Light energy utilization and microbial catalysis for enhanced biohydrogen: Ternary coupling system of triethanolamine-mediated Fe@C-Rhodobacter sphaeroides

Qiushi Jiang,Yanjing Li,Minmin Wang,Wen Cao,Xueying Yang,Sihu Zhang,Liejin Guo

This study investigated the mediating effect of Triethanolamine on Fe@C- Rhodobacter sphaeroides hybrid photosynthetic system to achieve efficient biohydrogen production. The biocompatible Fe@C generates excited electrons upon exposure to light, releasing ferrum for nitrogenase synthesis, and regulating the pH of the fermentation environment. Triethanolamine was introduced to optimize the electron transfer chain, thereby improving system stability, prolonging electron lifespan, and facilitating ferrum corrosion. This, in turn, stimulated the lactic acid synthetic metabolic pathway of Rhodobacter sphaeroides , resulting in increased reducing power in the biohybrid system. The ternary coupling system was analyzed through the regulation of concentration, initial pH, and light intensity. The system achieved the highest total H 2 production of 5410.9 mL/L, 1.29 times higher than the control (2360.5 mL/L). This research provides a valuable strategy for constructing ferrum-carbon-based composite-cellular biohybrid systems for photo-fermentation H 2 production.

Biohydrogen production from brown algae fermentation: Relationship between substrate reduction degree and hydrogen production

Weiming Li,Lihui Lu,Chi Cheng,Nanqi Ren,Shang-Tian Yang,Meng Liu

In this study, mannitol and mannitol-rich seaweed were fermented to investigate the relationship between substrate reduction degree and hydrogen production performance. The results showed that acetate was required in mannitol fermentation with an optimum acetate/mannitol mass ratio of 1:5. Hydrogen production and yield of mannitol fermentation reached 123.76 mL and 2.12 mol/mol-mannitol, respectively, 42.02 % and 26.95 % higher than that of glucose, respectively. The acetate was fully assimilated and the butyrate selectivity reached 100 % in the effluent. Redox potential and electron distribution showed that mannitol increased the overall electron input from mannitol and acetate, leading to the increase in hydrogen and butyrate generation. Hydrogen yield reached 2.33 mol/mol-mannitol with brown algae hydrolysate, which was the highest ever reported. This study demonstrated that substrate with a higher reduction degree could yield higher hydrogen and showed the great application potential of brown algae fermentation for the co-production of hydrogen and butyrate.

Variety of rumen microbial populations involved in biohydrogenation related to individual milk fat percentage of dairy cows

Zhang Lei,Shen Hong,Zhang Jiyou,Mao Shengyong

Our objective was to investigate the contribution of the rumen microbiome on the individual milk fat percentage (MFP) of Holstein dairy cows under the same nutritional and management conditions. From 92 early lactation dairy cows, the top 10 with the highest MFP (HF; n = 10) and the last 10 with the lowest MFP (LF; n = 10) were selected for the study. As a result, the milk trans-10, cis-12 C18:2 content was significant lower in the HF group than that in the LF group (P < 0.001). The rumen acetate to propionate ratio was significant higher in the HF group than that in the LF group (P = 0.035). According to the results of 16S rRNA gene sequencing, a minor but significant difference existed between the groups (P = 0.040). Three genera of the family Lachnospiraceae and four genera of the order Bacteroidales were identified to be the biomarkers for the LF group and HF group in the LEfSe analysis, respectively. Three microbial modules enriched by the family Lachnospiraceae were positively related to the milk trans-10, cis-12 C18:2 content (rs > 0.60, P < 0.05). According to the results of shotgun metagenome sequencing, three kinds of linoleic acid (LA) isomerase genes were present in the gene pools of the rumen microbiome. Among them, the relative abundance of Bifidobacterium LA isomerase (BBI) was higher in the HF group than that in the LF group (P = 0.007). Three metagenome-assembled genomes (MAGs) with LA isomerase genes were positively correlated to the milk trans-10, cis-12 C18:2 content (rs> 0.40, P < 0.05). Furthermore, all of these three MAGs were found to be able to produce lactate. Taken together, these results indicate that the increased relative abundance of microbial population with the trans-10 biohydrogenation pathway within the rumen microbiome contributes to the decrease of MFP via the increase of rumen trans-10, cis-12 C18:2 production. This study provides a new perspective for the development of measures for improving the milking performance of dairy cows.

