Pectic polysaccharides are the mainly bioactive components in Lithocarpus litseifolius (sweet tea) leaves. Nevertheless, owing to their diverse and complex chemical structures, the detailed structure-function relationships (SFR) of pectic polysaccharides from sweet tea (STP) are still unclear. Herein, the influence of STP's esterified degree on its diverse biological functions was uncovered. The results showed that the de-esterified STPs with a middle-esterified degree (33.53 %) and a low-esterified degree (7.66 %) were successfully prepared when compared with the original high-esterified STP (47.01 %), and their primary structural features were almost stable after the controllable de-esterification treatment. Furthermore, the findings inferred that STP's antioxidant, anti-diabetic, and immunostimulatory functions showed inverse correlations with the esterified degree. Moreover, the de-esterified STP with a lower-esterified degree could be more easily utilized by intestinal microorganisms to positively regulate the gut microbial composition. Overall, these findings can provide valuable insights for elucidating STP's precise SFR.

Although sweet tea is rich in bioactive polysaccharides , the knowledge regarding their structures, bioactivities, and gut microbial metabolism is still limited. Therefore, in order to promote the application of sweet tea polysaccharide (STP) in the food industry, the pressurized hot water extraction (PHWE) of STP was optimized, and its structural properties and biological effects as well as microbial fermentation characteristics were investigated. The maximum extraction yield (4.64 % ± 0.03 %) of STP extracted by PHWE was obtained under the optimal conditions. Both homogalacturonan and arabinogalactan might exist as major polysaccharide fragments in STP. Additionally, STP exerted obviously in vitro antioxidant, anti-diabetic, and immunostimulatory effects, which might be related to its chemical properties, such as uronic acids , conjugated polyphenolics, and esterification degree. Furthermore, STP could be consumed by intestinal microbiota , and its fermentability was about 54 % at the end stage of fecal fermentation. Indeed, STP could modulate the microbial composition via improving the growth of several beneficial microbes, causing the release of beneficial short-chain fatty acids. Collectively, the findings indicate that the PHWE is an efficient method for extracting bioactive polysaccharides from sweet tea, and results can also provide a scientific basis for developing STP into functional foods or functional ingredients.

In this study, different types of natural deep eutectic solvents (NADES) were used to extract flavonoids from sweet tea. A total of 7 flavonoids , primarily dihydrochalcones, were successfully extracted. Among the NADESs tested, a ternary deep eutectic solvent Bet-PG-Ur (betaine: propanediol : urea = 1: 2: 1, molar ratio) demonstrated the highest extraction efficiency. The chemical and morphological changes of both the NADES and extracted compounds were analyzed using Fourier transform infrared spectroscopy (FT-IR) and scanning electron microscopy (SEM). To optimize the extraction process and evaluate the total flavonoid yield, response surface methodology (RSM), artificial neural network (ANN), and random forest (RF) techniques were employed. The NADES demonstrated consistent extraction performance even after three cycles of recovery using the SPE method. The results revealed that all three models successfully predicted the TFC in the green extraction ultrasonic process. The ANN model exhibited superior result consistency compared to the others, which can be attributed to the enhanced statistical parameters obtained. The highest total flavonoid content (TFC) of 167.561 mg/g was predicted by ANN and achieved under the following conditions: liquid/solid ratio of 19, water content of 50 %, temperature of 45 °C, and ultrasonic power of 400 W.

Sweet tea is a functional herbal tea with anti-inflammatory, anti-diabetic, and other effects, in which phloridzin and trilobatin are two functional compounds. However, the current methods for their quantification are time-consuming, costly, and environmentally unfriendly. In this paper, we propose a rapid method that integrates online pressurized liquid extraction and high-performance liquid chromatography featuring a superficially porous column for fast separation. Moreover, we employ an equal absorption wavelength method to eliminate using multiple standard solutions and relative calibration factors. Our verification process corroborated the technique's selectivity, accuracy, precision, linearity, and detection limitations. Separately, our methodology demonstrated excellent analytical efficiency, cost-effectiveness, and environmental friendliness. Practical application using six distinct batches of sweet tea samples yielded results in congruence with the external standard method. The analytical rate of this technique is up to over 18 times faster than traditional methods, and organic solvent consumption has been reduced to less than 1.5 mL. Therefore, this method provides a valuable way to achieve quality control and green analysis of sweet tea and other herbal teas.

In this study, we investigated the effect of sweet tea polysaccharide (STP) on the physicochemical and structural properties of heat-induced whey protein isolate (WPI) gels, and explored the potential mechanism. The results indicated that STP promoted the unfolding and cross-linking of WPI to form a stable three-dimensional network structure, and significantly improved the strength , water-holding capacity and viscoelasticity of WPI gels. However, the addition of STP was limited to 2 %, too much STP would loosen the gel network and affect the gel properties. The results of FTIR and fluorescence spectroscopy suggested that STP affected the secondary and tertiary structures of WPI, promoted the movement of aromatic amino acids to the protein surface and the conversion of α-helix to β-sheet. In addition, STP reduced the surface hydrophobicity of the gel, increased the free sulfhydryl content, and enhanced the hydrogen bonding, disulfide bonding , and hydrophobic interactions between protein molecules . These findings can provide a reference for the application of STP as a gel modifier in the food industry.

Switchable deep eutectic solvents (SDESs) have recently been developed to improve the recovery and reuse of the extractants. In this work, temperature-induced SDESs were prepared and used for the extraction of flavonoids (representative phlorizin and trilobatin) from sweet tea ( Lithocarpus litseifolius (Hance) Chun). These SDESs solution can switch between solution and sediment depending on the temperature, and the phase diagram of SDESs solution had been drawn. Their temperature-responsive characteristics were studied by various characterizations, including variable-temperature FT-IR, variable-temperature 1 H NMR, and variable-temperature dynamic light scattering (DLS). SDES-1 (Benzamide: Malic acid = 2:1) was screened as the optimal extractant, and the major factors affecting the extraction process were studied by single-factor experiments and optimized by response surface methodology (RSM). The extraction yields for phlorizin and trilobatin were 114.72 and 12.56 mg/g, respectively. Moreover, the recoveries for phlorizin and trilobatin reached 98.46 % and 94.64 %, respectively. This study thus provides an environmentally sound strategy for extracting bioactive compounds from plant resources.
