Water depth alters the fate of estrone across the sediment–water interface in a typical inland lake

Linzhu Du,Shuhang Wang,Xia Jiang,Zhihao Wu,Arne Bratkic,Wei Guo

Water depth variation alters lake sediment environments, affecting the exchange of oxygen, trace elements, and nutrients at the sediment–water interface (SWI). However, there is still limited information on the in-situ fate of refractory emerging pollutants such as estrogenic compounds (ECs) at the SWI of different water depths in lakes. In this study, we integrated active sampling and diffusive gradients in thin film (DGT) technology to in-situ investigate the distribution, diffusion, and microbial responses of estrone (E1) at the SWI from the shallow-water zone (SZ), transition-water zone (TZ), and deep-water zone (DZ) in a typical inland lake of northern China. The surface sediment of the DZ accumulated most E1 (479.57 ± 194.90 ng/kg). In the vertical profile of the sediments, for SZ and TZ, E1 concentration in the sediment, porewater, and DGT fluctuated within a certain concentration range with increasing sediment depth. For the DZ, it exhibited a fluctuating and increasing trend with increasing depth. Dynamic migration analysis of E1 indicated partial resupply in each zone, and the resupply parameter R was 0.102 to 0.416 in the SZ, 0.111 to 0.384 in the TZ, and 0.084 to 0.374 in the DZ, respectively. The resupply capacity in the upper sediment layers (−1 to −4 cm) was higher for the TZ and DZ, whereas the SZ showed higher capacity in the lower layers (−5 to −9 cm). Based on the microbial sequencing results, Sulfuricurvum (typically present in microaerophilic and anoxic conditions) was predominant in the DZ (17.99 ± 15.4 %). Achromobacter (typical E1-degrading genus distributed in oxic environments) was more abundant in the SZ, which led to a higher E1 degradation potential in this zone. The results of this study shed light on the fate of E1 in the SWI of lakes at different water depths, facilitating a more precise control of estrogenic pollution in lake sediments.

A Bioenvironment-Responsive Versatile Nanoplatform Enabling Rapid Clearance and Effective Tumor Homing for Oxygen-Enhanced Radiotherapy

Qiuhong Zhang,Jingwen Chen,Ming Ma,Han Wang,Hangrong Chen

Nanoradiosensitizer-augmented cancer radiotherapy (RT) has been widely investigated, whereas the desirable nanoradiosensitizer with the characteristics of rapid clearance, effective tumor homing, and tumor hypoxia relief is still lacking. Herein, bismuth sulfide–albumin composite nanospheres followed by catalase conjugation (denoted as BSNSs-CAT) have been well constructed as a bioenvironment-responsive nanoradiosensitizer platform. The BSNSs-CAT phagocytosed by normal cells demonstrate architectural disintegration into small ones in response to physiological pH, achieving small size-favored rapid clearance, which largely mitigates the concern of long-term toxicity of BSNSs-CAT in normal tissues. More importantly, benefiting from their large size-favored enhanced permeability and retention effect, BSNSs-CAT accumulate efficiently in tumors and remain architecturally stable in a mildly acidic tumor microenvironment (TME), which strongly favors a response with H2O2 overproduced TME. As a result, the produced intratumor oxygen could overcome tumor hypoxia-associated RT resistance, together with the radiosensitization effect of bismuth, collectively enhancing RT efficacy. This research demonstrates a versatile material solution by fully exploring its bioenvironment-responsive nature, offering a new strategy for nanomedicine design and application.

Influence of sulfadiazine on anaerobic fermentation of waste activated sludge for volatile fatty acids production: Focusing on microbial responses

Jing Xie,Xu Duan,Leiyu Feng,Yuanyuan Yan,Feng Wang,Haiqing Dong,Renyong Jia,Qi Zhou