Steam explosion-ionic liquid pretreatments on wetland lignocellulosic biomasses of Phragmites (sp.) and Thalia dealbata for Bio H2 conversion

Hongyun Peng,Lingling Gao,Mengjiao Li,Yibin Shen,Qiongqiu Qian,Xia Li

Bio H2 conversion from wetland lignocellulosic biomass is one of the promising alternatives to fossil fuels. Both Phragmites (sp.) and Thalia dealbata are holocellulose-rich and lignin-rich wetland plant biomasses. Scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) demonstrated alteration of lignocellulosic structures due to hemicelluloses removal by steam explosion (SE) pretreatment, and further disruption of cellulose crystallinity after treatment with SE followed by [Bmim]Cl (SE-[Bmim]Cl). Thermogravimetry/differential thermogravimetry (TG/DTG) revealed an increase in amorphous cellulose and partial delignification in lignocellulosic structures as a consequence of SE-[Bmim]Cl. The pretreatment of both SE and SE-[Bmim]Cl yielded lignocellulosic substrates with improved properties in terms of their conversion into glucose and bio H2. Five-to-ten folds (by SE) and ten-to-twenty folds (by SE-[Bmim]Cl) more glucose was released from the lignocellulosic substrates of both plants than those of contrast samples. Compared to Phragmites (sp.), the greater destruction in lignocellulosic structure of Thalia dealbata as a result of SE and SE-[Bmim]Cl, increased the accessible surface area and disrupted the cellulose crystallinity much more, thus making it more efficient for bio H2 conversion. The bio H2 of 1.97 ± 0.14 mmol H2 g−1 dry weight (DW) was yielded after sludge anaerobic fermentation of Thalia dealbata treated with SE, and it increased to 4.79 ± 0.86 mmol H2 g−1 DW after SE-[Bmim]Cl treatment. For Phragmites (sp.) it was 1.45 ± 0.42 and 2.75 ± 0.76 mmol H2 g−1 DW after SE and SE-[Bmim]Cl pretreatment, respectively. Therefore, SE-[Bmim]Cl pretreatment can be developed for efficiently enhancing bio H2 conversion from wetland plant Thalia dealbata.

External Electrons Directly Stimulate Escherichia coli for Enhancing Biological Hydrogen Production

Wei Zhou,Wen Zhang,Wei Geng,Yaoqi Huang,Tong-Kai Zhang,Zi-Qian Yi,Yang Ge,Yao Huang,Ge Tian,Xiao-Yu Yang

External electric field has the potential to influence metabolic processes such as biological hydrogen production in microorganisms. Based on this concept, we designed and constructed an electroactive hybrid system for microbial biohydrogen production under an electric field comprised of polydopamine (PDA)-modified Escherichia coli (E. coli) and Ni foam (NF). In this system, electrons generated from NF directly migrate into E. coli cells to promote highly efficient biocatalytic hydrogen production. Compared to that generated in the absence of electric field stimulation, biohydrogen production by the PDA-modified E. coli-based system is significantly enhanced. This investigation has demonstrated the mechanism for electron transfer in a biohybrid system and gives insight into precise basis for the enhancement of hydrogen production by using the multifield coupling technology.

Biohydrogen Production from Hydrolysates of Selected Tropical Biomass Wastes with Clostridium Butyricum

Dan Jiang,Fang Zhen,Chin Siew-xian,Tian Xiao-fei,Su Tong-chao

Biohydrogen production has received widespread attention from researchers in industry and academic fields. Response surface methodology (RSM) was applied to evaluate the effects of several key variables in anaerobic fermentation of glucose with Clostridium butyrium and achieved the highest production rate and yield of hydrogen. Highest H 2 yield of 2.02 mol H 2 /mol-glucose was achieved from 24 h bottle fermentation of glucose at 35 °C, while the composition of medium was (g/L): 15.66 glucose, 6.04 yeast extract, 4 tryptone, 3 K 2 HPO 4 , 3 KH 2 PO 4 , 0.05 L-cysteine, 0.05 MgSO 4 ·7H 2 O, 0.1 MnSO 4 ·H 2 O and 0.3 FeSO 4 ·7H 2 O, which was very different from that for cell growth. Sugarcane bagasse and Jatropha hulls were selected as typical tropical biomass wastes to produce sugars via a two-step acid hydrolysis for hydrogen production. Under the optimized fermentation conditions, H 2 yield (mol H 2 /mol-total reducing sugar) was 2.15 for glucose, 2.06 for bagasse hydrolysate and 1.95 for Jatropha hull hydrolysate in a 3L fermenter for 24 h at 35 °C, with H 2 purity of 49.7–64.34%. The results provide useful information and basic data for practical use of tropical plant wastes to produce hydrogen.

