,Ruijuan Wang,Xuefeng Wu,Dongdong Mu,Li Guo,Xingjiang Li

Corn silk, an agricultural by-product rich in bioactive compounds, has long remained underutilized. In this study, we systematically examined how probiotic co-fermentation influences the bioactivity, flavor profile, and metabolic landscape of corn silk. The results demonstrated that fermentation significantly enhanced the total phenolic content by 73.31 % and flavonoid content by 80.20 %, along with a substantial improvement in antioxidant capacity. Volatile compound analysis identified 36 key aroma-related substances. Notably, 4-methylguaiacol and 4-ethylguaiacol contributed to both improved flavor and antioxidant function, highlighting a synergistic relationship between sensory quality and bioactivity during fermentation. Non-targeted metabolomics further revealed substantial changes in the metabolic profile post-fermentation, with 110 differential metabolites identified—75 upregulated and 35 downregulated. These metabolites were primarily enriched in the TCA cycle, phenylpropanoid biosynthesis, and amino acid metabolism pathways. Together, these findings uncover the molecular mechanisms by which probiotic fermentation remodels metabolic networks to enhance both the functional and sensory attributes of corn silk. This work offers a theoretical foundation and technical guidance for the high-value utilization of agricultural by-products and the development of functional foods.

Screening of uric acid-lowering active components of corn silk polysaccharide and its targeted improvement on renal excretory dysfunction in hyperuricemia mice

Xizhu Wang,Liyan Yuan,Zhijie Bao,Baoshang Fu,Pengfei Jiang,Tiecheng Ma,Songyi Lin

To screen the in vitro uric acid-lowering activity of the neutral polysaccharides (NCSP) and acid polysaccharides (ACSP) isolated from the corn silk were evaluated. Furthermore, preliminary structure-activity relationships for them were investigated. The molecular weight (Mw) of NCSP and ACSP were 74.1 and 109.4 kDa, respectively. NCSP consisted of mannose (Man), glucuronic acid (GlcA), glucose (Glc), galactose (Gal), and arabinose (Ara), in addition to these monosaccharides, the ACSP contains galactose acid (GalA). NMR spectroscopy showed that the NCSP main chain consisted of 1,4,6)- α -D-Glc p , 1,4)- α -D-Glc p , 1,3)- β -D-Gal p , and 1,3,5)- β -D-Man p . NCSP exhibited higher activity in anti-hyperuricemia (HUA). The results indicated that serum uric acid (UA) and liver xanthine oxidase (XO) activities levels decreased by 45.71% and 22.4% in the NCSP groups, respectively. In addition, NCSP could significantly improve the kidney damage and promote uric acid excretion. These findings provide evidence in support of NCSP as a potential dietary supplement for improving HUA.

Hypouricemia effects of corn silk flavonoids in a mouse model of potassium oxonated-induced hyperuricemia

Liyan Yuan,Zhijie Bao,Tiecheng Ma,Songyi Lin

The hypouricemic effect of corn silk flavonoids (CSFs) in vivo that were extracted by ethanol and fractionated by continuous elution with 40% (CSF-A) and 60% (CSF-B) ethanol solutions on polyamide column were investigated in this study. CSFs reduced serum uric acid (UA) level in a mouse model of potassium oxonate-induced hyperuricemia. CSF-B had the best hypouricemic effect, as it decreased the serum UA level by 26.69% and xanthine oxidase (XO) activity in the serum by 11.29%. The mechanism of action of CSF-B was related to the inhibition of XO activity and the promotion of UA excretion. CSF-B was found to contain 12 kinds of major flavonoids, five of which were speculated to influence its activity in the hyperuricemia mice. The five flavonoids were apigenin-6-C-glucoside-7-O-glucoside, kaempferol-3-O-rutinoside, luteolin-7-glucoside, luteolin-3′,7-di-O-glucoside, and naringenin, respectively. Structure analysis revealed that C-4′, C5 hydroxyl groups, and C2=C3 double bonds in CSF-B gave the latter its hypouricemic effect. Practical applications The prevalence of hyperuricemia has increased in recent times. Current hypouricemic drugs have side effects and can easily lead to various complications. Therefore, it is of great practical significance to find safer and more effective hypouricemic drugs. This study demonstrated that corn silk flavonoids may be used as a dietary supplement to manage hyperuricemia.