Extensive studies on anaerobic fermentation of waste activated sludge (WAS) for volatile fatty acids (VFAs) production focused on the effects of operating parameters and pretreatment methods, and little information is available for those of organic pollutants which were absorbed on sludge. The influence of sulfadiazine (SDZ), a typical antibiotic pollutant in WAS, on VFAs production during anaerobic fermentation was investigated in this study. The accumulation of VFAs was remarkably affected in the presence of SDZ. When the content of SDZ was 50 mg per kilogram dry sludge the concentration of VFAs from sludge was 2032.8 mg COD/L, much higher than that of control (1540.2 mg COD/L). Mechanism investigation revealed that the content of extracellular polymeric substances (EPS) from sludge was increased due to the presence of SDZ, which provided more substrates, i.e. , protein and carbohydrate, and created a favorable environment for anaerobes . The hydrolysis and acidification of WAS were stimulated by SDZ, and the functional microorganisms were advantageous to VFAs production. The activities of protease, α-glucosidase and acetate kinase were promoted when SDZ occurred, which were beneficial for hydrolysis and acidification. The effect of SDZ on pure strains further confirmed that the formation of VFAs during anaerobic fermentation was stimulated by SDZ.

Bio-responsive composite liposomes against Campylobacter jejuni in vitro and its application in chicken preservation

Lin Lin,Canchao Mei,Xiaochen Chen,Changzhu Li,Zichun Hua,Haiying Cui

The infection of Campylobacter jejuni ( C. jejuni ) in food can cause huge economic and health costs, and liposomes based on biological switch is a promising antibacterial strategy. In this study, a bio-responsive composite liposome was designed with silk fibroin and L-fucose (FLPs) based on the chemotaxis and protease of C. jejuni , and Litsea cubeba essential oil as antibacterial agent. The oil encapsulation and protein mosaic rates of the composite liposome were 70.6 ± 0.5% and 47.9 ± 1.0%, respectively. The in vitro release experiment verified that the release rates of essential oil from the liposome exceeded 95% at 5 days. Furthermore, FLPs possessed excellent antibacterial activity , the inhibition zone was 23.5 mm and 20% ( v /v) of the composite liposomes could inactivate 99% C. jejuni . Moreover, laser scanning confocal microscopy and scanning electron microscopy images confirmed the antimicrobial activity of FLPs was better than that of other liposomes, and it could attach to bacteria well. Sprayed liposomes revealed sustained ability of retarding bacterial reproduction within 5 days with the refrigerated chicken meat inoculated with C. jejuni , and had no impact on the color and texture of the food. Hence, the bio-responsive composite liposome has a promising application in inhibiting C. jejuni growth in food. Industrial relevance In this study, the composite liposome with high encapsulation efficiency shows significant and sustained antibacterial activity, and has no negative effects on the color and texture of chicken meat. The findings of this study will facilitate the development and application of liposomes as active antimicrobial packaging in food industry.

Bioresponsive nanotherapy for preventing dental caries by inhibiting multispecies cariogenic biofilms

Danfeng Liu,Xianbin Ma,Yaoting Ji,Rourong Chen,Shuhui Zhou,Hantao Yao,Zichen Zhang,Mengjie Ye,Zhigang Xu,Minquan Du

Early childhood caries (ECC) is a public healthcare concern that greatly reduces the quality of life of young children. As a leading factor of ECC, cariogenic biofilms are composed of acidogenic/aciduric pathogens and extracellular polysaccharides (EPSs), creating an acidic and protected microenvironment. Antimicrobial photodynamic therapy (aPDT) is a noninvasive, painless, and efficient therapeutic approach that is suitable for treating ECC. However, due to the hyperfine structure of cariogenic biofilms, most photosensitizers (PSs) could not access and penetrate deeply in biofilms, which dramatically hamper their efficiency in the clinic. Herein, bioresponsive nanoparticle loaded with chlorin e6 (MPP-Ce6) is developed, which largely increases the penetration depth (by over 75%) and retention (by over 100%) of PS in the biofilm compared with free Ce6. Furthermore, MPP-Ce6-mediated aPDT not only kills the bacteria in preformed biofilms but also inhibits multispecies biofilm formation. A rampant caries model is established to mimic ECC in vivo , where the population of cariogenic bacteria is decreased to 10% after MPP-Ce6-mediated aPDT. Importantly, the number and severity of carious lesions are efficiently reduced via Keyes’ scoring and micro-CT analysis. This simple but effective strategy can serve as a promising approach for daily oral hygiene in preventing ECC.

