Combined alkali pretreatment for enhanced enzymatic saccharification of sugarcane leaf

Yingxue Gong,Zhibin Fu,Menglian Liu,Yu Dai,Jianghai Lin,Zehuan Liu

The alkalis sodium ethoxide and calcium hydroxide were combined to pretreat the abundant agricultural waste sugarcane leaf (SCL). The pretreatment conditions were optimized using single factors and orthogonal array, and the results of material's chemical compositions and enzymatic hydrolysis were investigated. The optimized conditions were pretreating SCL with C 2 H 5 ONa:Ca(OH) 2 volume ratio of 4:1 at 121 °C for 30 min. Under these conditions, the lignin removal rate of SCL was 86.26%, while the total reducing sugars and total enzymatic efficiency of enzymatic hydrolysis could reach 14.82 g/L and 80.02% respectively. These pretreating and hydrolysis results were similar compared with those of SCL pretreated with strong base C 2 H 5 ONa alone. But the strong base dosage was saved 20% and the pretreatment time was shortened to 30 min. Therefore the combined alkali pretreatment could significantly save energy and reduce environmental pollution, which made it a promising method in large-scale production.

Preparation, characterization, and bioactivities of polysaccharides fractions from sugarcane leaves

MO Mengmiao,JIANG Fengyu,CHEN Weiming,DING Zhendong,BI Yongguang,KONG Fansheng

Three new polysaccharides (SLP-50, SLP-70, and SLP-90) were fractionated successively by gradient concentrations of ethanol. This study aimed to investigate the initial structural characterization and bioactivities of these sugarcane leaves polysaccharides (SLPs) obtained by ultrasound-assisted extraction. The results, which were further validated by IR spectrometry, revealed that the three SLPs contained uronic acids, proteins, and sulfate groups in addition to carbohydrate. SLP-50 (36.40 kDa), SLP-70 (12.97 kDa), and SLP-90 (3.52 kDa) were acidic heteropolysaccharides mainly comprised of mannose (Man), rhamnose (Rha), glucuronic acid (Glc A), galacturonic acid (Gal A), galactose (Gal), glucose (Glc), xylose (Xyl), and arabinose (Ara), in different molar ratios. SLP-90 exhibited higher antioxidant capacity than SLP-50 and SLP-70. In vitro, all fractions showed significant hypoglycemic potential and antibacterial activity against Gram-positive and Gram-negative bacteria. The IC50 values of SLP-50, SLP-70, and SLP-90 were 0.11, 0.05, and 0.67 mg/mL on α-glucosidase, respectively, which were significantly lower than that of acarbose (0.80 mg/mL). These findings could provide a reference for developing and applying SLP-based functional foods.

Study on the Extraction and Purification of Polysaccharides from Sugarcane Leaves by Ultrasound-Assisted Enzymatic Hydrolysis and the Antioxidant Activity

Yan-fei Zheng,Qiang Zhang

This study optimized the ultrasound-assisted enzymatic extraction (UAEE) of polysaccharides from sugarcane leaves and evaluated their structural and antioxidant properties. Response surface methodology was used to optimize the extraction, achieving a high polysaccharide yield of 4.31 ± 0.08%, which significantly surpassed that of conventional methods. Purification yielded two distinct, low-molecular-weight polysaccharide fractions (P-1 at 15 kDa and P-2 at 9.6 kDa). Structural characterization revealed significant differences in monosaccharide composition; P-1 was rich in glucose and xylose, while P-2 exhibited greater structural diversity, including a high content of uronic acid. Both fractions demonstrated dose-dependent antioxidant activities. Notably, P-1 showed superior hydroxyl radical scavenging activity (IC₅₀ = 4.3 mg/mL) compared to P-2 (IC₅₀ = 5.2 mg/mL), whereas P-2 exhibited slightly better DPPH radical scavenging and ferric reducing power. These findings demonstrate that UAEE is a highly efficient method for producing low-molecular-weight, bioactive polysaccharides from sugarcane leaves, which show promise as natural antioxidants for further investigation in functional food and pharmaceutical applications.

Characterization and biological activity of acidic sugarcane leaf polysaccharides by microwave-assisted hot alkali extraction

Fengyu Jiang,Yuanhao Sheng,Feilin Wang,Haihui Pan,Weiming Chen,Fansheng Kong

As a waste of sugarcane, the added value of sugarcane leaves has not been fully utilized, and sugarcane leaf polysaccharide is an important active component of sugarcane leaves. Using new extraction methods to study the structure and biological activity of polysaccharides from sugarcane leaves is of great significance for the development and application of sugarcane leaves. To determine the structural characteristics and biological activities of different acidic sugarcane leaf polysaccharides (ASLPs) extracted by microwave-assisted hot alkali extraction (MALE), three new acidic sugarcane leaf polysaccharides (ASLP-50, ASLP-70 and ASLP-90) were precipitated by ethanol. The structure of ASLPs extracted by MALE contains protein, sulfate and uronic acid . ASLP-50 (195.88 kDa), ASLP-70 (94.62 kDa) and ASLP-90 (16.37 kDa) were acidic polysaccharides composed of mannose , galacturonic acid, glucose, arabinose and fucose . The results of XRD and NMR showed that ASLPs were amorphous polysaccharides composed of α-L-Araf-(1→ main chain, and its branching points were composed of Arap and GalpA residues. In addition, ASLPs showed significant antioxidant, hypoglycemic, hypolipidemic and antibacterial activities in vitro . The research and application of ASLPs provide a reference for the recycling of sugarcane leaves, which is conducive to the development of food , agriculture and pharmaceutical industries.

Comparison of physicochemical properties and biological activities between two acidic sugarcane leaf polysaccharides

Fengyu Jiang,Jinhong Zhong,Huizhong Hu,Yufu Li,Yongguang Bi,Fansheng Kong

Sugarcane leaves are a common waste product of sugarcane but contain various active components. To make full use of the value of sugarcane leaves, it shall be urgently developed as a sustainable resource. To compare the differences in structure and activity between acidic sugarcane leaf polysaccharides (ASLs) extracted by different extraction methods, ASLW and ASLM polysaccharides were extracted using hot alkali extraction (HAE) and microwave-assisted hot alkali extraction (MHAE), respectively. The physicochemical properties were analyzed with high-performance liquid chromatography, scanning electron microscopy, infrared spectroscopy and X-ray diffraction. Then, the antioxidant, hypolipidemic, hypoglycemic and antibacterial activities of ASLW and ASLM were evaluated by the inhibition of oxygen free radicals, the inhibition of α-glucosidase and α-amylase, the binding ability with bile salts and the filter paper method. Molecular weight and monosaccharide composition analyses showed that ASLW (142.23 kDa) and ASLM (98.86 kDa) were heteropolysaccharides with a wide range of monosaccharide types and molar ratios. ASLM is an amorphous polysaccharide with stronger stability. Also in vitro bioactivity analysis showed that ASLM had stronger antioxidant, hypolipidemic, hypoglycemic and antibacterial effects than ASLW. As a sustainable active substance, the use of ASLM will benefit the pharmaceutical, industrial, agricultural, and food industries.
