Boron nitride/agarose hydrogel composites with high thermal conductivities

Yazdan Ali,Wang Ji-Zhe,Hu Bing-Kun,Xie Wen-Sheng,Zhao Ling-Yun,Nan Ce-Wen,Li Liang-Liang

Abstract Hydrogels are cross-linked polymers suitable for various applications, but the thermal conductivities of hydrogel-based composites have not been thoroughly investigated. In this study, agarose hydrogel-based composites with various boron nitride (BN) fillers were synthesized and their thermal conductivities were systematically investigated. With the increase in the agarose content from 1.5 wt% to 3.0 wt%, the thermal conductivity of the composite decreased. The composites with BN micropowder had larger thermal conductivities than those of the composites with BN nanopowder at the same filler loading, as the BN micropowder provided better thermal conduction pathways in the hydrogel matrix than those provided by the nanopowder. The maximum thermal conductivity of 2.69 W·m −1 ·K −1 was achieved when 15 wt% microscale BN fillers were added into 1.5 wt% agarose hydrogel, which was 3.5 times larger than that of the pure agarose hydrogel. Additionally, a theoretical model was used to calculate the thermal conductivities of the BN/agarose hydrogel composites; a good agreement was achieved between the experimental and fitting ones. This study demonstrated that the thermal conductivities of hydrogel-based materials can be efficiently and significantly enhanced using BN fillers. Graphic Abstract

Green Agarose Hydrogel Implants With High Biocompatibility for Skin Repair and Regeneration in Photoaged Skin

Ying Guo,Bei Tian,Yi Xie,Xiuxia Sun,Jianxi Xiao

Skin aging is a multifaceted process marked by the destruction of skin structure and the diminishment of skin function, significantly impacting both physical and mental health. Injectable hydrogels are promising for skin repair, but chemical crosslinking in most hydrogels can cause cytotoxicity, whereas agarose hydrogels avoid crosslinking yet face injectability challenges. We have herein developed green agarose hydrogel implants with high biocompatibility for skin repair and regeneration in photoaged skin. The hydrogel is prepared by utilizing agarose's ability to dissolve at high temperatures and gel at low temperatures. This process yields implants with particle sizes predominantly ranging from 100 to 150 μm. The agarose hydrogel implant exhibits excellent injectability, with a steady injection force curve consistently around 4 N. The agarose hydrogel implant promotes the proliferation of human dermal fibroblasts and does not induce hemolysis or pyrogenic reactions. In a photoaging mouse model, the agarose hydrogel implant improves skin density and hydration, reduces transepidermal water loss, and stimulates collagen regeneration. This agarose hydrogel implant provides a novel approach to repairing aged skin and holds significant potential in the fields of skin health and tissue regeneration. Graphical

Agarose hydrogel doped with gold nanobipyramids(AuNBPs@AG)as colorful height readout device for sensing hydrogen peroxide in complex sample matrix

Wen Shou,Shuangting Yang,Yueliang Wang,Bin Qiu,Zhenyu Lin,Longhua Guo

The portable and instrument-free sensing device for quantitative detection is of significant applications in resource constrained countries and regions. Here, a colorful height readout device formed by agarose hydrogel doped with gold nanobipyramids (AuNBPs@AG) is developed for the determination of hydrogen peroxide (H 2 O 2 ) content. The responsive mechanism is based on H 2 O 2 guided etching of AuNBPs, which causes the plasmon-mediated color change of the AuNBPs@AG hydrogel filled in plexiglass tube from top to bottom. We demonstrate the height of color-changed hydrogel is quantitatively related to the concentration of H 2 O 2 . The agarose hydrogel, as the carrier material for loading AuNBPs, guarantees the stability of AuNBPs and promotes the separation of complex samples. The method shows a good linear relationship in four ranges of H 2 O 2 concentrations of 20 − 100 μM, 200 − 1000 μM, 1 − 10 mM and 10 − 100 mM. Compared with previous methods, using this inexpensive and easy to operate device for H 2 O 2 detection can effectively reduce the interference of the color background, overcome the problem of the nanoparticles aggregation, and eliminate the need for expensive and sophisticated detection instruments. The practicality of the method is demonstrated via the analysis of commercial products such as contact lens solution and hair dye with satisfactory results. Our work provides a novel path for developing the height-readout quantitative devices with integrating hydrogels and plasmonic nanomaterials.

