The catabolism of fatty acid (FA) is regarded as a key pathway of aroma volatile compounds in oriental sweet melon (Cucumis melo var. makuwa). In our research, two cultivars of oriental sweet melon, Caihong7 and Tianbao, were employed to illuminate which step of the biosynthetic pathway of aroma compounds could be regulated by ethylene (ETH). The role of ETH in determining the profiles of straight-chain aroma volatile compounds, levels of FA as aroma precursors, activities of aroma-related enzymes derived from FA pathway, and expression patterns of key enzymes were investigated. Overall, exogenous application of ETH increased the production rates of endogenous ETH and levels of FA. Compared with control, the level of straight-chain esters, especially the acetate, hexanoate, and hexyl esters, was significantly increased by ETH, whereas the content of alcohol and aldehyde reduced. In addition, the metabolism of free FA included linoleic acid (LA), linolenic acid (LeA), and oleic acid (OA) appeared to be ETH-dependent. The activities of lipoxygenase (LOX), alcohol dehydrogenase (ADH), and alcohol acetyltransferase (AAT) as well as the expression patterns of Cm-ADH1, Cm-ADH2, Cm-AAT1, and Cm-AAT4 were positively regulated by ETH. In contrast, hydroperoxide lyase (HPL) and Cm-AAT2 and Cm-AAT3 seemed to be independent of ETH modulation. These results suggested that the dissimilation of FA included LA, LeA, and OA into the acetate, hexanoate, and hexyl esters mainly through ETH regulating the LOX pathway by enhancing the expression of particular members of aroma-related key enzyme gene families as well as the activities of dehydrogenation and esterification.

Simple SummaryHoneybees (Apis mellifera) and bumblebees (Bombus terrestris) are often used to pollinate melon flowers in facilities. The researchers identified the volatiles in male and female flowers of a common melon species (Cucumis melo) grown in facilities and measured the EAG and behavioral responses of honeybees and bumblebees when exposed to isolated volatiles from the melon flowers. These results provide basic data for the rational utilization of bees.AbstractAs important pollinators, honeybees and bumblebees present a pollination behavior that is influenced by flower volatiles through the olfactory system. In this study, volatile compounds from melon flowers were isolated and identified by headspace solid-phase microextraction (HS-SPME) and gas chromatography–mass spectrometry (GC-MS), and their effects onApis melliferaandBombus terrestriswere investigated by electroantennogram (EAG) and behavior tests (Y-tube olfactometer). The results showed that 77 volatile compounds were detected in melon flowers, among which the relative content of aldehydes was the highest (61.34%; 82.09%).A. melliferashowed a strong EAG response to e-2-hexenal, e-2-octenal, and 1-nonanal.B. terrestrisshowed a strong EAG response to e-2-hexenal, e-2-octenal, 2,5-dimethyl-benzaldehyde, benzaldehyde and benzenepropanal. In behavior tests, the volatiles with the highest attractive rate toA. melliferawere e-2-hexenal (200 μg/μL, 33.33%) and e-2-octenal (300 μg/μL, 33.33%), and those toB. terrestriswere e-2-hexenal (10 μg/μL, 53.33%) and 2,5-dimethyl-benzaldehyde (100 μg/μL, 43.33%). E-2-hexenal and e-2-octenal were more attractive toA. melliferathanB. terrestris, respectively (10 μg/μL, 10 μg/μL, 200 μg/μL). In conclusion, the volatiles of melon flowers in facilities have certain effects on the electrophysiology and behavior of bees, which is expected to provide theoretical and technical support for the pollination ofA. melliferaandB. terrestrisin facilities. Keywords:Apis mellifera;Bombus terrestris;flower volatiles;electroantennography (EAG);olfactory behavior;melon

Bacterial fruit blotch is one of the most destructing diseases of melon producing-regions. Here, zinc oxide quantum dots (ZnO QDs) were synthesized, and their antibacterial activity against Acidovorax citrulli was investigated. The results indicated that the obtained ZnO QDs displayed 5.7-fold higher antibacterial activity than a commercial Zn-based bactericide (zinc thiazole). Interestingly, the antibacterial activity of ZnO QDs irradiated with light was 1.8 times higher than that of the dark-treated group. It was because ZnO QDs could induce the generation of hydroxyl radicals and then up-regulate the expression of oxidative stress-related genes, finally leading to the loss of cell membrane integrity. A pot experiment demonstrated that foliar application of ZnO QDs significantly reduced the bacterial fruit blotch disease incidence (32.0%). Furthermore, the supply of ZnO QDs could improve the growth of infected melon seedlings by activating the antioxidant defense system. This work provides a promising light-activated quantum-bactericide for the management of pathogenic bacterial infections in melon crop protection.

Compared to other melon types, oriental sweet melon ( Cucumis melo var. makuwa Makino) is quite a different species with a poor shelf-life due to its typical climacteric behaviour, and thin pericarp and rind. In order to investigate the effect of ethanol application on postharvest storage at room temperature (24 °C) and quality of oriental sweet melon, we measured physiological and quality changes induced by ethanol vapor or ethanol injection. Both ethanol treatments significantly inhibited internal ethylene concentrations (IEC), and no ethylene peak occurred when compared with control fruit during storage. However, both ethanol treatments maintained firmness but without significant differences between ethanol-treated and the control melons, except with ethanol injection on day 4. Soluble solids contents were not affected by either treatments. Glucose, fructose and sucrose contents showed a greater increase in ethanol-treated melon flesh and peel tissues than in controls after four days, especially sucrose, the main sugar in melons. Compared with the control, both ethanol treatments resulted in different profiles and composition of aromatic volatile compounds during storage. Both ethanol treatments resulted in a significant increase in ethyl acetate, hexyl acetate and 2-methyl-1-butyle acetate, and a decrease of 2,3-butanediyl diacetate, phenylmethyl acetate and 2-phenethyl acetate. The contents of total esters, including eight newly detected ethyl esters, were 2–4 fold higher in ethanol-treated melons. Ethanol injection more effectively increased accumulation of aroma volatile compounds for a short time, while the effects of ethanol vapor persisted during the storage. Both ethanol treatments increased alcohol acyltransferase (AAT) activity in melon flesh within four days, and levels peaked three days earlier but lower than in the control. There were no significant differences in activities of alcohol dehydrogenase (ADH) and lipoxygenase (LOX) in ethanol-treated and control melons. In summary, postharvest ethanol application reduced the IEC of harvested oriental melons and improved levels of volatile aroma compounds, especially the ethyl esters.
