In the present study, we utilizedStevia rebaudianaL. (SRLe) extract to in situ biosynthesize nanoscale alpha hematite (α-Fe2O3) nanoparticles (NPs) with potent antioxidant, antimicrobial, and anticancer properties. SRLe-α-Fe2O3was characterized using physiochemical analyses, including UV/Vis, FTIR, XRD, DLS, EDX, SEM, and TEM studies. Among tested solvents, CHCl3/MeOH (2:1v/v) SRL extract (least polar solvent) contained the highest EY, TPC, and antioxidant capacity of ~3.5%, ~75 mg GAE/g extract, and IC50= 9.87 ± 0.7 mg/mL, respectively. FTIR confirmed the engagement of coating operation to the colloidal α-Fe2O3NPs. TEM, SEM, and DLS revealed that SRLe-α-Fe2O3has a spherical shape, uniform size distribution with aggregation for an average size of ~18.34 nm, and ζ = −19.4 mV, forming a repulsive barrier that helped to improve stability. The synthesized nanoparticles displayed considerable antibacterial activity againstE. coliandS. aureusbacterial growth, and exhibited superior activity against the A549 lung cancer cell lines. These findings indicate that the increased availability of bioactive substances with antioxidant properties of SRLe makes it a potentially interesting material for the preparation of biologically active compounds and green synthesis of nanoparticles. Keywords:Stevia rebaudianaL.;IONP green synthesis;antioxidant;cytotoxic;antitumor;extraction yield

Stevia rebaudiana , a perennial herb, is recognized not only for its sweet steviol glycosides but also for its rich flavonoid content, which confer pharmacological properties including anti-inflammatory, antimicrobial, and anticancer activities. However, the enzymatic basis underlying flavonoid modification in S. rebaudiana remains poorly understood. In this study, we identified, cloned, and heterologously expressed a novel flavonoid glycosyltransferase gene, SrUGT72B1 in E. coli . The recombinant SrUGT72B1 catalyzed the glycosylation of multiple flavonoids using UDP-glucose as the primary sugar donor, and exhibited broad substrate promiscuity toward apigenin, luteolin, phloretin and kaempferol. In addition to UDP-glucose, SrUGT72B1 also accepted UDP-xylose and UDP-rhamnose, with UDP-glucose exhibiting the highest catalytic efficiency. Biochemical characterization revealed that the enzyme functions optimally at pH 9.0 and 50 °C. Notably, SrUGT72B1 demonstrates regioselective 5-O-glycosylation toward apigenin, a rare activity among plant glycosyltransferases. Molecular docking and molecular dynamics simulations provided structural insights into this unique regioselectivity and substrate recognition. Together, these findings establish SrUGT72B1 as a previously uncharacterized flavonoid 5-O-glycosyltransferase, expanding the functional landscape of plant UGTs and offering potential applications in the biosynthesis of value-added flavonoid glycosides.

Stevia leaves have been claimed to possess the highest antioxidant capacity among botanic foods, yet the antioxidant activity has been attributed to the polyphenols from Stevia leaves (PPS). However, most studies about the PPS were based on Stevia extracts that contain abundant polyphenols and flavonoids. The PPS excluded flavonoids were found to possess antioxidant activity similar to that of epigallocatechin gallate, demonstrating stronger hydrophilic antioxidant activity and stability than that of ascorbic acid (VC), vitamin E, or chlorogenic acid under the assayed circumstances. Both of the PPS and their antioxidant activity are very stable in various physical environments, but less stable in the presence of potassium sorbate or sucrose. Moreover the combination of PPS and VC could increase the antioxidant stability of either VC or PPS. Therefore, the PPS have the potential to be a natural, inexpensive, and abundant antioxidant that can be utilized in the pharmaceutical and cosmetics. Stevioside, a natural high-intensity sweetener, is widely employed across the food, pharmaceutical, and daily chemical industries due to its intense sweetness and health benefits. However, traditional extraction and purification processes for steviol glycosides fromStevia rebaudianaare plagued by low efficiency, high energy consumption, substantial environmental impact, and inconsistent product quality. This study systematically optimized the extraction, decolorization, decontamination, and desalting processes to overcome these challenges. The extraction method was refined using 20% ethanol as the solvent, an optimal temperature of 50 °C, and a 1:10 material-to-liquid ratio, increasing the steviol glycoside yield from 32.0% to 49.1%. Decolorization employing a combination of resins D940 and T5 achieved decolorization rates of 89–92% with minimized steviol glycoside loss, surpassing the non-selective adsorption limitations of activated carbon. For decontamination, calcium hydroxide (Ca(OH)2) outperformed diatomaceous earth, attaining a 98% protein removal rate while maintaining steviol glycoside loss below 20%. The desalting resin LXP-016 demonstrated superior performance at 40 °C, enhancing the ability of ionic impurity removal. These optimizations collectively improve the efficiency, sustainability, and quality of steviol glycoside production, offering a promising framework for industrial-scale applications.
