Multifunctional Polyacrylonitrile-based Pearl Nanofibrous Membrane Prepared by Electrospinning

Yu Miao,Chen Zhuoming,Xin Binjie,Liu Xunlin,Xu Wenjun

Pearl is a pure natural material, which has multifunctional properties such as anti-ultraviolet, anti-static and far-infrared physiotherapy. In this study, a series of polyacrylonitrile/pearl nanofibrous membranes (PPFM) with different pearl powders were prepared by the electrospinning method. The membranes were then investigated by scanning electron microscopy (SEM) and fourier-transform infrared spectroscopy (FTIR), and their functional properties such as far-infrared emission, ultraviolet (UV) protection, and electrical resistivity were also studied. The experimental results reveal that pearl powders can significantly enhance the far-infrared emission of polyacrylonitrile (PAN), and the average emissivity was increased up to 0.795 in the wavelength range of 8–14 μm when the pearl powder mass fraction increased to 9 %. In addition, the UV protection factor (UPF) of the membrane with 9 % pearl powder could reach 968, while the UPF of the pristine PAN membrane was only 11, and the electrical resistivity of PPFM was also improved after blending with pearl powders. The study proves that pearl powder can efficiently endow the PAN with multifunctional properties for more promising applications.

Nano-pearl powder/chitosan-hyaluronic acid porous composite scaffold and preliminary study of its osteogenesis mechanism

Xiaoni Li,Pu Xu,Yanan Cheng,Wenbai Zhang,Bin Zheng,Qiuzi Wang

Bone tissue engineering using osteoinductive scaffolds has evolved into a promising approach for bone regeneration. Pearl powder has recently gained interest in bone regeneration due to their bioactive characteristics and favorable mechanical properties. In order to mimic bone tissue structurally and compositionally, chitosan-hyaluronic acid (C-HA) scaffolds containing nano-pearl powder (NPP) were fabricated in current study by using freeze-drying method. The microstructure, porosity, hydrophilia, and mechanical property of scaffolds were studied by scanning electron microscopy (SEM), mercury porosimeter, contact angles, and universal material experiment machine respectively. The crystal form of NPP, which was packed in scaffolds, was tested by X-ray diffraction (XRD), and the interaction between three constituents was studied by Fourier transformed infrared (FTIR). In the other hand, the biocompatibility, osteogenic characteristics, and osteogenic related gene expression of scaffolds were examined by cell counting kit-8 (CCK-8), alkaline phosphatase activity (ALP), RT-qPCR, and Western blotting. The results revealed that the NPP was successfully incorporated in the C-HA scaffolds. The hydrophilia and mechanical properties of scaffolds increased with NPP content, whereas the pore morphology was disturbed, especially for 25% scaffold. The scaffolds had a high porosity between 89% and 93%, and decreased slightly with the increase of NPP. Cell culture tests indicated that scaffolds with higher NPP proportion were beneficial for the proliferation and differentiation of MC3T3-E1 cells, and scaffolds with 10 wt% and 25 wt% nano-pearl powder were most effective. The differentiation might be promoted by upregulating the expression of Col αI, OCN, OPN and Runx2 genes. Therefore, the nano-pearl powder/chitosan-hyaluronic acid (NPP/C-HA) scaffolds may serve as a promising biomaterial for bone tissue engineering.

3D printing nacre powder/sodium alginate scaffold loaded with PRF promotes bone tissue repair and regeneration

Bin Liu,Cewen Hu,Xinyue Huang,Kaiqi Qin,Lei Wang,Zhilong Wang,Jiachen Liang,Fuqiang Xie,Zengjie Fan

Bone defect is a common complication of bone diseases, which often affects the quality of life and mental health of patients. The use of biomimetic bone scaffolds loaded with bioactive substances has become a focal point in the research on bone defect repair. In this study, composite scaffolds resembling bone tissue were created using nacre powder (NP) and sodium alginate (SA) through 3D printing. These scaffolds exhibit several physiological structural and mechanical characteristics of bone tissue, such as suitable porosity, appropriate pore size, applicable degradation performance of bone regeneration and reaching the mechanical requirements of cancellous bone etc. Then, platelet-rich fibrin (PRF), containing a mass of growth factors, was loaded on the NP/SA scaffolds. This aims to fully maximize the synergistic effect with NP , thereby accelerating bone tissue regeneration. Overall, this study marks the first instance of preparing a bionic bone structure scaffold containing NP by 3D printing technology, which combined with PRF to further accelerate bone regeneration. These findings offer a new treatment strategy for bone tissue regeneration in clinical applications.

Effects of Different Extraction Methods on Biological Activities of Pearl Powder Extracts

Wang Wenchao,An Lianhao,Lei Junrong,Li Dan,Shen Zheluan,Wang Hong,Li Qingyong

Pearl powder has been used in medicine and beauty treatment since ancient times in China. However, it is still unclear whether the biological activities of pearl powder are affected by different extraction methods or not. In this study, we determine the total protein yield, protein molecular weight and biological activities including antioxidant, tyrosinase inhibition and melanogenesis inhibition activities and cytotoxicity of different pearl powder extracts prepared by five different preparation methods. The ultrasonic-assisted extraction (UAE) and microwave-assisted extraction (MAE) of pearl powder were optimized by the response surface method based on total protein yield. Compared with solid-liquid extraction (SLE), the optimized UAE and MAE increased the total protein yield without changing protein molecular weight. In addition, the extract by MAE enhanced the tyrosinase inhibition activity and anti-melanogenesis activity slightly. Acidolysis hydrolyzes macromolecular protein into small molecules with obvious cytotoxicity and low activities. Enzymolysis can hydrolyze macromolecular protein into small molecules with high protein yield and improvement of bioactivities. In the determination of the antioxidant assay, the extract prepared by the enzymolysis showed significant activity. These results suggest that protein molecules of the pearl powder extract are degraded appropriately by enzymolysis to smaller molecules to obtain the enhanced biological activities that can be applied in the cosmetic industry.