New insights into the effects of antibiotics and copper on microbial community diversity and carbon source utilization

Zhang Wenjie,Wang Jinhua,Zhu Lusheng,Wang Jun,Mao Shushuai,Yan Xiaojing,Wen Shengfang,Wang Lanjun,Dong Zikun,Kim Young Mo

Residual antibiotics (ABs) and heavy metals (HMs) are continuously released from soil, reflecting their intensive use and contamination of water and soil, posing an environmental problem of great concern. Relatively few studies exist of the functional diversity of soil microorganisms under the combined action of ABs and HMs. To address this deficiency, BIOLOG ECO microplates and the Integrated Biological Responses version 2 (IBRv2) method were used to comprehensively explore the effects of single and combined actions of copper (Cu) and enrofloxacin (ENR), oxytetracycline (OTC), and sulfadimidine (SM2) on the soil microbial community. The results showed that the high concentration (0.80 mmol/kg) compound group had a significant effect on average well color development (AWCD) and OTC showed a dose–response relationship. The results of IBRv2 analysis showed that the single treatment group of ENR or SM2 had a significant effect on soil microbial communities, and the IBRv2 of E1 was 5.432. Microbes under ENR, SM2, and Cu stress had more types of available carbon sources, and all treatment groups were significantly more enriched with microorganisms having D-mannitol and L-asparagine as carbon sources. This study confirms that the combined effects of ABs and HMs can inhibit or promote the function of soil microbial communities. In addition, this paper will provide new insights into IBRv2 as an effective method to evaluate the impacts of contaminants on soil health.

Hybrid stem cell-derived bioresponsive vesicles for effective inflamed blood-brain barrier targeting delivery

Honghui Wu,Xinchi Jiang,Yaosheng Li,Yunfei Dong,Juanjuan Zheng,Lu Li,Ying Li,Jinqiang Wang,Xianming Lin,Xiangnan Zhang,Tianyuan Zhang,Zhen Gu,Jianqing Gao

Blood-brain barrier (BBB) dis-integrity, a characteristic of extensive inflammatory encephalopathy, provides the access for brain-targeted drug delivery. Inspired from the bioresponsiveness of stem cell, we conceived of new nano-scale delivery strategy via neural stem cell membrane-derived bioresponsive vesicles (NSC-Lipo) for inflamed BBB target delivery. After recombination of neural stem cell membranes with simple liposomes , the functional ligands, especially very late antigen-4 (VLA-4), were entirely engrafted into NSC-Lipo and equipped the NSC-Lipo with “pass order” to the “selective gate”. It bestowed the bioresponsive vesicles with selective target the injured brain microvessel endothelial cells (BMECs) and penetrate into the lesion through the vascular cell adhesion molecule-1 (VCAM-1)/VLA-4 interaction in the post-ischemic mice model. The released anti-inflammatory agent, metformin , could reverse the inflammatory response of BMECs and rapidly recovers BBB’s integrity. With the repair of BBB’s integrity and down regulation of VCAM-1 expression, the targeting ability of bioresponsive vesicles cease to be in force as well. The bioresponsive vesicles led to enhanced survival rate of ischemic stroke mice (from 30 % to 90 %) after just a single injection compared to metformin loaded bare liposomes.

Smart GSH/pH dual-bioresponsive degradable nanosponges based on β-CD-appended hyper-cross-linked polymer for triggered intracellular anticancer drug delivery

Yutong Dai,Qingman Li,Shurong Zhang,Shan Shi,Yang Li,Xudong Zhao,Liping Zhou,Xin Wang,Yijian Zhu,Wei Li

Efficient accumulation and on-demand intracellular drug release in the desired site are a crucial issue in developing ideal drug delivery systems (DDSs). Glutathione (GSH)/pH dual-bioresponsive degradable Nanosponges were developed based on β-CD-appended hyper-cross-linked polymer by one-pot polymerization of acryloyl-6- ethylenediamine-6-deoxy-β-Cyclodextrin (β-CD-NH-ACy), acrylic acid (AA) and N , N -bis(acryloyl)- cystamine (BACy) as cross-linker to deliver doxorubcin (DOX) and investigated for GSH/pH triggered DOX release, in which the massive carboxyl and amino groups, and disulfide bonds were used as pH and GSH bioresponsive fragments, respectively. In the proposed DDSs, DOX was readily incorporated into the three dimensional networks of the Nanosponges either as inclusion complexes or as non-inclusion complexes, with a high drug loading capacity of 22.6%. In vitro release studies suggested that the Nanosponges exhibited GSH/pH triggered disintegration and drug release performance, in which DOX release was significantly accelerated in acidic (pH5.0) and cytosolic reduction (10 mM GSH) conditions, with ~77.0% of DOX release. The morphology changes of DOX@Nanosponges in releasing media (pH5.0, 10 mM GSH) were further studied by TEM . Confocal microscopy observation demonstrated that DOX was delivered and released into cytoplasm and nucleus of A549 cells in 7 h incubation with DOX@Nanosponges. MTT assays manifested that the Nanosponges exhibited low cytotoxicity up to a concentration of 1000 μg/mL and DOX@Nanosponges had high anti-tumor activity. These findings demonstrated that the dual-bioresponsive Nanosponges may function as a promising platform for targeted delivery and intracellular drug controlled release in tumor therapy.

