Efficient Angiogenesis-Based Diabetic Wound Healing/Skin Reconstruction through Bioactive Antibacterial Adhesive Ultraviolet Shielding Nanodressing with Exosome Release

Min Wang,Chenggui Wang,Mi Chen,Yuewei Xi,Wei Cheng,Cong Mao,Tianzhen Xu,Xingxing Zhang,Cai Lin,Weiyang Gao,Yi Guo,Bo Lei

Diabetic wound healing and angiogenesis remain a worldwide challenge for both clinic and research. The use of adipose stromal cell derived exosomes delivered by bioactive dressing provides a potential strategy for repairing diabetic wounds with less scar formation and fast healing. In this study, we fabricated an injectable adhesive thermosensitive multifunctional polysaccharide-based dressing (FEP) with sustained pH-responsive exosome release for promoting angiogenesis and diabetic wound healing. The FEP dressing possessed multifunctional properties including efficient antibacterial activity/multidrug-resistant bacteria, fast hemostatic ability, self-healing behavior, and tissue-adhesive and good UV-shielding performance. FEP@exosomes (FEP@exo) can significantly enhance the proliferation, migration, and tube formation of endothelial cells in vitro. In vivo results from a diabetic full-thickness cutaneous wound model showed that FEP@exo dressing accelerated the wound healing by stimulating the angiogenesis process of the wound tissue. The enhanced cell proliferation, granulation tissue formation, collagen deposition, remodeling, and re-epithelialization probably lead to the fast healing with less scar tissue formation and skin appendage regeneration. This study showed that combining bioactive molecules into multifunctional dressing should have great potential in achieving satisfactory healing in diabetic and other vascular-impaired related wounds.

Resveratrol loaded native silk fiber-sericin hydrogel double interpenetrating bioactive wound dressing facilitates full-thickness skin wound healing

Yanwei Li,Susu Wang,Yurong Li,Guozheng Zhang,Tangfeng Wu,Yongkang Wei,Xinyu Cao,Hui Yan,Peisheng Liang,Zihan Yan,Yanan Guo,Yeshun Zhang

Wound repair is challenging for traditional wound dressings. New bioactive dressings need to be developed urgently. Herein, we reported a highly bioactive silk protein wound dressing (SPD) with natural silk fiber-sericin hydrogel interpenetrating double network structure, which combines the dual characteristics of natural silk and sericin hydrogel. Silk fiber scaffolds were secreted directly from silkworms bred by regulating their spinning behaviors. Sericin in SPD is obtained by dissolving silkworm cocoons at high temperature and high pressure, while it remains intact activities to self-assemble a hydrogel. To explore the effect of SPD, we first systematically evaluated its physicochemical properties and biological activities in vitro. The SPD exhibits high porosity, prominent mechanical strength, pH-responsive degradability, and excellent anti-oxidation and cell compatibility. Besides, SPD can load and maintain long-term drug release. Based on the satisfactory performance of SPD in vitro, effective in vivo treatment was achieved in a mouse full-thickness wound model, as demonstrated by a significantly accelerated wound healing process, promote the regeneration of hair follicles and sebaceous glands, increased expression of vascular endothelial growth factor, and reduced inflammation. Further, resveratrol was loaded into SPD to enhance the effects of anti-oxidation and anti-inflammation for wound healing. Our investigation shows that SPD with excellent physicochemical and biological properties applied in a murine full-thickness skin wound model resulted in remarkable and efficient acceleration of healing process, which may inspire the design of new, effective, and safer medical materials for tissue regeneration.

Enhanced wound healing in diabetic mice by hyaluronan/chitosan multilayer-coated PLLA nanofibrous mats with sustained release of insulin

Xiang Gao,Rui Huang,Yanpeng Jiao,Thomas Groth,Wufeng Yang,Chenlin Tu,Hailong Li,Fan Gong,Jiaqi Chu,Mingyan Zhao

The healing of wounds in diabetes mellitus patients remains a critical clinical challenge globally. To promote healing, the development of bioactive dressings that not only provide physical protection but also supply a favorable microenvironment for healing at the wound interface is urgently required. Herein, Michael addition reaction was first used for intrinsic-crosslinking of bioactive maleilated chitosan/thiolated hyaluronan (mCH/tHA) multilayer coatings on poly (L-lactic acid) (PLLA) nanofibrous mats (P-mCH/tHA) for a sustained release of insulin (IN) and compared that with pure PLLA and PLLA mats coated with native CH/HA (P-CH/HA) multilayers. The physicochemical studies showed that P-mCH/tHA have higher tensile strength, an accelerated degradation rate, more antibacterial activity and sustained release of insulin when compared to P-CH/HA and pure PLLA mats. Studies on bioactive properties of PLLA mats coated with either mCH/tHA or CH/HA indicated an improved adhesion, spreading and proliferation of mesenchymal stem cells on P-mCH/tHA_IN in comparison to the other PLLA mats. Most interestingly, in vivo investigations with a diabetes mouse model demonstrated fastest wound closure when using P-mCH/tHA_IN among all groups with facilitated re-epithelization, angiogenesis, and increased collagen deposition in the wounds, which makes this coating in combination with IN promising for development of new bioactive wound dressings.