Leveraging Microgels Prepared from Poly(ethylene glycol) Bisepoxide and Polyetheramine for Versatile Surface Structuring of Agarose Hydrogels

Xueying Yu,Xinnan Wang,Wei He

We demonstrate a macromer-type bisepoxide, poly(ethylene glycol) diglycidyl ether, polymerizing readily with a trifunctional polyetheramine Jeffamine T-403 in water to facilitate the development of a series of microgels abbreviated as PMG. Simply by varying the concentration of the as-prepared thermoresponsive intermediate prepolymer from 1 to 2 and 4%, hydrodynamic sizes of the resulting P1MG, P2MG, and P4MG are easily tuned in the submicrometer to micrometer range shown by the dynamic light scattering results. Besides size difference, these microgels also deform differently, where the drying-induced deformation effect is most severe for P1MG and least prominent for P4MG. Simple evaporative deposition of PMG into multilayer packing provides versatile and green options for microgel-mediated surface structuring of agarose hydrogels. Specifically, deformabile P1MG- and P2MG-derived coatings render agarose gel microwrinkle textures by buckling against swelling-induced surface instability. Conversely, stiffer P4MG microgels lead to a patchy patterned hierarchical coating on agarose, similar to the cracking effect in drying colloidal films. The straightforward microgel-on-macrogel strategy allows integration of both wrinkle and patchy patterns to generate Janus-type agarose gels, just by rationally arranging the coating sequence. Diversifying topographic features attainable through microgel-based coatings on hydrogels could potentially make the sustainable and biocompatible material of agarose a more compelling choice for bioapplications. Brief demonstrations of the broad applicability of P1MG toward wrinkling of proteinaceous and synthetic hydrogels further highlight promising prospects of the PMG microgel-on-macrogel functionalization strategy.

Nano-chitin reinforced agarose hydrogels: Effects of nano-chitin addition and acidic gas-phase coagulation

Jing Liao,Yijin Wang,Bo Hou,Jiamin Zhang,Huihua Huang

Hydrogels based on natural polymers such as agarose usually show low applicability due to their weak mechanical properties. In this work, we developed a dual cross-linked agarose hydrogel by adding different amounts of TEMPO-oxidized nano-chitin (0–0.2 %) to agarose hydrogel matrices and then physically cross-linked under acidic gas-phase coagulation. The prepared hydrogels were characterized by FTIR, XRD, TGA , and SEM. The effects of nano-chitin addition and acidic gas-phase coagulation on the properties of agarose hydrogels, such as gel strength, swelling degree, rheological properties, and methylene blue (MB) adsorption capacity, were also studied. Structural characterizations confirmed that nano-chitin was successfully introduced into agarose hydrogels. The gel strength, storage modulus , and MB adsorption capacity of agarose hydrogels gradually increased with the increasing nano-chitin addition, whereas the swelling degree decreased. After acidic gas-phase coagulation, agarose/nano-chitin nanocomposite hydrogels exhibited improved gel strength and storage modulus, while the swelling degree and MB adsorption capacity were slightly reduced. The combination of oxidized nano-chitin and acidic gas-phase coagulation is expected to be an effective way to improve the properties of natural polymer hydrogels.

A convenient purification method for silver nanoclusters and its applications in fluorescent pH sensors for bacterial monitoring

Huayu Xiong,Huiling Zheng,Wei Wang,Jichao Liang,Wei Wen,Xiuhua Zhang,Shengfu Wang

Herein, we report a convenient approach to purify water-soluble dihydrolipoic acid (DHLA)-capped Ag nanoclusters (Ag NCs) by pH-induced precipitation under acidic conditions. The fluorescence of Ag NCs could be completely recovered by re-dispersing the precipitate into a basic solution using DHLA and NaBH 4 as stabilizing ligands and etching reagent. DHLA-Ag NCs-doped agarose hydrogels have been prepared to monitor pH with a wide range from 8.0 to 4.0. When pH decreased, the fluorescence of the hydrogels under a UV lamp decreased and completely disappeared after pH 5. The DHLA-Ag NCs-doped agarose hydrogels biosensor showed low cytotoxicity and long stability. Accordingly, a fluorescent pH sensor for bacterial monitoring has been employed based on the “OFF–ON” signal switch of the Ag NCs-agarose hydrogel.