Hierarchically Releasing Bio-Responsive Nanoparticles for Complete Tumor Microenvironment Modulation via TGF-β Pathway Inhibition and TAF Reduction

Jun Zhang,Tiantian Zuo,Jie Yang,Zongwei Hu,Zhihua Wang,Rui Xu,Siyu Ma,Yawen Wei,Qi Shen

The aggressive progression of breast cancer is impacted significantly by the tumor microenvironment (TME). The current chemotherapy normally causes cytotoxicity to tumor cells, while does not effectively modulate the TME. Thus, the chemotherapy effect of breast cancer is usually dissatisfactory. In this study, a kind of hierarchically releasing bio-responsive nanoparticles (R(D)/H(S) NPs), constructed by β-cyclodextrin-grafted heparin and pH-sensitive pseudorotaxane, were investigated to enhance the breast cancer chemotherapeutic efficacy through TME modulation. Doxorubicin (DOX) and transforming growth factor-β (TGF-β) receptor inhibitor (SB431542) loaded onto R(D)/H(S) NPs were released rapidly for the respective response to low pH in endosomes/lysosomes and heparanase (HPSE) in TME. Our results showed that R(D)/H(S) NPs effectively inhibited the formation of tumor-associated fibroblasts (TAFs) and reduced TGF-β and collagen I secretion. Besides, the immunosuppressive microenvironment was effectively reversed into immunogenic, characterized by increased CD8+ and CD4+ T cell infiltration, which distinctly inhibited breast cancer metastasis. Therefore, R(D)/H(S) NPs remodeled the TME by downregulating TAFs, TGF-β, and collagen I; activating the immune microenvironment; and then amplifying the chemotherapeutic efficacy of DOX.

Implantable Bioresponsive Hydrogel Prevents Local Recurrence of Breast Cancer by Enhancing Radiosensitivity

Zhiguang Fu,Hongqi Li,Peng Xue,Hanying Yu,Shuo Yang,Cheng Tao,Wei Li,Yingjie Wang,Jianjun Zhang,Yu Wang

Breast cancer is one of the most common types of cancer. Patients are often concerned about regional recurrence after breast cancer surgery. Radiotherapy plays a vital role in reducing recurrence and prolonging the survival of patients undergoing breast-conserving surgery and high-risk mastectomy. However, 8–15% of patients still have disease progression due to radiation resistance. Therefore, new strategies for combination radiotherapy sensitization must be investigated. In this study, an implantable drug loading system, sunitinib nanoparticles @ matrix metalloproteinases -response hydrogel (NSMRH), uses enzyme-sensitive hydrogel as a carrier to load sunitinib nanoparticles, was identified. The releasing profile demonstrated that sunitinib nanoparticles may be continuously released from the hydrogels. Functional experiments revealed that, when paired with NSMRH, radiation may significantly inhibit tumor cell proliferation, migration, and invasion in vitro . Further animal experiments showed that NSMRH combined with radiotherapy could more effectively control the recurrence of subcutaneous xenograft tumors, prolong the survival time, and have no obvious toxicity in nude mice. Finally, by studying the molecular mechanism of NSMRH, it was hypothesized that in breast cancer cells, NSMRH cooperated with sensitized radiotherapy, mainly due to significantly blocking the G2/M phase, reducing the DNA repair efficiency, inhibiting tumor angiogenesis, promoting apoptosis, and reversing the abnormal expression of platelet-derived growth factor receptor alpha (PDGFRA) after radiotherapy. These findings suggest that NSMRH’s radiation sensitization and anti-tumor activity may aid in the development of a novel method in future clinical applications.