Structural characterization and hypolipidemic activities of purified stigma maydis polysaccharides

Wenwen Deng,Xia Yang,Yuan Zhu,Jiangnan Yu,Ximing Xu

This study aimed to investigate structural features and antihyperlipidemic effects of the stigma maydis polysaccharide, termed SMP-1. This polysaccharide was composed of D-mannose, L-rhamnose, D-glucose, D-galactose, L-arabinose, D-xylose, and D-galacturonic acid, with a molar ratio of 1.00:0.21:1.41:1.44:0.70:0.44:0.56. The SMP-1 was mainly bonded by (1 → 6) and (1 → 3) linkages, with various monosaccharides being evenly distributed in the main and side chains. Moreover, SMP-1 had neither triple-helical structure nor molecular aggregation. Importantly, the SMP-1 could effectively bind the bile acids in vitro and significantly lower the total cholesterol, triglyceride, low-density lipoprotein cholesterol levels, and moderately increase the high-density lipoprotein cholesterol level in poloxamer 407-induced hyperlipidemic mice. Moreover, pretreatment with SMP-1 (≥300 mg/kg) could remarkably reduce fat accumulation and restore hepatocyte morphology in the liver of hyperlipidemic mice. Altogether, these findings indicated that SMP-1 could be developed as a safe and effective food supplement for preventing and treating hyperlipidemic disorders.

Shankaranarayana Sahana Hosanagara,Gajanana Vijayalaxmi Karki,Chavan Mohan,Chavannavar Suvarna V.,Doddanagappa Shobha

Purpose To analyse phytochemical composition, antioxidant potential, antimicrobial activity and α-amylase, α-glucosidase and xanthine oxidase inhibitory potential of corn silk extracts derived from the Syngenta 5414 corn variety. Methods Corn silk sample was lyophilized and extracted using 80% ethanol, methanol, water or acetone. Phytochemicals were qualitatively screened and bioactive compounds were identified through GCMS. Antimicrobial activities were evaluated using the agar well method for bacteria and fungal biomass method for fungi. The extracts were subjected to evaluation for their antidiabetic activity through α-amylase and α-glucosidase inhibitory assays, while their anti-gout potential was assessed through xanthine oxidase inhibitory assay, encompassing all four solvent extracts in the study. Results Phytochemical screening revealed the presence of phenols, flavonoids, tannins, alkaloids, sterols, protein xanthoproteic, and cardiac glycosides in the extracts. The Folin-Ciocalteu and AlCl 3 assays demonstrated higher yields of phenolics and flavonoids in 80% ethanolic extract. Furthermore, the extracts remarkably scavenged reactive oxygen species like DPPH, ABTS and elicited considerable ferric ion reducing antioxidant power. GC–MS analysis identified 21 compounds in the 80% ethanolic extract, with 1,2,3-Propanetriol-1-acetate, n-Hexadecanoic acid, and 4H-Pyran-4-one,2,3-dihydro-3,5-dihydroxy-6-methyl- as major components. Antimicrobial assays demonstrated the ethanolic (80%) and methanolic extract's remarkable ability to inhibit the growth of Gram-positive bacteria and exhibit anti-fungal properties. The ethanolic (80%) extract exhibited an IC 50 value of 0.70 ± 0.02 and 0.71 ± 0.01 mg against α-amylase and α-glucosidase, respectively, and 33.76 ± 0.43 mg against xanthine oxidase. Conclusion Corn silk exhibits potential as a valuable reservoir of natural bioactive compounds endowed with a wide array of health-promoting properties. Graphical

A comparison of chemical composition, bioactive components and hypoglycemic activity of Stigma maydis obtained from different growing times

Yi Zhang,Jinxin Pang,Lianmou Yao,Chunfang Wang,Bingjie Chen,Qi Zheng,Yingxiong Hu,Yongjin Qiao

Stigma maydis (SM) possesses remarkable nutritional value due to the presence of health-protective chemical constituents in it. Despite its commercial uses, there is a lack of information on changes in the nutritional profile of SM during Shenke 601 waxy corn development by following four different maturity stages (silking stage; blister stage; milky stage; dough stage). In this study, the ideal times for the development of seven active compounds (polysaccharides, saponin, rutin, luteolin, quercetin, kaempferol and chlorogenic acid) were at the silking stage. The total flavonoid and total phenol content increased as the SM matured. Except for the above-mentioned active compounds, the content of major nutritional components of dough stage was higher than that of three other stages. The extract of Stigma maydis exhibited good α-amylase, α-glucosidase and pancreatic lipase inhibition activities. The ethanolic SM extracts showed better inhibitory effects than aqueous SM extracts with a dose–effect relationship. The hypoglycemic activity of ethanolic extract was higher than that of aqueous extract at each stage. This provided an important basis for the application of SM.
