Current continuous flow polymerase chain reaction (CF-PCR) microfluidic chips require external precision syringe pumps and off-line methods (e.g., electrophoresis and hybridization) to detect PCR products, resulting in complex operations and possible cross-contamination and consequently CF-PCR is still confined to laboratories. Herein, a portable all-in-one microfluidic device is fabricated for rapid diagnosis of pathogens based on an integrated CF-PCR and electrophoresis biochip. A new method was proposed for automatic sample injection into the chip which can substitute the costly external precision syringe pump. It not only achieves rapid DNA amplification and on-site PCR product detection, but also realizes automatic sample injection. As an application, three periodontal pathogens (e.g., Porphyromonas gingivalis, Treponema denticola and Tannerela forsythia) were successfully amplified in the device. Treponema denticola was amplified in as short as 2′31′′, and detection of PCR products was completed within 3′43′′. The minimum number of bacteria that can be amplified was 125 cfu per μl. The all-in-one device has the potential to be applied in point-of-care nucleic acid testing for diseases.

In most circumstances, wounds face the challenges of bacterial invasions and inappropriate inflammatory responses when they lack proper wound management. Endowing dressings with both antibacterial and anti-inflammatory functions is a compelling strategy for resolving the above issues. However, seizing the right moment to change the dressings and providing satisfactory management of wounds are still urgently required. Herein, an antibacterial and anti-inflammatory nanofibrous mat is proposed by encapsulating antibiotic gentamicin sulfate (GS) and anti-inflammatory drug ibuprofen (IB) into nanofibers via a coaxial electrospinning technique and is further decorated with Prussian blue nanocrystals (PBNCs) to enhance anti-inflammatory activity and, more importantly, to monitor bacterial infections and guide dressing changes in a timely manner. Such a nanofibrous mat releases most of the therapeutic drugs within 120 min and reveals excellent antibacterial activity and anti-inflammatory ability. Specifically, it can destroy both Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), as well as conspicuously reduce the production of reactive oxygen species (ROS) and the expression of pro-inflammatory cytokines in macrophages. In addition, the nanofibrous mat can be used for point-of-use diagnosis of living bacteria relying on the naked eye or color analysis, which exhibits the potential of monitoring wound infection and guiding dressing changes promptly. This finding demonstrates the theranostic applications of multifunctional nanofibrous mats in wound healing.

Furunculosis, which is caused by Aeromonas salmonicida, can induce septicemia, leading to the rapid death of fishes belonging to Salmonidae, Cyprinidae, and Fuscheridae, and lamprey. Targeting A. salmonicida, five DNAzyme sequences with the highest enrichment rates were selected through the Systematic Evolution of Ligands by Exponential Enrichment (SELEX). The enrichment rates were 34.78, 23.60, 8.91, 2.89, and 2.34%, respectively. The DNAzyme with the highest activity, named D-AS-2, showed specificity and sensitivity. D-AS-2 was combined with carboxyl-functionalized graphene to construct a biosensor, which showed good fluorescence response to scabies lesion samples. The diagnostic procedure was completed in <2 min and can be used for the on-site diagnosis of fish diseases. A low-cost, rapid, simple, and highly specific biosensor for the diagnosis of furunculosis was established based on DNAzyme and carboxyl-functionalized graphene.

Most of the current cathodic photoelectrochemical (PEC) bioassays either rely on dissolved oxygen as a signal reporting molecule or are interfered by atmospheric oxygen (if using other molecules than oxygen as the signaling molecule), which leads to their monotonous detection mechanism, vulnerable/nonadjustable performance, complicated instrument and time consuming operation. Herein, we develop an innovative, oxygen independent cathodic PEC bioassay with distinct advantages of robust performance, simple instrument/facile operation, as well as sensitive and prompt readout. Specifically, taking the ochratoxin A (OTA) as a model analyte, the detection combines the largely augmented photocurrent of flower-like bismuth oxyiodide (BiOI) microspheres by potassium ferricyanide (K 3 [Fe(CN) 6 ]) under aerobic conditions, the highly selective aptamer recognition as well as the powerful exonuclease-catalyzed target recycling amplification. Removal of the surface immobilized DNA under the cooperative effects of OTA induced double-stranded DNA (dsDNA) dissociation and RecJ f exonuclease mediated digestion of the released single-stranded DNA results in enhanced flux of K 3 [Fe(CN) 6 ] to the photoelectrode, enables a “signal-on” and “label-free” mode for OTA detection with high sensitivity and good selectivity. It also shows acceptable results for the detection of OTA spiked in wine samples. Since the detection takes place in ambient air and is not interfered by the atmospheric oxygen, this facile and robust cathodic PEC biosensing system displays promising applicability for on-site and real-time diagnosis. More significantly, it is extensible as a general strategy for versatile cathodic PEC bioanalysis.

The integration of smartphones with conventional analytical approaches plays a crucial role in enhancing on-site detection platforms for point-of-care testing. Here, we developed a simple, rapid, and efficient three-channel colorimetric sensor array, leveraging the peroxidase (POD)-like activity of polydopamine-decorated FeNi foam (PDFeNi foam), to identify antioxidants using both microplate readers and smartphones for signal readouts. The exceptional catalytic capacity of PDFeNi foam enabled the quick catalytic oxidation of three typical peroxidase substrates (TMB, OPD and 4-AT) within 3 min. Consequently, we constructed a colorimetric sensor array with cross-reactive responses, which was successfully applied to differentiate five antioxidants (i.e., glycine (GLY), glutathione (GSH), citric acid (CA), ascorbic acid (AA), and tannic acid (TAN)) within the concentration range of 0.1–10 μM, quantitatively analyze individual antioxidants (with AA and CA as model analytes), and assess binary mixtures of AA and GSH. The practical application was further validated by discriminating antioxidants in serum samples with a smartphone for signal readout. In addition, since pesticides could be absorbed on the surface of PDFeNi foam through π-π stacking and hydrogen bonding, the active sites were differentially masked, leading to featured modulation on POD-like activity of PDFeNi foam, thereby forming the basis for pesticides discrimination on the sensor array. The nanozyme-based sensor array provides a simple, rapid, visual and high-throughput strategy for precise identification of various analytes with a versatile platform, highlighting its potential application in point-care-of diagnostic, food safety and environmental surveillance.

We report a low-cost and convenient microchannel resistance (MCR) biosensing platform that uses current signal to report biorecognition. The biorecognition behavior between targets and biometric molecules (antigens, antibodies, or oligonucleotides) immobilized on magnetic beads and polystyrene (PS) microspheres induces a quantitative change in the unreacted PS microspheres. After magnetic separation, the unreacted PS microsphere solution is passed through the microchannel, leading to an obvious blocking effect, resulting in an increase in resistance, which can in turn be measured by monitoring the electric current. Thus, the biorecognition is directly converted into a detectable current signal without any bulky instruments or additional chemical reactions. The MCR biosensing platform is cost-effective and user-friendly with high accuracy. It can be an appropriate analysis technique for point-of-care testing in resource-poor settings.
