P-Doped Carbon Quantum Dots with Antibacterial Activity

Shuiqin Chai,Lijia Zhou,Shuchen Pei,Zhiyuan Zhu,Bin Chen

It is a major challenge to effectively inhibit microbial pathogens in the treatment of infectious diseases. Research on the application of nanomaterials as antibacterial agents has evidenced their great potential for the remedy of infectious disease. Among these nanomaterials, carbon quantum dots (CQDs) have attracted much attention owing to their unique optical properties and high biosafety. In this work, P-doped CQDs were prepared by simple hydrothermal treatment of m-aminophenol and phosphoric acid with fluorescence emission at 501 nm when excited at 429 nm. The P-doped CQDs showed effective antibacterial activity againstEscherichia coli(E. coli) andStaphylococcus aureus(S. aureus). The minimal inhibitory concentrations (MICs) of P-doped CQD were 1.23 mg/mL forE. coliand 1.44 mg/mL forS. aureus. Furthermore, the morphologies ofE. colicells were damaged andS. aureusbecame irregular when treated with the P-doped CQDs. The results of zeta potential analysis demonstrated that the P-doped CQDs inhibit antibacterial activity and destroy the structure of bacteria by electronic interaction. In combination, the results of this study indicate that the as-prepared P-doped CQDs can be a promising candidate for the treatment of bacterial infections. Keywords:carbon quantum dots;biocompatibility;antibacterial activity

Efficient Catalyst-Free One-Pot Synthesis of Polysaccharide-Polypeptide Hydrogels in Aqueous Solution

Xiaoyu Guo,Bin Yang,Junyi Chen

The preparation of polysaccharide-peptide hydrogels usually involves multiple synthetic steps, thus reducing the effectiveness and practicality of these approaches. Inspired by recent discoveries in aqueous N-carboxyanhydride (NCA) ring-opening polymerization (ROP) and ring-opening polymerization-induced nanogelation, we present an aqueous one-pot strategy to prepare polysaccharide-polypeptide hydrogels. In this study, water-soluble polysaccharide carboxymethyl chitosan is used as the macromolecular initiator to prepare polysaccharide-polypeptide copolymers through the aqueous ROP of NCA. The catalyst-free approach afforded hydrogels with properties that could be controlled by adjusting the type and amount of NCA used, with the elastic modulus ranging from 50 Pa to 18000 Pa. The hydrogen bond-cross-linked hydrogel exhibited self-healing and injectable properties. Morphology characterization revealed that micelles were formed in the early stage of reaction, suggesting that the polymerization follows an aqueous ring-opening polymerization-induced self-assembly (ROPISA) mechanism and that aggregation of micelles during the reaction caused the gelation. Moreover, the hydrogels displayed high swelling ratios (>95% water content), and hemolysis and cytotoxicity experiments demonstrated that the hydrogels had excellent biocompatibility, indicating their potential in medical applications.

PLLA–PEG/mPEG Copolymer with Improved Hydrophilicity, Crystallinity, and Biocompatibility: An In-Depth Study on the Crystallization Kinetics

Ruixian Lian,Dong Zhou,Lan Xiao,João Rodrigues,Ruilong Sheng,Zhishan Bai,Yulin Li,Changsheng Liu

Poly(lactic acid) (PLA) possesses excellent biocompatibility and biodegradability for the construction of biomaterials. However, its limited crystallinity largely restricts practical application. In this study, four poly(l-lactic acid) (PLLA) copolymers were synthesized by incorporating two different molecular weights of PEG/mPEG (1K and 2K) chains with l-lactide via ring-opening polymerization (ROP). The impact of PEG/mPEG chains on the hydrophilicity and mechanical properties of the resulting copolymers, their crystallization kinetics, and activation energy was also examined. The results demonstrate that introducing PEG/mPEG chains could significantly improve the hydrophilicity, crystallinity, and crystallization rate and reduce the maximum crystallization temperature of the copolymer materials. Among the PLLA copolymers, PLLA-mPEG (2K) exhibits the most remarkable improvements in polymer crystallinity, crystallization rate, and reduction of the maximum crystallization temperature. The advancements in copolymer material properties were anticipated to significantly expand the potential applications for PLLA-based polymer materials, which also greatly improved the applicability as sustainable and controllable biopolymer materials.