Microbial-Driven Synthesis and Hydrolysis of Neohesperidin Dihydrochalcone: Biotransformation Process and Feasibility Investigation

Yucong Zou,Xiaofeng Li,Xuan Xin,Haixia Xu,Guanglei Zhao

A novel yeast-mediated hydrogenation was developed for the synthesis of neohesperidin dihydrochalcone (NHDC) in high yields (over 83%). Moreover, whole-cell catalytic hydrolysis was also designed to hydrolyze NHDC into potential sweeteners, hesperetin dihydrochalcone-7-O-glucoside (HDC-G) and hesperetin dihydrochalcone (HDC). The biohydrogenation was further combined with whole-cell hydrolysis to achieve a one-pot two-step biosynthesis, utilizing yeast to hydrogenate C═C in the structure, while Aspergillus niger cells hydrolyze glycosides. The conversion of NHDC and the proportion of hydrolysis products could be controlled by adjusting the catalysts, the components of the reaction system, and the addition of glucose. Furthermore, yeast-mediated biotransformation demonstrated superior reaction stability and enhanced safety and employed more cost-effective catalysts compared to the traditional chemical hydrogenation of NHDC synthesis. This research not only provides a new route for NHDC production but also offers a safe and flexible one-pot cascade biosynthetic platform for the production of high-value compounds from citrus processing wastes.

The enhancement on biohydrogen production by the driving forces from extracellular iron oxide respiration

Lei Yu,Wei-xin Cheng,Quan Wang

Biohydrogen productions from xylose and glucose under dark condition were enhanced by the presence of natural Fe 3 O 4 . The electron equivalent of H 2 fractions accounted for 4.55 % and 5.69 % of the total given xylose and glucose in the experiments without Fe 3 O 4 , and that were correspondingly increased to 5.14 % and 6.50 % in the experiments with 100 mg/L of Fe 3 O 4 , respectively. Moreover, Fe 3 O 4 increased the total intracellular NAD(H) concentrations by 8.84 % and 8.37 %, and boosted the ratios of NADH/NAD + by 8.33 % and 17.72 % in xylose and glucose fermentation, respectively, comparing to the corresponding control experiments. The formation of electron couples of Fe(III)/Fe(II) during the iron oxide respiration and more generation of active extracellular polymeric substances components were determined as the important reasons for the improved biohydrogen production performance. Thus, a promotion mechanism of the internal “driving forces” from extracellular iron oxide respiration on the biohydrogen production was proposed.

Synergistic increase of bio-hydrogen production from sludge anaerobic fermentation by thermal hydrolysis coupled with calcium hypochlorite pretreatment

Qian Zhang,Jiawei Hu,Cheng Li

To solve the problem of low bio-hydrogen yield from anaerobic fermentation (AF) of sludge, a novel and efficient method by combining thermal hydrolysis (TH) with calcium hypochlorite (CH) for sludge pretreatment was proposed in this study. Experimental data demonstrated that TH + CH method synergistically promoted the bio-hydrogen production from sludge. The optimal condition of the combined pretreatment was TH at 70 °C plus 0.12 g/g volatile suspended solids (VSS) of CH, and relevant hydrogen yield was 22.52 mL/g VSS. Mechanism study exposed that TH + CH method effectively destructed extracellular polymeric substances (EPS) and sludge cells, causing the promotion of hydrolysis rate and generation of abundant fermentation substrates. In the several biochemical processes of sludge AF, acidogenesis and acetogenesis that responsible for hydrogen generation were not or only slightly suppressed by TH + CH pretreatment, while homoacetogenesis, sulfate-reducing and hydrogenotrophic methanogenesis that can consume hydrogen were severely inhibited under this condition, resulting in hydrogen accumulation. Microbiological analysis demonstrated that the functional bacteria (e.g. Rhodobacter , Proteiniborus and Petrimonas ) related to hydrogen generation were all enriched under TH + CH condition.

Poly(ionic liquid)-Flocculated Chlorella Loading Bactericidal and Antioxidant Hydrogel as a Biological Hydrogen Therapy for Diabetic Wound Dressing

Hui Xu,Xiaonan Yuan,Yu Rao,Shuna Gao,Jiangna Guo,Feng Yan

Infection and oxidative stress seriously hinder the healing of diabetic wounds, resulting in various serious health and clinical problems. Herein, a sustainable biological hydrogen (H2)-producing hyaluronic acid–based hydrogel patch (HAP-Chl) was constructed by loading an imidazolium-based poly(ionic liquid) (PIL) flocculated live Chlorella as a diabetic wound dressing. The PIL can flocculate Chlorella through electrostatic interactions between PIL and Chlorella to form Chlorella agglomerates, endowing the Chlorella in the central agglomerates with the ability to continuously produce H2 for 24 h under mild conditions. Combining the membrane disruption-related bactericidal mechanism of PIL and the antioxidant properties of the produced H2, HAP-Chl was determined to be antibacterial and antioxidant. In addition to exhibiting biocompatible and nontoxic activities, subsequent Staphylococcus aureus-infected chronic wound studies revealed that HAP-Chl is capable of promoting the healing of chronic wounds by effectively killing bacteria, reducing extensive ROS, relieving inflammation, and promoting the deposition of mature collagen and angiogenesis. This study provides a new strategy for constructing an in situ sustainable H2-producing hydrogel, enabling the formation of novel antibacterial and antioxidant material platforms with potential for wound dressing applications.