Wearable Biosensing of Caffeine via Microfluidic-Integrated Agarose Hydrogels for Personalized Metabolic Monitoring

Mengjie Su,Linlian Zhu,Yaqiu Zhou,Zhengyuan Ding,Yi Yao,Mengwei Li,Qianying Qiu,Wenjing Peng,Hui Gao,Chunmei Yu

Wearable sweat sensors have gained tremendous attention on account of performing noninvasive and real-time monitoring of sweat analytes, providing a view of the human physiological state at the molecular level. Caffeine is a methylxanthine alkaloid, and its levels in the human body are directly related to health. Developing a wearable device for dynamically assessing caffeine levels in sweat could facilitate real-time personalized health monitoring. In this study, we introduced a wearable agarose hydrogel based sensor integrated with a microfluidic module for the real-time tracking of caffeine in sweat. The hydrogel based caffeine sensor demonstrated highly sensitive detection of caffeine over a broad range from 4 nM to 18.2 μM and showed a low detection limit of 1 nM. On-body tests confirmed the ability of the hydrogel based sensor to detect sweat caffeine levels continuously and dynamically, which offers a novel strategy for the instantaneous monitoring of sweat biomarkers, making it promising for subsequent daily personalized health management.

Preparation of open porous polycaprolactone microspheres and their applications as effective cell carriers in hydrogel system

Qingchun Zhang,Ke Tan,Zhaoyang Ye,Yan Zhang,Wensong Tan,Meidong Lang

Common hydrogel, composed of synthetic polymers or natural polysaccharides could not support the adhesion of anchorage-dependent cells due to the lack of cell affinitive interface and high cell constraint. The use of porous polyester microspheres as cell-carriers and introduction of cell-loaded microspheres into the hydrogel system might overcome the problem. However, the preparation of the open porous microsphere especially using polycaprolactone (PCL) has been rarely reported. Here, the open porous PCL microspheres were fabricated via the combined emulsion/solvent evaporation and particle leaching method. The microspheres exhibited porous surface and inter-connective pore structure. Additionally, the pore structure could be easily controlled by adjusting the processing parameters. The surface pore size could be altered from 20   μm to 80   μm and the internal porosities were varied from 30% to 70%. The obtained microspheres were evaluated to delivery mesenchymal stem cells (MSCs) and showed the improved cell adhesion and growth when compared with the non-porous microspheres. Then, the MSCs loaded microspheres were introduced into agarose hydrogel. MSCs remained alive and sustained proliferation in microsphere/agarose composite in 5-day incubation while a decrement of MSCs viabilities was found in agarose hydrogel without microspheres. The results indicated that the microsphere/hydrogel composite had a great potential in cell therapy and injectable system for tissue regeneration.

Simultaneously colorimetric detection and effective removal of mercury ion based on facile preparation of novel and green enzyme mimic

Yanyun Zou,Qingli Chai,Ting Zhu,Xiaoxiao Yu,Guobin Mao,Ningxing Li,Jinyang Chen,Guosong Lai

In this work, an environmentally-friendly and cost-effective enzyme mimic was obtained by facile one-pot preparation of chitosan/Cu/Fe (CS/Cu/Fe) composite. This composite exhibited significantly enhanced oxidase-mimicking activity during catalyzing the oxidation of 3, 3′, 5, 5′-tetramethylbenzidine (TMB). The CS/Cu/Fe composite was comprehensively characterized and the possible catalytic mechanism was reasonably explored and discussed. Benefiting from the thermal stability and the compatibility with carbohydrate, the CS/Cu/Fe composite was further integrated with agarose hydrogel to fabricate a portable analytical tube containing oxidase mimic. Based on the inhibition of the catalytic oxidation of TMB in the presence of cysteine, as well as the recovery of oxidase-like activity of CS/Cu/Fe due to the specific complexation of cysteine and mercury ion (Hg 2+ ), the rapid colorimetric detection of Hg 2+ was successfully carried out in the analytical tube. This colorimetric method showed good linear response to Hg 2+ over the range from 40 nM to 8.0 μM with a detection limit of 8.9 nM. The method also revealed high selectivity and satisfactory results in recovery experiments of Hg 2+ detection in tap water and lake water. Furthermore, it was found that the effective removal of Hg 2+ could be realized in the analytical tube based on efficient Hg 2+ adsorption by CS/Cu/Fe composite and agarose hydrogel. This study not only prepared a robust and low-cost enzyme mimic, but also proposed a smart strategy to simultaneously monitor and remove toxic Hg 2+ from contaminated water.