Tough Hydrogel Reinforced by Meta-Aramid Nanofibers for Flexible Sensors

Zhiwen Hou,Yongzheng Li,Donghao Zhang,Cun Peng,Yan Wang,Kunyan Sui

Hydrogels exhibit significant promise for advanced flexible sensing applications owing to their intrinsic softness, biocompatibility, and customizable functionalities. Nevertheless, their limited mechanical strength poses a critical barrier to practical implementation. In this study, we engineered a mechanically robust alginate/chitosan (SA/CS) hydrogel reinforced with meta-aramid (PMIA) nanofibers. The resulting composite hydrogel achieves a tensile strength of 16.8 MPa, substantially exceeding the performance of conventional biomass-derived hydrogels. When employed as a flexible sensor, the hydrogel demonstrates exceptional pressure-sensing capabilities, featuring high sensitivity (178.41 MΩ/MPa below 5 kPa), rapid response kinetics (0.4–0.8 s), and sustained stability (>200 cycles). Leveraging these properties, we successfully monitored vocal cord vibrations and finger motion trajectories, highlighting their potential for biomechanical sensing applications.

Living organisms growth and release immobilized in an assembled dipeptide hydrogel

Lin An,Shuhao Wang,Bingyu Liao,Jin Liu,Xiaoyan Jin,Yuyang Cai,Zibo Li,Yue Li,Junfeng Ma,Junbai Li

Self-assembled dipeptide hydrogel provides three-dimensional networks through noncovalent interactions for the embedment and the release of multiple biocomponents as well as for the growth of live cells. Here we demonstrate that Caenorhabditis elegans ( C. elegans ) as model organisms can be immobilized with significant vital signs in N -fluorenylmethoxycarbonyl diphenylalanine (Fmoc-FF) dipeptide hydrogel. With the controllability of dipeptide, assembly conditions for immobilization, growth and imaging of C. elegans were optimized. Compared with conventional anesthesia method, dipeptide hydrogel minimized unintended interferences for long-termed imaging. Particularly, nanofiber-based hydrogel can be diluted in solution as nanofiber dispersions alongside the release of C. elegans . By further analyzing the physiological phenotypes including basic growth, movement and reproduction once released from hydrogel, we confirmed the practicality and biocompatibility of Fmoc-FF hydrogel especially towards repeated immobilization and release of small-scaled living organisms, further broadening the scope of dipeptide hydrogel usage.

Extraction of biocompatible hydroxyapatite from fish scales using novel approach of ionic liquid pretreatment

Nawshad Muhammad,Yanan Gao,Farasat Iqbal,Pervaiz Ahmad,Rile Ge,Umar Nishan,Abdur Rahim,Girma Gonfa,Zahoor Ullah

In this study the waste fish scales (FS) were dissolved in 1-butyl-3-methylimidazolium acetate ionic liquid to obtain a valuable product of hydroxyapatite (HAp). The HAp was obtained in the yield of 32   ±   2%. The obtained HAp was characterized using Fourier Transform Infrared Spectroscopy (FTIR), Powder X-rays Diffraction (PXRD), Thermal Gravimetric Analysis (TGA), Field Emission Scanning Microscopy (FE-SEM), Energy Dispersive X-rays spectroscopy (EDX), and Brunauer–Emmett–Teller (BET). The results of FTIR and XRD showed the characteristic peaks of the HAp. The thermal degradation temperature of the extracted HAp was relatively high. Furthermore, low weight loss was measured which confirmed the removal of organic part of FS during ionic liquid treatment. The FE-SEM result showed the particles with different morphologies and EDX analysis showed a Ca/P ratio of 1.60 for the extracted HAp. The biocompatibility of the extracted HAp was assessed through MTT cell viability assay using known Human Embryonic Kidney 293 cells (HEK cells) and epidermoid carcinoma cells (A431 cells).