Anaerobic Biohydrogenation of Isoprene by Acetobacterium wieringae Strain Y

Huijuan Jin,Xiuying Li,Hongyan Wang,Natalie L. Cápiro,Xiaocui Li,Frank E. Löffler,Jun Yan,Yi Yang

Isoprene is a ubiquitously distributed, biogenic, and climate-active organic compound. Microbial isoprene degradation in oxic environments is fairly well understood; however, studies exploring anaerobic isoprene metabolism remain scarce, with no isolates for study available. Here, we obtained an acetogenic isolate, designated Acetobacterium wieringae strain Y, which hydrogenated isoprene to a mixture of methyl-1-butenes at an overall rate of 288.8 ± 20.9 μM day−1 with concomitant acetate production at a rate of 478.4 ± 5.6 μM day−1. Physiological characterization demonstrated that isoprene was not utilized in a respiratory process; rather, isoprene promoted acetogenesis kinetically. Bioinformatic analysis and proteomics experiments revealed the expression of candidate ene-reductases responsible for isoprene biohydrogenation. Notably, the addition of isoprene to strain Y cultures stimulated the expression of proteins associated with the Wood-Ljungdahl pathway, indicating unresolved impacts of isoprene on carbon cycling and microbial ecology in anoxic environments (e.g., promoting CO2 plus H2 reductive acetogenesis while inhibiting methanogenesis). Our new findings advance understanding of microbial transformation of isoprene under anoxic conditions and suggest that anoxic environments are isoprene sinks.IMPORTANCE Isoprene is the most abundant, biologically generated, volatile organic compound on Earth, with estimated emissions in the same magnitude as methane. Nonetheless, a comprehensive knowledge of isoprene turnover in the environment is lacking, impacting global isoprene flux models and our understanding of the environmental fate and longevity of isoprene. A critical knowledge gap that has remained largely unexplored until recently is the microbiology and associated molecular mechanisms involved in the anaerobic biotransformation of isoprene. By integrating culture-dependent approaches with omics techniques, we isolated an acetogen, Acetobacterium wieringae strain Y, capable of anaerobic biohydrogenation of isoprene. We obtained the complete genome of strain Y, and proteomic experiments identified candidate ene-reductases for catalyzing the asymmetric reduction of the electronically activated carbon-carbon double bond of isoprene. We also demonstrated that isoprene biohydrogenation stimulates the expression of Wood-Ljungdahl pathway enzymes. This study emphasizes the ecological roles of specialized Acetobacterium on the natural cycling of isoprene in anoxic environments and the potential effects of isoprene biohydrogenation on acetogens and methanogens, which have implications for global climate change and bioenergy production.

Harnessing microbial hydrogen for remodeling tumor microenvironment to potentiate collaborative chemo-immunotherapy

Shi-Man Zhang,Ran Meng,Hong Chen,Hao Zhou,Xin-Chen Deng,Yan-Tong Lin,Wei-Qin Yao,Wei-Hai Chen,Xian-Zheng Zhang

Chemo-immunotherapy is fundamentally limited by insufficient tumor specificity and immunosuppressive tumor microenvironment (TME). Here, an engineered probiotic CB@PPH is elaborately constructed to remodel tumor microenvironment for potentiating collaborative chemo-immunotherapy. Specifically, phenylboronic acid-modified hyaluronic acid-coated paclitaxel (PTX)-loaded PCN-224 (PPH) nanoparticles are covalently conjugated with Clostridium butyricum (CB) to develop CB@PPH. Due to tumor hypoxia-tropism, CB@PPH facilitates the precise intratumoral delivery of PPH, and in situ kills tumor cells by inhibiting the depolymerization of tubulin. Moreover, the enriched CB@PPH at tumor tissues could persistently produce abundant microbial H 2 . On the one hand, the generated intratumoral H 2 would induce severe mitochondrial destruction and disrupt redox homeostasis within tumor cells, collaboratively amplifying immunogenic cell death signaling. On the other hand, microbial H 2 acts as an immune modulatory agent, remodeling the immunosuppressive TME into a pro-immunogenic milieu that activate robust antitumor immune responses. In combination with αPD-1 therapy, the engineered CB@PPH significantly inhibits the growth of subcutaneous tumors, as well as effectively suppresses the progression of distant tumors with a tumor inhibition rate of 91.0 %. This work underscores the innovative potential of microbial H 2 -assited chemo-immunotherapy, presenting a paradigm for the development of cancer therapeutic strategies.