Implantable Hydrogel-Protective DNA Aptamer-Based Sensor Supports Accurate, Continuous Electrochemical Analysis of Drugs at Multiple Sites in Living Rats

Shaoguang Li,Jun Dai,Man Zhu,Netzahualcóyotl Arroyo-Currás,Hongxing Li,Yuanyuan Wang,Quan Wang,Xiaoding Lou,Tod E. Kippin,Shixuan Wang,Kevin W. Plaxco,Hui Li,Fan Xia

The ability to track the levels of specific molecules, such as drugs, metabolites, and biomarkers, in the living body, in real time and for long durations, would improve our understanding of health and our ability to diagnose, treat, and monitor disease. To this end, we are developing electrochemical aptamer-based (EAB) biosensors, a general platform supporting high-frequency, real-time molecular measurements in the living body. Here we report that the use of an agarose hydrogel protective layer for EAB sensors significantly improves their signaling stability when deployed in the complex, highly time-varying environments found in vivo. The improved stability is sufficient that these hydrogel-protected sensors achieved good baseline stability and precision when deployed in situ in the veins, muscles, bladder, or tumors of living rats without the use of the drift correction approaches traditionally required in such placements. Finally, our implantable gel-protective EAB sensors achieved good biocompatibility when deployed in vivo in the living rats without causing any severe inflammation.

Blueberry anthocyanins-functionalized hydrogel labels for smartphone-assisted real-time visual freshness monitoring of perishable proteins

Yan Zhang,Danni Peng,Xiaohua He,Hehaoming Zheng,Jian Xiao,Duoxia Xu,Nan Yang

Given rising consumer demands for meat safety and quality assurance, developing an intuitive, cost-effective, and user-friendly sensor platform for real-time monitoring of perishable meat freshness is important. Herein, this study developed an innovative chitosan/agarose/blueberry anthocyanin (CS/AG/BA) hydrogel label system for visual real-time freshness tracking of perishable proteins through smartphone-assisted colorimetric analysis. Through systematic optimization of CS/AG compositional ratios (3:7–7:3) and pH conditions (2.0–4.0), we identified the CS:AG 3:7 -2.0 formulation as exhibiting optimal mechanical properties, water retention capacity, and ammonia sensitivity. The CS/AG/BA hydrogel label exhibited concentration-dependent chromatic transitions, progressively shifting from red to purplish-red and ultimately to blue-purple in response to increasing NH 3 concentrations within the packaged environment. The smart indicator label exhibits distinct color transitions from red to purplish-red and finally to blue-purple when monitoring refrigerated (4 °C) pork and shrimp spoilage. These visually observable color changes demonstrate excellent correlation with standard quality parameters (TVB-N content and pH values), confirming the hydrogel’s effectiveness in real-time freshness monitoring. Quantitative analysis established relationships between hydrogel RGB values and standard freshness parameters, enabling both qualitative visual assessment and quantitative evaluation of pork and shrimp quality. This dual-mode sensing platform combines the advantages of simple visual interpretation with smartphone-enabled digital quantification, offering a practical solution for real-time freshness monitoring throughout the cold chain while addressing critical needs for food safety, quality control, and consumer transparency in perishable protein products.

An Efficient, Recyclable, and Stable Immobilized Biocatalyst Based on Bioinspired Microcapsules-in-Hydrogel Scaffolds

Shaohua Zhang,Zhongyi Jiang,Jiafu Shi,Xueyan Wang,Pingping Han,Weilun Qian

Design and preparation of high-performance immobilized biocatalysts with exquisite structures and elucidation of their profound structure-performance relationship are highly desired for green and sustainable biotransformation processes. Learning from nature has been recognized as a shortcut to achieve such an impressive goal. Loose connective tissue, which is composed of hierarchically organized cells by extracellular matrix (ECM) and is recognized as an efficient catalytic system to ensure the ordered proceeding of metabolism, may offer an ideal prototype for preparing immobilized biocatalysts with high catalytic activity, recyclability, and stability. Inspired by the hierarchical structure of loose connective tissue, we prepared an immobilized biocatalyst enabled by microcapsules-in-hydrogel (MCH) scaffolds via biomimetic mineralization in agarose hydrogel. In brief, the in situ synthesized hybrid microcapsules encapsulated with glucose oxidase (GOD) are hierarchically organized by the fibrous framework of agarose hydrogel, where the fibers are intercalated into the capsule wall. The as-prepared immobilized biocatalyst shows structure-dependent catalytic performance. The porous hydrogel permits free diffusion of glucose molecules (diffusion coefficient: ∼6 × 10–6 cm2 s–1, close to that in water) and retains the enzyme activity as much as possible after immobilization (initial reaction rate: 1.5 × 10–2 mM min–1). The monolithic macroscale of agarose hydrogel facilitates the easy recycling of the immobilized biocatalyst (only by using tweezers), which contributes to the nonactivity decline during the recycling test. The fiber-intercalating structure elevates the mechanical stability of the in situ synthesized hybrid microcapsules, which inhibits the leaching and enhances the stability of the encapsulated GOD, achieving immobilization efficiency of ∼95%. This study will, therefore, provide a generic method for the hierarchical organization of (bio)active materials and the rational design of novel (bio)catalysts.