Balancing Bioresponsive Biofilm Eradication and Guided Tissue Repair via Pro-Efferocytosis and Bidirectional Pyroptosis Regulation during Implant Surgery

Zhengxi Wang,Xudong Zhang,Quan Liu,Xianli Hu,Jiawei Mei,Jun Zhou,Xianzuo Zhang,Dongdong Xu,Wanbo Zhu,Zheng Su,Chen Zhu

There is an increasingly growing demand to balance tissue repair guidance and opportunistic infection (OI) inhibition in clinical implant surgery. Herein, we developed a nanoadjuvant for all-stage tissue repair guidance and biofilm-responsive OI eradication via in situ incorporating Cobaltiprotoporphyrin (CoPP) into Prussian blue (PB) to prepare PB-CoPP nanozymes (PCZs). Released CoPP possesses a pro-efferocytosis effect for eliminating apoptotic and progressing necrotic cells in tissue trauma, thus preventing secondary inflammation. Once OIs occur, PCZs with switchable nanocatalytic capacity can achieve bidirectional pyroptosis regulation. Once reaching the acidic biofilm microenvironment, PCZs possess peroxidase (POD)-like activity that can generate reactive oxygen species (ROS) to eradicate bacterial biofilms, especially when synergized with the photothermal effect. Furthermore, generated ROS can promote macrophage pyroptosis to secrete inflammatory cytokines and antimicrobial proteins for biofilm eradication in vivo. After eradicating the biofilm, PCZs possess catalase (CAT)-like activity in a neutral environment, which can scavenge ROS and inhibit macrophage pyroptosis, thereby improving the inflammatory microenvironment. Briefly, PCZs as nanoadjuvants feature the capability of all-stage tissue repair guidance and biofilm-responsive OI inhibition that can be routinely performed in all implant surgeries, providing a wide range of application prospects and commercial translational value.

Harnessing the dual immunomodulatory function of myeloid-derived suppressor cells to reshape the inflammatory microenvironment for osteoarthritis therapy

Zili Guo,Tao Chen,Xuehuan Wen,Qiangchang Fu,Jiabin Lai,Jiajin Li,Jiayu Chen,Jie Weng,Wang Wen,Qihua Cao,Junxiong Zhu,Zhaoming Ye,Xiaohua Yu,Lifeng Jiang

Osteoarthritis (OA) pathogenesis is profoundly influenced by dysregulated immune dynamics, where persistent interleukin-17 (IL-17)/T helper 17 (Th17) cell mediated inflammation coordinates with failed regenerative processes to perpetuate joint destruction. Here, we unveil the role of myeloid-derived suppressor cells (MDSCs) as dual-phase regulators that paradoxically orchestrate both inflammatory escalation and tissue repair in OA progression. Intra-articular administration of MDSCs in OA mice amplified IL-17 dependent inflammatory cascades and chemokine-driven leukocyte recruitment, revealing a context-dependent pro-inflammatory phenotype. Unexpectedly, MDSC depletion failed to attenuate joint damage, implying their indispensable yet multifaceted role in OA pathogenesis. Mechanistically, MDSCs exhibited functional plasticity by upregulating arginase-1 to polarize M2 macrophages, fostering a regenerative niche alongside their inflammatory activity. To resolve this duality, we developed a bio-responsive hydrogel-microsphere system integrating transforming growth factor β1 (TGF-β1) and interleukin-1 β1 antibody (anti–IL-1β) loaded mesoporous silica nanoparticles (MSNs). This spatiotemporally controlled platform selectively suppressed MDSC-mediated Th17 cell expansion while harnessing their intrinsic capacity to drive M2 macrophage polarization and chondrogenesis. The resultant shift from a pro-inflammatory to pro-regenerative microenvironment significantly attenuated cartilage erosion and restored joint integrity in OA models. Our findings redefine MDSCs as bifunctional immune orchestrators in OA and establish precision biomaterial guided immune decoding as a paradigm-shifting therapeutic strategy. By engineering MDSC plasticity through antagonistic cytokine delivery, this work provides a blueprint for microenvironment remodeling in degenerative joint diseases.