Antimicrobial Polymer with Enhanced Activity and Reduced Toxicity upon Grafting to Chitosan Oligosaccharide

Liu Xiaoli,Xie Wancui,Yang Xihong,Zhan Xiaobei,Xia Wenshui

The aim of this work was to develop a novel, nontoxic, bioactive antimicrobial material based on a natural polymer, chitosan oligosaccharides (COS), a natural pyrone compound, kojic acid (KA), and piperazine. The COS-based polymer (COS-O-MB) has been synthesized by selective partial alkylation reaction to graft kojic acid, KA, and piperazine onto the COS molecular chain. Then, physicochemical properties and bioactivity of the COS-O-MBs were evaluated and it was characterized by FT-IR, NMR spectroscopy, Mw, PID, antimicrobial activity, hemolysis assay, biocompatibility, animal toxicity, and antimicrobial action mode studies. The results revealed that the alkylation reaction took place at the C-6 position of COS, and the as-prepared COS-O-MB significantly enhanced the antimicrobial activity and biocompatibility and reduced the hemolytic activity in vitro in comparison with that of the free COS and KA. Meanwhile, the antimicrobial action mode studies proved that COS-O-MB can pass through the anionic microbial membrane, destroy the cytomembrane integrity, and potentially affect protein denaturation, demonstrating that exposure to COS-O-MB results in the death of microbial cells. But importantly, COS-O-MB is not toxic to human cells. Hence, low hemolytic activity to human red blood cells, and nontoxic nature to female mice of SLAC KM strain, made this novel polymer material a promising and effective compound for food and pharmaceutical industries.

Enhanced compressive strength and biocompatibility of porous PVAf/MPC composite scaffolds featuring directional lamellar pore channels

Yufei Tang,Hao Zhang,Yue Qiao,Bo Zhang,Yani Sun,Zixiang Wu,Kang Zhao

Magnesium phosphate bone cement (MPC) is one of the most important candidates for bone implant materials due to its properties such as no need for sintering, self-curing formation, high early compressive strength and rapid degradation. However, traditional methods for creating porous structures for MPC, such as foaming or incorporating degradable materials, often result in closed pores or pores predominantly localized on the surface, severely limiting its comprehensive degradation and effective formation of new bone. In this study, porous MPC scaffolds with directional layered channels were successfully prepared using innovative directional freeze-drying and in situ hydration techniques. The key findings of the study were that the incorporation of degradable acidic PVA staple fibers not only significantly improved the mechanical properties of the porous MPC scaffolds, but also effectively addressed the issue of high alkaline cell toxicity upon degradation. When the fiber content is 3 wt%, the compressive strength of the porous scaffold increases from 4.67 to 8.83 MPa, which was attributed to both the high tensile strength displayed by PVA fibers and the presence of multiple fiber forms within the scaffold. And the acidification of PVA fibers during degradation resulting in a decrease in the pH value of the SBF from 8.93 to 7.67, which reduced the cytotoxicity of the scaffolds. Furthermore, through a 7-day microzone in situ hydration process, the porous MPC scaffold with a Mg/P molar ratio of 1.45:1 transformed into KMgPO 4 ·6H 2 O. The pH value of simulated body fluid (SBF) showed an increasing trend with the rising Mg/P molar ratio following degradation of porous MPC scaffolds. The conclusions of the study showed that the prepared porous PVA f /MPC scaffolds with oriented hierarchical channels exhibit non-cytotoxicity and provide an optimal environment for cell survival and proliferation, and demonstrated great potential applications in the field of biodegradable bone implantation materials.

Novel β-TCP/PVA bilayered hydrogels with considerable physical and bio-functional properties for osteochondral repair

Hang Yao,Junpei Kang,Weichang Li,Jian Liu,Renjian Xie,Yingjun Wang,Sa Liu,Dong-An Wang,Li Ren

Cartilage repairing grafts have been widely studied, and osteochondral replacement hydrogels have proven to be an excellent method in research and clinical fields. However, it has been difficult to simultaneously solve three main issues in osteochondral replacement preparation: surface lubrication, overall mechanical support and good simulations of cell regeneration. A novel integrated bilayered hydrogel osteochondral replacement was constructed by blending polyvinyl alcohol (PVA) and β-tricalcium phosphate (β-TCP) in this study. Separated nano-ball milling with ultrasound dispersion prepared β-TCP demonstrated suitable properties of tiny particle size, high purity and ideal distribution, improving the mechanical properties of the novel integrated hydrogel, and providing a cartilage-like lubrication effect and high biocompatibility, including cytocompatibility and osteogenesis. The reinforcement of β-TCP and integrated molding technology enabled the hydrogel to demonstrate excellent component compatibility and good bonding between the two layers, which promoted the strengthening of the compression modulus and tensile modulus up to three times by mechanical testing. The surface lubrication properties of the novel osteochondral hydrogel were similar to the natural cartilage by friction coefficient characterization. The two layers of the novel integrated graft provided a considerable bio-function by co-culturing with chondrocytes and synovium mesenchymal stem cells: chondrocytes promoted adherence achieved by the upper density layer and better osteogenesis performance of the porous lower layer. The design of the bilayered β-TCP/PVA osteochondral hydrogel is promising for use in articular cartilage repair.

Highly-stable polymer-crosslinked 2D MXene-based flexible biocompatible electronic skins for in vivo biomonitoring

Lianjia Zhao,Lili Wang,Yiqiang Zheng,Shufang Zhao,Wei Wei,Dawei Zhang,Xiyao Fu,Kai Jiang,Guozhen Shen,Wei Han

The ability of electronic skins to identify physiological and physical signals accurately is the key to human-computer interaction. However, it is still a great challenge to improve stability of electronic-skin when maintaining the high-level pressure-response for applications in harsh conditions. Here, a highly-stable electronic skin is designed by synergistically incorporating strong hydrogen-bonds in MXene and polyvinyl alcohol (PVA). Molecular dynamics calculations show that the introduction of PVA can reduce the diffusion coefficient of MXene in acidic and basic solutions, increasing the adsorption (free) energies and providing hybrid film with good chemical stability. Therefore, the film provides stable performance recordings that last for over half a year in harsh environments. The film also shows good biocompatibility and function in an in-vivo study conducted on mice. To enhance functionality of soft robots and personal protective equipment used in harsh environments, it is important that the film has persistent and stable performance, all-weather usability, and good in vivo compatibility.

Degradable UV-crosslinked hydrogel for the controlled release of triclosan with reduced cytotoxicity

Yunfeng Hu,Guangli Ren,Liehua Deng,Jinglin Zhang,Huidi Liu,Shansong Mu,Ting Wu

In order to realize the controlled release and limited toxicity of triclosan (TC), a degradable UV-crosslinked hydrogel was prepared. For this purpose, the acryloyl chloride-modified polyactide-poly(ethylene glycol)-polylactide block copolymer was firstly synthesized and then mixed with photoinitiator to form the hydrogel rapidly under a low-intensity UV light. The properties of hydrogel, such as hydrogel strength, swelling and degradation, had been studied. The results showed that the properties of hydrogel could be modulated by the concentration of copolymer. After the encapsulation of TC, the sustained-release capability and antibacterial effect of the obtained hydrogel were studied. It was found that the sustained release of TC could potentially reduce the side effects of TC. With controlled antibacterial activity, the use of TC would be safer and more efficient. Moreover, the biocompatibility of hydrogel was also evaluated by in vitro and in vivo assays, suggesting the great potential application in the delivery of TC. The novel aspects of this study include the cytotoxicity study and the in vitro and in vivo assays, which might be useful for other researchers in this field.
